The chloroplast-mitochondria interaction under water extremes: Insights into metabolic tug-of-war.

Plants respond to water extremes through dynamic, organelle-level reprogramming of resource allocation rather than metabolic shutdown. Recent advances suggest that drought and flooding induce coordinated metabolic reorganization between chloroplasts and mitochondria, shifting energy distribution from growth to stress acclimation via retrograde signaling pathways. These signals are integrated by the trehalose-6-phosphate (Tre6P)-sucrose non-fermenting 1-related protein kinase 1 (SnRK1)-target of rapamycin (TOR) network, a central regulatory hub that interprets carbon and energy status to modula

Sensitive detection of chloroplast movements through changes in leaf cross-polarized reflectance.

We show that the positioning of chloroplasts in plant cells affects the ability of photosynthetic organs to depolarize linearly polarized light through diffuse reflection. We apply this phenomenon for the detection of blue-light-induced chloroplast movements using light reflected from leaves and green stems. High-light-induced chloroplast avoidance leads to an increase in reflectance, while chloroplast accumulation in low light has the opposite effect. The changes in reflectance are correlated with an increase in the leaf's ability to depolarize linearly polarized light. Inclusion of polarizat

Thylakoid membrane transporter SecE1 is required for chloroplast development and photosynthetic function in Arabidopsis.

Chloroplast biogenesis depends on protein transport, pigment biosynthesis, and thylakoid membrane assembly, which are coordinated through anterograde (nucleus-to-plastid) and retrograde (plastid-to-nucleus) signaling. The chloroplast Sec translocon mediates the import of nuclear-encoded precursor proteins across the thylakoid membrane, yet the regulatory role of its core subunit SecE1 in chloroplast development and photosynthetic performance remains incompletely understood. Here, we characterized a chlorotic Arabidopsis mutant with partial loss of SecE1 function. Compared with the wild type an

RNA-Guided Engineering of the Chloroplast Genome Enabled by Plastid-Expressed Guide RNAs.

Our goal is to develop RNA-guided engineering of the chloroplast genome using the CRISPR/Cas9 system. We designed chloroplast minigenes to obtain properly sized single guide RNAs (sgRNAs) in tobacco chloroplasts. The sgRNA 5' end is defined by transcription from an rRNA operon promoter, and its 3' end by processing a downstream tRNA (trnG) or a hepatitis delta virus (HDV) ribozyme. Cas9 is expressed from a nuclear gene and is targeted to chloroplasts by fusion to a transit peptide. Cas9 incorporated the sgRNA and introduced double-strand breaks in the plastid DNA (ptDNA). We report here that t

Comparative genomics and phylogenetic analysis of three Malvaceae species on the basis of chloroplast genomes.

INTRODUCTION: The Malvaceae family shows rich species diversity and has substantial economic and medicinal value. However, the frequent interspecific hybridization among members of this family has resulted in confused phylogenetic relationships among the groups, limiting the usefulness of traditional classification methods. METHODS: This study aimed to investigate the phylogenetic relationships among selected taxa of Malvaceae by evaluating 23 chloroplast (CP) genomes, including three newly assembled CP genomes. Among these three genomes, the CP genome of Hibiscus schizopetalus L. was reporte

Conserved DNA architect couples chloroplast development to cell cycle in developing cotyledons.

Light orchestrates cotyledon greening and growth by coordinating chloroplast development with cell division and expansion, yet the molecular integrator of these processes remains unknown. Here we identify RDE (REGULATOR OF DG1 EXPRESSION), a conserved DNA architectural factor that synchronizes thylakoid protein complex biogenesis during chloroplast development with the G1-S transition of cell cycle progression in Arabidopsis thaliana. RDE mediates promoter DNA bending to sequester DPa in stable nucleoprotein complexes, precluding DPa-E2Fa heterodimer formation. This repression mechanism contro

Chloroplast reconstitution of heterologous Rubisco and carboxysome enables photoautotrophic growth of Chlamydomonas reinhardtii.

