The role of residual (veterinary) antibiotics in chemical exposome analysis: Current progress and future perspectives.

Humans are exposed to a complex mixture of environmental and food-related chemicals throughout their lifetime. Exposome research intends to explore the nongenetic, that is, environmental causes of chronic disease and their interactions comprehensively. Residual antibiotics can enter the human body through therapeutics, foods of animal origin, aquatic products, or drinking water. In the last decade, significant levels of residual antibiotics in human urine have been described, demonstrating frequent exposure throughout populations. To which extent they contribute to human health risks is debate

The soil nematode exposome: Unraveling the impacts of particulate plastics from agroecosystems to one health.

The escalating accumulation of particulate plastics in terrestrial ecosystems demands innovative, biologically informed approaches to evaluate their ecological and human health impacts. Soil nematodes are gaining recognition as sensitive and ecologically relevant bioindicators for assessing the effects of plastic pollution in soils. Their widespread distribution, trophic adaptability, and close association with soil particles make them ideal sentinels for detecting subtle yet significant biological responses to micro- and nanoplastics. Plastic contamination can disrupt soil structure, microbio

Impact of Food Exposome on Atherosclerotic Plaque Stability: Metabolomic Insights from Human Carotid Endarterectomy Specimen.

Carotid atherosclerotic stenosis (CAS) is a leading cause of ischemic stroke. Current understanding of plaque vulnerability remains largely confined to histopathological characterization. Consequently, identifying molecular determinants of plaque stability represents a major challenge to advance prevention strategies. Untargeted metabolomic analysis was performed using mass spectrometry coupled to liquid chromatography on carotid plaques removed from patients with CAS undergoing endarterectomy. To identify factors influencing plaque stability, we compared 42 asymptomatic with 30 symptomatic CA

Impact of Exposome Factors on Epidermal Barrier Function in Patients with Obstructive Sleep Apnea Syndrome.

Exposome factors, such as sleep deprivation and diet, could affect skin barrier function. The objectives of this study are to compare skin barrier function between patients with Obstructive Sleep Apnea Syndrome (OSAS) and healthy individuals, and to evaluate the effect of other exposome factors on skin. A cross-sectional study was conducted. Patients with OSAS and healthy volunteers matched by age and sex were included. OSAS severity was assessed by the Apnea-Hypopnea Index (AHI). Validated questionnaires were used to assess diet, anxiety, depression, and psychological stress. Skin barrier fun

Adult skin acute stress responses to short-term environmental and internal aggression from exposome factors.

Exposome factors that lead to stressed skin can be defined as any disturbance to homeostasis from environmental (meteorological factors, solar radiation, pollution or tobacco smoke) and/or internal exposure (unhealthy diet, hormonal variations, lack of sleep, psychosocial stress). The clinical and biological impact of chronic exposome effects on skin functions has been extensively reviewed, whereas there is a paucity of information on the impact of short-term acute exposure. Acute stress, which would typically last minutes to hours (and generally no more than a week), provokes a transient but

Exploration of preterm birth rates using the public health exposome database and computational analysis methods.

Recent advances in informatics technology has made it possible to integrate, manipulate, and analyze variables from a wide range of scientific disciplines allowing for the examination of complex social problems such as health disparities. This study used 589 county-level variables to identify and compare geographical variation of high and low preterm birth rates. Data were collected from a number of publically available sources, bringing together natality outcomes with attributes of the natural, built, social, and policy environments. Singleton early premature county birth rate, in counties wi

Matrix-Resolved Profiling of the Prenatal Chemical Exposome Reveals Selective Placental Retention and Fetal Transfer.

Chemical exposure during human development, especially in utero, can disrupt many biological processes and increase disease vulnerability. Assessing the chemical landscape across the maternal-fetal interface is challenging. Here we address this challenge by conducting an exposome-based analysis across a suite of integrated maternal-fetal biological compartments: maternal urine, maternal serum, placental tissue, and umbilical cord serum. Using wide-scope liquid chromatography-high-resolution mass spectrometry (LC-HRMS) in 23 mother-infant dyads from the INSULIN cohort (Spain), we profiled 659 c

Neurodegeneration in Sub-Saharan Africa: Intersections of Biological Ageing, Genomic Diversity and the Lifetime Exposome.

Neurodegenerative diseases are emerging as an important consequence of population ageing in sub-Saharan Africa (SSA), but their scientific relevance extends beyond a projected rise in case numbers. SSA combines exceptional human genomic diversity with marked heterogeneity in vascular and metabolic risk, infection, nutrition, occupational exposures, air pollution, and access to health care. This combination creates an underused setting in which to study why biological ageing produces neurodegeneration in some individuals but relative resilience in others. This review critically synthesises evid

Chemical exposome patterns in mothers and children across urbanisation levels in five European birth cohorts.

BACKGROUND: Urbanisation can be an important determinant of human exposure to synthetic chemical pollutants. The impact of contaminant exposures on health is of particular concern during susceptible periods of life, such as in utero and during childhood, when exposure may lead to adverse health effects in childhood and later adulthood. OBJECTIVE: We aimed to examine how contaminant exposures vary between urban and non-urban areas across five different European birth cohorts in Spain, France, Greece, the UK, and Lithuania. METHODS: Urine and blood samples were collected from a total of 1021 m

Exposome-wide association study and predictive risk score for incident Alzheimer's disease and related dementias.

INTRODUCTION: Environmental exposures contribute substantially to Alzheimer's disease and related dementias (ADRD), yet comprehensive exposome-wide investigations systematically evaluating multi-domain environmental factors remain limited and lacking utility for precision medicine. METHODS: We conducted an exposome-wide association study (ExWAS) among 499,992 UK Biobank participants, examining 193 exposures across five domains (social, physical, chemical, lifestyle, and ecosystems). Cox proportional hazard models with Bonferroni correction identified ADRD-associated exposures. We used the XGB

Urban Exposome and Skin Microbiome in Atopic Dermatitis: A Comparative Study Between Two Brazilian Municipalities.

