Development of Functional or Medical Foods for Oral Administration of Insulin for Diabetes Treatment: Gastroprotective Edible Microgels.

Insulin and an antacid [Mg(OH)2] were co-encapsulated inside calcium alginate microgels (diameter = 280 μm) using a vibrating nozzle injector. Confocal microscopy indicated that insulin was successfully encapsulated inside the microgels and remained inside them after they were exposed to simulated gastric conditions. Localized fluorescence intensity measurements indicated that the internal pH of the antacid-loaded microgels was around pH 7.4 after incubation in acidic gastric fluids but below the limit of detection (pH < 4) in the antacid-free microgels. After incubation in small intestine con

Biphasic microgel-based bioinks with balanced printability, mechanical stability, and cytocompatibility for extrusion-based bioprinting.

A primary challenge in extrusion-based bioprinting using microgel-based bioink is balancing printability, mechanical stability, and biological functionality. In this study, we developed a microgel-based biphasic bioink that leverages microgels' advantages while maintaining porosity to enhance cellular performance. This biphasic bioink consists of densely packed physically crosslinked silk acid-based microgels (SA-MG) suspended in a secondary hydrogel precursor of alginate/carboxymethyl cellulose (Alg/CMC), which crosslinks physically after printing. The resulting dynamic, hybrid matrix enhance

Refractive-index-matched jammed microgel assemblies enable volumetric printing of porous hydrogel architectures.

Jammed microgels form porous hydrogel architectures in which packing-derived interstitial voids provide transport-accessible pathways while retaining the processability of soft granular matter. However, the same microgel-continuous phase interfaces scatter and redistribute projected light, complicating volumetric printing. Here, we develop refractive-index-matched jammed microgel assemblies by independently tuning the refractive indices of gelatin/acrylamide microgels and an immiscible silicone-oil continuous phase. Cooling-induced gelatin gelation stabilizes discrete microgels, whereas acryla

Sprayable Dual-Network Microgel Adhesive for Robust Wet Adhesion and Prolonged Analgesia in Complex Wounds.

Managing deep, irregular wounds remains challenging due to poor suturability, persistent bleeding-induced wet environments, and neuropathic pain. Here, we have developed a sprayable dual-network microgel adhesive (PR) that rapidly covers complex wound geometries, combining strong wet tissue adhesion with sustained ropivacaine release. The sprayable powder format allows PR microgels to penetrate and adhere to irregular wound beds that conventional dressings cannot reach, with uniform particle size ensuring complete wound protection. Notably, PR maintains exceptional wet adhesion (∼40 kPa, over

Fabrication of Single-Bacterium Microgel with Gas-Shearing Strategy for Precision Probiotic Delivery in IBD Therapy.

The human gut microbiome is essential for maintaining health, as it substantially impacts immune regulation and overall balance within the body. Accordingly, disruptions in this microbial community are associated with various diseases. Probiotics offer a promising solution, but their effectiveness is often hampered by challenges related to gastrointestinal delivery. To overcome the issue of probiotic survival in the gastrointestinal system, researchers have explored various encapsulation techniques. However, traditional coarse encapsulation techniques lack precision and effective targeting, li

Electrochemical Controlling of Double Microgel Layer Formation on an Electrode Surface via an Electrosensitive Inclusion Complex.

In this study, we demonstrate the formation of a self-assembled microgel double layer on an electrode surface, utilizing the ability to form electro-responsive, reversible inclusion complexes between microgels modified with ferrocene and β-cyclodextrin in these systems. The bottom layer was based on microgels containing ferrocene moieties and derivatives of cysteine. The presence of the amino acid derivative enabled the formation of the well-packed monolayer on the gold surface through chemisorption, while ferrocene was responsible for electroactivity. The addition of βCD-modified microgel led

Adhesive Nanoparticle-in-Microgel System with ROS Scavenging Capability and Hemostatic Activity for Postoperative Adhesion Prevention.

