Expression of the KAL gene in multiple neuronal sites during chicken development.

The human KAL gene is responsible for the X chromosome-linked Kallmann syndrome. A partial cDNA sequence from the chicken KAL homologue was determined and used to study expression of the KAL gene, by in situ hybridization, during chicken development, from day 6 of incubation. The KAL gene is mainly expressed in neurons of the central nervous system during the second half of embryonic life. High levels of transcript were detected in mitral neurons of the olfactory bulbs, in striatal neurons, in Purkinje cells of the cerebellum, in retinal neurons, and in isolated neurons of the brainstem and sp

Interpersonal motor resonance in autism spectrum disorder: evidence against a global "mirror system" deficit.

The mirror neuron hypothesis of autism is highly controversial, in part because there are conflicting reports as to whether putative indices of mirror system activity are actually deficient in autism spectrum disorder (ASD). Recent evidence suggests that a typical putative mirror system response may be seen in people with an ASD when there is a degree of social relevance to the visual stimuli used to elicit that response. Individuals with ASD (n = 32) and matched neurotypical controls (n = 32) completed a transcranial magnetic stimulation (TMS) experiment in which the left primary motor cortex

Dissociation of mirroring and mentalising systems in autism.

The role of mirror neuron systems and mentalising systems in causing poor social and communication skills in individuals with autistic spectrum conditions is hotly debated. We studied 18 adults with autistic spectrum conditions in comparison to 19 age and IQ matched typical individuals. Behavioural assessments revealed difficulties in mental state attribution and action comprehension in the autism sample. We examined brain responses when observing rational and irrational hand actions, because these actions engage mirror and mentalising components of the social brain respectively. Both typical

Dissociating neural correlates of meaningful emblems from meaningless gestures in deaf signers and hearing non-signers.

Emblems are meaningful, culturally-specific hand gestures that are analogous to words. In this fMRI study, we contrasted the processing of emblematic gestures with meaningless gestures by pre-lingually Deaf and hearing participants. Deaf participants, who used American Sign Language, activated bilateral auditory processing and associative areas in the temporal cortex to a greater extent than the hearing participants while processing both types of gestures relative to rest. The hearing non-signers activated a diverse set of regions, including those implicated in the mirror neuron system, such a

Synesthesia, sensory-motor contingency, and semantic emulation: how swimming style-color synesthesia challenges the traditional view of synesthesia.

Synesthesia is traditionally regarded as a phenomenon in which an additional non-standard phenomenal experience occurs consistently in response to ordinary stimulation applied to the same or another modality. Recent studies suggest an important role of semantic representations in the induction of synesthesia. In the present proposal we try to link the empirically grounded theory of sensory-motor contingency and mirror system based embodied simulation/emulation to newly discovered cases of swimming style-color synesthesia. In the latter color experiences are evoked only by showing the synesthet

Language, gesture, and handedness: Evidence for independent lateralized networks.

Language, gesture, and handedness are in most people represented in the left cerebral hemisphere. To explore the relations among these attributes, we collected fMRI images in a large sample of left- and right-handers while they performed language tasks and watched action sequences. Regions of interest included the frontal and parietal areas previously identified as comprising an action-observation network, and the frontal and temporal areas comprising the primary areas for language production and comprehension. All of the language areas and most of the action-observation areas showed an overal

Imagining triadic interactions simultaneously activates mirror and mentalizing systems.

Coordinated triadic interactions, involving oneself, another person, and an external object, are considered a uniquely human skill. However, the exact mechanisms underlying the ability to engage in such social interactions remain hitherto unknown. We used functional neuroimaging to investigate the neural signature of triadic interactions. For this purpose, participants viewed pictures of objects in a 3T functional magnetic resonance imaging scanner and were asked whether they could imagine this object in a social interaction with another person. We also aimed to dissociate this process from, a

Action imagery combined with action observation activates more corticomotor regions than action observation alone.

BACKGROUND AND PURPOSE: Both action observation (AO) and action imagery have been proposed as therapeutic options for stroke rehabilitation. Currently, it is not clear to what extent their underlying neuronal mechanisms differ from each other and whether one of these therapeutic options might be preferable for this purpose. METHODS: Twenty-six neurologically healthy subjects were investigated using functional magnetic resonance imaging during AO alone and during AO with additional action imagery of video clips showing simple, object-related hand actions. RESULTS: The blood oxygenation level

[Evolution of speech and hearing].

Actual spoken language of man developed only approximately 200,000 to 100,000 years ago. As a result of natural selection, man has developed hearing, which is most sensitive in the frequency regions of 200 to 4000 Hz, corresponding to those of spoken sounds. Functional hearing has been one of the prerequisites for the development of speech, although according to current opinion the language itself may have evolved by mimicking gestures with the so-called mirror neurons. Due to hearing, gesticulation was no longer necessary, and the hands became available for other purposes.

Investigating action understanding: inferential processes versus action simulation.

In our daily life, we continuously monitor others' behaviors and interpret them in terms of goals, intentions, and reasons. Despite their central importance for predicting and interpreting each other's actions, the functional mechanisms and neural circuits involved in action understanding remain highly controversial. Two alternative accounts have been advanced. Simulation theory assumes that we understand actions by simulating the observed behavior through a direct matching process that activates the mirror-neuron circuit. The alternative interpretive account assumes that action understanding

ALS mouse model SOD1G93A displays early pathology of sensory small fibers associated to accumulation of a neurotoxic splice variant of peripherin.

