Absorption of anthocyanins from blueberry extracts by caco-2 human intestinal cell monolayers.

Recent studies have shown that dietary polyphenols may contribute to the prevention of cardiovascular disease and cancer. Anthocyanins from different plant sources including blueberries have been shown to possess potential anticancer activities. One of the key factors needed to correctly relate the in vitro study results to human disease outcomes is information about bioavailability. The objectives of the current study were to evaluate the absorption of blueberry anthocyanin extracts using Caco-2 human intestinal cell monolayers and investigate the effects of different aglycones, sugar moietie

Absorption, metabolism, and transport of carotenoids.

Carotenoids are currently under intense scrutiny regarding their potential to modulate chronic disease risk and prevent vitamin A deficiency, and renewed emphasis has been placed on achieving a better understanding of the metabolic fate of these compounds in humans. The development of new animal models, and use of human metabolic studies and stable tracer methods have greatly improved our knowledge of how carotenoids are absorbed, metabolized, and transported to tissues; however, many important issues remain unresolved. For example, intestinal uptake of carotenoids occurs by passive diffusion,

Toxicokinetics of soman stereoisomers after subcutaneous administration to atropinized guinea pigs.

The toxicokinetics of the four stereoisomers of the nerve agent C(+/-)P(+/-)-soman were investigated after subcutaneous administration of a 6 LD50 dose (148 micrograms/kg) to anaesthetized, atropinized, and artificially ventilated guinea pigs. Whereas the relatively nontoxic C(+/-)P(+)-isomers were not detected in blood, the highly toxic C(+/-)P(-)-isomers appeared within 1 min in the general circulation and reached maximum levels of 10-15 ng/ml blood within a period of ca. 7 min. In this absorption phase the blood levels of the C(+)P(-)-isomer lag clearly behind those of the C(-)P(-)-isomer.

Combining multi-surface and biotoxicity models to predict cadmium bioavailability and accumulation in a soil collembolan.

The link between internal metal concentrations in soil animals and external metal concentrations is a critical issue in soil ecotoxicity tests and involves metal transfer from solid-liquid interfaces to target soil animals. Soil cadmium (Cd) pollution is a major concern, the bioavailability of Cd to the model soil collembolan Folsomia candida was therefore determined in naturally Cd-contaminated soils by chemical extraction methods and mechanism-based multi-surface models (MSMs). Three combined models were also developed by combining MSMs with a free ion activity model (MSMs-FIAM), a modified

Dermal bioavailability of perfluoroalkyl substances using in vitro 3D human skin equivalent models.

Perfluoroalkyl substances (PFAS) have been identified in various products that come in contact with human skin, ranging from school uniforms to personal care products. Despite this, knowledge on human dermal uptake of PFAS is lacking. Thus, the human dermal absorption of 17 PFAS was assessed, for the first time, using in vitro 3D-human skin equivalent models exposed to 500 ng/cm2 PFAS dissolved in methanol over 24-36 h. The distribution of target PFAS is presented, based on three fractions: absorbed, un-absorbed, and retained within skin tissue (absorbable dose). Perfluoropentanoic acid (PFPeA

Enhanced Curcumin Bioavailability through Nonionic Surfactant/Caseinate Mixed Nanoemulsions.

The potential of heterogeneous systems like oil-in-water (O/W) nanoemulsions is exploited as an oral delivery system for curcumin, a natural lipophilic compound with numerous health benefits. Two types of O/W nanoemulsions, one stabilized by sodium caseinate (Cas-O/W), a surface-active and emulsifying protein, and the other stabilized by a blend of caseinate and Tween 20 (Mix-O/W), were loaded with the bioactive compound and tested through a simulated gastrointestinal digestion process to evaluate their effects on delivering of curcumin. It was first demonstrated that the amount of curcumin so

Physicochemical characterization of mineral (iron/zinc) bound caseinate and their mineral uptake in Caco-2 cells.

Milk proteins (especially caseins) are widely accepted as good vehicle for the delivery of various bioactive compounds including minerals. Succinylation is one of the most acceptable chemical modification techniques to enhance the mineral binding ability of caseins. Addition of minerals to succinylated proteins may alter their physicochemical and biochemical properties. Physicochemical characteristics of succinylated sodium caseinate (S.NaCN)-mineral (iron/zinc) complexes were elucidated. Chromatographic behaviour and fluorescence intensity confirmed the structural modification of S.NaCN upon

Mineral Dietary Supplement To Decrease Cadmium Relative Bioavailability in Rice Based on a Mouse Bioassay.

