Applications of electrochemical biosensors based on 2D materials and their hybrid composites in hematological malignancies diagnosis.

Hematological malignancies encompass a wide variety of severe diseases that pose a serious threat to human health. Given the fact that hematological malignancies are difficult to treat due to their unpredictable and rapid deterioration and high rates of recurrence, growing attention has been paid to their early screening and diagnosis. However, developing a rapid and effective diagnostic tool featuring a noninvasive sampling technique is still extremely challenging. In recent years, novel nanomaterials-based electrochemical biosensors have attracted great interest because of such advantages as

Self-Photocatalysis Boosted Electrochemiluminescence Signal Amplification via In Situ Generation of the Coreactant.

The classic luminol-based electrochemiluminescence (ECL) platform generally suffers from self-decomposition of the coreactant (i.e., H2O2) during the reaction process, seriously hampering the luminous signal stability, as well as its practical application. To address this issue, apart from the introduction of complex exogenous species, preoxidation of the luminophore, and electrocatalysis for ECL signal amplification, we proposed a novel ECL model to realize the signal enhancement via in situ self-photocatalytic generation of the coreactant H2O2. Interestingly, the luminescence of luminol was

Novel approach to the measurement of antithyroglobulin antibodies in human serum - application of the quartz crystal microbalance sensors.

Measurement of antithyroglobulin antibodies (TgAb) is an inevitable laboratory tool in the management of thyroid gland diseases. Currently available immunoassays still have limitations underlying the necessity of the introduction of fast, sensitive, and label-free technologies. Our aim was to develop a method for TgAb measurement in human serum based on the quartz crystal microbalance (QCM) technology. We immobilized thyroglobulin on the surface of Attana LNB Carboxyl sensor chip®, prepared standard curve covering the range of 1-50000 kIU/L, and established optimal measurement conditions. The

An electrochemical Hg2+ sensor based on signal amplification strategy of target recycling.

In this work, an unmodified homogeneous electrochemical sensor based on electrochemical bonding and catalytic hairpin assembly (CHA) was first constructed for the high sensitivity detection of Hg2+. Herein, tetraferrocene, a synthesized compound, was used as a signal marker that modified both ends of the hairpin probe to amplify the electrochemical signal. The interaction of T-Hg2+-T could induce the catalytic self-assembly of hairpins by means of auxiliary DNA. The rigid DNA triangle that was formed easily reaches the electrode and induced Au-S self-assembly assisted by potential, allowing te

Quaternary Ammonium Salt-Functionalized Tetraphenylethene Derivative Boosts Electrochemiluminescence for Highly Sensitive Aqueous-Phase Biosensing.

Aggregation induced emission active compounds (AIEgens) have appeared as a new kind of electrochemiluminescence (ECL) emitters due to their bright emission in the aggregated state but lack functional groups. Herein, we report a quaternary ammonium salt groups-functionalized AIEgen (QAU-1) and discover that coating QAU-1 on the indium tin oxide (ITO) surface (QAU/ITO) enabled QAU-1 to display significant cathodic ECL emission compared with that of QAU-1 in the dissolved state. Inspired by this, we applied QAU-1 as emitters to develop a novel ECL biosensor (Fc-DNA/QAU/ITO) through electrostatic

Designed miniaturization of microfluidic biosensor platforms using the stop-flow technique.

Here, we present a novel approach to increase the degree of miniaturization as well as the sensitivity of biosensor platforms by the optimization of microfluidic stop-flow techniques independent of the applied detection technique (e.g. electrochemical or optical). The readout of the labeled bioassays, immobilized in a microfluidic channel, under stop-flow conditions leads to a rectangular shaped peak signal. Data evaluation using the peak height allows for a high level miniaturization of the channel geometries. To study the main advantages and limitations of this method by numerical simulation

Large-Diameter DNA-Scaffolded Nanopores Enabled by Loosely Packed Peptides for Single-Molecule Sensing.

Creating alternative nanopores is crucial for advancing single-molecule methodologies. Peptide nanopores scaffolded by DNA nanostructures have emerged as a promising class of chemically synthesizable nanopores with organized geometries. However, no such nanopores have yet achieved single-molecule detection due to size limitations. Here, we overcome this limitation by employing the alamethicin (ALM) peptide, based on the hypothesis that peptides with looser helix-helix packing are advantageous for constructing large-diameter nanopores. The resulting DNA-ALM nanopores exhibited long-lived open s

Long-Term Stable Biosensing Using Multiscale Biostructure-Preserving Metal Thin Films.

