Analysis of sputum/tracheal aspirate and nasopharyngeal samples for SARS-CoV-2 detection by laboratory-developed test and Panther Fusion system.

In this study, 127 sputum/tracheal aspirate specimens were evaluated by a laboratory-developed real-time RT-PCR method and Fusion SARS-CoV-2 assay. These specimens were collected from the patients who have nasopharyngeal swab (NPS) samples being used for SARS-CoV-2 detection previously or simultaneously. The overall agreement was 96% between the lower respiratory tract (LRT) and NPS samples, suggesting that LRT specimens could be an option for patients who develop a productive cough or those receiving invasive mechanical ventilation.

COVID-19: Therapeutics and Their Toxicities.

SARS-CoV-2 is a novel coronavirus that emerged in 2019 and is causing the COVID-19 pandemic. There is no current standard of care. Clinicians need to be mindful of the toxicity of a wide variety of possibly unfamiliar substances being tested or repurposed to treat COVID-19. The United States Food and Drug Administration (FDA) has provided emergency authorization for the use of chloroquine and hydroxychloroquine. These two medications may precipitate ventricular dysrhythmias, necessitating cardiac and electrolyte monitoring, and in severe cases, treatment with epinephrine and high-doses of diaz

Colorimetric tetra-combo loop-mediated isothermal amplification assay for efficient SARS-CoV-2 detection.

Coronavirus disease 2019 (COVID-19), which is caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) has spread rapidly and caused a pandemic in 2020. Both pharmaceutical approaches, such as vaccinations and non-pharmaceutical approaches such as social distancing through lockdown and international border restrictions had been implemented to control the outbreak and the transmission of SARS-CoV-2. However, the prolonged implementation of these measures negatively impacted the population and global economy, while the continuously emerging SARS-CoV-2 variants led to breakthrough i

The establishment of a universal standard for both viral antigen and nucleid acid detection based on digital PCR.

BACKGROUND: Viral antigen rapid detection tests (Ag-RDTs) and PCR-based nucleic acid amplification tests (NAATs) are essential for diagnosing infections like SARS-CoV-2. However, unlike nucleic acids which can be quantified precisely in copies, viral antigens lack a standardized unit of measurement, hindering precise analytical evaluation of Ag-RDTs and comparison with NAATs. METHODS: We established a universal national standard for SARS-CoV-2 antigen and nucleic acid based on an Omicron BA.1 strain, inactivated using β-propiolactone (BPL). Its concentration was assigned via multi-laboratory

DNA Nanobridge with Cascade Operation Logic on a Transistor for Comprehensive Evaluation of Respiratory Infection.

Acute respiratory infections may cause a cytokine storm that poses a great threat to life. Thus, on-site pathogen screening and infection severity assessment are highly desired for immediate medical intervention. Here, we report the comprehensive monitoring of respiratory infection by a graphene field-effect transistor (g-FET) modified with a DNA nanobridge that consists of two DNA tetrahedrons linked by a bridge strand. The recognition of target viral nucleic acid triggers bridge dissociation, releasing the probe to detect the inflammatory cytokine IL-6. Such a cascade logical operation enabl

Design of a lateral flow assay targeting the conserved NIID_2019-nCoV_N gene region for molecular viral diagnosis.

SARS-CoV-2, the causative agent of the COVID-19 pandemic, remains a significant threat to global public health. Therefore, rapid and accurate detection of the virus continues to be of critical importance. Among the specific gene regions of SARS-CoV-2, the Nucleocapsid (N) protein gene is one of the most frequently targeted for viral identification, with NIID_2019-nCOV_N being a notable example. While reverse transcriptase polymerase chain reaction (RT-PCR) remains the gold standard for diagnosis, alternative molecular detection methods are still limited. In this study, a lateral flow assay (LF

Lateral flow strip assay of a gene segment in the COVID-19 virus with combined dual readout mode and preliminary multisite hybrid chain reaction amplification.

The past and present scenario of COVID-19 has revealed the necessity of simple point-of-care tests. When combined with the great advantages of amplification, lateral flow assay nucleic acid analysis represents a more sensitive molecular diagnostic technique compared to universal protein analysis. Room temperature operation, an enzyme-free nature, and in situ elongation make hybrid chain reaction amplification (HCR) a good candidate for amplified combined lateral flow assays (LFAs). Since dual modes of detection can not only satisfy different application scenarios, but also reduce the false-neg

Hand-held all-in-one (HAO) self-test kit for rapid and on-site detection of SARS-CoV-2 with colorimetric LAMP.

Throughout the COVID-19 pandemic, individuals potentially infected with severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) were forcibly recalled to local or central hospitals, where the diagnostic results were obtained a couple of days after the liquid biopsies were subjected to conventional polymerase chain reaction (PCR). This slow output of such a complex and time-consuming laboratory procedure hindered its widespread application. To overcome the limitations associated with such a centralized diagnostic system, we developed a hand-held and all-in-one type test kit in which the an

An at-home and electro-free COVID-19 rapid test based on colorimetric RT-LAMP.

INTRODUCTION: In the fight against virus-caused pandemics like COVID-19, diagnostic tests based on RT-qPCR are essential, but they are sometimes limited by their dependence on expensive, specialized equipment and skilled personnel. Consequently, an alternative nucleic acid detection technique that overcomes these restrictions, called loop-mediated isothermal amplification following reverse transcription (RT-LAMP), has been broadly investigated. Nevertheless, the developed RT-LAMP assays for SARS-CoV-2 detection still require laboratory devices and electrical power, limiting their widespread us

Colorimetric RT-LAMP Detection of Multiple SARS-CoV-2 Variants and Lineages of Concern Direct from Nasopharyngeal Swab Samples without RNA Isolation.

Since, during the Coronavirus disease 19 (COVID-19) pandemic, a large part of the human population has become infected, a rapid and simple diagnostic method has been necessary to detect its causative agent, the Severe Acute Respiratory Syndrome-related Coronavirus-2 (SARS-CoV-2), and control its spread. Thus, in the present study, we developed a colorimetric reverse transcription-loop-mediated isothermal amplification (RT-LAMP) kit that allows the detection of SARS-CoV-2 from nasopharyngeal swab samples without the need for RNA extraction. The kit utilizes three sets of LAMP primers targeting
