Stereochemical identification of glucans by oligothiophenes enables cellulose anatomical mapping in plant tissues.

Efficient use of plant-derived materials requires enabling technologies for non-disruptive composition analysis. The ability to identify and spatially locate polysaccharides in native plant tissues is difficult but essential. Here, we develop an optical method for cellulose identification using the structure-responsive, heptameric oligothiophene h-FTAA as molecular fluorophore. Spectrophotometric analysis of h-FTAA interacting with closely related glucans revealed an exceptional specificity for β-linked glucans. This optical, non-disruptive method for stereochemical differentiation of glycosid

Heterodera schachtii nematodes interfere with aphid-plant relations on Brassica oleracea.

Aboveground and belowground herbivore species modify plant defense responses differently. Simultaneous attack can lead to non-additive effects on primary and secondary metabolite composition in roots and shoots. We previously found that aphid (Brevicoryne brassicae) population growth on Brassica oleracea was reduced on plants that were infested with nematodes (Heterodera schachtii) prior (4 weeks) to aphid infestation. Here, we examined how infection with root-feeding nematodes affected primary and secondary metabolites in the host plant and whether this could explain the increase in aphid dou

Intra-specific variation in wild Brassica oleracea for aphid-induced plant responses and consequences for caterpillar-parasitoid interactions.

Herbivore-induced plant responses not only influence the initiating attackers, but also other herbivores feeding on the same host plant simultaneously or at a different time. Insects belonging to different feeding guilds are known to induce different responses in the host plant. Changes in a plant's phenotype not only affect its interactions with herbivores but also with organisms higher in the food chain. Previous work has shown that feeding by a phloem-feeding aphid on a cabbage cultivar facilitates the interaction with a chewing herbivore and its endoparasitoid. Here we study genetic variat

Anti-ulcer activity of Cyrtocarpa procera analogous to that of Amphipterygium adstringens, both assayed on the experimental gastric injury in rats.

ETHNOPHARMACOLOGICAL RELEVANCE: The bark of Amphipterygium adstringens (Aa) is commonly mixed or adulterated with the bark of Cyrtocarpa procera (Cp) and sold in Mexican markets. Aa is a well known species in Mexico used as decoction to relieve ulcers. Scientific reports reinforcing the anti-ulcer activity of Aa have been previously described, but those describing the anti-ulcer properties of Cp as a substitute for Aa in folk medicine are scarce. AIM OF THE STUDY: To investigate anatomical and phytochemical differences between these species, as well as to assess the anti-ulcer effect of Cp ex

Assessing the Likelihood of Transmission of Candidatus Liberibacter solanacearum to Carrot by Potato Psyllid, Bactericera cockerelli (Hemiptera: Triozidae).

'Candidatus Liberibacter solanacearum' (Lso) is a phloem-limited bacterium that severely affects important Solanaceae and Apiaceae crops, including potato, tomato, pepper, tobacco, carrot and celery. This bacterium is transmitted to solanaceous species by potato psyllid, Bactericera cockerelli, and to Apiaceae by carrot psyllids, including Trioza apicalis and Bactericera trigonica. Five haplotypes of Lso have so far been described, two are associated with solanaceous species and potato psyllids, whereas the other three are associated with carrot and celery crops and carrot psyllids. Little is

A genetic basis for the manipulation of sink-source relationships by the galling aphid Pemphigus batae.

We examined how the galling aphid Pemphigus batae manipulates resource translocation patterns of resistant and susceptible narrowleaf cottonwood Populus angustifolia. Using carbon-14 ((14)C)-labeling experiments in common garden trials, five patterns emerged. First, although aphid galls on resistant and susceptible genotypes did not differ in their capacity to intercept assimilates exported from the leaf they occupied, aphids sequestered 5.8-fold more assimilates from surrounding leaves on susceptible tree genotypes compared to resistant genotypes. Second, gall sinks on the same side of a shoo

The hexose transporters CsHT3 and CsHT16 regulate postphloem transport and fruit development in cucumber.

