We aimed to assess the mechanisms responsible for hyperphagia and metabolic alterations caused by maternal moderate caloric restriction during gestation. Male and female offspring of control and 20% caloric-restricted rats (CR) were studied. They were fed a normal-fat diet until 4 months of age and then moved to a high-fat diet until 6 months of age. Blood parameters and expression of selected genes in hypothalamus, retroperitoneal white adipose tissue (rWAT) and liver were analyzed at 25 days and 6 months of age. Plasma leptin was measured during suckling. Levels of proteins involved in insul

The present study is part of a larger project that investigates the effect of caloric restriction on longevity in the rhesus monkey. The purpose of the present study was to document presbycusis and the effect of caloric restriction on presbycusis in monkeys. The control group had 35 monkeys allowed to eat freely and the caloric-restricted group (CR) had 33 monkeys with a 30% reduction in caloric intake. Monaural and binaural auditory brainstem response (ABR) and middle latency response (MLR) were obtained from 27 female and 41 male monkeys that were 11-23 years of age and had been in the study

The current study was conducted to determine if physical activity and/or weight control could influence the age-related decrease in beta cell insulin response noted in earlier studies. As such, virgin, male Sprague-Dawley rats were maintained in our facility for 1 yr on three differential experimental programs; in the first group, control rats lived under standard laboratory conditions; the second group of rats ran several miles a day in exercise wheels, and the third group was given a calorie-restricted diet designed to keep the rats weight-matched with the exercising rats. The results showed

Cerebrovascular dysfunction, including reduced cerebral blood flow (CBF) and cerebrovascular reactivity (CVR), contributes to Alzheimer's disease (AD). Caloric restriction (CR) and resveratrol (Rsv) benefit vascular health, yet their impact on CBF and CVR in AD remains unclear. Here, we investigated the cerebrovascular effects of 40% CR or Rsv supplementation in WT and TgF344-AD rats. Using pseudo-continuous arterial spin labelling (pCASL) MRI, we observed that male Tg control (Ctrl) rats exhibited reduced relative CBF (rCBF) and absolute CBF (aCBF) in the caudate-putamen at baseline compared

UNLABELLED: Nutrient metabolism influences HIV-1 replication, antiviral immunity, and chronic inflammation, yet is difficult to leverage for therapeutic gain. We sought to modulate metabolism in the non-human primate model of HIV-1 by caloric restriction (CR), a modality canonically known for its antiaging benefits. Four months of 30% CR was safe and resulted in broad and systemic metabolic reprogramming in healthy adult male and female rhesus macaques. Relative to that of ad libitum- fed animals, CR lowered the frequencies of target CCR5+ CD4 T cells in the gut mucosa. Upon infection with SIV

Background: Global dietary patterns have gradually shifted toward a 'western type' with progressive increases in rates of metabolic imbalance. Recently, animal and human studies have revealed positive effects of caloric restriction (CR) on many health domains, giving new knowledge for prevention of ill and health promotion; Methods: We conducted a systematic review (SR) of randomized controlled trials (RCTs) investigating the role of CR on health status in adults. A meta-analysis was performed on anthropometric, cardiovascular and metabolic outcomes; Results: A total of 29 articles were retrie

Cognitive deficits may occur after mild traumatic brain injury (mTBI), but effective treatment modalities are presently unavailable. Caloric restriction (CR) has beneficial effects on neurodegenerative diseases and brain injury. However, the underlying mechanisms have not yet been clearly defined. Therefore, the aim of the present study was to investigate the short-term effects of CR treatment on cognitive function in mice after mTBI. Forty-five 12-week-old C57/BL6 mice were subjected to closed-head mTBI using a weight drop device. The mice were then randomly divided into three groups accordin

Caloric restriction and intermittent fasting are known to improve glucose homeostasis and insulin resistance in several species including humans. The aim of this study was to unravel potential mechanisms by which these interventions improve insulin sensitivity and protect from type 2 diabetes. Diabetes-susceptible New Zealand Obese mice were either 10% calorie restricted (CR) or fasted every other day (IF), and compared to ad libitum (AL) fed control mice. AL mice showed a diabetes prevalence of 43%, whereas mice under CR and IF were completely protected against hyperglycemia. Proteomic analys

OBJECTIVE: Individuals with high levels of orthorexic eating behavior (OEB) show a (pathological) fixation on healthy eating. Shared characteristics of OEB with individuals with anorexia nervosa (AN) raised debate over whether orthorexia nervosa constitutes a distinct disorder. Distinctions may lie in preference for and approach behavior toward foods differing in calorie content and healthiness. We expected reduced liking and weaker approach biases in AN and OEB compared with controls; specifically for high-caloric foods in AN and for unhealthy foods in OEB.
METHOD: Thirty-five self-reported

Alzheimer's disease (AD) is regarded as a disease of the brain. Cumulative evidence increasingly supports a full-body view on this disorder, with the liver and kidneys playing an important role in amyloid clearance. The latter is likely potentiated by caloric restriction (CR), whose impact on the metabolism of amyloid-handling tissues is poorly understood. We studied the sex-specific effects of amyloidosis and CR on oxidative and metabolic processes in APPPS1 mice that express amyloidogenic proteins. Wild-type (WT) and APPPS1 mice were either fed ad libitum (AL) or received 70% of their AL cal