INTRODUCTION: Atopic dermatitis (AD) is thought to arise from the interplay of skin barrier dysfunction, type 2 immune dysregulation, and cutaneous dysbiosis, the last of these characterized by reduced bacterial diversity and predominance of Staphylococcus aureus. The urban exposome, including air pollution and reduced environmental microbial diversity, may modulate this dysbiosis. We hypothesized that patients with AD residing in a metropolitan area with a higher pollutant burden would show greater skin dysbiosis, reflected by a higher relative abundance of S. aureus, than patients residing i

Meta-ERS: an exposome-based risk score using non-genetic factors to guide osteoporosis prevention.

BACKGROUND: Osteoporosis is influenced by both genetic and environmental factors, yet the relative contribution of the exposome remains unclear. This study aimed to systematically identify non-genetic exposures related to osteoporosis and develop an exposome risk score (ERS) to evaluate individual osteoporosis susceptibility. METHODS: We conducted an exposome-wide analysis of 477,792 UK Biobank participants to identify key exposures associated with osteoporosis. The selected exposures were combined into a weighted Meta-ERS and validated in the Scotland/Wales cohort. The Meta-ERS was further c

Exposome variations affect Drosophila bristle patterning via the regulation of proneural genes through distinct mechanisms.

How developing organisms respond to a changing environment is a fundamental question. Pollutants and temperature are major environmental factors. Using the bristle patterning of Drosophila as a model system, we observed that cold temperature and methotrexate, a medical drug that contaminates wastewaters, increase dorsocentral (DC) bristle number, a trait normally robust. The patterning of bristles is well understood and involves the achaete-scute (ac-sc) proneural genes. Modular enhancers activate ac-sc expression in groups of cells, called proneural clusters, from which bristle precursors are

The Tissue-Specific Eco-Exposome: Differential Pharmaceutical Bioaccumulation and Disposition in Fish among Trophic Positions.

Though bioaccumulation of pharmaceuticals by aquatic organisms continues to receive scientific attention, the internal disposition of these contaminants among different tissue compartments of fish species has been infrequently investigated, particularly among fish at different trophic positions. We tested a human to fish biological read-across hypothesis for contaminant disposition by examining tissue-specific accumulation in three understudied species, longnose gar (Lepisosteus osseus; piscivore), gizzard shad (Dorosoma cepedianum; planktivore/detritivore), and smallmouth buffalo (Ictiobus bu

Fungal exposome, human health, and unmet needs: A 2022 update with special focus on allergy.

Humans inhale, ingest, and touch thousands of fungi each day. The ubiquity and diversity of the fungal kingdom, reflected by its complex taxonomy, are in sharp contrast with our scarce knowledge about its distribution, pathogenic effects, and effective interventions at the environmental and individual levels. Here, we present an overview of salient features of fungi as permanent players of the human exposome and key determinants of human health, through the lens of fungal allergy and other fungal hypersensitivity reactions. Improved understanding of the fungal exposome sheds new light on the e

Parental, prenatal, and neonatal associations with ball skills at age 8 using an exposome approach.

There is little consistency in the literature concerning factors that influence motor coordination in children. A hypothesis-free "exposome" approach was used with 7359 children using longitudinal information covering 3 generations in regard to throwing a ball accurately at age 7 years. The analyses showed an independent robust negative association with mother's unhappiness in her midchildhood (6-11 years). No such association was present for study fathers. The offspring of parents who described themselves as having poor eyesight had poorer ability. This hypothesis-free approach has identified

An exploration of the breast milk nutriome, exposome and microbiome and their links to early growth in preterm infants.

Breastfeeding promotes improved growth and development in preterm infants, yet the mechanisms underlying these benefits remain unclear. This study explored the interplay of breast-milk nutritional, microbiological, and environmental chemical exposure on early preterm infant growth. In the prospective LACTACOL-cohort, growth was assessed in 137 exclusively breastfed preterm infants (including 40 twins) using Z-scores of discharge weight and fat-free mass (FFM, by air-displacement plethysmography). Breast-milk samples were analyzed for their nutriome (targeted and untargeted metabolomic and lipi

Challenges and Good Practices in Preprocessing and Normalization of Untargeted DNA Adductomics Data in Exposomics Research.

DNA adductomics is the study of the whole of DNA adducts in a biological sample and is a valuable asset to exposomics research. To date, a clear view on how to analyze larger sample series is lacking in DNA adductomics, and the preprocessing of untargeted DNA adductomics data is seldom applied. This work aimed to optimize a DNA adductomics data preprocessing workflow (in true untargeted mode). Building upon the xcms R package, we optimized parameters for peak detection, retention time alignment, and peak grouping to reliably detect and integrate putative DNA adduct LC-MS peaks. Next, to ensure

Exposome and cognition in psychosis: an exploratory study in sex-specific patterns in patients and their unaffected siblings.

BACKGROUND: The exposome is strongly linked to psychosis risk and may affect cognition in first-episode psychosis (FEP). Studying FEP patients and their unaffected siblings provides a framework to explore this relationship. This study examined whether environmental risk, measured by the Maudsley Environmental Risk Score (ERS), is associated with sex differences in cognitive performance in FEP patients and their unaffected siblings. METHODS: 72 individuals with FEP (43 males and 29 females) and 98 siblings (33 males and 65 females) from the PAFIP and PAFIP-FAMILIAS cohorts were assessed. Parti