Growing evidence suggests that amyotrophic lateral sclerosis (ALS) is a multisystem neurodegenerative disease that primarily affects motor neurons and, though less evidently, other neuronal systems. About 75% of sporadic and familial ALS patients show a subclinical degeneration of small-diameter fibers, as measured by loss of intraepidermal nerve fibers (IENFs), but the underlying biological causes are unknown. Small-diameter fibers are derived from small-diameter sensory neurons, located in dorsal root ganglia (DRG), whose biochemical hallmark is the expression of type III intermediate filame

Müllerian inhibiting substance contributes to sex-linked biases in the brain and behavior.

Many behavioral traits and most brain disorders are common to males and females but are more evident in one sex than the other. The control of these subtle sex-linked biases is largely unstudied and has been presumed to mirror that of the highly dimorphic reproductive nuclei. Sexual dimorphism in the reproductive tract is a product of Müllerian inhibiting substance (MIS), as well as the sex steroids. Males with a genetic deficiency in MIS signaling are sexually males, leading to the presumption that MIS is not a neural regulator. We challenge this presumption by reporting that most immature ne

Antinociceptive action of botulinum toxin type A in carrageenan-induced mirror pain.

"Mirror pain" or mirror-image pain (MP) is pain opposite to the side of injury. Mechanism and frequency in humans are not known. There is no consent on therapy. Here we report that unilaterally injected botulinum toxin type A (BT-A) has bilateral effect in experimental MP, thus deserves to be investigated as therapy for this condition. We examined the localization of BT-A's bilateral antinociceptive action in MP induced by 3 % carrageenan intramuscular injection in Wistar rats. BT-A was applied peripherally (5 U/kg), into ipsilateral or contralateral hind paw pad (i.pl.) and centrally (1 U/kg)

The small GTPase Rheb affects central brain neuronal morphology and memory formation in Drosophila.

Mutations in either of two tumor suppressor genes, TSC1 or TSC2, cause tuberous sclerosis complex (TSC), a syndrome resulting in benign hamartomatous tumors and neurological disorders. Cellular growth defects and neuronal disorganization associated with TSC are believed to be due to upregulated TOR signaling. We overexpressed Rheb, an upstream regulator of TOR, in two different subsets of D. melanogaster central brain neurons in order to upregulate the Tsc-Rheb-TOR pathway. Overexpression of Rheb in either the mushroom bodies or the insulin producing cells resulted in enlarged axon projections

The Motor Basis for Misophonia.

Misophonia is a common disorder characterized by the experience of strong negative emotions of anger and anxiety in response to certain everyday sounds, such as those generated by other people eating, drinking, and breathing. The commonplace nature of these "trigger" sounds makes misophonia a devastating disorder for sufferers and their families. How such innocuous sounds trigger this response is unknown. Since most trigger sounds are generated by orofacial movements (e.g., chewing) in others, we hypothesized that the mirror neuron system related to orofacial movements could underlie misophoni

Action observation network in childhood: a comparative fMRI study with adults.

Very little is known about the action observation network and the mirror neuron system (AON/MNS) in children and its age-related properties compared with those observed in adults. In the present fMRI study we explored the activation of areas belonging to the AON/MNS in children and adults during observation of complex hand-grasping actions, as compared to observation of simple grasping acts executed with the left and the right hand, seen from a first person perspective. The results indicate that during the action observation tasks in children there was activation of a cortical network similar

The Effects of Maternal Mirroring on the Development of Infant Social Expressiveness: The Case of Infant Cleft Lip.

Parent-infant social interactions start early in development, with infants showing active communicative expressions by just two months. A key question is how this social capacity develops. Maternal mirroring of infant expressions is considered an important, intuitive, parenting response, but evidence is sparse in the first two months concerning the conditions under which mirroring occurs and its developmental sequelae, including in clinical samples where the infant's social expressiveness may be affected. We investigated these questions by comparing the development of mother-infant interaction

Reduced functional connectivity between V1 and inferior frontal cortex associated with visuomotor performance in autism.

Some recent evidence has suggested abnormalities of the dorsal stream and possibly the mirror neuron system in autism, which may be responsible for impairments of joint attention, imitation, and secondarily for language delays. The current study investigates functional connectivity along the dorsal stream in autism, examining interregional blood oxygenation level dependent (BOLD) signal cross-correlation during visuomotor coordination. Eight high-functioning autistic men and eight handedness and age-matched controls were included. Visually prompted button presses were performed with the prefer

Coregistration of heading to visual cues in retrosplenial cortex.

Spatial cognition depends on an accurate representation of orientation within an environment. Head direction cells in distributed brain regions receive a range of sensory inputs, but visual input is particularly important for aligning their responses to environmental landmarks. To investigate how population-level heading responses are aligned to visual input, we recorded from retrosplenial cortex (RSC) of head-fixed mice in a moving environment using two-photon calcium imaging. We show that RSC neurons are tuned to the animal's relative orientation in the environment, even in the absence of he