To determine the effectiveness of mineral dietary supplements to modulate cadmium (Cd) exposure, an in vivo mouse bioassay was conducted to determine Cd relative bioavailability (Cd-RBA) in Cd-contaminated rice (0.80 mg Cd kg-1) with and without Zn, Fe, or Ca supplements as nitrate or chloride salts. Without mineral supplements, Cd-RBA was 43 ± 5.3% based on average Cd accumulation in the liver plus kidneys as the end point. Among Ca(NO3)2, Zn(NO3)2, and Fe(NO3)2 supplements, 150-5000 mg kg-1 Ca was the most effective in reducing rice Cd-RBA by 31-80% to 8.5-29%, while 30-200 mg kg-1 Zn supple

Influence of bread crust-derived Maillard reaction products on phosphorus balance in rats.

BACKGROUND: Maillard reaction products (MRP) improve food palatability and are linked to some positive biological actions. However, diverse negative consequences, some related to protein damage and mineral availability, have been established. AIM OF STUDY: We investigated the effects of MRP, from a bread crust diet, on phosphorus bioavailability and tissue distribution in rats to determine whether these effects are related to the molecular weight of browning products. METHODS: During a study period of 88 days, rats were fed either a control diet or one of the following: with bread crust as a

[Possibilities for the in vitro evaluation of bioavailability. 5. Compact peroral agents with basic drugs].

Lipophilic, basic drugs can be transported across lipoid-membrane even if the part of the substance being not ionized in the solution is very low. Therefore the resorption model can be used for the control of drugs containing the substances as salts. In the case of 4 substances no differences could be found concerning half transport time between the aqueous solutions and the drugs.

Bean protein-based micro and nano-delivery systems for food bioactives: enhancing stability, absorption, and food matrix compatibility.

Many health-beneficial bioactive compounds exhibit poor solubility, chemical instability, and limited bioavailability within food matrices. Encapsulation protects bioactive compounds from environmental conditions and facilitates their controlled release and absorption in the human body. Bean proteins have recently gained attention as sustainable carrier materials for the delivery of bioactive compounds through micro- and nanoencapsulation methods. Their amphiphilic structure and ability to form films, gels, and emulsions make them a promising alternative to animal- and soy-based proteins. This

A Framework for Developing Intake and Use Guidance for Dietary Supplement Bioactives.

Dietary bioactives, which are commonly found in food, beverages, and dietary supplements, include nonessential food components that can support health beyond preventing nutrient deficiencies. Despite their known health benefits and potential for adverse effects, there is limited guidance on appropriate intake levels, safety, and efficacy of these ingredients, particularly those present in dietary supplements. This lack of guidance can be challenging for individuals attempting to make informed decisions about products marketed for health benefits. A previously published perspective proposed a 4

State-of-the-art on absorption mechanisms and factors affecting the bioavailability of preformed vitamin A.

Vitamin A deficiency remains a major health issue in many countries. Two types of vitamin A are naturally present in our diet, provitamin A carotenoids from plant sources and preformed vitamin A, mainly retinyl esters and retinol, found in animal products, fortified foods and supplements. The objective was to review current knowledge and gaps in understanding the mechanism and factors influencing preformed vitamin A bioavailability. The results show that the fate of retinyl esters in the intestine is well described, but that of retinol deserves further investigation. Preformed vitamin A presen

Bioavailability of Food Polyphenols: Current State of Knowledge.

(Poly)phenols, including flavonoids, phenolic acids, and tannins, are a diverse class of compounds found in plant-based foods and beverages. Their bioavailability has been extensively described and detailed metabolic pathways elucidated. Although some parent (poly)phenols are absorbed intact in the small intestine, most pass to the colon where they are extensively catabolized and their microbial products absorbed into the circulation. The sum of the metabolites absorbed can reach almost 100% in some cases and in some individuals. In recent years, there have been three major areas of advancemen

Unlocking Polyphenol Efficacy: The Role of Gut Microbiota in Modulating Bioavailability and Health Effects.

In humans, the bioactivity of polyphenols is highly dependent on dose intake and their interactions with the gastrointestinal tract and gut microbiota, which metabolize polyphenols into bioactive or inactive derivatives. Polyphenols are only partially absorbed in the small intestine, where enzymatic hydrolysis releases aglycone forms that may cross the gut barrier. A significant proportion of polyphenols escapes absorption and reaches the colon, where resident microbes convert them into simpler phenolic metabolites. Such molecules are often more bioavailable than the parent compounds and can e

The effect of highly bioavailable forms of curcumin on lipoprotein(a) plasma levels: A systematic review and meta-analysis of randomized clinical studies.

Curcumin is a bioactive compound derived from the rhizome of Curcuma longa (turmeric) that has garnered increasing attention for its potential health benefits. However, its use in clinical practice is limited due to its generally poor bioavailability. This issue can be overcome using novel delivery systems that enhance curcumin's solubility, extend its residence time in plasma, improve its pharmacokinetic profile, and increase its cellular uptake. Novel curcumin formulations with improved bioavailability have been suggested to elevate plasma concentrations of lipoprotein(a) (Lp(a)), but there
