Studies on the status of lysine residues in phospholipase A2 from Naja naja atra (Taiwan cobra) snake venom.

Phospholipase A2 (PLA2) from Naja naja atra (Taiwan cobra) snake venom was subjected to lysine modification with trinitrobenzene sulphonic acid (TNBS), and two major trinitrophenylated (TNP) derivatives, TNP-1 and TNP-2, were separated by h.p.l.c. TNP-1 contained only one TNP group on Lys-6 and showed a marked decrease in enzymic activity, but still retained 45% of the lethal toxicity. Both Lys-6 and Lys-65 were modified in TNP-2, and modification of Lys-65 caused a further reduction of the lethal toxicity to 12.6%. However, the antigenicity of both TNP-1 and TNP-2 remained unchanged. The reac

EB1010, a putative Christensenellaceae-derived microbial peptide, exhibits anti-inflammatory activity and therapeutic potential in ulcerative colitis.

The gut microbiota plays a pivotal role in modulating the immune system, fostering immune tolerance, and defending against pathogens. Among its bioactive products, microbiota-derived peptides hold therapeutic promise for inflammatory diseases. In this study, we present EB1010, a 28-residue peptide derived from an unknown protein of a Christensenellaceae bacterium, as potential treatment of ulcerative colitis (UC). EB1010 exhibit strong anti-inflammatory effects in vitro by reducing proinflammatory mediator secretion and inhibiting NF-κB signaling in immune cells. Ex vivo human intestinal model

Investigation of ameliorative effects of indole-3 carbinol on TNBS-induced ulcerative colitis in rats.

Ulcerative colitis is a chronic inflammatory condition of the gastrointestinal tract that can predispose patients to colonic neoplasms. Various natural compounds have been explored for their therapeutic potential. Indole-3-carbinol (I3C), a natural compound derived from cruciferous vegetables, is recognized for its tissue-protective and regenerative properties. This study aimed to investigate the effects of I3C on experimental ulcerative colitis in rats. Thirty-two Wistar rats were randomly assigned to four groups: a control group receiving isotonic saline, a TNBS group administered trinitrobe

Mesenchymal Stem Cells Alleviate Inflammatory Bowel Disease Via Tr1 Cells.

Mesenchymal stem cells (MSCs) have been used to achieve exciting therapeutic outcomes in many animal studies and clinical trials for various autoimmune diseases, including inflammatory bowel disease (IBD). Type 1 regulatory T (Tr1) cells are the main source of interleukin (IL) 10 in the intestine. Whether Tr1 cells are involved during MSC-mediated IBD treatment is unclear. We treated a murine model of 2,4,6-trinitrobenzene sulfonic acid (TNBS)-induced colitis with human umbilical cord-derived MSCs (hUCMSCs) and found that the disease severity was alleviated significantly in a dose-dependent ma

A fumigaclavine C isostere alleviates Th1-mediated experimental colitis via competing with IFN-γ for binding to IFN-γ receptor 1.

Interferon gamma (IFN-γ) signaling in T cells plays an important role in developing T helper 1 (Th1)-mediated inflammation. Selective regulation of IFN-γ signaling is an attractive strategy for treating Th1-mediated immune diseases. In this study, we aimed to explore possible means of targeting IFN-γ signaling by using small molecule compound. A synthetic small molecule FC9 was identified as it selectively inhibited IFN-γ signaling in T cells without suppressing interleukin 4 (IL-4) signaling. Furthermore, FC9 inhibited IFN-γ-induced Janus kinase 2 (JAK2) activation via competing with IFN-γ fo

The correlation between NF-κB inhibition and disease activity by coadministration of silibinin and ursodeoxycholic acid in experimental colitis.

NF-κB is one of the most important nuclear factors responsible for overexpression of proinflammatory cytokines. This is demonstrated by increased NF-κB activity and other dependent immune factors in inflammatory bowel disease (IBD). Anti-inflammatory effects of silibinin and ursodeoxycholic acid (UDCA) along with their NF-κB inhibitory property are thought to be beneficial in colitis. Trinitrobenzene sulfonic acid was used to induce colitis rat models. After instillation, 48 rats were treated with oral silibinin, UDCA alone or a combination of both. Intraperitoneal dexamethasone was used in th

Extrinsic surgical denervation ameliorates TNBS-induced colitis in rats.

