We investigated the therapeutic effects of a craniosynostosis-associated molecule, NEL-like molecule-1 (NELL1; NEL [a protein strongly expressed in neural tissue encoding the epidermal growth factor-like domain]), on osteolysis induced by polyethylene (PE)-particle debris. We used a murine calvarial osteolysis model with in vivo adenovirus (Ad)-mediated gene transfer. In total, 76 female Balb/c mice were randomly assigned to four groups for treatment 1 day postoperation: SHAM (injected with 0.1 mL saline without implantation of particles); PE control (injected with 0.1 mL saline after implanta

This study was conducted to evaluate the effect of polyphosphoric acid (PPA) treatment on bone regeneration around titanium (Ti) implants in vivo. Adsorption of PPA by Ti was achieved by immersing Ti implants (2 mm in diameter, 4 mm in length) in different concentrations of PPA solution (0, 1 and 10 wt%) for 24 h at 37 degrees C after proper Ti surface cleaning. The treated Ti implants were implanted on 8-week-old-male rat (n = 30) tibiae. Two or four weeks after implantation, all animals were deeply anaesthetized and underwent perfusion fixation. Ten specimens in each condition were further i

Bone remodeling is the restructuring process of existing bone, which is in constant resorption and formation. Under normal conditions, this balanced process allows the renewal of 5-10% of bone volume per year. At the microscopic level, bone remodeling is produced in basic multicellular units, where osteoclasts resorb a certain quantity of bone and osteoblasts form the osteoid matrix and mineralize it to fill the previously created cavity. These units contain osteoclasts, macrophages, preosteoblasts and osteoblasts, and are controlled by a series of factors, both general and local, allowing nor

Cell-mediated gene therapy is one of the new modalities branching out from the wide-ranging field of gene transfer and therapy. When applied to bone formation and regeneration, it has particular advantages depending on the type of cell used as a platform for gene delivery. When utilizing adult mesenchymal stem cells or osteoprogenitor cells for the expression of bone-promoting osteogenic factors, the cells not only express the factors promoting bone growth, but can respond, differentiate and participate in the bone formation process. The ability of engineered cells to respond to the transgene,

Joint cartilage injury remains a major problem in orthopaedics with more than 500,000 cartilage repair procedures performed yearly in the United States at a cost of hundreds of millions of dollars. No consistently reliable means to regenerate joint cartilage currently exists. The technologies of gene therapy and tissue engineering were combined using a retroviral vector to stably introduce the human bone morphogenic protein-7 complementary deoxyribonucleic acid into periosteal-derived rabbit mesenchymal stem cells. Bone morphogenic protein-7 secreting gene modified cells subsequently were expa

Bone marrow contains a population of rare progenitor cells capable of differentiating into bone, cartilage, muscle, tendon, and other connective tissues. These cells, referred to as MSCs, can be purified and culture expanded from animals and humans. This review summarizes recent experimentation focused on characterizing the cellular aspects of osteogenic differentiation, and exploration of the potential for using autologous stem cell therapy to augment bone repair and regeneration. The authors have completed an array of preclinical studies showing the feasibility and efficacy of MSC based impl

Mesenchymal stem cells (MSCs) contribute substantially to bone regeneration, yet osteogenic cues alone do not fully explain how their reparative capacity is established, maintained, or lost. Prior studies show that organelles, especially mitochondria, play critical roles in MSC osteogenesis. Recent advances reveal that individual organelle states, inter-organelle coordination within MSCs, and reciprocal organelle-related intercellular communication together constitute an integrated regulatory network. In this review, we introduce 'organellar governance' as a framework for understanding MSC-med

After condylar fracture surgery, the temporomandibular joint undergoes varying degrees of self-repair. Usually, slow bone remodeling occurs. However, 2 abnormal patterns of condylar remodeling after condylar fracture surgery are found: one is in situ condylar bone regeneration, and the other is ectopic bone formation within the soft tissue of the joint capsule. Both of these abnormal patterns of bone formation require increased caution. Timely mouth-opening training can prevent the occurrence of joint ankylosis. Regular follow-up is necessary.

