Effects of Cross-Presentation, Antigen Processing, and Peptide Binding in HIV Evasion of T Cell Immunity.

Unlike cytosolic processing and presentation of viral Ags by virus-infected cells, Ags first expressed in infected nonprofessional APCs, such as CD4+ T cells in the case of HIV, are taken up by dendritic cells and cross-presented. This generally requires entry through the endocytic pathway, where endosomal proteases have first access for processing. Thus, understanding virus escape during cross-presentation requires an understanding of resistance to endosomal proteases, such as cathepsin S (CatS). We have modified HIV-1MN gp120 by mutating a key CatS cleavage site (Thr322Thr323) in the V3 loop

Estimating the In Vivo Killing Efficacy of Cytotoxic T Lymphocytes across Different Peptide-MHC Complex Densities.

Cytotoxic T lymphocytes (CTLs) are important agents in the control of intracellular pathogens, which specifically recognize and kill infected cells. Recently developed experimental methods allow the estimation of the CTL's efficacy in detecting and clearing infected host cells. One method, the in vivo killing assay, utilizes the adoptive transfer of antigen displaying target cells into the bloodstream of mice. Surprisingly, killing efficacies measured by this method are often much higher than estimates obtained by other methods based on, for instance, the dynamics of escape mutations. In this

High-sensitive troponin T and N-terminal-brain-natriuretic-peptide predict outcome in symptomatic aortic stenosis.

OBJECTIVES: Aortic stenosis (AS) and atherosclerosis share similarities when it comes to risk factors and disease progression. Like in other heart diseases, we hypothesized that biomarkers like high-sensitive troponin T (hsTnT), N-terminal-pro-brain-natriuretic-peptide (NT-proBNP) and high-sensitive C-reactive protein (hsCRP) could be useful in risk stratification. DESIGN: A total of 136 patients (57% men, mean age 74 years), referred for evaluation of AS (valve area 0.62 cm(2), left ventricular ejection fraction 64%) were consecutively enrolled in the study. The relationship between hsTnT, h

Vm24, a natural immunosuppressive peptide, potently and selectively blocks Kv1.3 potassium channels of human T cells.

Blockade of Kv1.3 K(+) channels in T cells is a promising therapeutic approach for the treatment of autoimmune diseases such as multiple sclerosis and type 1 diabetes mellitus. Vm24 (α-KTx 23.1) is a novel 36-residue Kv1.3-specific peptide isolated from the venom of the scorpion Vaejovis mexicanus smithi. Vm24 inhibits Kv1.3 channels of human lymphocytes with high affinity (K(d) = 2.9 pM) and exhibits >1500-fold selectivity over other ion channels assayed. It inhibits the proliferation and Ca(2+) signaling of human T cells in vitro and reduces delayed-type hypersensitivity reactions in rats in

Design of peptide-based inhibitors for human immunodeficiency virus type 1 strains resistant to T-20.

Enfuvirtide (T-20) is a fusion inhibitor that suppresses replication of human immunodeficiency virus (HIV) variants with multi-drug resistance to reverse transcriptase and protease inhibitors. It is a peptide derived from the C-terminal heptad repeat (C-HR) of HIV-1 gp41, and it prevents interactions between the C-HR and the N-terminal HR (N-HR) of gp41, thus interfering with conformational changes that are required for viral fusion. However, prolonged therapies with T-20 result in the emergence of T-20-resistant strains that contain primary mutations such as N43D in the N-HR of gp41 (where T-

Cytotoxic T cell-mediated diabetes in RIP-CD80 transgenic mice: autoantigen peptide sensitivity and fine specificity.

Rodent immune-mediated diabetes model studies have advanced understanding of beta cell-specific T cell responses, and the testing of therapeutic approaches. We have used an inducible diabetes model based on rat insulin promotor (RIP)-driven expression of CD80 (B7-1) on pancreatic beta cells. Using these mice, we have established that immunizing with a single autoantigen can promote progressive islet inflammation and eventually T cell-mediated diabetes. We now describe a potent immunization protocol using peptide-pulsed mature dendritic cells (DCs) to examine peptide epitopes derived from endog

Reversal of CD8+ T cell ignorance and induction of anti-tumor immunity by peptide-pulsed APC.

