Heterologous expression of a nonconventional terpene biosynthetic gene cluster from the deepsea-derived Streptomyces amphotericinicus DS22-01 led to the production of a novel cyclic diterpene, ostamycin A (1). Anti-influenza A virus activity evaluation revealed that compound 1 showed significant activity with an IC50 value of 4.72 μM, which was much stronger than that of the positive control ribavirin (IC50 = 20.80 μM). Inspired by its intriguing activity, yield optimization was achieved through a combined approach involving promoter engineering and codon modification in a stepwise manner. Thi

Salvia divinorum is a hallucinogenic plant native to the Oaxaca in Mexico. The active ingredient for psychotropic effects in this plant is salvinorin A, a potent and highly selective κ-opioid receptor agonist. Salvinorin A is distinct from other well-known opioids, such as morphine and codeine, in that it is a non-nitrogenous diterpenoid with no affinity for μ-opioid receptor, the prime receptor of alkaloidal opioids. A terpene opioid that selectively targets a new opioid receptor (κ-opioid receptor) can be instrumental in developing alternative analgesics. Elucidation of the salvinorin A bios

Hypaconitine, a neuromuscular blocker, is a diterpene alkaloid found in the root of Aconitum carmichaelii. Although hypaconitine was shown to affect various physiological responses in neurological models, the effect of hypaconitine on cell viability and the mechanism of its action of Ca2+ handling is elusive in cortical neurons. This study examined whether hypaconitine altered viability and Ca2+ signalling in HCN-2 neuronal cell lines. Cell viability was measured by the cell proliferation reagent (WST-1). Cytosolic Ca2+ concentrations [Ca2+ ]i was measured by the Ca2+ -sensitive fluorescent dy

A new regular diterpene possessing an unusual 1,6-anti-3-methylcyclohex-2-en-1-ol ring system, dactyloditerpenol acetate (1), has been extracted from the tropical sea hare Aplysia dactylomela and its stereostructure elucidated by spectroscopic methods. The absolute configuration of 1 was determined as 1S, 6S, 7R, 10S, and 11R by application of Kishi's method for the assignment of absolute configuration of alcohols. The new diterpene potently inhibited in vitro thromboxane B2 (TXB2) (IC50 0.4μM) and superoxide anion (O2(-)) (IC50 1μM) generation from Escherichia coli lipopolysaccharide (LPS)-ac

From the dichloromethane extract obtained from the gum resin of Boswellia serrata Roxb. (Burseraceae), a well-known medicinal plant resin ("Indian Olibanum"), the cembrane-type diterpene serratol was isolated in high yield. Its structure, previously reported without clear specification of double-bond geometry and without specification of stereochemistry, was reanalysed by means of spectroscopic measurements and unambiguously assigned as S(-)-cembra-3E,7E,11E‑triene-1-ol. Full assignment of all NMR data is reported for the first time. The compound was found to be identical with a cembrenol prev

Aparisthmium cordatum (Juss.) Bail. (Euphorbiaceae), known in the State of Pará, Brazil as "ariquena queimosa", is a medium-sized tree which is native to the North Brazilian coastal region. Previous phytochemical studies of the bark of A. cordatum yielded a furan diterpenoid with a clerodane skeleton, called aparisthman. Recently, we reported the antiulcerogenic activity of trans-dehydrocrotonin (DHC), a furan diterpene isolated from Croton cajucara bark, in different ulcerogenic models in mice and rats. The aim of the present study was to assess the possible antiulcerogenic activity of aparis

BACKGROUND: Drugs used for the treatment of diseases associated with chronic inflammation, such as cancer and rheumatoid arthritis have the potential to cause undesirable side-effects, which might result in patients ending treatment prematurely. However, plants are a viable option for the treatment of inflammatory diseases. In this study, we assessed the in vivo and in vitro anti-inflammatory activity, and the antitumor effects of the chloroform extract of Salvia ballotiflora (ECL). The pro-apoptotic effects of ECL in CT26 cells were also determined.
METHODS: The chloroform extract of Salvia

Plant abietane diterpenoids (e.g. aethiopinone, 1- oxoaethiopinone, salvipisone and ferruginol), synthesized in the roots of several Salvia spp, have antibacterial, antifungal, sedative and anti-proliferative properties. Recently we have reported that content of these compounds in S. sclarea hairy roots is strongly depending on transcriptional regulation of genes belonging to the plastidial MEP-dependent terpenoid pathway, from which they mostly derive. To boost the synthesis of this interesting class of compounds, heterologous AtWRKY18, AtWRKY40, and AtMYC2 TFs were overexpressed in S. sclare

