Microbial-Driven Butyrate Regulates Jejunal Homeostasis in Piglets During the Weaning Stage.

Microbe-derived butyrate plays an important role in the gut health of young mammals during the weaning stage. A greater understanding of how butyrate regulates intestinal development is necessary for overcoming post-weaning diarrheal diseases. We aimed to investigate whether jejunal microbial metabolite butyrate modulates the apoptosis/proliferation balance and immune response in piglets during the post-weaning period of the first 3 weeks of life. On the one hand, during the first week post-weaning, the relative abundances of the dominant bacterial families Erysipelotrichaceae (P < 0.01) and L

Protective effects of butyrate-based compounds on a mouse model for spinal muscular atrophy.

Proximal spinal muscular atrophy (SMA) is a childhood-onset degenerative disease resulting from the selective loss of motor neurons in the spinal cord. SMA is caused by the loss of SMN1 (survival motor neuron 1) but retention of SMN2. The number of copies of SMN2 modifies disease severity in SMA patients as well as in mouse models, making SMN2 a target for therapeutics development. Sodium butyrate (BA) and its analog (4PBA) have been shown to increase SMN2 expression in SMA cultured cells. In this study, we examined the effects of BA, 4PBA as well as two BA prodrugs-glyceryl tributyrate (BA3G)

The effects of prebiotics on microbial dysbiosis, butyrate production and immunity in HIV-infected subjects.

Altered interactions between the gut mucosa and bacteria during HIV infection seem to contribute to chronic immune dysfunction. A deeper understanding of how nutritional interventions could ameliorate gut dysbiosis is needed. Forty-four subjects, including 12 HIV+ viremic untreated (VU) patients, 23 antiretroviral therapy-treated (ART+) virally suppressed patients (15 immunological responders and 8 non-responders) and 9 HIV- controls (HIV-), were blindly randomized to receive either prebiotics (scGOS/lcFOS/glutamine) or placebo (34/10) over 6 weeks in this pilot study. We assessed fecal microb

Sodium butyrate and its synthetic amide derivative modulate nociceptive behaviors in mice.

In the present study we investigated the role of sodium butyrate (butyrate), and its more palatable derivative, the N-(1-carbamoyl-2-phenyl-ethyl) butyramide (FBA), in animal models of acute and chronic pain. We found that oral administrations of butyrate (10-200mg/Kg) or equimolecular FBA (21.2-424mg/Kg) reduced visceral pain in a dose- and time-dependent manner. Both drugs were also effective in the formalin test, showing an antinociceptive effect. This analgesic effect was blocked by glibenclamide, suggesting the involvement of ATP-dependent K(+) channels. Moreover, following repeated admin

The niacin/butyrate receptor GPR109A suppresses mammary tumorigenesis by inhibiting cell survival.

GPR109A, a G-protein-coupled receptor, is activated by niacin and butyrate. Upon activation in colonocytes, GPR109A potentiates anti-inflammatory pathways, induces apoptosis, and protects against inflammation-induced colon cancer. In contrast, GPR109A activation in keratinocytes induces flushing by activation of Cox-2-dependent inflammatory signaling, and the receptor expression is upregulated in human epidermoid carcinoma. Thus, depending on the cellular context and tissue, GPR109A functions either as a tumor suppressor or a tumor promoter. However, the expression status and the functional im

Cyclic AMP synergizes with butyrate in promoting β-defensin 9 expression in chickens.

Host defense peptides (HDP) have both microbicidal and immunomodulatory properties. Specific induction of endogenous HDP synthesis has emerged as a novel approach to antimicrobial therapy. Cyclic adenosine monophosphate (cAMP) and butyrate have been implicated in HDP induction in humans. However, the role of cAMP signaling and the possible interactions between cAMP and butyrate in regulating HDP expression in other species remain unknown. Here we report that activation of cAMP signaling induces HDP gene expression in chickens as exemplified by β-defensin 9 (AvBD9). We further showed that, albe

Role of clevidipine butyrate in the treatment of acute hypertension in the critical care setting: a review.

Acutely elevated blood pressure in the critical care setting is associated with a higher risk of acute end-organ damage (eg, myocardial ischemia, stroke, and renal failure) and perioperative bleeding. Urgent treatment and careful blood pressure control are crucial to prevent significant morbidity. Clevidipine butyrate (Cleviprex) is an ultrashort-acting, third-generation intravenous calcium channel blocker. It is an arterial-selective vasodilator with no venodilatory or myocardial depressive effects. Clevidipine has an extremely short half-life of approximately 1 minute as it is rapidly metabo

Butyrate specifically down-regulates salmonella pathogenicity island 1 gene expression.

Invasion of intestinal epithelial cells by Salmonella enterica is decreased after exposure to butyric acid. To understand the molecular mechanisms of this phenomenon, a comparative transcriptomic analysis of Salmonella enterica serovar Enteritidis and Salmonella enterica serovar Typhimurium grown in medium supplemented with butyrate was performed. We found that butyrate down-regulated the expression of 19 genes common to both serovars by a factor of twofold or more, and 17 of these genes localized to the Salmonella pathogenicity island 1 (SPI1). These included the SPI1 regulatory genes hilD an

The importance of colonic butyrate transport to the regulation of genes associated with colonic tissue homoeostasis.

The transition from normality to malignancy in colorectal cancer is characterized by alterations in the expression of genes associated with the maintenance of tissue homoeostasis. Butyrate, a product of microbial fermentation of dietary fibre in the colon, is known to regulate a number of genes associated with the processes of proliferation, differentiation and apoptosis of colonic epithelial cells, and, hence, homoeostasis of colonic tissue. We have shown previously that the transport of butyrate into colonocytes is of fundamental importance to butyrate's regulatory ability, and therefore sou

[Treatment of unspecific ulcerative rectocolitis in a child with enemas containing butyrate. Case report].

