Subacute sclerosing panencephalitis (SSPE) is a lethal slow viral disease of the central nervous system caused by a defective measles virus. The onset is mostly in childhood, manifesting clinically as decline in academic performance, behavioural changes, motor dysfunction and myoclonus. Adult-onset SSPE is rare and can present as rapidly progressive dementia. We present a young man of Indian origin with adult-onset SSPE with rapidly progressive dementia but no localising neurological signs. The diagnostic clues were parieto-occipital white matter changes on MR brain scan and history of childho

Persistence of measles virus in the brains of patients with subacute sclerosing panencephalitis (SSPE) is accompanied by changes in the viral matrix (M) protein. To understand the significance of these changes, cell culture and cell-free assays were developed to compare the functions of the M proteins of an SSPE virus Biken strain and its acute measles virus progenitor Nagahata strain. The Nagahata viral M protein is associated with the intracellular viral nucleocapsids and the plasma membrane, whereas the Biken viral M protein is localized mainly in the cytosol. The lack of M protein in the B

PURPOSE/AIM OF THE STUDY: Subacute sclerosing panencephalitis (SSPE) is a degenerative disease of the brain caused by a persistent measles virus infection occurring mostly in childhood or early adolescence. The spectrum of epileptic phenomena associated with SSPE is wide, varying from partial or generalized tonic-clonic seizures and atypical absences to myoclonic-atonic attacks. Tonic seizures are very rare in SSPE.
MATERIALS AND METHODS: Herein, we discuss a case of 25 years old male that presented with adult-onset SSPE with tonic seizures accompanying myoclonic seizures.
RESULTS: Patient w

Long-term persistent measles virus (MeV) infection of the central nervous system (CNS) can result in subacute sclerosing panencephalitis (SSPE), an invariably fatal late neurological complication of measles. Analogous SSPE-like chronic diseases have also been reported in adult dogs, cetaceans, and more recently harbor seals following infection by canine distemper virus (CDV), dolphin morbillivirus (DMV), and phocine distemper virus (PDV), respectively. Here, we characterize different animal morbilliviruses (CDVlynx, PDV2001, PDV2014, and DMV232-18) that persisted in the CNS of their respective

The isolation of a measles-like virus in CV-1 cell cultures from a biopsy specimen obtained from a case of subacute sclerosing panencephalitis (SSPE) is reported. Evidence of the presence of cell-free virus was obtained from the fifth cell culture passage of the isolated virus. The virus was identified as measles-related by standard immune sera. IF study did not show relevant differences between the isolated virus and the Edmonston strain of measles virus, even though the proportion of cells showing nuclear antigen was higher following the infection with the isolated virus than with the Edmons

Rat glioma C6 cells persistently infected with measles subacute sclerosing panencephalitis (SSPE) virus (C6/SSPE) express the viral membrane proteins haemagglutinin (HA) and F on their cell surface as well as the intracellular proteins N, P and M. Previously we have shown that the addition of a polyclonal antibody against the HA antigen to the growth medium of C6/SSPE cells led to a gradual loss of all viral antigens. Here we show that the addition of a monoclonal antibody (MAb K83) leads only to a transient decrease in viral antigens during the first three passages. After the third passage vi

We determined the nucleotide sequence of the fusion (F) gene of three strains (Osaka-1, -2, and -3) of nonproductive variants of measles virus (MV). These viral strains were isolated in Osaka, Japan, from brain tissues of patients with subacute sclerosing panencephalitis (SSPE). Phylogenetic analysis revealed a close relationship among the three strains of SSPE virus. The cytoplasmic tail of the F protein, predicted from sequence analysis of the gene, is altered in all three SSPE strains when compared to the MV field strains. However, the extent and mode of alteration are different in each str

Matched serum and cerebrospinal fluid (CSF) samples from eight cases of subacute sclerosing panencephalitis (SSPE) and 15 cases of multiple sclerosis (MS) were characterized in neutralization, hemolysis-inhibition (HLI), hemagglutination-inhibition (HI) with Tween 80-ether-treated antigen, complement-fixation (CF), and immunodiffusion tests. CF tests were carried out with crude virus material, purified nucleocapsids, and small particle hemagglutinin as antigens. A certain diversity in the relative content of antibodies against different virus products in various sera was found. There was a hig

Many animal models using experimental small animals for subacute sclerosing panencephalitis (SSPE) had been reported. But these models were not enough for understanding of pathogenesis of SSPE. After pathogenic measles virus was isolated with highly susceptible B95a cells, mimic infection of measles in human beings became easily produced in non-human primates. In this article, our attempt to develop SSPE model using cynomolgus monkeys will be introduced.

