One new cassane diterpenoid(1) and six known compounds(2-7) were isolated from the 95% ethanol extract of fresh Caesalpinia decapetala pods by silica gel column chromatography, gel filtration chromatography, and semi-preparative HPLC. Compound 1 was identified as a highly oxidized cassane diterpenoid, and its absolute configuration was determined by comprehensive spectroscopic analysis and electronic circular dichroism(ECD) calculations, being named caesalpin L. In this study we compared horseradish peroxidase (HRP)-labeled rabbit antihuman immunoglobulin G (IgG) conjugates, prepared by a one-step and a two-step method. When activity of peroxidase in adult Paragonimus westermani was monitored using o-dianisidine and H2O2 as substrates, its specific activity was 1.5 times higher in excretory-secretory product (ESP) than in crude extract. The enzyme was purified by two purification steps of Sephacryl S-300 Superfine gel permeation and DEAE-Trisacryl M anion exchange chromatographies. Immunoelectrophoresis, gel filtration-electrophoresis and immuno-gel filtration of cerebral fluid samples obtained from five patients with subacute sclerosing leucoencephalitis and known to contain an abnormal gamma-globulin fraction (gammac) produced strong evidence that the gammac-globulin is made up of several components of small molecular size. This fraction is obviously not homogeneous, since some of the cases show more than one precipitate on immuno-gel filtration with immune serum against y-globulin. Heat-labile toxin from a cell sonicate of a virulent type-D strain of Pasteurella multocida was purified by ammonium sulfate precipitation followed by ion exchange chromatography, gel filtration chromatography, and polyacrylamide gel electrophoresis. Toxic activity was assayed during toxin purification by cytopathic effect in Vero or bovine embryonic lung cell cultures. Toxicity for cells correlated with dermonecrosis in guinea pig skin. The taxane derivatives 13-dehydroxybaccatin III (13-DHB III) and 10-deacetylpaclitaxel (10-DAP) can be used as semi-synthetic precursors of paclitaxel. We have fractionated tau isoforms by elution at increasing pH values using iron-chelated affinity chromatography, which discriminates between isoforms phosphorylated to different extents.
