Inhibition of beta-galactosidase biosynthesis in Escherichia coli by tetracycline residues in milk.

Low levels of tetracyclines found as residues in milk inhibited the biosynthesis of beta-galactosidase in Escherichia coli. To produce the same effect, other antibacterials had to occur in concentrations that were more than 10-fold higher. This relative selectivity was exploited for the development of a screening test for tetracyclines in milk based on a chemiluminometric assay of beta-galactosidase. The method was validated with spiked samples of raw milk and applied to field samples contaminated with tetracyclines.

Ganglioside GM1 metabolism in living human fibroblasts with beta-galactosidase deficiency.

The uptake and catabolism of [3H-ceramide]-GM1 was followed in living fibroblasts from patient with different forms of beta-galactosidase deficiency. Gangliosides are identified according to the nomenclature of Svennerholm (1963). A total inability to metabolize the ingested substrate was found in infantile GM1-gangliosidosis whereas cells from an adult GM1-gangliosidosis variant showed a slower rate of degradation, compared with controls. Morquio B fibroblasts had a comparable catabolism of GM1 as controls. Fibroblasts from different types of galactosialidosis, a recessive disease associated

The relation between human lysosomal beta-galactosidase and its protective protein.

Cultured skin fibroblasts from patients with the lysosomal storage disease galactosialidosis lack a 54-kDa protein which is a precursor of 32-kDa and 20-kDa proteins, which immunoprecipitate with human anti-beta-galactosidase antiserum. The lack of a 32-kDa "protective protein" results in a combined deficiency of beta-galactosidase and sialidase. The mechanism of protection of lysosomal beta-galactosidase against proteolytic degradation is elucidated by sucrose density gradient centrifugation and immunoprecipitation studies. In normal fibroblasts at the low intralysosomal pH, more than 85% of

Dog GM1 gangliosidosis: characterization of the residual liver acid beta-galactosidase.

The residual liver acid beta-galactosidase activity from the first documented case of GM1 gangliosidosis in dogs was partially purified and characterized with respect to kinetic properties, thermostability, isoelectric point, molecular weight, and antigenicity. The GM1 dog liver beta-galactosidase appears to be identical with the normal dog liver enzyme in all properties examined. The canine disease is strikingly different from the human disease in the amount of enzyme that is present in the tissue. Unlike the human disease, in which normal amounts of catalytically defective beta-galactosidase

Globoid cell leucodystrophy (Krabbe's disease): deficiency of galactocerebroside beta-galactosidase.

Profound deficiency of a specific enzyme, galactocerebroside beta-galactosidase, has been demonstrated in the brains, liver, and spleen of three patients with Krabbe's globoid cell leucodystrophy. The activity of this enzyme was normal in a variety of other cerebral diseases, including those with similarly devasted white matter. The lack of enzyme activity was not due to an inhibitor in the tissue, nor is it due to a shift in the pH optimum. The deficiency of galactocerebroside beta-galactosidase as the primary enzymatic defect can account for the morphological and biochemical characteristics

Synthesis of C-5a-chain extended derivatives of 4-epi-isofagomine: Powerful β-galactosidase inhibitors and low concentration activators of GM1-gangliosidosis-related human lysosomal β-galactosidase.

From an easily available partially protected formal derivative of 1-deoxymannojirimycin, by hydroxymethyl chain-branching and further elaboration, lipophilic analogs of the powerful β-d-galactosidase inhibitor 4-epi-isofagomine have become available. New compounds exhibit improved inhibitory activities comparable to benchmark compound NOEV (N-octyl-epi-valienamine) and may serve as leads towards improved and more selective pharmacological chaperones for GM1-gangliosidosis.

Evaluation of lactulose synthesis using immobilized β-galactosidase in different reactor configurations.

Lactulose is a disaccharide composed of galactose and fructose. In the human body, it functions as a prebiotic, promoting the growth of beneficial bacteria, such as bifidobacteria and lactobacilli, in the gastrointestinal tract. Lactulose is produced via a transgalactosylation reaction between lactose and fructose, catalyzed by β-galactosidase. Prominent β-galactosidases used in lactulose synthesis include those from Aspergillus oryzae and Kluyveromyces lactis. Various methodologies have been developed to enhance the efficiency of enzymatic lactulose production. The objective of this study was

Novel Galactosidase-Beta-1 Variant in Infantile GM1 Gangliosidosis: A Case Report.

