Trypanosoma cruzi strain-specific monoclonal antibodies: identification of Colombian strain flagellates in the insect vector.

Spleen cells from mice immunized with insect-derived Trypanosoma cruzi metacyclic trypomastigotes were used to obtain Colombian strain-specific monoclonal antibodies. At least 4 different strain-specific antigens were recognized by the monoclonal antibodies on epimastigotes or metacyclic trypomastigotes. There was no reactivity with other stages of Colombian strain T. cruzi, nor with any stage of 15 other T. cruzi strains or isolates, nor with 22 other Trypanosomatidae. One of the monoclonal antibodies was used to identify, by indirect immunofluorescence, Colombian strain flagellates in cryost

Colonization of the rectum of Triatoma infestans by Trypanosoma cruzi studied by scanning electron microscopy: influence of blood uptake by the bug.

The colonization of the different regions of the rectum of Triatoma infestans by Trypanosoma cruzi was studied in unfed larvae, during and after feeding of the bug by scanning electron microscopy. The rectal pads always possessed the highest population densities, but in some bugs the main rectal sac and the region around the anus were also covered by a "carpet" of flagellates. Such high densities were never observed at the midgut/rectal junction. A slight decrease in this region might be caused by blood ingestion and the resulting excretion of urine. However, the flagellates in this region can

Overview of Chagas disease in Amazonas: an integrated approach.

In the Amazon, Chagas disease (CD) transmission is associated with the consumption of contaminated foods -most notably açaí-and constitutes an important public health concern. In Brazil, the state of Amazonas ranks third in acute-phase notifications among states in the region, with 25% of municipalities having already reported acute CD cases, predominantly in outbreaks in rural areas. This study describes the current overview of knowledge on CD in Amazonas, bringing together research findings from the Fundação de Medicina Tropical Doutor Heitor Vieira Dourado (FMT-HVD), developed in partnershi

New findings on the functional morphology of the male genitalia of the blood-feeding bug Rhodnius prolixus in the light of a different observation on living tissue.

Among insect species, the male genitalia evolved many different anatomical features to ensure reproductive success. Males of primitive insects do not copulate but transfer sperm packaged in a spermatophore to the female, whereas males of the most advanced insects copulate and deliver free sperm to the storage organ, the spermatheca, in the female reproductive tract. Based on previous studies on several species of triatomine bugs (Rhodnius prolixus included), Triatominae are thought to use an intermediate form of sperm transfer in which the male genitalia have evolved to carry out both copulati

First report of colonies of sylvatic Triatoma infestans (Hemiptera: Reduviidae) in the Paraguayan Chaco, using a trained dog.

In the Gran Chaco region, control of Triatoma infestans has been limited by persistent domestic infestations despite the efforts of the Vector Control Services. In Paraguay, this region is the highest endemic area in the country, showing high levels of indoor and outdoor infestation. Although sylvatic T. infestans have been found in the Bolivian and Argentine Chaco, similar searches for sylvatic populations of this species in Paraguay had been unsuccessful over the last 20 years. Here we present a new approach to detecting sylvatic Triatominae, using a trained dog, which has successfully confi

Surgery of chagasic megacolon.

Chagas' disease is an endemic clinical entity caused by Trypanosoma cruzi, a parasite that is transmitted to humans by the hematophagic Triatominae insects. It affects several million persons in Latin America, mostly in Brazil, Argentina, Chile, Paraguay, and Bolivia. Megacolon, the most common complication of intestinal trypanosomiasis, results in severe constipation, for which surgery is indicated. A variety of procedures have been proposed for the correction of this disabling condition including sigmoidectomy, abdominal rectosigmoidectomy, left colectomy, and subtotal colectomy. On long-ter

Molecular epidemiology of Chagas disease in the wild transmission cycle: the evaluation in the sylvatic vector Mepraia spinolai from an endemic area of Chile.

