Comparative analysis of immunogenicity for viral hemorrhagic septicemia virus (VHSV) vaccines inactivated by different methods.

Viral hemorrhagic septicemia virus (VHSV) belonging to the genus Novirhabdovirus and family Rhabdoviridae is a major viral disease that causes economic loss in the olive flounder (Paralichthys olivaceus). In this study, we produced formalin-, binary ethylenimine (BEI)-, β-propiolactone (BPL)-, and heat-treated, inactivated VHSV vaccines; and observed the glycoprotein changes in the inactivated VHSV vaccines and confirmed their correlation with vaccine efficacy. The control group showed a 100 % cumulative mortality rate, but the groups immunized with formalin- and BPL-inactivated VHSV vaccines

Efficacy of a Combined Natural Extract of Turmeric (Curcuma longa) and Liquorice (Glycyrrhiza uralensis) Against Viral Haemorrhagic Septicaemia Virus in Olive Flounders (Paralichthys olivaceus).

Viral haemorrhagic septicaemia virus (VHSV) causes a high mortality rate in cultured olive flounder, and effective treatment for VHSV infection is urgently required. Turmeric (Curcuma longa ) and liquorice (Glycyrrhiza uralensis) are well-known medicinal plants with a variety of biological activities. Turmeric and liquorice extract combination (TLex), containing 12 g glycyrrhizin and 0.72 g curcumin per litre, exhibited low cytotoxicity (CC50: 7903.08 ± 6.26 μg/mL) and potent antiviral activity (EC50: 149.4 ± 2.34 μg/mL) in hirame natural embryo (HINAE) cells. Plaque reduction assays indicated

Umbelliferone immersion therapy controls viral hemorrhagic septicemia virus in olive flounder (Paralichthys olivaceus) via direct virucidal and immunomodulatory effects.

Viral hemorrhagic septicemia virus (VHSV) is a major threat to global aquaculture, but no effective therapeutics currently exist. In this study, we identified umbelliferone (7-hydroxycoumarin) as a potent immersion-based antiviral agent against VHSV. Umbelliferone showed complete direct virucidal effects in vitro concentration- and time-dependently. Molecular docking predicted umbelliferone may bind to the pleckstrin homology domain of VHSV glycoprotein, a possible mechanism for viral inhibition. Immersion treatment of olive flounder (Paralichthys olivaceus) decreased the VHSV replication in n

In vivo effects of NPFF and NPAF peptides on gene expression in the brain-pituitary-gonadal axis of female half-smooth tongue soles.

Neuropeptide FF (NPFF) is one of the members of RF-amide peptide family, which has been implicated in diverse physiological processes across mammals. However, there is scarce information regarding the biological function of NPFF in fish reproduction. In the current study, npff was identified and characterized in the half-smooth tongue sole. The NPFF precursor consisted of one signal peptide and two mature peptides, designated as NPFF and NPAF. Analysis of tissue distribution indicated that npff exhibited high expression levels in the brain and pituitary, while its mRNA levels were much lower i

Kinetics of viral hemorrhagic septicemia virus (VHSV) and immune response in olive flounder (Paralichthys olivaceus) at altered temperature.

Viral hemorrhagic septicemia virus (VHSV) is a pathogen that causes hemorrhagic septicemia in olive flounder at water temperatures below 15 °C, leading to symptoms such as abdominal swelling due to ascites and muscle hemorrhaging, and in severe cases, mortality. In this study, we investigated the proliferation of NCCs, the transcriptional analysis of CD4 and CD8, and the expression of pro-inflammatory cytokines (IL-1β, IL6, TNFα) and the cytokines (IL12, IL15, IFN-1β, IFNγ) involved in cytotoxic cell activation in the kidney of olive flounder during VHSV infection at suboptimal temperature (17

Gene network analysis reveals a core set of genes involved in the immune response of Japanese flounder (Paralichthys olivaceus) against Vibrio anguillarum infection.

Japanese flounder (Paralichthys olivaceus) is one of the most economically important marine fish cultured in north Asia. Vibrio anguillarum is a severe bacterial pathogen to Japanese flounder and many other aquaculture species. In order to understand the immune response of flounder during bacterial infection, we systematically examined the transcriptome profiles of flounder spleen at three time points after V. anguillarum challenge. More than one billion high quality reads were obtained, approximately 80.70% of which were successfully mapped to the reference genome of flounder. A total of 6060

Dynamic distribution of formalin-inactivated Edwardsiella tarda in flounder (Paralichthys olivaceus) post intraperitoneal vaccination.

In order to investigate the dynamic distribution of antigen in different tissues post vaccination, an absolute real-time quantitative PCR was employed to detect the amount of antigen in flounder (Paralichthys olivaceus) post intraperitoneal (i.p.) injection with three concentrations (107, 108, 109 CFU ml-1) of formalin-inactivated Edwardsiella tarda bacterin. The results showed that the amount of uptaken antigen quickly increased and then decreased in different tissues. The peak occurred first in the spleen and head kidney at 6-9 h after injection, and in the liver and blood at 9-15 h, then in

Epibiotic green alga (Ulva spp.) on the skin of turbot (Scophthalmus maximus) with chronic aeromoniasis.

Chronic aeromoniasis in turbot (Scophthalmus maximus), caused by Aeromonas salmonicida, provokes cutaneous lesions that occasionally expose dermal tubercles, which may act as unconventional ecological niches. A storm in February 2024 caused severe damage at a turbot farm in Galicia (Northwestern Spain), leaving various tanks uncovered and subsequently exposing fish with chronic aeromoniasis to several days of intense solar irradiation. Afterward, farm staff observed green tufts over the animals. Two affected turbots were submitted for necropsy. Morphological features identified the epibiont as

Comparison of toxic effects of phenanthrene and perfluorooctane sulfonate on Chinese tongue sole Cynoglossus semilaevis larva in a muti-biomarker approach.

