Atypical PI3Ks coordinate chemotaxis, signaling dynamics, and multicellular development in Dictyostelium.

Phosphoinositide 3-kinase (PI3K) signaling regulates protrusion, polarity, membrane uptake, and multicellular development in Dictyostelium discoideum, but these functions have been interpreted largely through canonical Class I PI3Ks and PI(3,4,5)P₃ production. This framework does not fully explain how PI3K-dependent pathways restrain Ras activity, organize relay signaling, or coordinate the transition from single-cell migration to multicellular aggregation. Here, we show that three atypical PI3K-family enzymes, PikF, PikG, and PikH, define genetically separable functions within this broader PI

Photon and particle radiotherapy induce redundant modular chemotaxis of human lymphocytes.

Radiotherapy triggers chemokine release and leukocyte infiltration in preclinical models through activation of the senescence-associated secretory phenotype (SASP). However, effects of irradiation on senescence and SASP in human tissue and in the context of particle radiotherapy remain unclear. Here, we analyzed chemokine patterns after radiotherapy of human pancreatic tumors and cancer cell lines. We show that irradiated tumor cells coexpressed SASP chemokines in defined modules. These chemokine modules correlated with infiltration of distinct leukocyte subtypes expressing cognate receptors.

Artemisinin inhibits neutrophil and macrophage chemotaxis, cytokine production and NET release.

Immune cell chemotaxis to the sites of pathogen invasion is critical for fighting infection, but in life-threatening conditions such as sepsis and Covid-19, excess activation of the innate immune system is thought to cause a damaging invasion of immune cells into tissues and a consequent excessive release of cytokines, chemokines and neutrophil extracellular traps (NETs). In these circumstances, tempering excessive activation of the innate immune system may, paradoxically, promote recovery. Here we identify the antimalarial compound artemisinin as a potent and selective inhibitor of neutrophil

Mesenchymal stem cells require MARCKS protein for directed chemotaxis in vitro.

Mesenchymal stem cells (MSCs) reside within tissues such as bone marrow, cord blood, and dental pulp and can differentiate into other mesenchymal cell types. Differentiated MSCs, called circulating fibrocytes, have been demonstrated in human lungs and migrate to injured lung tissue in experimental models. It is likely that MSCs migrate from the bone marrow to sites of injury by following increasing chemokine concentrations. In the present study, we show that primary mouse bone marrow mesenchymal stem cells (BM-MSCs) exhibit directed chemotaxis through transwell inserts toward increasing concen

Anthrax edema toxin inhibits endothelial cell chemotaxis via Epac and Rap1.

Angiogenesis involves the assembly of endothelial cells into capillaries from a pre-existing vasculature. Because abnormal angiogenesis is a hallmark of many cancers, it is critical to find factors that control this process. Endothelial cells are enriched in the anthrax receptor; we therefore determined the effect of anthrax edema toxin (ET), an adenylyl cyclase, on chemotaxis. cAMP generated by ET does not block proliferation or survival but causes cytoskeletal changes and inhibits chemotaxis by primary human microvascular endothelial cells (HMVECs). These effects are due to the action of a d

Quantification of bacterial chemotaxis in porous media using magnetic resonance imaging.

Bacterial chemotaxis has the potential to enhance biodegradation of organic contaminants in polluted groundwater systems. However, studies of bacterial chemotaxis in porous media are scarce. In this study we use magnetic resonance imaging (MRI) for the noninvasive measurement of changes in bacterial-density distributions in a packed column at a spatial resolution of 330 microm as a function of time. We analyze both the diffusive and the chemotactic behavior of Pseudomonas putida F1 in the presence of the chemical stimulus trichloroethylene (TCE). The migration of motile bacteria in experiments

Recurrent staphylococcal abscesses associated with defective neutrophil chemotaxis and allergic rhinitis.