In the present report, we have studied the potential of naive and activated effector CD8(+) T cells to function as anti-tumor T cells to a solid tumor using OVA-specific T cells from TCR-transgenic OT-I mice. Adoptive transfer of naive OT-I T cells into tumor-bearing syngeneic mice did not inhibit tumor cell growth. The adoptively transferred OT-I T cells did not proliferate in lymphoid tissue of tumor-bearing mice and were not anergized by the tumor. In contrast, adoptive transfer of preactivated OT-I CTL inhibited tumor growth in a dose-dependent manner, indicating that E.G7 was susceptible

T lymphocyte responses to a synthetic peptide from the human intestinal adenovirus 12 in coeliac disease.

Peripheral blood mononuclear cells of treated coeliac patients are known to liberate leucocyte migration inhibition factor when challenged with a synthetic dodecapeptide of the E1b protein of adenovirus 12 sharing sequence homology with A gliadin. This study has compared the response of the unfractionated peripheral blood mononuclear cells with a highly purified T cell population. Coeliac mononuclear cells and T lymphocytes showed identical leucocyte migration inhibition activity. The migration indices for the coeliac group were significantly different than for the control group irrespective o

DC-CD8+ T cell-associated inflammation and sustained indel-derived candidate peptide exposure in radiation enteritis.

PURPOSE: Radiation enteritis (RE) is a frequent and challenging complication of radiotherapy for abdominal and pelvic malignancies, characterized by intestinal epithelial damage, immune dysregulation, and inflammation. Current treatments are limited by poor efficacy and significant side effects. Radiation-induced DNA damage can generate neoantigens, but their role in RE pathogenesis and therapeutic potential remains unclear. METHODS: An irradiation-induced RE mouse model was established. The dependency on dendritic cell (DCs) and CD8+ T cells was examined using depletion models. Indel-derived

Treatment of multiple sclerosis with peptide-coupled red blood cells induces antigen-specific T regulatory cells.

Inducing antigen-specific immune tolerance is a long-standing aim in organ-specific autoimmune diseases including multiple sclerosis (MS). We developed a therapy based on autologous red blood cells (RBCs) as tolerizing carriers of autoantigens and tested these in an open-label, dose-escalation, phase Ib clinical trial with clinical, imaging, and mechanistic readouts. RBCs coupled with seven immunodominant myelin peptides (pcRBCs) were safe and well tolerated. Several clinical and imaging readouts remained stable, which was supported by a reduction of neurofilament light chain indicating reduce

Development of a peptide-based vaccine using a T cell epitope derived from SARS-CoV-2.

Follicular helper T (Tfh) cells are a subset of CD4+ T cells that help B cells to produce high-affinity antibodies. Efficient Tfh cell induction by vaccines is critical for protective efficacy against diseases. A Tfh epitope, identified from the SARS-CoV-2 spike protein, has been shown to bind to the corresponding HLA and activates Tfh cells. Here, we assessed the efficacy of a peptide vaccine conjugated with SARS-CoV-2 Tfh epitope against experimental hypertension in a mouse model. The Tfh-angiotensin II (Tfh-Ang II) vaccine activated Tfh and germinal center B cells and induced antibodies aga

T cell proliferative response to a homocitrullinated peptide correlates with joint pathology in collagen induced arthritis.

Rheumatoid arthritis (RA) is characterized by autoimmune responses against citrullinated and homocitrullinated proteins/peptides (CitP and HomoCitP respectively), resulting in joint pain, inflammation, and damage. Although the collagen induced arthritis (CIA) model shares pathological features with RA, it remains unclear whether this model is suitable for studying RA-specific immune responses. Therefore, we investigated whether immune responses to CitP and HomoCitP influence arthritis severity in CIA. Male DBA/1J mice were immunized with bovine type II collagen (CII) (N = 58) or PBS (N = 14) i

Identification of multifunctional T-cell peptide epitopes for the development of DNA vaccines against dengue virus.

Dengue virus (DENV) is an important arthropod-borne virus that poses a global health threat, with half of the world's population at risk of infection. Currently, there is a lack of safe and effective vaccines for its prevention. Antibody-dependent enhancement (ADE) occurs when cross-reactive antibodies fail to neutralize heterologous DENV serotypes effectively, facilitating viral entry into Fc receptor-bearing cells and leading to more severe disease. It is reported that multifunctional T cells are closely related to the protective effects of the vaccine. We selected 25 peptide candidates base

Ectromelia-encoded virulence factor C15 specifically inhibits antigen presentation to CD4+ T cells post peptide loading.

