Comparison of the Properties of PARP1 and HPF1 in Naked Mole-Rat and Human.

DNA repair is the main mechanism for maintaining genome stability in living organisms and is considered one of the key factors influencing the maximum lifespan of mammals. Two proteins are of particular interest in the context of studying molecular basis of longevity: the key DNA repair regulator poly(ADP-ribose) polymerase 1 (PARP1) and the recently discovered modulator of its activity, histone poly(ADP-ribosyl)ation factor 1 (HPF1). This work analyzes the results of comparative analysis of the primary structures of PARP1 and HPF1, as well as examples of computer modeling of the orthologous p

Synergistic interplay and dual-target therapeutics: unraveling the coordinated network of SIK2 and PARP-1 in cancer development.

Salt-inducible kinase 2 (SIK2) and poly (ADP-ribose) polymerase 1 (PARP-1) form an unexpected, intricate synergistic network driving tumorigenesis, therapy resistance, and immune evasion, positioning their co-inhibition as a transformative strategy to overcome fundamental limitations in current cancer therapy. Mounting mechanistic evidence underscores their synergistic interplay in perpetuating genomic instability, enhancing tumor cell survival under replicative stress, and evading immune surveillance. Critically, emerging data suggest that SIK2 and PARP-1 engage in a functionally integrated r

Uro-protective role of chrysin against cyclophosphamide-induced hemorrhagic cystitis in rats involving the turning-off NF-κB/P38-MAPK, NO/PARP-1 and STAT-3 signaling cascades.

BACKGROUND: Chemotherapeutic agents are used to treat a wide range of cancer types, but they cause serious side effects which must be managed after treatment. Cyclophosphamide (CYP) is one of chemotherapeutic drugs that causes hemorrhagic cystitis (HC) induced by acrolein. OBJECTIVE: The current investigation intended to uncover the role of chrysin (CHR) in CYP-induced HC in rats and explore the signaling pathway beyond this effect. ANALYSIS: process: A single dose of CYP (200 mg/kg/IP) was injected, meanwhile CHR (25, 50 and 100 mg/kg, P.O) was administered respectively for 7 days prior to

PARP1 controls the transcription of CD24 by ADP-ribosylating the RNA helicase DDX5 in pancreatic cancer.

The PARP1 protein plays a key role in DNA damage repair and ADP-ribosylation to regulate gene expression. Strategies to target PARP1 have rapidly been developed for cancer treatment. However, the role of the innate immune response in targeted anti-PARP1 therapy remains poorly understood. In this work, we aimed to elucidate the regulatory mechanism underlying the immunogenicity of PARP1 and explore efficient therapeutic strategies to enhance the antitumor effect of PARP inhibitors. The relationships between PARP1 expression and immunosuppressive factors were examined by qRTPCR and immunoblot an

6,(5H)-phenanthridinone protects against carbon tetrachloride-induced cytotoxicity in human HepG2 cells.

Carbon tetrachloride (CCl4) is a compound associated with free radical mediated hepatotoxicity in humans and laboratory animals. Previous research indicates that the cytotoxicity caused by CCl4 may be mediated by the rapid induction of PARP-1, a nuclear repair enzyme, which results in celluar depletion of NAD+ and ATP. Animal models indicate that the inhibition of PARP-1 after CCl4 exposure will attenuate cytotoxicity in mouse hepatocytes. In this investigation, the potential hepatoprotective effects of the PARP-1 inhibitor 6,(5H)-phenanthridinone against CCl4-induced hepatotoxicity was tested

G-quadruplex homeostasis is a determinant of PARP inhibitor toxicity in BRCA2-deficient cells.

Inhibition of PARPs is a key strategy to treat tumours with defects in homologous recombination (HR), including those with mutations in the tumour suppressor gene BRCA2. PARP inhibitors generate replication stress, creating a dependence on HR to repair the resulting DNA damage. However, the DNA lesions generated upon PARP inhibition that impede replication fork progression and trigger a requirement for BRCA2 in cell survival are poorly defined. Here, we demonstrate that elevated levels of G-quadruplex (G4) DNA structures is a determinant of genome instability and PARP inhibitor toxicity, while

Role of PARP-1 in Human Cytomegalovirus Infection and Functional Partners Encoded by This Virus.