OBJECTIVE: To report a promising therapy to ulcerative colitis in children. METHODS: We report a case of ulcerative colitis in 4 years old female, who had this diagnosis since she was 1 year old. The treatment of RCU1 had an unsatisfactory response to conventional therapies (rectal and oral corticosteroids and immunosuppressive agent). RESULTS: She showed improvement, using enemas containing butyrate, in clinical, endoscopic and histological parameters. CONCLUSIONS: Although the role of butyrate in the treatment of ulcerative colitis still remains to be determined, we find effective respons

Arginine butyrate-induced susceptibility to ganciclovir in an Epstein-Barr-virus-associated lymphoma.

Lymphoproliferative disorders associated with Epstein-Barr virus (EBV) infections can occur in the setting of immunosuppression. In some patients, the lymphoproliferative disorder can resemble an aggressive monoclonal non-Hodgkins lymphoma (NHL). These NHL are poorly responsive to conventional therapy. Similarly, antiviral therapy with synthetic nucleosides such as ganciclovir are ineffective because the genes that render the virus susceptible to therapy are not expressed in EBV+ lymphomas. Using a cell line derived from a lung transplant recipient with an EBV+ immunoblastic NHL, we studied th

Acetate, butyrate and proline uptake in the caecum and colon of prairie voles (Microtus ochrogaster).

We have measured unidirectional uptake (not transmural flux) of acetate, butyrate and proline by everted sleeves of intact tissue from the jejunum, caecum, proximal colon and distal colon of prairie voles (Microtus ochrogaster). There was active (i.e. Na(+)-dependent) transport of L-proline in the jejunum, but we found no evidence for it in any region of the hindgut (i.e. the caecum, proximal colon and distal colon). Uptake of acetate was carrier-mediated in all three regions of the hindgut, but the Jmax and apparent Km (< or = 1.5 mmol l-1) were low, and uptake was primarily passive over the

Mechanism of L-triiodothyronine (T3) uptake by glial C6 cells: regulation by butyrate.

The mode of entry of triiodothyronine (T3) and its regulation by butyrate was studied in cultured glial C6 cells. Uptake of [125I]T3 increases for at least 60 min in C6 cells. The amount of cell-associated radioactivity is 2- to 4-fold higher during the entire time-course in cells previously exposed to 2 mM butyrate for 48 h. Uptake was non-saturable since uptake velocity was linearly related to the extracellular hormone concentration between 0.2 and 800 nM T3 in control and butyrate-treated cells. Uptake velocity increased by more than 3-fold in the cells incubated with the fatty acid. T3 upt

Butyrate-producing Eubacterium rectale inhibits gastric carcinogenesis and augments the efficacy of immunotherapy.

BACKGROUND: Immunotherapy has transformed gastric cancer (GC) treatment, but therapeutic resistance remains a major limitation. Recent evidence suggests that the gut microbiota can modulate anti-tumor immunity and enhance immunotherapy response, offering a promising avenue to overcome resistance. METHODS: Using a prospective GC cohort, we identified Eubacterium rectale (E. rectale) as a bacterium associated with improved efficacy of tislelizumab plus chemotherapy and longer progression-free survival (PFS). We then conducted in vitro and in vivo experiments to evaluate the effects of E. rectale

The effect of adding colostrum or sodium butyrate to the diet on the intestinal barrier of weaned piglets.

Weaning is a critical stage for piglets, often leading to intestinal barrier disruption, impaired nutrient absorption, and growth reduction. This study investigated the effects of dietary supplementation with dried bovine colostrum or sodium butyrate on intestinal barrier function, nutrient absorption, and growth performance in weaned piglets. Eighteen DanBred Hybrid piglets (28 days old) were allocated to three groups (n = 6): control (no additive), colostrum, and sodium butyrate. Piglets were fed standardized prestarter and starter diets for 28 days. Growth performance and feed conversion ra

Methylation and expression dynamics in broiler liver following in-ovo sodium butyrate administration.

INTRODUCTION: Butyrate is one of the three main short-chain fatty acids, and it provides energy, controls the state of the intestinal microbiota and mediates the immune response. Sodium butyrate supplementation improves poultry production and changes the intestinal microbiota dynamically. These changes may affect the liver directly and indirectly through pathways in the gut-liver axis, the bidirectional relationship between the liver and intestines. The study analysed gene expression and methylation in the broiler liver after in-ovo stimulation by sodium butyrate. MATERIAL AND METHODS: Incuba

Mechanisms of Butyrate in Diabetes Through the Metabolism-Immunity Axis and Targeted Therapeutic Strategies.

This review summarizes the impact of butyrate on the development of diabetes, particularly its mechanisms of action through the metabolism-immunity axis. Butyrate, a short-chain fatty acid produced by the fermentation of dietary fibers by gut microbiota, plays a crucial role in maintaining gut health and metabolic homeostasis. Recent studies have highlighted the significance of butyrate in regulating metabolic functions, including glucose metabolism, while also affecting immune responses. However, the precise mechanisms by which butyrate influences these pathways in the context of diabetes rem

Therapeutic potential of butyrate supplementation in sepsis: a review of preclinical evidence and translational perspectives.

Sepsis remains a major global health problem and is responsible for millions of deaths annually despite significant progress in antimicrobial therapy and organ support. Increasing evidence highlights the role of the gut-immune axis in shaping host responses during sepsis, with particular interest in microbiota-derived metabolites such as short-chain fatty acids (SCFAs). Among these, butyrate has emerged as a promising candidate due to its anti-inflammatory, immunomodulatory, and intestinal barrier-preserving properties. This narrative review summarizes current evidence regarding the biological