Serum and cerebrospinal fluid (CSF) measles and distemper antibody levels were quantified in a series of twenty patients (four subacute sclerosing panencephalitis (SSPE); ten multiple sclerosis (MS); six non-MS neurological cases) using independent competitive inhibition radioimmunoassays. These results were used in a Tourtellotte calculation to measure the intracerebral IgG synthesis to each virus. The results confirmed that in SSPE there is a greatly enhanced intracerebral measles antibody synthesis (6.0 mg/day). However, it was found that in SSPE this represents only part of a general syste

Measles virus generally produces acute illness. Rarely, however, persistent infection of brain cells occurs, resulting in a chronic and fatal neurological disease, subacute sclerosing panencephalitis (SSPE). Evidence indicates that expression of the measles virus matrix protein is selectively restricted in this persistent infection, but the mechanism underlying this restriction has not been identified. Defective translation of matrix messenger RNA has been described in one SSPE cell line. This report presents evidence that in a different SSPE tissue culture cell line IP-3-Ca, the matrix protei

Subacute sclerosing panencephalitis (SSPE) is a rare but fatal late neurological complication of measles, caused by persistent measles virus (MeV) infection of the central nervous system. There are no drugs approved for the treatment of SSPE. Here, we followed the clinical progression of a 5-year-old SSPE patient after treatment with the nucleoside analog remdesivir, conducted a post-mortem evaluation of the patient's brain, and characterized the MeV detected in the brain. The quality of life of the patient transiently improved after the first two courses of remdesivir, but a third course had

The complete nucleotide sequence of cloned cDNAs corresponding to the full-length mRNA encoding the NP protein of the Yamagata-1 strain of subacute sclerosing panencephalitis (SSPE) virus was determined. The gene is composed of 1683 nucleotides and contains a single large open reading frame, which is capable of encoding 525 amino acids with a molecular weight of 58,399. Comparison of the nucleotide and predicted amino acid sequences with those of the Edmonston strain of measles virus (MV) showed that the gene and the protein were highly conserved. However, the antigenic sites on the NP protein

Studies on the immunoprecipitation of the proteins of several different measles and subacute sclerosing panencephalitis (SSPE) virus strains from lysates of infected cells have revealed a relative lack of antibodies to the nonglycosylated viral membrane protein (M) in the sera of patients with SSPE, in contrast to a high level of antibodies to other viral proteins. No evidence was found for the existence of a large number of antigenically unique M proteins. In contrast to SSPE sera, other hyperimmune human or rabbit sera precipitated M protein as well as the other viral proteins. The results s

Subacute sclerosing panencephalitis (SSPE) is a chronic encephalitis of childhood and young adolescence due to persistent measles virus infection of the central nervous system. In majority of cases onset occurs from 5-15 years of age. In a nonimmunized population the average onset is 8 years. Children with SSPE had experienced natural infection with the rubeola virus at an early age, half before age 2 years. SSPE generally occurs 5-10 years after measles infection. In the early stages of the disease behavioral and personality changes is followed by myoclonic jerks and convulsions. In late stag

Subacute sclerosing panencephalitis (SSPE) is a lethal disease induced by the persistence of measles virus in the human brain. In many SSPE cases, the viral matrix (M) protein cannot be detected; in others, M proteins of the expected size are found and sequence analysis of M cDNAs has confirmed that the reading frames are intact, showing only several missense mutations. To determine whether these alterations result in nonfunctional proteins, we have replaced the M gene of an infectious full-length genomic cDNA (from vaccine strain Edmonston) with different M genes derived from four patients wi

Dravet syndrome, characterized predominantly by myoclonus, has a striking clinical resemblance to subacute sclerosing panencephalitis (SSPE). Patients with Dravet syndrome develop significant mental decline with advancing age of affected child like in SSPE. It is well established that SCN1A gene mutations are associated with Dravet syndrome. Even periodic EEG complexes have been described in Dravet syndrome. In addition to Dravet syndrome, several other types of acute and subacute encephalopathic syndromes having clinical and electroencephalographic resemblance to SSPE are associated with SCN1

Subacute sclerosing panencephalitis(SSPE) is a progressive and fatal neurological illness without established treatment. As the precise mechanism how measles virus (MV) causes SSPE is still unknown, full clarification of pathogenesis and pathophysiology is essential to establish effective strategy to treat the illness. Viral, host and environmental factors are involved in the development of SSPE. As host factors, immaturity of immune system and central nervous system are well recognized. Recently, we demonstrated that functional polymorphisms of MxA, interleukin(IL)-4 and interferon regulatory

Subacute sclerosing panencephalitis (SSPE) patients carry persistent measles virus infection in the brain. Furthermore, the blood lymphocytes contain viral RNA. Lymphocytes derived from 6 SSPE patients were stimulated with Epstein-Barr virus (EBV). Production of antibodies against measles virus of the IgG isotype was detected in the supernatants of cell cultures of all patients, regardless of the disease's activity, duration or interferon therapy. In contrast, only some of these cell cultures also produced antibodies against myelin.

Two to five separate gel-precipitating (GP) antibody activities to sonicated measles virus cell pack antigen were detected by double immunodiffusion and immunoelectrophoresis of sera and cerebrospinal fluids (CSF) from patients with subacute sclerosing panencephalitis (SSPE). The major GP activity was identified as antibody to measles virus nucleocapsids, and a weaker activity as antibody to the virus haemagglutinin. The specificities of the other activities were not determined. Evidence is presented that measles GP antibody activities of both serum and CSF from patients with SSPE are carried