Phenanthrene (Phe) and perfluorooctane sulfonate (PFOS) are among the most frequently detected contaminants in the coastal environment of China. However, comparative studies on the toxic effects of these two chemicals in fish are limited. This study examined biomarker responses in Chinese tongue sole (Cynoglossus semilaevis) larvae exposed to sublethal Phe/PFOS (10 and 100 μg/L) for 7 days. The results showed that both Phe and PFOS significantly altered antioxidant indexes, such as activities of superoxide dismutase (SOD), catalase (CAT) and peroxidase (POD), and the detoxification metabolic p

Glial cell line-derived neurotrophic factor (GDNF) is essential for colonization and expansion of turbot (Scophthalmus maximus) germ cells in recipients and in vitro culture.

Germ cell transplantation (GCT) is a promising biotechnology that enables the production of donor-derived gametes in surrogate recipients. It plays a crucial role in the protection of endangered species, the propagation of elite species with desired traits, and long-term preservation of genetic resources. This significance is particularly pronounced when GCT is synergistically employed with cryopreservation techniques. However, GCT often encounters challenges due to low colonization rates and, in some cases, complete loss of donor cells in recipients. Glial cell line-derived neurotrophic facto

The role of marine biotoxins on the trophic transfer of Mn and Zn in fish.

Essential nutrients are critical for physiological processes of organisms. In fish, they are obtained primarily from the diet, and their transfer and accumulation are known to be impacted by environmental variables such as water temperature, pH and salinity, as well as by diet composition and matrices. Yet, prey items consumed by fish may also contain toxic compounds such as marine toxins associated with harmful algae. These biotoxins have the potential to affect essential trace element assimilation in fish through chemical interactions such as the formation of trace element-toxin complexes or

Transcriptome profiling provides gene resources for understanding gill immune responses in Japanese flounder (Paralichthys olivaceus) challenged with Edwardsiella tarda.

Marine organisms are commonly under threats from various pathogens. Edwardsiella tarda is one of the fish pathogens that seriously infect cultured and wild fish species. Bacteremia caused by E. tarda can be a fatal disease in humans. Fish gill is a mucosa-associated lymphoid tissue that directly contacted with sea water. Generating gill transcriptomic resources that challenged by E. tarda is crucial for understanding the molecular mechanisms underlying gill immune responses. In this study, we performed transcriptome profiling of gene expression in Japanese flounder gills (Paralichthys olivaceu

Evolution of the Proto Sex-Chromosome in Solea senegalensis.

Solea senegalensis is a flatfish belonging to the Soleidae family within the Pleuronectiformes order. It has a karyotype of 2n = 42 (FN = 60; 6M + 4 SM + 8 St + 24 T) and a XX/XY system. The first pair of metacentric chromosomes has been proposed as a proto sex-chromosome originated by a Robertsonian fusion between acrocentric chromosomes. In order to elucidate a possible evolutionary origin of this chromosome 1, studies of genomic synteny were carried out with eight fish species. A total of 88 genes annotated within of 14 BACs located in the chromosome 1 of S. senegalensis were used to elabor

Hind-casting the quantity and composition of discards by mixed demersal fisheries in the North Sea.

Many commercial fisheries seek to maximise the economic value of the catch that they bring ashore and market for human consumption by discarding undersize or low value fish. Information on the quantity, size and species composition of discarded fish is vital for stock assessments and for devising legislation to minimise the practice. However, except for a few major species, data are usually extremely sparse and reliant on observers aboard a small sample of fishing vessels. Expanding these data to estimate total regional discards is highly problematic. Here, we develop a method for utilising ad

Isolation and characterization of Toll-like receptor 9 in half-smooth tongue sole Cynoglossus semilaevis.

Toll-like receptors (TLRs) are considered as key sensors to trigger the host's innate immune system and adaptive immune responses by recognizing various PAMPs and initiating signal transduction. TLR9, as a member of TLR family, mediates the recognition of unmethylated CpG dinucleotide motifs commonly found in both bacterial and viral genomes. In the current study, the TLR9 gene was isolated from one of flatfish species, half-smooth tongue sole (Cynoglossus semilaevis). In the 4588 bp genomic sequence, three exons, two introns, and 5' UTR of 23 bp and 3' UTR of 342 bp were identified. Putative

Chromatophore distribution and inferior performance of albino Japanese flounder Paralichthys olivaceus with special reference to different chromatophore expression between albinism and pseudo-albinism.

Albinism with a large variation in body color was found in a hatchery population of Japanese flounder. In addition to albinism, ambicoloration and pseudo-albinism were simultaneously observed in some individuals. Albinos had a remarkably lower number of melanophores on the scales of ocular side than wild-type individuals did, although no significant difference was observed in the numbers of xanthophores and iridophores. The intensity of body color significantly correlated with the number of melanophores among the albinos. No significant differences were observed in the intensity of body color

Identification and Functional Analysis of tgfb2b Gene in Ovarian Development of Chinese Tongue Sole (Cynoglossus semilaevis).

Transforming growth factor β (TGF-β) superfamily members are critical in teleost sex determination and differentiation. Tgfb2b is an important TGF-β ligand gene exhibiting dominant expression in the ovary of Chinese tongue sole (Cynoglossus semilaevis), yet its function in sex regulation remains unclear. In the present study, the gene expression pattern, transcriptional regulation, and knockdown effect were examined. Its expression persisted and showed a gradual increase throughout ovarian development from 3 months to 1.5 years post-hatching. In situ hybridization (ISH) revealed that the gene