Preventing tumor recurrence while promoting bone regeneration remains a major challenge for osteosarcoma implants after limb-salvage surgery. Herein, we developed a DNA molecular robot (DMR) that converts tumor microenvironment (TME) and near-infrared (NIR) stimuli into integrin-mediated mechanical forces, generating microtubule-targeting activity without drugs. The DNA-polypeptide chimera contains RGD peptides for integrin anchoring and allosteric DNA switches with tunable piconewton sensitivity. Under acidic TME and NIR irradiation, DMR modulates microtubule dynamics, suppresses adhesion pro

Retrograde peri-implantitis (RPI) is characterized by atypical and often progressive bone loss at the periapex of a dental implant. Although RPI reportedly has a lower prevalence than marginal peri-implantitis, if left untreated it can lead to loss of dental implants. In this clinical case series, two patients with different presentations of RPI were treated with guided bone regeneration (GBR) using bone graft and collagen membrane. Longterm follow-up showed clinical and radiographic improvements that resulted in radiographic bone fill and reduced probing depths around implants. Observations r

The skeleton is highly innervated by numerous nerve fibers. These nerve fibers, in addition to transmitting information within the bone and mediating bone sensations, play a crucial role in regulating bone tissue formation and regeneration. Traditional bone tissue engineering (BTE) often fails to achieve satisfactory outcomes when dealing with large-scale bone defects, which is frequently related to the lack of effective reconstruction of the neurovascular network. In recent years, increasing research has revealed the critical role of nerves in bone metabolism. Nerve fibers regulate bone cells

The extracellular matrix (ECM) of tissues progressively changes its mechanical properties in processes such as tissue development, repair, and disease progression. While stiffness has become a key design parameter of biomaterials, most synthetic biomaterials employed in cell culture or tissue regeneration do not display these gradual changes in mechanical properties. Here, we report on a hydrogel platform with the capacity to exhibit progressive stiffening from 0.8 to 7.4 kPa within a ∼48 h time period. The material integrates the tyramine derivative of hyaluronic acid (HAT) and Laponite® (Lap

Proteolysis-targeting chimeras (PROTACs) degrade target proteins through the ubiquitin-proteasome system. To date, PROTACs are primarily used to treat various diseases; however, they have not been applied in regenerative therapy. Herein, this work introduces MDM2-targeting PROTACs customized for application in bone regeneration. An MDM2-PROTAC library is constructed by combining Nutlin-3 and CRBN ligands with various linker designs. Through a multistep validation process, this work develops MDM2-PROTACs (CL144 and CL174) that presented potent degradation efficiency and a robust inductive effec

Sclerostin is a key inhibitor of the Wnt signaling pathway, functioning by binding to the LRP5/6 receptor. This interaction inhibits beta-catenin expression, resulting in the downregulation of osteogenic markers, which contributes to the promotion of osteoporosis and an increase in osteoclast numbers. The primary objective of this research was to investigate the effects of sclerostin antibody (Scl-ab) on bone formation utilizing graft materials in tooth sockets, and to analyze the regulatory interaction between sclerostin and bone tissue through targeted sclerostin inhibition and stimulation o

Extensive research has delved into the multifaceted roles of osteoclasts beyond their traditional function in bone resorption in recent years, uncovering their significant influence on bone formation. This shift in understanding has spurred investigations into engineering strategies aimed at leveraging osteoclasts to not only inhibit bone resorption but also facilitate bone regeneration. This review seeks to comprehensively examine the mechanisms by which osteoclasts impact bone metabolism. Additionally, it explores various engineering methodologies, including the modification of bioactive mat

Bone regeneration requires a well-orchestrated cellular and molecular response including robust vascularization and recruitment of mesenchymal and osteogenic cells. In femoral fractures, angiogenesis and osteogenesis are closely coupled during the complex healing process. Here, we show with advanced longitudinal intravital multiphoton microscopy that early vascular sprouting is not directly coupled to osteoprogenitor invasion during calvarial bone regeneration. Early osteoprogenitors emerging from the periosteum give rise to bone-forming osteoblasts at the injured calvarial bone edge. Microves