BACKGROUND/AIMS: Neurogenic inflammation refers to an inflammatory reflex arc by sensory neurons which transmit nocious stimulus centrally and results in both pain perception and intense local inflammatory reaction. Specific neurons, receptors, and their respective neurotransmitters have been studied in numerous organ systems including the gastrointestinal tract. Neurogenic inflammation has been suggested to play a key role in the pathogenesis of inflammatory bowel disease. In this study, we studied the effect of surgical denervation of specific somatosensory neurons in a well-established anim

A traditional Japanese medicine mitigates TNBS-induced colitis in rats.

OBJECTIVE: Differences in dietary habits may be one of the reasons that the incidence of inflammatory bowel disease has remained lower in Japan than in Western countries. We investigated whether a traditional Japanese medicine (Strong Wakamto), based on Aspergillus oryzae koji, would exert any effect on experimental colitis in rats. MATERIAL AND METHODS: Colitis was induced using an enema of trinitrobenzene sulfonic acid (TNBS) and ethanol. Strong Wakamto was administered for 28 days before induction of colitis and for 7 days thereafter, and the effect of this medicine was evaluated. RESULTS

Effects of plain and controlled-ileal-release budesonide formulations in experimental ileitis.

BACKGROUND: Budesonide combines a topical anti-inflammatory activity with high first-pass hepatic extraction. This study compared the effects of plain and controlled-ileal-release (CIR) formulations of budesonide on intestinal inflammation. METHODS: Ileitis was induced in hamsters by an intraluminal injection of trinitrobenzene sulphonic acid. Inflammation was assessed histologically and by measuring mastocytosis and myloperoxidase activity. Adrenal-pituitary axis suppression was assessed by radio-immunoassay of plasma cortisol. Animals received budesonide (200 or 800 micrograms/kg/day), CIR

Loss of free amino groups in the water-insoluble fraction of nuclear senile cataracts.

Free amino groups were analyzed in the water-insoluble fraction (WIF) of cataractous human lenses of the nuclear sclerosis, pigmented type. Two modified versions of the trinitrobenzene sulfonic acid (TNBS) procedure for the determination of free amino groups in proteins (AFSA Habeeb, Anal Biochem 14:328-336, 1966) were used for this purpose. The concentration of free amino groups in WIF was found to be inversely related to the dry weight of this fraction. Taking WIF weight as a measure of the severity of nuclear cataract, it can be thus said that a relative loss of free amino groups apparently

Proliferation of anti-DNA-producing NZB B cells in a non-autoimmune environment.

This study demonstrates that purified NZB B cells, but not other NZB spleen cell populations, are capable of transferring anti-DNA antibody production into unirradiated H-2-compatible xid recipients. The number of autoantibody-producing B cells and the concentration of anti-DNA antibody found in the recipients correlated directly with the number of NZB B cells transferred. In addition, the number of anti-DNA-secreting lymphocytes found in the xid hosts increased exponentially with time post cell transfer. Several lines of evidence suggest that this phenomenon reflected the rapid proliferation

Cellular and genetic control of antibody responses. VIII. MHC restricted recognition of accessory cells, not B cells, by parent-specific subpopulations of normal F1 T helper cells.

Normal F1 helper T cell populations were analyzed under conditions in which the number of T cells was strictly limiting so that anti-hapten PFC responses to TNP-KLH increased linearly with increasing numbers of T cells. The slope of the response line is a measure of the helper activity of the added T cell population and reflects the number of helper T cells activated. In this way, the recognition and activation requirements of normal F1 helper T cell subpopulations were analyzed. It was determined that the F1 helper T cells specific for recognizing antigen in the context of the MHC determinant

Split unresponsiveness to trinitrophenyl (TNP) determinant. Suppression of anti-TNP antibody responses by sensitization with picryl chloride.