GM1 gangliosidosis is an autosomal recessive lysosomal storage disorder caused by pathogenic GLB1 variants that impair β-galactosidase activity, resulting in GM1 ganglioside accumulation. The infantile (type I) form is the most severe. We describe the case of a one-year-old girl born to consanguineous parents who presented with developmental regression, hypotonia, coarse facial features, hepatosplenomegaly, macular cherry-red spots, Mongolian spots, and sensorineural hearing loss. Whole-exome sequencing revealed a novel homozygous GLB1 variant, NM_000404.4:c.1525T>A (p.Trp509Arg), absent from

CRISPR-Cas9 engineering for high-level β-galactosidase production in Saccharomyces boulardii.

Saccharomyces boulardii is a clinically validated probiotic yeast widely used to treat gastrointestinal disorders. Its natural probiotic effects, combined with the capacity of surviving the harsh gastrointestinal conditions, makes it an attractive chassis for therapeutic protein delivery. However, its engineering has relied on a limited set of genetic tools and lacks systematic validation, which has constrained its development as a next generation engineered probiotic. In this study, the compatibility of the EasyClone-MarkerFree vector system with CRISPR-Cas9 editing in S. boulardii was system

Immobilization of the β-galactosidase enzyme by encapsulation in polymeric matrices for application in the dairy industry.

Lactose intolerance affects ∼65% of the global adult population, leading to the demand for lactose-free products. The enzyme β-galactosidase (βG) is commonly used in the industry to produce such products, but its recovery after lactose hydrolysis is challenging. In this scenario, the study aims to encapsulate βG within capsules, varying in dimensions and wall materials, to ensure their suitability for efficient industrial recovery. The enzyme βG was encapsulated through ionic gelation using alginate and its blends with pectin, maltodextrin, starch, or whey protein as wall materials. The capsul

The development of a broad-spectrum retaining β-exo-galactosidase activity-based probe.

Acid β-galactosidase (GLB1) and galactocerebrosidase (GALC) are retaining exo-β-galactosidases involved in lysosomal glycoconjugate metabolism. Deficiency of GLB1 may result in the lysosomal storage disorders GM1 gangliosidosis, Morquio B syndrome, and galactosialidosis, and deficiency of GALC may result in Krabbe disease. Activity-based protein profiling (ABPP) is a powerful technique to assess the activity of retaining glycosidases in relation to health and disease. This work describes the use of fluorescent and biotin-carrying activity-based probes (ABPs) to assess the activity of both GLB1

Caffeine and Theophylline Inhibit β-Galactosidase Activity and Reduce Expression in Escherichia coli.

The β-galactosidase enzyme is a common reporter enzyme that has been used extensively in microbiological and synthetic biology research. Here, we demonstrate that caffeine and theophylline, common natural methylxanthine products found in many foods and pharmaceuticals, negatively impact both the expression and activity of β-galactosidase in Escherichia coli. The β-galactosidase activity in E. coli grown with increasing concentrations of caffeine and theophylline was reduced over sixfold in a dose-dependent manner. We also observed decreasing lacZ mRNA transcript levels with increasing methylxa

Spray congealed lipid microparticles for the local delivery of β-galactosidase to the small intestine.

Oral local delivery of therapeutic biologics is generally limited due to the multiple obstacles of the gastrointestinal (GI) tract, mainly represented by acidic stomach pH and digestive enzymes. In the present study, spray congealing was used to prepare solid lipid microparticles (SLMs) loaded with β-galactosidase (lactase), an enzyme used for the treatment of lactose intolerance, to achieve a local drug delivery to the small intestine. Lactase was characterized in terms of activity at different pH, kinetic parameters and proteolytic degradation by digestive enzymes. Then, five lipid excipient

Affective and cognitive behavior in the alpha-galactosidase A deficient mouse model of Fabry disease.

Fabry disease is an X-linked inherited lysosomal storage disorder with intracellular accumulation of globotriaosylceramide (Gb3) due to α-galactosidase A (α-Gal A) deficiency. Fabry patients frequently report of anxiety, depression, and impaired cognitive function. We characterized affective and cognitive phenotype of male mice with α-Gal A deficiency (Fabry KO) and compared results with those of age-matched male wildtype (WT) littermates. Young (3 months) and old (≥ 18 months) mice were tested in the naïve state and after i.pl. injection of complete Freund`s adjuvant (CFA) as an inflammatory
