Selective Intrinsically Photosensitive Retinal Ganglion Cells Ablation Regulates Myopia Development Through the Choroid/MAPK Pathway.

PURPOSE: This study investigates the role of selective ablation of intrinsically photosensitive retinal ganglion cells (ipRGCs) in regulating axial length (AL) and scleral remodeling during myopia progression. METHODS: Three-week-old pigmented guinea pigs were randomly assigned to five experimental groups, with form-deprivation myopia (FDM) models receiving intravitreal melanopsin-saporin (MEL-SAP) injections. Molecular analyses (Western blot, RT-qPCR) quantified expression of retina-, choroid-, and sclera-related factors. Structural changes were assessed via immunohistochemistry, optical coh

Brain functional MRI responses to blue light stimulation in Leber's hereditary optic neuropathy.

Melanopsin retinal ganglion cells (mRGCs) are intrinsically photosensitive photoreceptors contributing both to image and non-image-forming (NIF) functions of the eye. They convey light signal to the brain to modulate circadian entrainment, sleep, alertness, cognition, brightness perception and coarse vision. Given that rods and cones also contribute to all these impacts of light, isolating mRGC visual and NIF roles in humans is challenging so that mRGC functions remains to be fully characterized. Here, we evaluated light-driven visual and cognitive brain responses in Leber's Hereditary Optic N

Degeneration of ipRGCs in Mouse Models of Huntington's Disease Disrupts Non-Image-Forming Behaviors Before Motor Impairment.

Intrinsically photosensitive retinal ganglion cells (ipRGCs), which express the photopigment melanopsin, are photosensitive neurons in the retina and are essential for non-image-forming functions, circadian photoentrainment, and pupillary light reflexes. Five subtypes of ipRGCs (M1-M5) have been identified in mice. Although ipRGCs are spared in several forms of inherited blindness, they are affected in Alzheimer's disease and aging, which are associated with impaired circadian rhythms. Huntington's disease (HD) is an autosomal neurodegenerative disease caused by the expansion of a CAG repeat i

Pupillary response to blue light as a biomarker of seasonal pattern in Major Depressive Episode: A clinical study using pupillometry.

Depressive disorders are characterized by disturbances in light signal processing. More specifically, an alteration of the melanopsin response is suggested. The post-illumination pupillary response (PIPR) to blue light (post-blue PIPR) is increasingly used as a marker of the activity of intrinsically photosensitive melanopsin ganglion cells (ipRGCs). We hypothesized that individuals with Major Depressive Episode (MDE) who exhibited a higher vulnerability to season patterns showed a decreased ability to transmit light signals to the brain. We explored the correlation between the post-blue PIPR

Contribution of the eye and of opn4xa function to circadian photoentrainment in the diurnal zebrafish.

The eye is instrumental for controlling circadian rhythms in mice and human. Here, we address the conservation of this function in the zebrafish, a diurnal vertebrate. Using lakritz (lak) mutant larvae, which lack retinal ganglion cells (RGCs), we show that while a functional eye contributes to masking, it is largely dispensable for the establishment of circadian rhythms of locomotor activity. Furthermore, the eye is dispensable for the induction of a phase delay following a pulse of white light at CT 16 but contributes to the induction of a phase advance upon a pulse of white light at CT21. Chromatic Pupillometry Findings in Alzheimer's Disease.

Intrinsically photosensitive melanopsin retinal ganglion cells (mRGCs) are crucial for non-image forming functions of the eye, including the photoentrainment of circadian rhythms and the regulation of the pupillary light reflex (PLR). Chromatic pupillometry, using light stimuli at different wavelengths, makes possible the isolation of the contribution of rods, cones, and mRGCs to the PLR. In particular, post-illumination pupil response (PIPR) is the most reliable pupil metric of mRGC function. We have previously described, in post-mortem investigations of AD retinas, a loss of mRGCs, and in th

Profound defects in pupillary responses to light in TRPM-channel null mice: a role for TRPM channels in non-image-forming photoreception.

TRPM1 is a spontaneously active non-selective cation channel that has recently been shown to play an important role in the depolarizing light responses of ON bipolar cells. Consistent with this role, mutations in the TRPM1 gene have been identified as a principal cause of congenital stationary night blindness. However, previous microarray studies have shown that Trpm1 and Trpm3 are acutely regulated by light in the eyes of mice lacking rods and cones (rd/rd cl), a finding consistent with a role in non-image-forming photoreception. In this study we show that pupillary light responses are signif

Chromatic Pupillometry - a New Technique for Assessing Function in Glaucoma?

Chromatic pupillometry allows quantification of photoreceptor-driven (extrinsic) and melanopsin-driven (intrinsic) responses of the intrinsic-photosensitive retinal ganglion cells (ipRGCs). This small subpopulation of retinal ganglion cells is also affected by glaucoma, making chromatic pupillometry a potential diagnostic tool. Studies show reduced phasic and tonic responses in glaucoma patients. The diagnostic value in earlier studies depended on the technical details and the study design. The purpose of this article is to give an introduction into the principles of chromatic pupillometry and

Retino-retinal projection in juvenile and young adult rats and mice.

Identification of retino-retinal projecting RGCs (ret-ret RGCs) has been accomplished by tracing RGCs in one retina after intravitreal injection of different tracers in the other eye. In mammals, rabbit and rat, ret-ret RGCs are scarce and more abundant in newborn than in adult animals. To our knowledge, ret-ret RGCs have not been studied in mice. Here we purpose to revisit the presence of ret-ret RGCs in juvenile and young adult rats and mice by using retrograde tracers applied to the contralateral optic nerve instead of intravitreally. In P20 (juvenile) and P60 (young adult) animals, the lef

A Role for Thalamic Projection GABAergic Neurons in Circadian Responses to Light.

