rTMS-induced motor cortex activation drives neural network tissueoid mediated spinal motor neural pathway reconstruction.

Rationale: The integration of biological and physical interventions represents a promising therapeutic strategy for spinal cord injury (SCI), offering a novel approach to restore disrupted motor pathways. This study investigates whether repetitive transcranial magnetic stimulation (rTMS) can prevent cerebral neuroapoptosis and promote the regeneration and integration of brain-derived nerve fibers with neural network tissueoids (NNToids) following SCI. Methods: Neural stem cell-derived NNToids were transplanted into rats with complete SCI and simultaneously treated with 10 Hz rTMS. Neuroinflamm

Slow-Oscillatory Transcranial Alternating Current Stimulation Over the Primary Motor Cortex Improves Motor Skill Acquisition.

Slow-oscillatory brain activity (< 4 Hz) is crucial in memory consolidation and motor performance. Although transcranial alternating current stimulation (tACS) can modulate cortical oscillatory activity and influence motor learning, the effects of slow oscillation modulation via tACS remain unclear. This study investigated the effects of 0.75-Hz tACS applied to the primary motor cortex (M1) on the acquisition and retention of a visuomotor tracking task. Fifty-four right-handed healthy adults (mean age: 21.2 ± 0.6 years) were assigned to one of three groups: M1-tACS, Cz-tACS, or sham. In the M1

Bilateral Transcranial Direct Stimulation Over the Primary Motor Cortex Alters Motor Modularity of Multiple Muscles.

Transcranial direct current stimulation (tDCS) over the primary motor cortex (M1) has been demonstrated to modulate the motor performance of both healthy individuals and patients with neuromuscular disorders. However, the effect of tDCS on motor control of multiple muscles, which is a prerequisite to change in motor performance, is currently unknown. Using dimensionality reduction analysis, we investigated whether bilateral tDCS over M1 modulates the coordinated activity of 12 muscles. Fifteen healthy men participated in this randomized, double-blind crossover study. Each participant received

Anodal tDCS over Primary Motor Cortex Provides No Advantage to Learning Motor Sequences via Observation.

When learning a new motor skill, we benefit from watching others. It has been suggested that observation of others' actions can build a motor representation in the observer, and as such, physical and observational learning might share a similar neural basis. If physical and observational learning share a similar neural basis, then motor cortex stimulation during observational practice should similarly enhance learning by observation as it does through physical practice. Here, we used transcranial direct-current stimulation (tDCS) to address whether anodal stimulation to M1 during observational

Localized Tuning Fields for 3D Hand Position in the Primary Motor Cortex and Premotor Cortex of Macaques.

A central question in motor neuroscience is how the brain represents the state of the limbs to guide volitional movements. While the primate motor cortex is known to encode movement kinematics, such as velocity and direction, whether it also maintains a direct and explicit representation of hand position in 3D space remains debated. To address this, we recorded the activity of single neurons in the primary motor cortex (M1) and dorsal premotor cortex (PMd) of two male rhesus macaques performing a naturalistic, self-paced 3D reach-and-grasp task. We found significant populations of neurons in b

Topographic distribution of output neurons in cerebellar nuclei and cortex to somatotopic map of primary motor cortex.

To investigate the somatotopic organization of the cerebellum, we analysed multisynaptic inputs to the primary motor cortex (MI) using retrograde transneuronal transport of rabies virus. At 3 days after rabies injections into proximal forelimb, distal forelimb and hindlimb representations of the macaque MI, second-order neurons via the thalamus were labeled in the deep cerebellar nuclei, including the dentate (DN), anterior interpositus (AIN) and posterior interpositus nuclei. In the DN, the labeling of both the forelimb and hindlimb was seen mainly in the dorsal aspect. The labeling of the hi

Muscle and timing-specific functional connectivity between the dorsolateral prefrontal cortex and the primary motor cortex.

The pFC has a crucial role in cognitive control, executive function, and sensory processing. Functional imaging, neurophysiological, and animal studies provide evidence for a functional connectivity between the dorsolateral pFC (DLPFC) and the primary motor cortex (M1) during free choice but not instructed choice selection tasks. In this study, twin coil, neuronavigated TMS was used to examine the precise timing of the functional interaction between human left DLPFC and ipsilateral M1 during the execution of a free/specified choice selection task involving the digits of the right hand. In a th

Temporal lobe perceptual predictions for speech are instantiated in motor cortex and reconciled by inferior frontal cortex.

Humans use predictions to improve speech perception, especially in noisy environments. Here we use 7-T functional MRI (fMRI) to decode brain representations of written phonological predictions and degraded speech signals in healthy humans and people with selective frontal neurodegeneration (non-fluent variant primary progressive aphasia [nfvPPA]). Multivariate analyses of item-specific patterns of neural activation indicate dissimilar representations of verified and violated predictions in left inferior frontal gyrus, suggestive of processing by distinct neural populations. In contrast, precen

Motor Cortex VIP Interneurons Participate in Dexmedetomidine-Associated Sleep Modulation.

Vasoactive intestinal peptide (VIP)-expressing neurons within the motor cortex (MC) exhibit diurnal variation in excitability and may participate in the modulation of sleep architecture. In this study, we found that MCVIP neurons displayed significantly higher excitability during the light phase. Using chemogenetic approaches, we examined their contribution to sleep-wake modulation. Given the sleep-modulating actions of the α2-adrenergic receptor agonist dexmedetomidine (Dex), we further tested whether Dex engages MCVIP neurons. Dex increased MCVIP neuronal excitability in slice recordings in

Motor Cortex Disinhibition Correlates with Olfactory Dysfunction in Parkinson's Disease.