Hexoses are essential for plant growth and fruit development. However, the precise roles of hexose/H+ symporters in postphloem sugar transport and cellular sugar homeostasis in rapidly growing fruits remain elusive. To elucidate the functions of hexose/H+ symporters in cucumber (Cucumis sativus L.) fruits, we conducted comprehensive analyses of their tissue-specific expression, localization, transport characteristics, and physiological functions. Our results demonstrate that CsHT3 (C. sativus hexose transporter), CsHT12, and CsHT16 are the primary hexose/H+ symporters expressed in cucumber fru

Mechanisms of Fe biofortification and mitigation of Cd accumulation in rice (Oryza sativa L.) grown hydroponically with Fe chelate fertilization.

Cadmium contaminated rice from China has become a global food safety issue. Some research has suggested that chelate addition to substrates can affect metal speciation and plant metal content. We investigated the mitigation of Cd accumulation in hydroponically-grown rice supplied with EDTANa2Fe(II) or EDDHAFe(III). A japonica rice variety (Nipponbare) was grown in modified Kimura B solution containing three concentrations (0, 10, 100 μΜ) of the iron chelates EDTANa2Fe(II) or EDDHAFe(III) and 1 μΜ Cd. Metal speciation in solution was simulated by Geochem-EZ; growth and photosynthetic efficiency

Calcium in agriculture and crop production.

Calcium (Ca2+) is an essential mineral for plants, functioning both as a nano- to micromolar signaling ion in gene regulation and, at millimolar levels, in nutrition-supporting structural integrity and enzyme activation. These functions depend on adequate fertilization. However, Ca2+ problems stem mainly from hydraulic access and local availability within specific tissues and compartments rather than bulk supply; thus, deficiency can occur even in Ca2+-rich soils due to soil immobilization and physiological constraints. Because Ca2+ is poorly phloem-mobile and moves mainly via transpiration-dr

Plant defence against aphids: the PAD4 signalling nexus.

In Arabidopsis thaliana, PHYTOALEXIN DEFICIENT 4 (PAD4) functions as a key player in modulating defence against the phloem sap-feeding aphid Myzus persicae (Sülzer), more commonly known as the green peach aphid (GPA), an important pest of a wide variety of plants. PAD4 controls antibiosis and antixenosis against the GPA. In addition, PAD4 deters aphid feeding from sieve elements on Arabidopsis. In the past few years, substantial progress has been made in dissecting the role of PAD4 and its interaction with other signalling components in limiting aphid infestation. Several key genes/mechanisms

Drought and sugar transport: From turgor sensing to carbon allocation.

Drought triggers reprogramming of sugar transport, which acts as a central hub linking stress perception to carbon allocation. Under drought, FERONIA acts as a cell wall-plasma membrane turgor sensor that activates SnRK2s via outside-in signaling, and stress-activated kinases (SnRK2s, CPKs, and CIPKs) directly phosphorylate SWEET, SUT1, and TST transporters to enhance long-distance sucrose transport and vacuolar sugar accumulation. Recent cross-species single-cell atlases have identified phloem foundational genes as new targets for improving sugar transport. Meanwhile, deep mutational scanning

Panel statement on young pine plants used for afforestation or reforestation as pathway for pinewood nematode (PWN) Bursaphelenchus xylophilus.

Pinewood nematode (PWN) (Bursaphelenchus xylophilus) is a destructive pest of Pinus trees worldwide which depends for transmission on the cerambycid insects of the genus Monochamus. EFSA was asked to assess the potential role of young host plants for afforestation or reforestation planting as a pathway for PWN. According to legislation (EU COM/2023/415 final), these seedlings are not older than 2 years. Based on communication with SNPF (Syndicat National des Pépiniéristes Forestiers), such pine seedlings are assumed to have a collar diameter no more than 12 mm. It was evaluated that seedlings,

Formation of metal-nicotianamine complexes as affected by pH, ligand exchange with citrate and metal exchange. A study by electrospray ionization time-of-flight mass spectrometry.