Four patients with recurrent staphylococcal furonculosis and deep abscess formation were evaluated to determine if a defect in the host defense mechanism could account for the unusual incidence of infection. Each also suffered repeated allergic rhinitis, often preceding the onset of infection. A marked defect in neutrophil granulocyte chemotaxis occurred when the patients were symptomatic with rhinitis and abscess formation. Their mean chemotactic index (+/- SD) was 16 +/- 6, while that of 25 control subjects was 70 +/- 11. Neutrophil random migration, phagocytosis, bactericidal activity, and

Infection, dermatitis, increased IgE, and impaired neutrophil chemotaxis. A possible relationship.

Eleven patients with recurrent bacterial infections were found to have impaired neutrophil chemotaxis. The infections were primarily staphylococcal abscesses involving the skin and the respiratory tract. All patients had chronic eczematour dermatitis and an elevated level of serum IgE. The peripheral blood eosinophil count was equal to or greater than 10% of the circulating leukocytes in 10 of 11 patients. Associated findings in some patients included urticaria and oral or cutaneous Candida infection. These observations suggest a relationship between chronic dermatitis, elevated serum IgE leve

Semaglutide Reduces Odorant-Driven Chemotaxis and Reproductive Output in Caenorhabditis elegans.

Glucagon-like peptide-1 receptor agonists (GLP-1RAs) reduce food intake and body weight in humans, partly via neural circuits governing reward and energy homeostasis. Using Caenorhabditis elegans as a tractable model, we examined neurobehavioral and physiological effects of semaglutide, a long-acting GLP-1RA. Wild-type N2 animals exposed to semaglutide (1 μM and 10 μM) showed significantly reduced chemotaxis toward the volatile odorant diacetyl. Semaglutide also reduced egg-laying and delayed larval development at both concentrations tested. These phenotypes demonstrate that semaglutide alters

Foraging Drosophila suzukii larvae exhibit chemotaxis and invade intact, uninfested fruit.

Spotted-wing drosophila, Drosophila suzukii (Matsumura) (Diptera: Drosophilidae), is a destructive pest of ripening berries and stone fruits, where larval feeding rapidly renders fruit unmarketable. Larvae are widely assumed to remain inside their natal fruit until pupation, limiting exposure to external management tactics. Here, we challenge this paradigm by demonstrating that D. suzukii larvae exhibit complex chemotactic capabilities and dynamic interactions with host fruit throughout development. Third-instar larvae responded in a dose-dependent manner to fermentation-associated volatiles a

KMT5a-Mediated IRF3 Stabilization Enhances Macrophage Chemotaxis and Renal Fibrosis Progression.

While the histone lysine methyltransferase (KMT) family responsible for posttranslational modification has been reported to regulate multiple diseases progression like tumor and cardiovascular disease, its role in renal fibrosis progression is not completely understood. Here, via TGF-β-treated HK-2 cells and an in vivo unilateral ureteral obstruction mice model, the histone lysine methyltransferase family was screened and the upregulated expression of KMT5a was identified in these renal fibrosis models. Interference assays using lentivirus transfection in HK-2 cells and transgenic mice display

Regulator of G protein signaling 5 restricts neutrophil chemotaxis and trafficking.

Neutrophils are white blood cells that are mobilized to damaged tissues and to sites of pathogen invasion, providing the first line of host defense. Chemokines displayed on the surface of blood vessels promote migration of neutrophils to these sites, and tissue- and pathogen-derived chemoattractant signals, including N-formylmethionylleucylphenylalanine (fMLP), elicit further migration to sites of infection. Although nearly all chemoattractant receptors use heterotrimeric G proteins to transmit signals, many of the mechanisms lying downstream of chemoattractant receptors that either promote or

IL-17 Enhances Chemotaxis of Primary Human B Cells during Asthma.

IL-17 is a pro-inflammatory mediator that is believed to play a critical role in regulating tissue inflammation during asthma, COPD, as well as other inflammatory disorders. The level of expression of IL-17 has been shown to be upregulated in lung bronchial tissue of asthmatic patients. Several reports have provided further evidence that this cytokine could play a key role in enhancing the migration of inflammatory as well as structural cells of the bronchial lung tissue during asthma and COPD. B cell infiltration to sites of inflammation during inflammatory disorders such as bowel disease, as

Syk regulates multiple signaling pathways leading to CX3CL1 chemotaxis in macrophages.