BACKGROUND: Motor cortex disinhibition, as measured by impaired short-interval intracortical inhibition (SICI) using transcranial magnetic stimulation (TMS), is a well-established feature of Parkinson's disease (PD). However, its substantial variability among patients remains unexplained, prompting questions about its origin, clinical relevance, and connection to disease heterogeneity. OBJECTIVE: Based on biological links between olfaction and motor function, we aimed to investigate the possible relationship between motor cortex disinhibition and olfactory dysfunction in PD. METHODS: We asse

Inconsistent primary motor cortex glucose hypometabolism in primary lateral sclerosis.

OBJECTIVE: Primary lateral sclerosis (PLS) is a rare neurodegenerative disorder and a model for upper motor neuron degeneration, which is believed to begin in the primary motor cortex. However, clinical observation suggests that not all PLS cases show primary motor cortex glucose hypometabolism on 2-deoxy-2-[18F]fluoro-D-glucose positron emission tomography (FDG-PET). We aimed to assess the reliability of FDG-PET in identifying motor cortex hypometabolism over disease course in a sample of patients with PLS. METHODS: Baseline FDG-PET data from nine consecutive PLS patients were analyzed. At l

Dynamic spatial coding in parietal cortex mediates tactile-motor transformation.

Movements towards touch on the body require integrating tactile location and body posture information. Tactile processing and movement planning both rely on posterior parietal cortex (PPC) but their interplay is not understood. Here, human participants received tactile stimuli on their crossed and uncrossed feet, dissociating stimulus location relative to anatomy versus external space. Participants pointed to the touch or the equivalent location on the other foot, which dissociates sensory and motor locations. Multi-voxel pattern analysis of concurrently recorded fMRI signals revealed that tac

Primary motor cortex crucial for action prediction: A tDCS study.

The neural network underlying action observation - i.e., the action observation network - forms an anticipatory representation of observed actions. Although correlational studies suggest that the motor cortex (M1) might be involved in this anticipatory coding, it is unclear whether M1 is also causally essential for making accurate predictions about observed actions. To test the functional relevance of M1 to action prediction, we used offline monopolar transcranial direct current stimulation (tDCS). In four tDCS groups of healthy participants, we administered 15 min of anodal or cathodal consta

Parietal transcranial direct current stimulation modulates primary motor cortex excitability.

The posterior parietal cortex is part of the cortical network involved in motor learning and is structurally and functionally connected with the primary motor cortex (M1). Neuroplastic alterations of neuronal connectivity might be an important basis for learning processes. These have however not been explored for parieto-motor connections in humans by transcranial direct current stimulation (tDCS). Exploring tDCS effects on parieto-motor cortical connectivity might be functionally relevant, because tDCS has been shown to improve motor learning. We aimed to explore plastic alterations of pariet

Primary motor cortex and movement prevention: where Stop meets Go.

Processes that engage frontal cortex and the basal ganglia are responsible for the prevention of planned movements. Here, we review the role of primary motor cortex (M1) in this function. M1 receives and integrates input from a range of cortical and subcortical sites. It is also the final cortical processing site for voluntary motor commands, before they descend to the spinal cord. Inhibitory networks within M1 may be an important mechanism for the prevention or suppression of movement. Transcranial magnetic stimulation (TMS) has been used to evaluate corticospinal excitability and intracortic

Motor cortex changes in spinal cord injury: a TMS study.

Using paired pulse transcranial magnetic stimulation (TMS) paradigms, we studied cortical excitability in a patient with spinal cord lesion. During posterior tibial nerve stimulation, the contextual flexion of hand fingers contralateral to the stimulated lower limb had suggested a change in motor cortex excitability. Results showed a decrease in the activity of motor cortex inhibitory circuits. This could suggest that in spinal cord injury, just as in stroke and peripheral deafferentation, a disinhibition of latent synapses within the motor cortex and the rewriting of a new motor map can occur

Decreased thickness of primary motor cortex in primary lateral sclerosis.

BACKGROUND AND PURPOSE: Primary lateral sclerosis (PLS) is a rare form of motor neuron disease characterized by upper motor neuron dysfunction. Because pathologic examination has revealed a loss of neurons in the motor cortex of patients with PLS, we sought to confirm and extend this finding by using MR imaging to measure cortical thickness. METHODS: Seven patients with PLS and 7 age-matched neurologically normal control subjects were examined with heavily T1-weighted short-tau inversion recovery (STIR) MR imaging performed at 3T. Cortical thickness in the anterior and posterior banks of both

Acamprosate reduces motor cortex excitability determined by transcranial magnetic stimulation.

Acamprosate is effective in reducing alcohol intake in weaned alcoholics. We were interested if acamprosate had an effect on the excitability of cortical motoneurons determined by transcranial magnetic stimulation (TMS). We studied 12 male healthy volunteers (mean age 29.5 years, SD = 4.8) who were either treated with 6 tablets of acamprosate (each containing 333 mg verum) per day or placebo (randomized cross-over design) for 1 week. TMS was performed after each treatment session including a paired stimulation paradigm. Motor evoked potentials (MEPs) of the placebo and verum group did not diff

Enhanced beta power emerges from simulated parkinsonian primary motor cortex.

Primary motor cortex (M1) layer 5B pyramidal tract (PT5B) neurons develop intrinsic pathology in rodent and primate Parkinson's disease (PD) models. We used computer simulation to predict how decreased PT5B neuron excitability, identified with current injection in vitro, would change activity patterns of the M1 network. Using NEURON/NetPyNE, we implemented computer simulations of PT5B neurons based on control and 6-OHDA-treated mouse slice data. Parkinsonian PT5B neurons, in an otherwise unmodified simulated M1 network, produced major changes in LFP oscillatory power: an order of magnitude inc