Nicotianamine (NA) is considered as a key element in plant metal homeostasis. This non-proteinogenic amino acid has an optimal structure for chelation of metal ions, with six functional groups that allow octahedral coordination. The ability to chelate metals by NA is largely dependent on the pK of the resulting complex and the pH of the solution, with most metals being chelated at neutral or basic pH values. In silico calculations using pKa and pK values have predicted the occurrence of metal-NA complexes in plant fluids, but the use of soft ionization techniques (e.g. electrospray), together

A shoot derived long distance iron signal may act upstream of the IMA peptides in the regulation of Fe deficiency responses in Arabidopsis thaliana roots.

When plants suffer from Fe deficiency, they develop morphological and physiological responses, mainly in their roots, aimed to facilitate Fe mobilization and uptake. Once Fe has been acquired in sufficient quantity, the responses need to be switched off to avoid Fe toxicity and to conserve energy. Several hormones and signaling molecules, such as ethylene, auxin and nitric oxide, have been involved in the activation of Fe deficiency responses in Strategy I plants. These hormones and signaling molecules have almost no effect when applied to plants grown under Fe-sufficient conditions, which sug

An Indicating Role of Antioxidant System Enzymes at the Stage of Active Structural Anomalies Formation in Karelian Birch (Betula pendula Roth var. carelica (Mercl.) Hämet-Ahti).

INTRODUCTION: A complex study of the antioxidant system enzymes (AOS) is an important subject of biochemical research; changes in the activity of these enzymes can be used as a biochemical marker of various processes in plants. At the same time, practically little attention has been paid to describing the regularities of these enzymatic reactions in different wood formation processes, such as xylogenesis. This article discusses the outcomes of different behaviors of AOS enzymes, which are involved in both the redistribution of the ROS balance and phenolic compounds at the early stages of wood

Translocation of sugar and tritiated water in squash plants.

When (14)C-sugar and THO were simultaneously introduced through a cut side vein or flap of a squash leaf (Cucurbita melopepo, Bailey cv. torticollis) concurrent translocation of (14)C-sugars, T-photosynthates and THO with parallel, almost flat, gradients was observed in the petiole for periods of 1 to 3 hr. Parallel translocation gradients were not observed when (14)C was introduced as (14)CO(2) and T by painting a leaf with THO. Autoradiography of frozen sections to locate the tissues in which THO was moving was unsuccessful. Steam-girdling blocked the movement of (14)C and T when (14)C-gluco

Interactive regulation of immune-related resistance genes with salicylic acid and jasmonic acid signaling in systemic acquired resistance in the Xanthomonas-Brassica pathosystem.

Pathogen-responsive immune-related genes (resistance genes [R-genes]) and hormones are crucial mediators of systemic acquired resistance (SAR). However, their integrated functions in regulating SAR signaling components in local and distal leaves remain largely unknown. To characterize SAR in the Xanthomonas campestris pv. campestris (Xcc)-Brassica napus pathosystem, the responses of R-genes, (leaf and phloem) hormone levels, H2O2 levels, and Ca2+ signaling-related genes were assessed in local and distal leaves of plants exposed to four Xcc-treatments: Non-inoculation (control), only secondary

Non-coding RNAs: cross-kingdom regulators of plant-virus-vector interactions in complex agroecosystems.

Plant viruses depend on insect vectors for dissemination in complex agroecosystems and often exploit plant and insect physiology to enhance transmission. This review synthesizes recent advances in understanding cross-kingdom exchange of non-coding RNAs (ncRNAs) as a central mechanism shaping plant-virus-vector interactions and broader agroecosystem dynamics. ncRNAs are exchanged among plants, insect vectors, and viruses via the phloem, insect saliva, and external plant surfaces through exudates. These molecules can suppress plant antiviral and anti-insect plant responses, modulate vector biolo

Tissue-specific nuclear spin-spin relaxation anisotropy and water nano-confinement in celery stalks.

Recent magnetic resonance imaging (MRI) measurements have revealed proton spin-spin relaxation anisotropy in only one component of the celery stalk, even though water accounts for up to 95% of its weight and is present throughout all its tissues. Our analysis shows that this anisotropy arises from nanoconfined water within the collenchyma cell walls. In these regions, restricted molecular motion and hydrogen bonding between water molecules and cell-wall constituents give rise to residual anisotropic dipolar interactions. In contrast, water in the xylem, phloem, and parenchyma occupies larger c