Microparticle detection technology uses materials that can specifically recognize complex biostructures, such as antibodies and aptamers, as trapping agents. The development of antibody production technology and simplification of sensing signal output methods have facilitated commercialization of disposable biosensors, making rapid diagnosis possible. Although this contributed to the early resolution of pandemics, traditional biosensors face issues with sensitivity, durability, and rapid response times. We aimed to fabricate microspaces using metallic materials to further enhance durability of

Structural Hairpin Anchoring-Mediated TtAgo Activity Regulation for Programmable Biosensing.

Thermus thermophilus Argonaute (TtAgo) has shown exceptional promise in biosensing and molecular diagnostics owing to its programmable and DNA-guided specific cleavage activity. However, the potential of regulating TtAgo activity through precise structural modification of guide DNA (gDNA) remains to be fully utilized, thereby limiting the flexible expansion of TtAgo-based biosensing strategies. In this work, TtAgo achieves optimal cleavage activity strictly with a specific linear single-stranded gDNA (linear gDNA) length range (about 15-21 nt), whereas an excessively long linear gDNA fails to

Decoding the molecular mechanisms of sour and salty sensation using biomimetic taste-based biosensors.

This critical review examines the molecular processes that govern the perception of sour and salty tastes and their application in biomimetic sensor technologies. Sour and salty tastes are unique among the five basic taste modalities because they are mediated by ion channels rather than G-protein-coupled receptors. This critical review examines how proton (H+)-dependent sour mechanisms (OTOP1, KIR2.1) and sodium (Na+) dependent salty mechanisms (ENaC) have inspired four classes of biomimetic taste sensors: membrane-mimetic, ion-channel-based, cell-based, and nanomaterial enhanced. Low-Cost Self-Driven Liquid Biosensor Based on Metamaterials for Glioblastoma-Related Sample Detection.

A self-driven terahertz metamaterial liquid biosensor composed of channel-structured metamaterials and a quartz microcavity is proposed for glioblastoma sample detection. The device transports liquid samples via capillary force. The permittivity (εeff) of liquid suspensions is comprehensively analyzed to provide theoretical support for the detection. For suspensions with the same contents, εeff decreases with increasing concentration, while under the same concentration condition, εeff decreases as particle size grows. An electric dipole resonance is excited at ~1.54 THz with theoretical sensit

In Vivo Study on the Safe Use of a Novel Intraoperative Sensing Tool for Tissue Stiffness Assessment in Endoscopic Surgery.

A novel endoscopic palpation tool (EPT), designed for tactile and stiffness sensing using fiber Bragg gratings (FBGs) was evaluated in a surgical environment for intraoperative safety and effectiveness. The EPT consisted of four FBGs arranged in a cross pattern and embedded within an elastic, hollow, silicone hemispherical dome designed to deform upon contact with soft tissue. The EPT was employed to scan both in vivo and ex vivo tissue samples. Monitoring of porcine vital signs during minimally invasive and open surgical procedures showed no significant changes during use of the EPT. Perioper

A Linear and High-Sensitivity Microwave Biosensor on a FR-4 Substrate for Aqueous Glucose Monitoring Using a Concentric Square-Shaped Split-Ring Resonator.

Non-invasive glucose monitoring remains a significant challenge in diabetes management, with existing approaches often limited by poor accuracy, high cost, or patient discomfort. Microwave-based biosensors offer a promising label-free alternative by exploiting the dielectric contrast between glucose and water. This paper presents a compact, dual-band concentric square-shaped split-ring resonator (SRR-type) biosensor fabricated on a low-cost FR-4 substrate for aqueous glucose detection. The sensor leverages electric field confinement in inter-ring gaps to transduce glucose-induced permittivity

Copper Hexacyanoferrates Obtained via Flavocytochrome b2 Assistance: Characterization and Application.

Artificial enzymes or nanozymes (NZs) are gaining significant attention in biotechnology due to their stability and cost-effectiveness. NZs can offer several advantages over natural enzymes, such as enhanced stability under harsh conditions, longer shelf life, and reduced production costs. The booming interest in NZs is likely to continue as their potential applications expand. In our previous studies, we reported the "green" synthesis of copper hexacyanoferrate (gCuHCF) using the oxidoreductase flavocytochrome b2 (Fcb2). Organic-inorganic micro-nanoparticles were characterized in detail, incl