Human cytomegalovirus (HCMV) is a ubiquitous pathogen that threats the majority of the world's population. Poly (ADP-ribose) polymerase 1 (PARP-1) and protein poly (ADP-ribosyl)ation (PARylation) regulates manifold cellular functions. The role of PARP-1 and protein PARylation in HCMV infection is still unknown. In the present study, we found that the pharmacological and genetic inhibition of PARP-1 attenuated HCMV replication, and PARG inhibition favors HCMV replication. PARP-1 and its enzymatic activity were required for efficient HCMV replication. HCMV infection triggered the activation of P

Parthanatos participates in glutamate-mediated HT22 cell injury and hippocampal neuronal death in kainic acid-induced status epilepticus rats.

AIMS: Epileptic seizures or status epilepticus (SE) can cause hippocampal neuronal death, which has detrimental effects. Parthanatos, a new form of programmed cell death, is characterized by hyperactivation of poly (ADP-ribose) polymerase-1 (PARP-1), excessive synthesis of poly ADP-ribose polymer, mitochondrial depolarization, and nuclear translocation of apoptosis-inducing factor, observed in various neurodegenerative disorders but rarely reported in epilepsy. We aimed to investigate whether parthanatos participates in the mechanism of seizure-induced hippocampal neuronal death. METHODS: Glu

HDAC inhibitors suppress protein poly(ADP-ribosyl)ation and DNA repair protein levels and phosphorylation status in hematologic cancer cells: implications for their use in combination with PARP inhibitors and chemotherapeutic drugs.

The therapeutic efficacy of histone deacetylase inhibitors (HDACi) for hematologic malignancies and solid tumors is attributed to their ability to remodel chromatin, normalize dysregulated gene expression, and inhibit repair of damaged DNA. Studies on the interactions of HDACi with PARP inhibitors in hematologic cancers are limited, especially when combined with chemotherapeutic agents. Exposure of hematologic cancer cell lines and patient-derived cell samples to various HDACi resulted in a significant caspase-independent inhibition of protein PARylation, mainly catalyzed by PARP1. HDACi affec

Parthanatos-associated proteins are stimulated intraocularly during development of experimental murine cytomegalovirus retinitis in mice with retrovirus-induced immunosuppression.

The mechanisms that contribute to retinal tissue destruction during the onset and progression of AIDS-related human cytomegalovirus (HCMV) retinitis remain unclear. Evidence for the stimulation of multiple cell death pathways including apoptosis, necroptosis, and pyroptosis during the pathogenesis of experimental murine cytomegalovirus (MCMV) retinitis in mice with retrovirus-induced immunosuppression (MAIDS) has been reported. Parthanatos is a caspase-independent cell death pathway mediated by rapid overactivation of poly (ADP-ribose) polymerase-1 (PARP-1) and distinct from other cell death p

Aldehyde dehydrogenase 2 and PARP1 interaction modulates hepatic HDL biogenesis by LXRα-mediated ABCA1 expression.

HDL cholesterol (HDL-C) predicts risk of cardiovascular disease (CVD), but the factors regulating HDL are incompletely understood. Emerging data link CVD risk to decreased HDL-C in 8% of the world population and 40% of East Asians who carry an SNP of aldehyde dehydrogenase 2 (ALDH2) rs671, responsible for alcohol flushing syndrome; however, the underlying mechanisms remain unknown. We found significantly decreased HDL-C with increased hepatosteatosis in ALDH2-KO (AKO), ALDH2/LDLR-double KO (ALKO), and ALDH2 rs671-knock-in (KI) mice after consumption of a Western diet. Metabolomics identified A

ClickGen: Directed exploration of synthesizable chemical space via modular reactions and reinforcement learning.