The sylvatic transmission cycle of Chagas disease in Chile is composed of wild mammals and insects of the genus Mepraia. We determined infection rates and Trypanosoma cruzi genotypes in Mepraia spinolai. We collected 227 insects from two ecologically contrasting areas to assess T. cruzi infection. Polymerase chain reaction (PCR)-amplified minicircle DNAs were characterized by Southern blot and hybridization tests with genotype-specific probes. Infection in insects from the more fertile area was almost 2-fold higher than in the poorer area. The genotype TCI was the most prevalent and other geno

Triatomines as experimental vectors of trypanosomes of Asian monkeys.

Systematic surveys of the wild macaques of South Asia by blood culture resulted in the discovery that trypanosomiasis is enzootic in the simians of Indonesia, Malaysia, India, and Thailand. The isolates obtained differ in morphology, metabolism, and ability to multiply in arthropods. Following this discovery, interest focused on possible transmissions of these trypanosomiases. Laboratory-reared and wild-caught insects were studied to determine which are satisfactory intermediate hosts and potential natural vectors. Successful results were obtained with insectary-reared reduviids and Indonesian

Potential geographic displacement of Chagas disease vectors under climate change.

Climate change is projected to profoundly alter global biodiversity with significant implications for vector-borne disease dynamics. In tropical regions, rising temperatures and shifting precipitation patterns influence the distribution and behaviour of insect disease vectors, thereby affecting disease transmission cycles. Chagas disease, caused by the Trypanosoma cruzi and transmitted by triatomine bugs, is a major public health concern in Latin America. Brazil is particularly vulnerable to climate-driven vector redistribution due to its vast land area, diverse ecosystems and rapid land-use c

Trypanosoma cruzi/Triatomine Interactions-A Review.

This review summarizes the interactions between Trypanosoma cruzi, the etiologic agent of Chagas disease, and its vectors, the triatomines, and highlights open questions. Four important facts should be emphasized at the outset: (1) The development of T. cruzi strains and their interactions with the mammalian host and the insect vector vary greatly. (2) Only about 10 of over 150 triatomine species have been studied for their interactions with the protozoan parasite. (3) The use of laboratory strains of triatomines makes generalizations difficult, as maintenance conditions influence the interact

Spatial modeling and risk assessment of chagas disease vector distribution in Espírito Santo, Brazil: A comprehensive approach for targeted control.

Chagas disease, a persistent and life-threatening infection caused by the protozoan Trypanosoma cruzi, remains a significant public health concern in Latin America. Despite the Brazilian State of Espírito Santo (ES) not being classified as a high-risk area, the presence of epidemiologically significant triatomines like Panstrongylus megistus suggests a latent risk of T. cruzi transmission. This study, employing spatial modeling, assesses the distribution of key triatomine species in ES and predicts areas at risk for Chagas disease transmission. Our models, constructed with Maxent, KUENM, and Q

A cost-benefit analysis of Chagas disease control.

Chagas disease transmission can be effectively interrupted by insecticidal control of its triatomine bug vectors. We present here a simple model comparing the costs and benefits of such a programme, designed to eliminate domestic populations of Triatoma infestans throughout its known area of distribution over the seven southernmost countries of Latin America. The model has been simplified to require only four financial estimates relating to the unit cost of housing spraying and benefits due to avoidance of premature death in the acute phase of the disease, avoidance of supportive treatment and

Toxicity of methyl bromide and other gaseous insecticides to Triatoma infestans.

Cuticular penetration of lipid soluble substances in Triatoma infestans is very poor (less than 1% in 24 hr), while the respiratory route is particularly efficient (28% in 5 min). Accordingly, several noxious gases killed the insect at short periods of time. CH3Br and SO2 were particularly effective and they also evidenced ovicide action. With the former compound this effect is also observed at low concentrations. The CH3Br hydrolysis products or its metabolites were not similarly effective, while other methylating agent, diazomethane, was very active. The irreversible binding of 14CH3Br to pr

The role of CYP4Gs involved in hydrocarbon formation in the survival, reproduction, and pyrethroid resistance of Triatoma infestans, a major Chagas disease vector.