Smallpox and monkeypox pose severe threats to human health. Other orthopoxviruses are comparably virulent in their natural hosts, including ectromelia, the cause of mousepox. Disease severity is linked to an array of immunomodulatory proteins including the B22 family, which has homologs in all pathogenic orthopoxviruses but not attenuated vaccine strains. We demonstrate that the ectromelia B22 member, C15, is necessary and sufficient for selective inhibition of CD4+ but not CD8+ T cell activation by immunogenic peptide and superantigen. Inhibition is achieved not by down-regulation of surface

SAG4 DNA and Peptide Vaccination Provides Partial Protection against T. gondii Infection in BALB/c Mice.

Toxoplasma gondii can lead to congenital infections in human. Surface antigen protein 4 (SAG4) of T. gondii is a potential stimulator for humoral and cellular immune responses. In the present study, a DNA vaccine encoding SAG4 from T. gondii was constructed and used to immunize BALB/c mice with peptide to evaluate the protective efficacy of the vaccine. The productions of IgG antibodies and cytokines (gamma interferon) from the vaccine (pSAG4/peptide) group were significantly higher than pSAG4 or peptide groups. After a lethal challenge by 1 × 104 tachyzoites from the I strain (RH), the surviv

B- and T-cell responses in group a streptococcus M-protein- or Peptide-induced experimental carditis.

The etiology of rheumatic fever and rheumatic heart disease (RF/RHD) is believed to be autoimmune, involving immune responses initiated between streptococcal and host tissue proteins through a molecular mimicry mechanism(s). We sought to investigate the humoral and cellular responses elicited in a Lewis rat model of group A streptococcus M-protein- or peptide-induced experimental valvulitis/carditis, a recently developed animal model which may, in part, represent human rheumatic carditis. Recombinant streptococcal M5 protein elicited opsonic antibodies in Lewis rats, and anti-M5 antisera recog

Elucidation and role of critical residues of immunodominant peptide associated with T cell-mediated parasitic disease.

Granulomatous inflammation in schistosomiasis is strictly dependent on CD4+ Th lymphocytes sensitized to egg Ags, but its intensity is genetically regulated. C3H and CBA (H-2k) are strains of mice that develop large granulomas; they also strongly respond to the major egg Ag Sm-p40. We now show that the immunodominant epitope recognized by CD4+ Th cells from infected H-2k mice is confined to 13-mer peptide 234-246 (PKSDNQIKAVPAS), which elicits an I-Ak-restricted Th1-type response. Using a panel of alanine-monosubstituted peptides, we identified Asp237 as the main contact residue with I-Ak. On

Peptide vaccination breaks tolerance to HER-2/neu by generating vaccine-specific FasL(+) CD4(+) T cells: first evidence for intratumor apoptotic regulatory T cells.

BALB/c mice transgenic (Tg) for the transforming rat neu oncogene (BALB-neuT) are genetically predestined to develop mammary carcinogenesis in a process similar to that in humans. We crossed HLA-A2.1/HLA-DR1 (A2.1/DR1) Tg mice with BALB-neuT mice to generate A2.1/DR1 x BALB-neuT triple Tg (A2.1/DR1 x neuT(+)) mice, which represent an improvement over BALB-neuT mice for evaluating vaccination regimens to overcome tolerance against HER-2/neu. Targeted T-cell therapy for human leukemia: cytotoxic T lymphocytes specific for a peptide derived from proteinase 3 preferentially lyse human myeloid leukemia cells.

Proteinase 3 is present in high concentration in the primary granules of acute and chronic myeloid leukemia blasts, and may represent a potential T-cell target antigen. We screened proteinase 3 against the binding motif of HLA-A2.1. Based on its high predicted binding, a 9-mer peptide, "PR-1," was synthesized and tested for binding to HLA-A2.1 using the T2 cell line. PR-1 at 100 micrograms/mL significantly increased expression of HLA-A2.1, with median channel of fluorescence increasing from 22 to 294. Binding half-life was determined to be 1,460 minutes by I125-labeled beta 2-microglobulin inc
