Thyroid hormone controls the onset of androgen sensitivity in the developing larynx of Xenopus laevis.

Gonadal differentiation, the onset of androgen-stimulated laryngeal growth and the genesis of a sex difference in laryngeal innervation, all temporally coincide with thyroid hormone (TH)-induced metamorphosis in Xenopus laevis. To explore the role TH plays in the ontogeny of the Xenopus androgen-sensitive vocal neuromuscular system, we examined gonadal and laryngeal development in tadpoles in which metamorphosis had been blocked by treatment with the thyroxine synthesis inhibitor propylthiouracil (PTU). PTU treatment did not arrest gonadal differentiation. Testes from PTU-treated male tadpoles

Molecular analysis and developmental expression of the focal adhesion kinase pp125FAK in Xenopus laevis.

Integrin-mediated cellular adhesion to components of the extracellular matrix (ECM) is important in a number of morphogenetic events that occur during vertebrate embryogenesis. Recent studies suggest that the focal adhesion kinase pp125FAK is involved in the regulation of integrin-dependent signaling processes triggered by cell adhesion to the ECM. We report the cDNA cloning and sequence analysis of the Xenopus homolog of pp125FAK. We also describe temporal and spatial patterns of FAK expression during early development. Xenopus FAK shares greater than 90% identity with its avian and mammalian

Development of the olfactory nerve in the clawed frog, Xenopus laevis: II. Effects of hypothyroidism.

Quantitative and morphological data were obtained on developing olfactory axons in normal and hypothyroid larvae of the African clawed frog Xenopus laevis. Hypothyroid larvae were produced by rearing the animals, beginning at stage 48, in a 0.01% solution of propylthiouracil (PTU), a treatment that blocks synthesis of thyroid hormone. These PTU-treated larvae were compared to their age-matched siblings when these siblings reached stage 52 (premetamorphic larvae; prior to synthesis of thyroid hormone), stage 57 (late premetamorphic larvae; after the onset of thyroid hormone synthesis), or stage

Single olfactory organ associated with prosencephalic malformation and cyclopia in a Xenopus laevis tadpole.

A cyclops Xenopus laevis tadpole with a single olfactory organ is described. At a stage comparable to 48, the telencephalon was severely atrophic and only the region where the olfactory fibres terminated appeared to have the cytoarchitecture of the olfactory bulb. In this animal the central nervous system (CNS) appeared normally developed only posterior to the preoptic area. The hypothesis of a diencephalic origin of the region where the olfactory fibres terminated is discussed in the light of our previous results of olfactory placode transplantation. By analogy between this case and other mal

Craniofacial malformation in Xenopus laevis tadpoles caused by the exposure of early embryos to ethanol.

The principal features of fetal alcohol syndrome (FAS) include microcephaly, reduced body size, and characteristic facial malformation, most of which are associated with hypoplasticity of structures around the mouth. A recent study using pregnant mice suggested that the malformations in FAS are caused by the effects of ethanol on early embryos during gastrulation and neurulation. Exposure of Xenopus laevis early embryos to 1-2% ethanol until they developed to the late neurula stage produced craniofacial malformations in tadpoles which have many similarities with the described features of FAS.

Locations of androgen-concentrating cells in the brain of Xenopus laevis: autoradiography with 3H-dihydrotestosterone.

The distribution of hormone-concentrating cells in the brains of South African clawed frogs, Xenopus laevis, was examined autoradiographically after the administration of 3H-dihydrotestosterone. Hormone-accumulating cells were found in cranial nerve nucleus IX-X and adjacent smaller cells, a presumed medullary vestibular nucleus, a presumed sensory nucleus of cranial nerve V, dorsal tegmental area of the medulla, laminar nucleus of the torus semicircularis, ventral thalamus, and anterior pituitary. The pattern of dihydrotestosterone-labelled cells differs from previously reported results follo

The formation of the gonadal ridge in Xenopus laevis. II. A scanning electron microscope study.

This paper studies the surface morphology of the developing gonadal ridge in X. laevis between stages 44 and 49 (Nieuwkoop & Faber, 1956). During this period the primordial germ cells (PGCs) move laterally from the dorsal mesentery of the gut to the position of the presumptive gonadal ridge. As they do so the coelomic lining cells lateral to the mesentery differentiate into a specialized, longitudinally orientated band, stretching nearly the full length of the dorsal mesentery on each side. The PGCs migrate beneath this band of cells, which thus becomes the germinal epithelium of the gonadal r

Studies on the liver of Xenopus laevis. II. The ultrastructure of the peritoneal cover and the perihepatic layer.

The peritoneal cover and the subcapsular region of the liver of Xenopus laevis were examined with electron microscopy. In the subcapsular region a prominent perihepatic layer two to ten cells wide was found. This perihepatic layer is mainly composed of granulocytopoietic tissue. Other cell types found in the perihepatic layer were lymphocytes, pigment cells and fat storing cells. The presence of a perihepatic granulocytopoietic layer in the liver of Xenopus laevis is in contrast to current opinion that such a layer is a characteristic typical of urodeles. The phylogenetic significance of such

A comparison of amphibian (Xenopus laevis) peritoneal- and bone marrow-derived mast cells.

Mast cells are tissue-resident granulocytes that are often viewed through a negative lens due to their roles in allergies and hypersensitivity. While central to these conditions, mast cells have broader roles in vertebrate immune systems that are often overlooked. Whereas most mast cell research to date has been conducted in mammalian models, mast-like cells are found across vertebrates, presumably with conserved and divergent functions in disparate immune systems. Our past studies defined a role of amphibian (Xenopus laevis) mast cells in skin antifungal defenses but stopped short of function

Identification and Characterization of Static Craniofacial Defects in Pre-Metamorphic Xenopus laevis Tadpoles.