Very long-chain normal and branched hydrocarbons are essential for insect physiology and ecology. The final metabolic step of hydrocarbon formation involves an oxidative decarbonylation carried out by cytochrome P450 from family 4, subfamily G (CYP4G). Triatoma infestans Klug (Hemiptera: Reduviidae: Triatominae) has two CYP4G genes, named CYP4G106 and CYP4G107. In this study, we have silenced in T. infestans the expression of both genes by RNA interference in order to assess the relevance of hydrocarbons to survival, reproductive physiology, and insecticide susceptibility. Gene knockdown led t

Morphometric variability in the Chagas' disease vector Rhodnius prolixus.

Morphometric variability was studied in six domestic Venezuelan populations of the blood-sucking bug Rhodnius prolixus Stal 1897 (Reduviidae, Triatominae) and in a sylvatic population identified as R. robustus Larrousse 1927. Evidence is here provided by both uni- and multifactorial analyses of extensive variation of morphological traits between the R. prolixus populations studied. Regardless the geographic or climatic environmental factor tested, none can be retained in a selective model accounting for the morphological variability observed. Moreover, the results failed to support any correla

Challenges and opportunities for primary, secondary, and tertiary prevention of Chagas' disease.

A century after its discovery, Chagas' disease still represents a major public health challenge in Latin America. Moreover, because of growing population movements, an increasing number of cases of imported Chagas' disease have now been detected in non-endemic areas, such as North America and some European countries. This parasitic zoonosis, caused by Trypanosoma cruzi, is transmitted to humans by infected Triatominae insects, or occasionally by non-vectorial mechanisms, such as blood transfusion, mother to fetus, or oral ingestion of materials contaminated with parasites. Following the acute

Trypanosoma cruzi (Chagas, 1909) transmission among captive wild mammals, triatomines and free-living opossums from surrounding areas in the São Paulo Zoological Park, Brazil.

BACKGROUND: Trypanosoma cruzi is a multi-host parasite that infects dozens of mammalian species in the most variable wild environments in Americas. Between 2013 and 2018, autochthonous infections by T. cruzi were suspected in three European wolves (Canis lupus) and an orange dwarf porcupine (Coendou spinosus) from Fundação Parque Zoológico de São Paulo (FPZSP), Brazil, current Coordenadoria de Fauna Silvestre (CFS), which is inserted in a remnant of the Atlantic rainforest inside one of the biggest and most populous municipality of the world. This study aims to detect T. cruzi infections in ca

Modelling geospatial distributions of the triatomine vectors of Trypanosoma cruzi in Latin America.

Approximately 150 triatomine species are suspected to be infected with the Chagas parasite, Trypanosoma cruzi, but they differ in the risk they pose to human populations. The largest risk comes from species that have a domestic life cycle and these species have been targeted by indoor residual spraying campaigns, which have been successful in many locations. It is now important to consider residual transmission that may be linked to persistent populations of dominant vectors, or to secondary or minor vectors. The aim of this project was to define the geographical distributions of the community

Chagas disease vector blood meal sources identified by protein mass spectrometry.

Chagas disease is a complex vector borne parasitic disease involving blood feeding Triatominae (Hemiptera: Reduviidae) insects, also known as kissing bugs, and the vertebrates they feed on. This disease has tremendous impacts on millions of people and is a global health problem. The etiological agent of Chagas disease, Trypanosoma cruzi (Kinetoplastea: Trypanosomatida: Trypanosomatidae), is deposited on the mammalian host in the insect's feces during a blood meal, and enters the host's blood stream through mucous membranes or a break in the skin. Identifying the blood meal sources of triatomin

[Transmission of chagasic infection by oral route in the natural history of Chagas disease].

The enzootic cycle of Trypanosoma cruzi in the nature is maintained for millions of years, in great part by oral route, through the ingestion of infected triatomines by edentates(Xenarthra), marsupials and primates and by carnivores eating other infected mammals. The other phases of the natural history of Chagas disease--Anthropozoonosis, Zoonosis or Anphixenosis and Zooanthroponosis are more recent. The most ancient human mummies found with DNA of T. cruzi by PCR, from northern Chile and southern Peru, dated of 9.000 years and the infection must be acquired from the forest or in the caves as