The nutritional composition of the diet and food choices can influence weight loss program outcomes, although caloric deficit remains the main strategy for reducing body fat. This study compared weight loss, hematological parameters, biochemical profile, and behavior of overweight and obese dogs (n = 34) fed under caloric restriction using commercial dry food (n = 9), homemade food (n = 9), or mixed feeding (n = 16). Each dog was monitored for approximately three months (104 days) with monthly consultations for clinical evaluation, caloric intake adjustments, blood collection, and morphometric

Caloric restriction (CR) alters energy metabolism and increases systemic glucocorticoid production, contributing to bone loss and bone marrow adipose tissue (BMAT) expansion. The enzyme 11β-hydroxysteroid dehydrogenase type 1 (11β-HSD1) mediates the local amplification of glucocorticoids within these tissues, but its contribution to CR-induced skeletal alterations remains unclear. Eight-week-old male and female WT and global 11β-HSD1 KO mice were assigned to ad libitum feeding or 30% CR for 8 wk. Body composition was assessed by DXA, bone microarchitecture by micro-CT (μCT), biomechanics by 3-

BACKGROUND: Psoriasis is a chronic immune‑mediated inflammatory skin disease that is strongly associated with metabolic dysregulation and excess adiposity. Dietary strategies that reduce energy intake either by continuous daily caloric restriction (CCR) or time‑limited/periodic intake such as intermittent fasting (IF) or ketogenic diets (Keto) may improve psoriasis severity through weight loss, metabolic improvements, alterations in gut microbiota, and immune-metabolic effects.
OBJECTIVE: We aimed to compare the therapeutic effectiveness of IF, Keto, and CCR in reducing the severity of psoria

Age-related obesity is a growing healthcare burden. Effectiveness of body weight reducing mediators changes with aging and these alterations may contribute to aging obesity. Our previous results suggested a potential contributions of urocortin 2 (UCN2) to aging obesity and age-related cachexia. Lifestyle interventions, such as caloric restriction or physical activity can improve body weight and body composition. Previous observations suggest that lifestyle interventions may also influence age-associated regulatory changes in energy balance. We aimed to study the effects of a 12-week treadmill

The geroscience hypothesis proposes that therapy to slow or reverse molecular changes that occur with aging can delay or prevent multiple chronic diseases and extend healthy lifespan1-3. Caloric restriction (CR), defined as lessening caloric intake without depriving essential nutrients4, results in changes in molecular processes that have been associated with aging, including DNA methylation (DNAm)5-7, and is established to increase healthy lifespan in multiple species8,9. Here we report the results of a post hoc analysis of the influence of CR on DNAm measures of aging in blood samples from t

The origins of theories specifying dietary restraint as a cause of eating disorders can be traced to the 1970s. This paper will present an overview of the origins of dietary restraint theories and a brief historical review of evidence will be summarized. Recent research will be presented, including the results from the CALERIE Phase 1 study, as well as CALERIE Phase 2, which were NIH-sponsored randomized controlled trials. CALERIE 2 provided a test of the effect of two years of caloric restriction (CR) on the development of eating disorder syndromes and symptoms in comparison to a control grou

Caloric restriction (CR) or Dietary restriction (DR) is known to improve health and in many cases increases lifespan. However, its negative effect on reproduction has not been fully studied. Practicing CR/DR without adequate knowledge on its side effect may risk complications such as infertility, birth defect, or malnutrition. In this study, by using several CR strategies in C. elegans, we examine key functions of reproduction including embryonic development and larvae growth. We find that CR significantly decreases the survival of embryos and slows the growth of the offspring. We further dete

Postoperative cognitive dysfunction is common in the elderly. Endoplasmic reticulum stress (ER-stress) increases neuronal apoptosis after surgery, and chaperone molecules, such as heat shock proteins (HSPs), help reduce unfolded protein reactions, thereby promoting protein homeostasis. Mammal sirtuin1 (SIRT1)-mediated deacetylation of heat shock factor 1 (HSF1) upregulates HSF1 binding to the HSP70 promoter. Caloric restriction (CR) improves cognition in many neurodegenerative models. In this study, we evaluated whether CR improves impaired learning and memory after surgery by attenuating ER-s

Ames dwarf (Prop1 (df/df) ) mice are remarkably long-lived and exhibit many characteristics of delayed aging and extended healthspan. Caloric restriction (CR) has similar effects on healthspan and lifespan, and causes an extension of longevity in Ames dwarf mice. Our study objective was to determine whether Ames dwarfism or CR influence neuromusculoskeletal function in middle-aged (82 ± 12 weeks old) or old (128 ± 14 w.o.) mice. At the examined ages, strength was improved by dwarfism, CR, and dwarfism plus CR in male mice; balance/ motor coordination was improved by CR in old animals and in mi

Rodents are commonly used as models for human aging because of their relatively short life span, the ease of obtaining age-specific tissue samples, and lower cost. However, age-associated disease may confound inbred animal studies. For example, numerous physiologically significant lesions, such as chronic nephropathy, are more common in aged Fischer 344 (F344) rats than in other strains (Bronson RT, Genetic Effects of Aging, 1990). Conversely, F344 x Brown-Norway F1-hybrid (F344BN) rats, developed by the National Institute on Aging for aging research, live considerably longer and have fewer pa