Postoperative adhesion is a noteworthy clinical complication in abdominal surgery due to the existing physical barriers are unsatisfactory and inefficient in preventing its occurrence. In this work, an elaborate nanoparticle-in-microgel system (nMGel) is presented for postoperative adhesion prevention. nMGel is facilely formed by crosslinking manganese dioxide (MnO2) nanoparticles-loaded gelatin microspheres with polydopamine using a modified emulsification-chemical crosslinking method, generating a nano-micron spherical hydrogel. After drying, powdery nMGel with sprayability can perfectly cov

FLIM nanoscopy resolves the structure and preferential adsorption in the co-nonsolvency of PNIPAM microgels in methanol-water.

Polymer microgels are swollen macromolecular networks with a typical size of hundred of nanometers to several microns that show an extraordinary open and responsive architecture to different external stimuli, being therefore important candidates for nanobiotechnology and nanomedical applications such as biocatalysis, sensing and drug delivery. It is therefore crucial to understand the delicate balance of physical-chemical interactions between the polymer backbone and solvent molecules that to a high extent determine their responsivity. In particular, the co-nonsolvency effect of poly(N-isoprop

Janus-Structured Microgel Barrier with Tissue Adhesive and Hemostatic Characteristics for Efficient Prevention of Postoperative Adhesion.

Postoperative adhesion (POA) is a common and serious complication following various types of surgery. Current physical barriers either have a short residence time at the surgical site with a low tissue attachment capacity or are prone to undesired adhesion formation owing to the double-sided adhesive property, which limits the POA prevention efficacy of the barriers. In this study, Janus-structured microgels (Janus-MGs) with asymmetric tissue adhesion capabilities are fabricated using a novel bio-friendly gas-shearing microfluidic platform. The anti-adhesive side of Janus-MGs, which consists o

Microgel Encapsulated Mesoporous Silica Nanoparticles for Releasing Wnt16 to Synergistically Treat Temporomandibular Joint Osteoarthritis.

Temporomandibular joint osteoarthritis (TMJOA) is a commonly encountered degenerative joint disease in oral and maxillofacial surgery. Recent studies have shown that the excessive unbalanced activation of Wnt/β-catenin signaling is connected with the pathogenesis of TMJOA and due to the inability to inhibit the over-activated Wnt pathway, while Wnt16-deficient mice has a more severe Knee OA. However, the efficacy of direct intra-TMJ injection of Wnt16 for the relief of TMJOA is still not directly confirmed. Moreover, small-molecule drugs such as Wnt16 usually exhibit short-lived efficacy and p

Gelatin microgel-coated balloon catheter with enhanced delivery of everolimus for long-term vascular patency.

In-stent restenosis (ISR) after percutaneous coronary intervention is a major reason for limited long-term patency due to complex neointimal proliferation caused by vascular injury. Drug-coated balloon (DCB) has been developed to treat various cardiovascular diseases including ISR by providing anti-proliferative drugs into blood vessel tissues. However, a significant proportion of the drug is lost during balloon tracking, resulting in ineffective drug delivery to the target region. In this study, we report an everolimus-coated balloon (ECB) using everolimus-loaded gelatin-hydroxyphenyl propion

Graphene Oxide Functionalized Gelatin Methacryloyl Microgel for Enhanced Biomimetic Mineralization and in situ Bone Repair.

INTRODUCTION: The formation of bone-like apatite (Ap) on natural polymers through biomimetic mineralization using simulated body fluid (SBF) can improve osteoconductivity and biocompatibility, while lowering immunological rejection. Nonetheless, the coating efficiency of the bone-like Ap layer on natural polymers requires improvement. Carbonyls (-COOH) and hydroxyls (-OH) are abundant in graphene oxide (GO), which may offer more active sites for biomimetic mineralization and promote the proliferation of rat bone marrow stromal cells (BMSCs). METHODS: In this study, gelatin methacryloyl (GelMA

4D Printing of Extrudable and Degradable Poly(Ethylene Glycol) Microgel Scaffolds for Multidimensional Cell Culture.