The thalamus is an important hub for sensory information and participates in sensory perception, regulation of attention, arousal and sleep. These functions are executed primarily by glutamatergic thalamocortical neurons that extend axons to the cortex and initiate cortico-thalamocortical connectional loops. However, the thalamus also contains projection GABAergic neurons that do not extend axons toward the cortex. Here, we have harnessed recent insight into the development of the intergeniculate leaflet (IGL) and the ventral lateral geniculate nucleus (LGv) to specifically target and manipula

Effect of light on blinking in patients with idiopathic isolated blepharospasm.

OBJECTIVE: Melanopsin may be involved in the pathophysiology of photophobia in idiopathic isolated blepharospasm. We assessed the efficacy of blocking wavelengths of melanopsin absorption to reduce blinking in blepharospasm as a possible surrogate for photophobia. METHODS: Twenty-one participants (11 blepharospasm and 10 healthy controls) were studied. There were three sessions: (1) a baseline condition to measure the blink rate (BR) without intervention; (2) two conditions where the participants received intermittent light stimuli with high or low intensity without wearing study lenses; (3)

Biological rhythms in the context of light at night (LAN).

In mammals including man, the most important zeitgeber for endogenous rhythms is the environmental light/dark cycle. Mammals perceive light through the eyes and that perception is relayed to the suprachiasmatic nucleus (SCN) by means of neuronal signals. The SCN, in turn, innervates the pineal gland, resulting in the production and release of melatonin almost exclusively during night-time hours. Thus, besides object recognition, eyes serve as the sensory organ for detecting the presence or absence of light. The way that light entrains the SCN is still a matter of intense research. It has been

[Not Available].

In addition to the rods and cones, the melanopsin-containing intrinsically photosensitive retinal ganglion cells (ipRGC) are responsible for encoding ambient illumination conditions. The main task of the ipRGC is to signal information about illumination to the brain. Via the retinohypothalamic pathway, light synchronises our internal biological clock and sets it to follow the environment's light-dark cycle. Light is the most important zeitgeber for our inner clock and the ipRGC are therefore highly important for the field of chronobiology. Many ocular and retinal functions also follow a diurna

Roles of the nonvisual opsins in the mammalian retina.

In addition to the rods and cones that subserve vision, several classes of mammalian retinal ganglion cells are also directly photoreceptive. These "intrinsically photosensitive" retinal ganglion cells ("ipRGCs") utilize the nonvisual photopigments melanopsin (Opn4), neuropsin (Opn5), and encephalopsin (Opn3), which are expressed in largely nonoverlapping ganglion cell populations. Remarkably, all three nonvisual opsins appear to influence the circadian clock, but in different ways. Opn4-expressing cells project from the retina to brain regions regulating circadian rhythms, such as the suprach

Chloroxylenol impairs the visual system, induces visually-mediated behavioral abnormalities and circadian rhythm disorders in zebrafish (Danio rerio).

p-Chloro-m-xylenol (PCMX) is a widely used broad-spectrum antimicrobial agent. Its extensive production, utilization and discharge have led to frequent detection in aquatic environments and biological samples, posing potential ecological risks. However, its toxic effects on the visual system and visual-related behaviors of aquatic organisms remain poorly understood. Here, zebrafish larvae were exposed to 0.4, 1.2, 2 and 2.8 mg/L PCMX to investigate impacts on ocular morphology, retinal development, visual-mediated behaviors and circadian rhythms. The results demonstrated that PCMX at ≥0.4 mg/L

[Sleep disturbance associated with Smith-Magenis syndrome].

Smith-Magenis syndrome (SMS) (OMIM #182290) is a rare genetic disorder with a prevalence of 1 in 25 000 live births. Approximately 90% of SMS patients have harbored a 3.7 Mb interstitial 17p11.2 deletion involving the RAI1 gene, while 10% of cases have carried pathogenic variants of the RAI1 gene. SMS is characterized by sleep disturbance, intellectual impairment, developmental delay, craniofacial and cardiovascular anomalies, obesity, self injury, aggressive and autistic-like behaviors. Most SMS patients have sleep disorders such as short total sleep time, frequent night waking, short sleep o

Flp-recombinase mouse line for genetic manipulation of ipRGCs.

Light has myriad impacts on behavior, health, and physiology. These signals originate in the retina and are relayed to the brain by more than 40 types of retinal ganglion cells (RGCs). Despite a growing appreciation for the diversity of RGCs, how these diverse channels of light information are ultimately integrated by the ~50 retinorecipient brain targets to drive these light-evoked effects is a major open question. This gap in understanding primarily stems from a lack of genetic tools that specifically label, manipulate, or ablate specific RGC types. Here, we report the generation and charact

Effect of monochromatic light on the circadian clock of cultured chick retinal tissue.

The central biological clock system of bird is formed by hypothalamus suprachiasmatic nucleus, pineal gland and retina thereby interacting with each other in a neuroendocrine loop. Previous results have confirmed that monochromatic light can influence the clock genes in the pineal gland, hypothalamus and retina of chicks in vivo. The present work was conducted to study whether the cultured retinal tissue of chick could maintain the circadian oscillation and whether the monochromatic light affect the expression level of cultured retinal circadian clock in vitro. Retinal tissues of 0-day-old chi
