Alkaliphilic microbial cellulases: sources, structural insights, and bioengineering approaches for industrial applications.

As the biofuel/biorefinery/green economy advances quickly, the need for durable cellulases to operate under varying conditions in the industrial sector has increased. A specific type of cellulase that has emerged from this demand is alkaliphilic cellulase (ALKC). Due to their stability and high catalytic activity at elevated pH ranges (e.g., pH 8-11), ALKCs are being developed as biocatalysts for alkaline pretreatment and other harsh, high-pH industrial processes. The purpose of this review is to provide an updated overview of ALKCs focusing on their microbial origins, structural characteristic

Bioengineering approaches to dynamic impact analysis for cranial fracture interpretation in arcaheology.

Cranial fractures are widely documented in archaeological contexts, yet the application of fracture mechanics to differentiate traumatic events remains limited. This study analyses a dataset of 234 human cadavers subjected to 329 experimentally controlled blunt-impact tests, examining mechanical variables and fracture patterns that could be relevant to archaeological interpretation. The results show substantial methodological variability across the analysed studies. Analysis of these studies indicates that impact energy is the most reliable parameter for assessing fracture severity, suggesting

Focused ultrasound in modern medicine: bioengineering interfaces, molecular effects, and clinical breakthroughs.

Ultrasound technology, first utilized in 1947-1948 for diagnostic applications in obstetrics and gynecology, has significantly expanded its scope to include both diagnostic and therapeutic uses in modern medicine. The advent of continuous therapeutic ultrasound has allowed for its application in treating musculoskeletal pathologies, enhancing fracture healing, and even facilitating tumor treatment when paired with MRI. Ultrasonic cavitation, gas body activation, and mechanical stress are primary non-thermal mechanisms responsible for its biological effects. Recent advancements have expanded ul

Stem cells, cell therapies, and bioengineering in lung biology and diseases 2023.

Repair and regeneration of a diseased lung using stem cells or bioengineered tissues is an exciting therapeutic approach for a variety of lung diseases and critical illnesses. Over the past decade, increasing evidence from preclinical models suggests that mesenchymal stromal cells, which are not normally resident in the lung, can be used to modulate immune responses after injury, but there have been challenges in translating these promising findings to the clinic. In parallel, there has been a surge in bioengineering studies investigating the use of artificial and acellular lung matrices as sc

Bioengineering of a human physiologically relevant microfluidic blood-cerebrospinal fluid barrier model.

The human blood-cerebrospinal fluid barrier (hBCSFB) plays a crucial role in regulating brain interstitial fluid homeostasis, and disruption of the hBCSFB is associated with various neurological diseases. Generation of a BCSFB model with human physiologically relevant structural and functional features is crucial to reveal the cellular and molecular basis of these diseases and discover novel neurologic therapeutic agents. Unfortunately, thus far, few humanized BCSFB models are available for basic and preclinical research. Here, we demonstrate a bioengineered hBCSFB model on a microfluidic devi

Bioengineering human intestinal mucosal grafts using patient-derived organoids, fibroblasts and scaffolds.

Tissue engineering is an interdisciplinary field that combines stem cells and matrices to form functional constructs that can be used to repair damaged tissues or regenerate whole organs. Tissue stem cells can be expanded and functionally differentiated to form 'mini-organs' resembling native tissue architecture and function. The choice of the scaffold is also pivotal to successful tissue reconstruction. Scaffolds may be broadly classified into synthetic or biological depending upon the purpose of the engineered organ. Bioengineered intestinal grafts represent a potential source of transplanta

A tissue-bioengineering strategy for modeling rare human kidney diseases in vivo.

The lack of animal models for some human diseases precludes our understanding of disease mechanisms and our ability to test prospective therapies in vivo. Generation of kidney organoids from Tuberous Sclerosis Complex (TSC) patient-derived-hiPSCs allows us to recapitulate a rare kidney tumor called angiomyolipoma (AML). Organoids derived from TSC2-/- hiPSCs but not from isogenic TSC2+/- or TSC2+/+ hiPSCs share a common transcriptional signature and a myomelanocytic cell phenotype with kidney AMLs, and develop epithelial cysts, replicating two major TSC-associated kidney lesions driven by genet

Bioengineering a non-genotoxic vector for genetic modification of mesenchymal stem cells.

Vectors used for stem cell transfection must be non-genotoxic, in addition to possessing high efficiency, because they could potentially transform normal stem cells into cancer-initiating cells. The objective of this research was to bioengineer an efficient vector that can be used for genetic modification of stem cells without any negative somatic or genetic impact. Two types of multifunctional vectors, namely targeted and non-targeted were genetically engineered and purified from E. coli. The targeted vectors were designed to enter stem cells via overexpressed receptors. The non-targeted vect

Bioengineering the Endocrine Pancreas: Intraomental Islet Transplantation Within a Biologic Resorbable Scaffold.

Transplantation of pancreatic islets is a therapeutic option to preserve or restore β-cell function. Our study was aimed at developing a clinically applicable protocol for extrahepatic transplantation of pancreatic islets. The potency of islets implanted onto the omentum, using an in situ-generated adherent, resorbable plasma-thrombin biologic scaffold, was evaluated in diabetic rat and nonhuman primate (NHP) models. Intraomental islet engraftment in the biologic scaffold was confirmed by achievement of improved metabolic function and preservation of islet cytoarchitecture, with reconstitution

Human-scale whole-organ bioengineering for liver transplantation: a regenerative medicine approach.