Despite the significant potential of generative models, low synthesizability of many generated molecules limits their real-world applications. In response to this issue, we develop ClickGen, a deep learning model that utilizes modular reactions like click chemistry to assemble molecules and incorporates reinforcement learning along with inpainting technique to ensure that the proposed molecules display high diversity, novelty and strong binding tendency. ClickGen demonstrates superior performance over the other reaction-based generative models in terms of novelty, synthesizability, and docking

Targeting ZDHHC12-mediated PARP1 palmitoylation potentiates PARP inhibitor cytotoxicity.

Poly(ADP-ribose) polymerase inhibitors (PARPi) elicit cytotoxicity by trapping PARP1 at DNA lesions, but clinical resistance remains a major challenge. Here, we identify reversible S-palmitoylation as a negative regulator of PARP1 chromatin engagement. Mass spectrometry reveals PARP1 palmitoylation at conserved cysteines within its DNA-binding domains. DNA damage reduces PARP1 palmitoylation, enhancing DNA binding, whereas inhibition of depalmitoylases APT1/2 elevates palmitoylation and suppresses DNA binding. The palmitoyltransferase ZDHHC12 catalyzes PARP1 palmitoylation, and its inhibition,

Arsenite interacts selectively with zinc finger proteins containing C3H1 or C4 motifs.

Arsenic inhibits DNA repair and enhances the genotoxicity of DNA-damaging agents such as benzo[a]pyrene and ultraviolet radiation. Arsenic interaction with DNA repair proteins containing functional zinc finger motifs is one proposed mechanism to account for these observations. Here, we report that arsenite binds to both CCHC DNA-binding zinc fingers of the DNA repair protein PARP-1 (poly(ADP-ribose) polymerase-1). Furthermore, trivalent arsenite coordinated with all three cysteine residues as demonstrated by MS/MS. MALDI-TOF-MS analysis of peptides harboring site-directed substitutions of cyst

Proteomic Analysis of the Downstream Signaling Network of PARP1.

Poly-ADP-ribosylation (PARylation) is a protein posttranslational modification (PTM) that is critically involved in many biological processes that are linked to cell stress responses. It is catalyzed by a class of enzymes known as poly-ADP-ribose polymerases (PARPs). In particular, PARP1 is a nuclear protein that is activated upon sensing nicked DNA. Once activated, PARP1 is responsible for the synthesis of a large number of PARylated proteins and initiation of the DNA damage response mechanisms. This observation provided the rationale for developing PARP1 inhibitors for the treatment of human

In Vitro Evaluation of PARP1 Inhibitor Olaparib-Cyanine Dye Conjugate for the Treatment of Glioblastoma.

Inhibition of DNA damage repair is a common mechanism to induce cancer cell death and, therefore, to treat cancers such as breast, prostate and ovarian cancer. This approach, although very attractive, has not been feasible for treating brain tumours due to the stringent structural features necessary for pharmaceutical agents to cross the blood-brain barrier (BBB). This study presents the proof of concept in vitro cellular activity of PARP1 (Poly(ADP-ribose) polymerase 1) enzyme inhibitor olaparib conjugated with a known BBB-crossing heptamethine cyanine dye. We also present evidence for cellul

JPI-547, a novel dual inhibitor of PARP1/2 and tankyrase is more effective than first-generation PARP inhibitors in preclinical BRCA1/2-mutated cancer models.

BACKGROUND: Poly(ADP-ribose) polymerase (PARP) inhibitors are highly effective therapies for BRCA1/2-mutated tumors. However, most patients eventually develop acquired resistance. Here, we report that JPI-547, a second-generation PARP inhibitor targeting both PARP1/2 and tankyrase, demonstrates potent antitumor activity in olaparib-sensitive and resistant BRCA1/2 mutant models. METHODS: Olaparib-resistant (OR) models were generated using BRCA-mutated human ovarian and breast cancer cell lines and ovarian Patient-Derived Tumor Xenograft (PDTX) by exposing to olaparib.