Granular synthetic hydrogels are useful bioinks for their compatibility with a variety of chemistries, affording printable, stimuli-responsive scaffolds with programmable structure and function. Additive manufacturing of microscale hydrogels, or microgels, allows for the fabrication of large cellularized constructs with percolating interstitial space, providing a platform for tissue engineering at length scales that are inaccessible by bulk encapsulation where transport of media and other biological factors are limited by scaffold density. Herein, synthetic microgels with varying degrees of de

Cyclic Micropipette Aspiration Reveals Viscoelastic Change of a Gelatin Microgel Prepared Inside a Lipid Droplet.

Gelatin microgels prepared inside lipid droplets have a much higher elasticity than that of bulk gels because of their differences in nanostructure. This nanostructural difference in gelatin microgels is expected to provide the microgels with unique viscoelastic properties that differ from the bulk gels. To clarify this hypothesis, here we evaluated the frequency-dependent viscoelasticity of gelatin gels by developing a cyclic micropipette aspiration. The frequency-dependent relationship between storage modulus E' (reflecting elasticity) and loss modulus E″ (reflecting viscosity) was compared

Targeted Treatment of Ischemic and Fibrotic Complications of Myocardial Infarction Using a Dual-Delivery Microgel Therapeutic.

Myocardial infarction (MI), commonly known as a heart attack, affects millions of people worldwide and results in significant death and disabilities. A major cause of MI is fibrin-rich thrombus formation that occludes the coronary arteries, blocking blood flow to the heart and causing fibrin deposition. In treating MI, re-establishing blood flow is critical. However, ischemia reperfusion (I/R) injury itself can also occur and contributes to cardiac fibrosis. Fibrin-specific poly( N-isopropylacrylamide) nanogels (FSNs) comprised of a core-shell colloidal hydrogel architecture are utilized in th

"Targeted Delivery-Long-Acting Adhesion-Synergistic Treatment" Integrated Adhesive Bilayer Microgels Loaded with Costunolide for Ulcerative Colitis Therapy.

Oral colon-targeted drug delivery platforms are highly important for the treatment of ulcerative colitis (UC). In this study, an adhesive bilayer microgel (Cp@GTL) integrating time-lag positioning, interfacial mucoadhesive retention, and enzyme-triggered release was engineered for targeted colonic delivery. The adhesive bilayer microgels were loaded with costunolide (Cos), a natural sesquiterpene lactone exhibiting potent anti-inflammatory activity. The inner gel layer was constructed by crosslinking gelatin and low-methoxyl pectin through tannic acid and Ca2+ chelation, forming an interpenetr

Stem Cells from the Apical Papilla-Laden Microgels in Combination with Mandibular Advancement for Adult Condylar Cartilage Development.

Angle's Class II malocclusion, characterized by mandibular retraction, significantly impacts occlusal function and facial aesthetics. In adults, the limited development potential of the condylar cartilage poses challenges to effective mandibular remodeling. Clinically, mandibular advancement (MA) can promote the development of condylar cartilage in adolescents, but its effect on adult condylar cartilage remains controversial. Herein, MA is achieved via three methods in adult rats to investigate the effect of MA on adult condylar cartilage development. MA could stimulate the growth of adult con

Injectable microgels loaded with programmable WELNs based on mendelian randomization macroanalysis alleviate osteoarthritis by restoring the circadian rhythm.

The development of interventional therapies that can effectively alleviate joint wear and promote cartilage regeneration is a major challenge in the clinical management of osteoarthritis (OA). This study aimed to develop an injectable microgel system with synergistic therapeutic functions. For decades, plant-derived exosome-like nanovesicles (PELNs) have gradually become an emerging direction in the research and development of disease intervention methods. To determine the most suitable PELNs in the treatment of OA, this study conducted mendelian randomization (MR) analysis to reveal the prote