Contact sensitization by epicutaneous application of picryl chloride causes in mice a significant reduction of the antibody responses to immunogenic TNP-conjugates. This split unresponsiveness is along-lasting. It was found that hapten applied on the skin became attached to the serum proteins and the transfer of such a serum into normal recipients, while not influencing the ability of these animals to become contact-sensitized to PCl, rendered them unable to mount the anti-TNP antibody response. Possible mechanisms of split unresponsiveness to the TNP determinant induced by PCl treatment are s

Engineering Cooperative Microenvironments in a Nitro-Functionalized Trinuclear Copper Metal-Organic Framework for High-Efficiency Heterogeneous Catalysis.

The deliberate engineering of cooperative catalytic microenvironments within metal-organic frameworks (MOFs) offers a powerful strategy for promoting sustainable catalysis. Herein, we report a robust nitro-functionalized trinuclear metal-organic framework, formulated as {[Cu3(NTCB)2(4,4'-bip)(DMF)2]·4DMF·3H2O}n (NUC-180; H3NTCB = 1,3,5-tri(4-carboxy-2-nitrophenyl)-2,4,6-trinitrobenzene, 4,4'-bip = 4,4'-bipyridine), featuring a high void fraction and dual nanoscale channel systems. Upon activation, NUC-180a exposes coordinatively unsaturated metal centers and strongly electron-withdrawing nitro

Involvement of Periostin in Intestinal Fibrosis Associated With Crohn's Disease.

BACKGROUND AND AIM: Crohn's disease (CD) is a chronic inflammatory bowel disease characterized by excessive intestinal fibrosis. We previously demonstrated that periostin is highly expressed in various fibrosis-related diseases and is important in the development of fibrosis. In this study, we aimed to investigate the role of periostin in the pathogenesis of CD. METHODS: We evaluated periostin expression in the terminal ileum of CD patients. We examined serum periostin levels in 75 CD patients and 150 healthy controls. We applied periostin-deficient (PNKO) and wild-type (WT) mice to a mouse m

Eosinophil-derived COX-2 protects against experimental colitis through the PGE2-IL-22 axis.

Inflammatory bowel disease (IBD) is driven by a breakdown in immune regulation and epithelial barrier function, yet the contribution of eosinophils to this process has remained poorly defined and controversial. While eosinophils infiltrate the intestinal mucosa during both flares and remission, their role in shaping disease outcomes is unclear. Our RNA-seq analyses of colonic eosinophils isolated from dextran sulfate sodium (DSS)-treated mice revealed a significant upregulation of cyclooxygenase (COX)-2 (gene name, Ptgs2). Eosinophil-specific deletion of COX-2 (Ptgs2fl/fleoCre+/-) reduced IL-2

Peptidylarginine deiminase 2 contributes to pathogenesis in trinitrobenzenesulfonic acid-induced colitis through macrophage extracellular trap-independent pathways.

Peptidylarginine deiminase 2 (PAD2) is an enzyme that converts arginine to citrulline and is involved in diseases, such as Alzheimer's diseases, fibrosis and cancer. However, its role in inflammatory bowel disease remains unclear. In this study, we investigated the pathogenic effects of PAD2 on inflammatory bowel disease using a trinitrobenzene sulfonic acid (TNBS)-induced murine colitis model. PAD2-deficient (PAD2KO) mice were generated using CRISPR/Cas9-mediated genomic editing. TNBS injection resulted in body weight loss, extensive colonic erosion, and ulceration in wild-type (WT) mice. D-mannose reduces oxidative stress, inhibits inflammation, and increases treg cell proportions in mice with ulcerative colitis.

BACKGROUND: Regulatory T (Treg) cells is required to dampen immune responses against intestinal microbiota, which aid in a healthy body to promise that the resident gut microbiota should not attract the attention of the immune system. Inflammation and inflammatory bowel disease (IBD) can be induced if the immune system fails to ignore the resident gut microbiota and targets them instead. D-mannose, a common monosaccharide in nature, has been shown to ameliorate multiple autoimmune diseases. This study aimed to investigate the therapeutic effect of D-mannose on mice ulcerative colitis (UC) indu