Several studies have clearly established the importance of the interaction between macrophages and CX3CL1 in the progression of disease. A previous study demonstrated that Syk was required for CX3CL1-mediated actin polymerization and chemotaxis. Here, we delineated the signaling cascade of Syk-mediated cell migration in response to CX3CL1. Inhibition of Syk in bone marrow-derived macrophages or reduction of Syk expression using siRNA in RAW/LR5 cells indicated that Syk was required for the activation of PI3K, Cdc42, and Rac1. Also, reduction in WASP or WAVE2 levels, common downstream effectors

β1 integrins regulate fibroblast chemotaxis through control of N-WASP stability.

Chemotactic migration of fibroblasts towards growth factors, such as during development and wound healing, requires precise spatial coordination of receptor signalling. However, the mechanisms regulating this remain poorly understood. Here, we demonstrate that β1 integrins are required both for fibroblast chemotaxis towards platelet-derived growth factor (PDGF) and growth factor-induced dorsal ruffling. Mechanistically, we show that β1 integrin stabilises and spatially regulates the actin nucleating endocytic protein neuronal Wiskott–Aldrich syndrome protein (N-WASP) to facilitate PDGF recepto

Microfluidic monitoring of Pseudomonas aeruginosa chemotaxis under the continuous chemical gradient.

This study presents a microfluidic approach for the rapid analysis of bacterial chemotaxis in response to chemical gradients. The diffusional mixing of laminar flow continuously generates a stable chemical gradient in a microfluidic device. For the proof of concept, we have investigated the effects of the attractant peptone and repellent trichloroethylene (TCE) on chemotactic responses of wild type Pseudomonas aeruginosa PAO1 and chemotactic mutant PC4. The microfluidic method clearly demonstrates that P. aeruginosa PAO1 is attracted to peptone and repelled from TCE, whereas PC4 shows non-chem

CX3CL1/fractalkine is released from apoptotic lymphocytes to stimulate macrophage chemotaxis.

Cells undergoing apoptosis are efficiently located and engulfed by phagocytes. The mechanisms by which macrophages, the professional scavenging phagocytes of apoptotic cells, are attracted to sites of apoptosis are poorly defined. Here we show that CX3CL1/fractalkine, a chemokine and intercellular adhesion molecule, is released rapidly from apoptotic lymphocytes, via caspase- and Bcl-2-regulated mechanisms, to attract macrophages. Effective chemotaxis of macrophages to apoptotic lymphocytes is dependent on macrophage fractalkine receptor, CX3CR1. CX3CR1 deficiency caused diminished recruitment

Phosphorylation assays of chemotaxis two-component system proteins in Borrelia burgdorferi.

Borrelia burgdorferi has a complex chemotaxis signal transduction system with multiple chemotaxis gene homologs similar to those found in Escherichia coli and Bacillus subtilis. The B. burgdorferi genome sequence encodes two cheA, three cheY, three cheW, two cheB, two cheR, but no cheZ genes. Instead of cheZ, B. burgdorferi contains a different CheY-P phosphatase, referred to as cheX. The multiple B. burgdorferi histidine kinases (CheA1 and CheA2) and response regulators (CheY1, CheY2, and CheY3) possess all the domains and functional residues found in E. coli CheA and CheY, respectively. Anthrax toxins inhibit immune cell chemotaxis by perturbing chemokine receptor signalling.

Pathogenic strains of Bacillus anthracis produce two potent toxins, lethal toxin (LT), a metalloprotease that cleaves mitogen-activated protein kinase kinases, and oedema toxin (ET), a calcium/calmodulin-dependent adenylate cyclase. Emerging evidence indicates a role for both toxins in suppressing the initiation of both innate and adaptive immune responses, which are essential to keep the infection under control. Here we show that LT and ET inhibit chemotaxis of T-cells and macrophages by subverting signalling by both CXC and CC chemokine receptors. The data highlight a novel strategy of immun