Craniofacial development is a complex, highly conserved process involving multiple tissue types and molecular pathways, with perturbations resulting in congenital defects that often require invasive surgical interventions to correct. Remarkably, some species, such as Xenopus laevis, can correct some craniofacial abnormalities during pre-metamorphic stages through thyroid hormone-independent mechanisms. However, the full scope of factors mediating remodeling initiation and coordination remain unclear. This study explores the differential remodeling responses of craniofacial defects by comparing

The olfactory network of larval Xenopus laevis regenerates accurately after olfactory nerve transection.

Across vertebrate species, the olfactory epithelium (OE) exhibits the uncommon feature of lifelong neuronal turnover. Epithelial stem cells give rise to new neurons that can adequately replace dying olfactory receptor neurons (ORNs) during developmental and adult phases and after lesions. To relay olfactory information from the environment to the brain, the axons of the renewed ORNs must reconnect with the olfactory bulb (OB). In Xenopus laevis larvae, we have previously shown that this process occurs between 3 and 7 weeks after olfactory nerve (ON) transection. In the present study, we show t

Investigation of Potassium Chloride for Euthanasia of Anesthetized African Clawed Frogs (Xenopus laevis).

Euthanasia is frequently performed in amphibians, but techniques are currently limited in number and variable in effectiveness. The current study examined the use of potassium chloride (KCl) for euthanasia of anesthetized African clawed frogs (Xenopus laevis). Twenty adult, female African clawed frogs were anesthetized by immersion in buffered tricaine methanesulfonate (MS-222) for 5 min beyond loss of righting reflex. Frogs were then randomly assigned to receive one of 4 treatments: KCl via intracardiac injection (10 mEq/kg; IC, n = 5), intracoelomic injection (100 mEq/kg; ICe, n = 5), immers

Influenza D virus M2 protein exhibits ion channel activity in Xenopus laevis oocytes.

BACKGROUND: A new type of influenza virus, known as type D, has recently been identified in cattle and pigs. Influenza D virus infection in cattle is typically asymptomatic; however, its infection in swine can result in clinical disease. Swine can also be infected with all other types of influenza viruses, namely A, B, and C. Consequently, swine can serve as a "mixing vessel" for highly pathogenic influenza viruses, including those with zoonotic potential. Currently, the only antiviral drug available targets influenza M2 protein ion channel is not completely effective. Thus, it is necessary to

Acute effects of Fe₂O₃, TiO₂, ZnO and CuO nanomaterials on Xenopus laevis.

Metal oxide nanomaterials have exhibited toxicity to a variety of aquatic organisms, especially microbes and invertebrates. To date, few studies have evaluated the toxicity of metal oxide nanomaterials on aquatic vertebrates. Therefore, this study examined effects of ZnO, TiO(2), Fe(2)O(3), and CuO nanomaterials (20-100 nm) on amphibians utilizing the Frog Embryo Teratogenesis Assay Xenopus (FETAX) protocol, a 96 h exposure with daily solution exchanges. Nanomaterials were dispersed in reconstituted moderately hard test medium. These exposures did not increase mortality in static renewal expos

Skin impulse excitation of spinal sensory neurons in developing Xenopus laevis (Daudin) tadpoles.

Responses to gentle touch in young Xenopus tadpoles are mediated by spinal cord sensory Rohon-Beard neurons. Tadpoles also respond to noxious stimuli that elicit 'skin impulses', which propagate between epithelial cells over the whole body surface, somehow entering the CNS to generate a response. After hatching (~48 h post-fertilization), skin impulse signals enter the CNS only via cranial nerves, but previous evidence suggested the possibility of direct entry to the spinal cord before this (~24 h). We have used behavioural and electrophysiological methods to explore the developmental pattern

Ovarian hyperstimulation syndrome in gonadotropin-treated laboratory South African clawed frogs (Xenopus laevis).

Ovarian hyperstimulation syndrome (OHS) is a rare but sometimes fatal iatrogenic complication of ovarian stimulation associated with the administration of exogenous gonadotropins to women undergoing treatment for infertility. Laboratory Xenopus spp are commonly treated with human chorionic gonadotropin (hCG) to stimulate ovulation and optimize the number of oocytes harvested for use in biomedical research. Here we report cases of OHS in 2 gonadotropin-treated laboratory Xenopus laevis. After receiving hCG, the frogs developed severe subcutaneous accumulation of fluid, coelomic distention, and

Hypergravity susceptibility of ventral root activity during fictive swimming in tadpoles (Xenopus laevis).

1. Fictive swimming is an experimental model to study early motor development. As vestibular activity also affects the development of spinal motor projections, the present study focused on the question whether in Xenopus laevis tadpoles, the rhythmic activity of spinal ventral roots (VR) during fictive swimming revealed age-dependent modifications after hypergravity exposure. In addition, developmental characteristics for various features of fictive swimming between stages 37/38 and 47 were determined. Parameters of interest were duration of fictive swimming episodes, burst duration, burst fre

Comparative developmental toxicity of nickel to Gastrophryne carolinensis, Bufo terrestris, and Xenopus laevis.

The early embryo-larval developmental toxicity of nickel (Ni) to 3 amphibian species, Xenopus laevis (South African clawed frog), Bufo terrestris (southern toad), and Gastrophryne carolinensis (eastern narrow-mouthed toad), was evaluated using a modified FETAX model. Studies were initiated from late blastulae stage (Nieuwkoop and Faber [NF] stage 10 or Gosner stage 12) and completed at a common embryological-based test termination point, which represented the completion of the major stages of organogenesis (NF stage 46 for Xenopus or Gosner stage 26 for the toads). Results indicated that, in t