INTRODUCTION: Kaurene diterpenes (KDs) constitute a chemical class often found in the genus Annona with interesting biological activities. To date, chromatographic tools have been mostly used to determine KDs. Quantitative nuclear magnetic resonance (qNMR) has distinguished itself in quantitative estimation of natural products and is an interesting choice to assess total KD contents.
OBJECTIVE: To establish a 1 H qNMR method for determining the total KD contents in extracts and fractions obtained from Annona vepretorum stems.
METHODOLOGY: Stems were extracted with hexane and methanol, result

Premna, a genus consisting of approximately 200 species, predominantly thrives in tropical and subtropical areas. Many of these species have been utilized in ethnopharmacology for diverse medicinal applications. In Saudi Arabia, Premna resinosa (Hochst.) Schauer (Lamiaceae) grows wildly, and its slightly viscid leaves are attributed to the production of leaf accession. In this study, we aimed to extract the surface accession from fresh leaves using dichloromethane to evaluate the anticancer potential. The plant exudate yielded two previously unknown labdane diterpenes, Premnaresone A and B, in

ETHNOPHARMACOLOGICAL RELEVANCE: Boswellia serrata oleo-gum resin, known as Kundur in Unani medicine, has traditional applications in neurological and cognitive disorders, including Majoon-e-Nisyan and Dawa-ul-Kibreet for Nisyan (amnesia) and Muqawwi-e-Asab (nerve-strengthening) respectively. Serratol is a cembrane diterpene constituent of B. serrata resin, while related frankincense cembranes exhibit neuroactive properties. However, the contribution of serratol to the neurological activities of B. serrata remains poorly explored.
AIM OF THE STUDY: To isolate serratol from B. serrata resin, ev

The class II diterpene cyclase from pleuromutilin biosynthesis contains a unique variant of the otherwise highly conserved DxDD motif (DxDM), which cooperatively serves as the catalytic acid, and uniquely produces an "A" ring contracted product, mutildienyl pyrophosphate (MPP). The correlation between these features was investigated here via substitution of aspartate for methionine, which largely blocks the production of MPP and leads to a novel hydroxylated product, syn-halima-13E-en-5β-ol-15-PP, providing insight into this unique reaction.

Cutibacterium acnes is a Gram-positive bacterium implicated in acne vulgaris, a common inflammatory skin disease. Rising antibiotic resistance in C. acnes highlights the need for new therapeutic agents. Callicarpa americana L., a shrub native to the southeastern United States, has a history of use in Native American traditional medicine. Bioassay-guided fractionation of C. americana leaf extracts led to the isolation of 12(S),16ξ-dihydroxycleroda-3,13-dien-15,16-olide (compound 1), a clerodane diterpene. Compound 1 exhibited growth-inhibitory activity against C. acnes (MIC: 8 μg/mL). A route t

The fruits of Amomum villosum are often considered a medicinal and food homologous material and have been found to have therapeutic effects in chronic enteritis, gastroenteritis, and duodenal ulcer. The aim of this study is to discover the anti-inflammatory active ingredients from dried ripe fruits of A. villosum and to elucidate the molecular mechanisms. We verified that the inhibitory activity of the ethyl acetate extract was superior to Dexamethasone (Dex), so we ultimately chose to study the ethyl acetate extract from the fruits of A. villosum. A total of 33 compounds were isolated from it

INTRODUCTION: The production of marine drugs in its normal habitats is often low and depends greatly on ecological conditions. Chemical synthesis of marine drugs is not economically feasible owing to their complex structures. Biotechnology application via elicitation represents a promising tool to enhance metabolites yield that has yet to be explored in soft corals.
OBJECTIVES: Study the elicitation impact of salicylic acid (SA) and six analogues in addition to a systemic acquired resistance inducer on secondary metabolites accumulation in the soft coral Sarcophyton ehrenbergi along with the

Antiproliferative bioassay-guided fractionation of the ethanolic extract of the endemic Madagascan plant Metaporana sericosepala led to the first natural product isolation of a butenolide diterpene, which was synthesized during an anti-inflammatory study in 1988. The structure of the compound was elucidated as 3-homofarnesyl-4-hydroxybutenolide (1) by analysis of its spectroscopic data, including 1D- and 2D-NMR data and chemical evidence. The once synthetic compound can now also be considered as a natural product. Compound 1 had modest antiproliferative activity towards the A2780 ovarian cance