The model green alga Chlamydomonas reinhardtii has a pyrenoid within chloroplast for photosynthetic CO2 fixation under limiting CO2 conditions and its chloroplast is an ideal chassis for engineering photosynthetic modules. In contrast, carboxysomes are bacterial microcompartments that encapsulate Rubisco for CO2 fixation. The aim of the present study was to determine if a bacterial carboxysome or its components can replace pyrenoid and function to allow Chlamydomonas growth powered by photosynthesis. We replaced the Chlamydomonas endogenous RbcL gene with cbbL and cbbS from the chemoautotrophi

Plant chloroplast stress response: insights from mass spectrometry metabolites analysis.

Plant chloroplasts produce excess reactive oxygen species (ROS) during photosynthesis, particularly under biotic and abiotic stress conditions. These adverse environmental stresses lead to significant alterations in various cellular components, especially within the chloroplast, which serves as a key stress-sensor organelle. The stress response of chloroplasts can trigger plastid-to-nucleus retrograde signaling and enhance the biosynthesis of biologically active compounds and phytohormones, which are mechanisms that aid plants in acclimating to environmental stress. While ROS act as signaling

The Mars1 kinase confers photoprotection through signaling in the chloroplast unfolded protein response.

In response to proteotoxic stress, chloroplasts communicate with the nuclear gene expression system through a chloroplast unfolded protein response (cpUPR). We isolated Chlamydomonas reinhardtii mutants that disrupt cpUPR signaling and identified a gene encoding a previously uncharacterized cytoplasmic protein kinase, termed Mars1-for mutant affected in chloroplast-to-nucleus retrograde signaling-as the first known component in cpUPR signal transmission. Lack of cpUPR induction in MARS1 mutant cells impaired their ability to cope with chloroplast stress, including exposure to excessive light.

Limited Responsiveness of Chloroplast Gene Expression during Acclimation to High Light in Tobacco.

Acclimation to changing light intensities poses major challenges to plant metabolism and has been shown to involve regulatory adjustments in chloroplast gene expression. However, this regulation has not been examined at a plastid genome-wide level and for many genes, it is unknown whether their expression responds to altered light intensities. Here, we applied comparative ribosome profiling and transcriptomic experiments to analyze changes in chloroplast transcript accumulation and translation in leaves of tobacco (Nicotiana tabacum) seedlings after transfer from moderate light to physiologica

Effects of Cr2O3 nanoparticles on the chlorophyll fluorescence and chloroplast ultrastructure of soybean (Glycine max).

Chromic oxide nanoparticles (Cr2O3 NPs) are widely used in commercial factories and can cause serious environmental problems. However, the mechanism behind Cr2O3 NP-induced phytotoxicity remains unknown. In this study, the effects of Cr2O3 NPs on the growth, chlorophyll fluorescence, SEM-EDS analysis, and chloroplast ultrastructure of soybean (Glycine max) were investigated to evaluate its phytotoxicity. The growth of soybean treated with various Cr2O3 NP suspensions (0.01, 0.05, 0.1, and 0.5 g L-1) was significantly inhibited. Specially, shoot and root biomass decreased by 9.9 and 46.3%, resp

Integration of chloroplast nucleic acid metabolism into the phosphate deprivation response in Chlamydomonas reinhardtii.

Cell survival depends on the cell's ability to acclimate to phosphorus (P) limitation. We studied the chloroplast ribonuclease polynucleotide phosphorylase (PNPase), which consumes and generates phosphate, by comparing wild-type Chlamydomonas reinhardtii cells with strains with reduced PNPase expression. In the wild type, chloroplast RNA (cpRNA) accumulates under P limitation, correlating with reduced PNPase expression. PNPase-deficient strains do not exhibit cpRNA variation under these conditions, suggesting that in the wild type PNPase limits cpRNA accumulation under P stress. PNPase levels