At this time, the only definitive treatment of hepatic failure is liver transplantation. However, transplantation has been limited by the severely limited supply of human donor livers. Alternatively, a regenerative medicine approach has been recently proposed in rodents that describe the production of three-dimensional whole-organ scaffolds for assembly of engineered complete organs. In the present study, we describe the decellularization of porcine livers to generate liver constructs at a scale that can be clinically relevant. Adult ischemic porcine livers were successfully decellularized usi

Skin irritation in man: a comparative bioengineering study using improved reflectance spectroscopy.

Variable types of skin irritation were induced in 8 human female volunteers, ranging from subclinical to visible erythema with slight oedema. Skin reactions were graded clinically and objectively using transepidermal water loss (TEWL), laser Doppler flowmetry (LDF) and improved reflectance spectroscopy. This last technique enables separation of in vivo erythemas into relatively deoxygenated (venous--deoxy hem) and oxygenated (arterial--oxy hem) haemoglobin components. Compared to uninvolved skin, an empty patch increased oxy hem by 197% +/- 121% (p < 0.05). Exposure to vehicles also changes sk

Applications of Mesenchymal Stromal Cells in Hematologic Disease: Advances, Challenges, and Bioengineering Strategies.

Mesenchymal stromal cells (MSCs) are multipotent cells of mesodermal origin capable of self-renewal and multilineage differentiation. Characterized by low immunogenicity and tropism toward injury sites, MSCs exhibit critical properties including hematopoietic support, immunomodulation, and tissue regeneration. These unique attributes position MSCs as promising therapeutic tools for hematologic diseases, where disruption of the bone marrow niche impairs normal hematopoiesis. Co-transplantation of MSCs with hematopoietic stem cells (HSCs) facilitates HSC homing to the bone marrow niche and signi

Advances in Thyroid Gland Regeneration: The Integrated Approach of Cell Biology and Bioengineering.

The thyroid gland is an endocrine organ responsible for production of triiodothyronine and thyroxine, essential hormones that regulate human metabolism. A wide range of conditions can impair its function, leading to potential life-threatening consequences such as myxedema coma. The standard treatment for hypothyroidism is lifelong levothyroxine supplementation, which, despite being a significant therapeutic breakthrough, has notable limitations and does not fully restore quality of life for many patients. Biomimetic thyroid gland has emerged as a promising alternative treatment strategy for pa

Tracheal Regeneration: Recent Progress in the Application of Stem Cells in Tracheal Bioengineering.

Traumatic injury, stenosis, and malignancy involving large segments of the airway are difficult to reconstruct and require novel solutions. Despite advances in surgical techniques, the reconstruction of long-segment tracheal defects remains a significant challenge. Several bioengineering approaches have been explored for tracheal regeneration in vitro and in vivo, using cells in combination with three dimentional (3D) biological or synthetic scaffolds. This paper reviews recent advances in developing bioengineered trachea and the technologies utilized toward generating transplantable tracheal

Engineering the future of advanced therapy medicinal products: a bioengineering call to action.

Advanced Therapy Medicinal Products - cell therapies, gene therapies, and tissue-engineered products - are beginning to deliver on the promise of curative medicine: CAR-T therapies double survival in chemotherapy-refractory lymphomas, gene therapies reverse the natural history of spinal muscular atrophy and hemoglobinopathies, and Pluripotent Stem Cell (PSC)-derived islet transplantation renders type 1 diabetic patients insulin-independent. Yet the trajectory from proof-of-concept to equitable, scalable deployment is consistently impeded not only by unresolved biology but also by engineering,

Bioengineering Approaches for the 3D Blood-Retinal Barrier: Disease Modeling and Drug Development.

The integrity and functionality of the blood-retina barrier (BRB) are essential for maintaining homeostasis of the retina. Disruption of the BRB is a key feature in different stages of retinal diseases including diabetic retinopathy, age-related macular degeneration (AMD), and inherited retinal diseases such as retinitis pigmentosa. In order to understand and recapitulate various retinal diseases, there has been significant interest in the progress of in vitro 3D-BRB models as they may address limitations associated with traditional 2D cultures and xenogeneic animal models. Therefore, it is cr

Polydopamine-assisted biomineralized fibrous membrane and short fiber-reinforced spheroids for bone bioengineering.

Self-assembled spheroids provide substantial cell-cell interactions and cytoarchitectural complexity. However, the lack of bio-instructive cues can hinder cell differentiation, prompting the adoption of fiber-reinforced spheroids in tissue engineering. Herein, electrospun nanofibrous membrane were photochemically crosslinked, fragmented into short nanofibers (SNFs), and then surface-modified with mussel-inspired polydopamine (pDA). pDA-coated SNFs (pDA-SNFs) were incubated in simulated body fluid to induce nucleation and growth of a nanoscale hydroxyapatite layer in order to form mineralized p

A 3D cell culture system for bioengineering human neuromuscular junctions to model ALS.

The signals that coordinate and control movement in vertebrates are transmitted from motoneurons (MNs) to their target muscle cells at neuromuscular junctions (NMJs). Human NMJs display unique structural and physiological features, which make them vulnerable to pathological processes. NMJs are an early target in the pathology of motoneuron diseases (MND). Synaptic dysfunction and synapse elimination precede MN loss suggesting that the NMJ is the starting point of the pathophysiological cascade leading to MN death. Therefore, the study of human MNs in health and disease requires cell culture sy
