Effect of glycerol on cochlea microcirculation.

Microcirculation in the cochlea of l2 normal anesthetized guinea pigs was investigated after intravenous infusion of 2.4 ml/kg of 40% glycerol. In these experiments endolymphatic PO2, blood pressure, cochlear microphonics, and endocochlear potentials were recorded. Administration of glycerol caused a significant increase in endocochlear PO2 for about 30 minutes. The improvement of hearing in patients with Meniere's disease is thus not only a result of the reduced hydrostatic pressure, as has been demonstrated by others, but may also be the result of vasodilatation, which improves microcirculat

Electrocochleography of ears with mumps deafness.

Electrocochleography was performed on 16 affected ears of 15 deaf patients whose deafness had resulted from the mumps. Although pure-tone audiometry showed no response in each case, the cases of mumps deafness can be classified into the following three types of cochlear impairment according to the cochlear microphonic (CM) response: (1) no action potential (AP) response but a well-developed CM response, with impairment at the neural level and probable functioning of Corti's organ; (2) absence of both the AP CM responses, with severe impairment of both the neural regions and Corti's organ; (3)

Effects of electrical stimulation on the acoustically evoked auditory-nerve response in guinea pigs with a high-frequency hearing loss.

Criteria for cochlear implantation keep expanding and people with substantial residual low-frequency hearing are considered candidates for implantation nowadays. Therefore, electro-acoustical stimulation in the same ear (EAS) is receiving increasing interest. We have investigated the effects of intracochlear electrical stimulation on acoustically evoked auditory-nerve activity, using a forward masking paradigm. The stimulation electrode was placed in the basal turn of the cochlea. Compound action potential (CAP) recordings were performed in guinea pigs with severe high-frequency hearing loss a

Application of intraoperative round window electrocochleography for screening the patients with auditory neuropathy.

BACKGROUND: Most patients with auditory neuropathy (AN) could receive good even the best effects after cochlear implantation. How to diagnose AN objectively and accurately is very important. In this study, we screened the patients with AN according to the presence or absence of compound action potential (CAP) of intraoperative round window electrocochleography (RW ECochG). METHODS: Intraoperative RW ECochG was performed on 32 patients with profound sensorineural deafness, who had normal cochlea during cochlear implantation surgery under general anesthesia in the standard operating room. The c

Trimethyltin disrupts loudness recruitment and auditory threshold sensitivity in guinea pigs.

Trimethyltin (TMT) impairs auditory thresholds within minutes of systemic administration. However, there are no data which relate to the output of the auditory nerve at sound levels above threshold. In this experiment, we evaluated the functional effects of TMT on the auditory threshold by identifying the sound level which just produced a detectable compound action potential (CAP). We also assessed outer hair cell function by measuring the cochlear microphonic (CM), a nonpropagated ac potential which is phase-locked to the stimulus. Finally, we measured the growth of the N1 amplitude as a func

Effects of acute styrene and simultaneous noise exposure on auditory function in the guinea pig.

Although styrene has been demonstrated to disrupt vestibular function acutely, parallel studies have not been conducted in the auditory system. This article presents data on the effects of acute styrene administration by injection and inhalation on cochlear function. No deleterious effect of the maximally tolerated styrene dose on hearing was identified when cochlear function was assessed using a within-subjects design. When guinea pigs were administered styrene by inhalation during a single 7-h period, normal auditory function was observed both 1 and 7 days later as compared to chamber contro

The active process is affected first by intense sound exposure.

Evidence exists to suggest that intense sound releases excess neurotransmitter from the inner hair cells. However, it has been previously reported that intense sound affects the cochlear micromechanics by altering the stereocilia. Therefore, we tested the hypothesis that intense sound affects structures involved in transduction before it affects the nerve endings. In order to test this hypothesis, we examined the interaction of intense sound with kynurenate which blocks the action of the neurotransmitter on the afferent nerve endings. Neurophysiologic assessment of endolymphatic hydrops.

An electrophysiological method for assessing endolymphatic hydrops of the cochlea in an animal model of Meniere's disease is described. Eighth nerve gross action potentials (AP) were evoked by tone bursts presented at discrete phases of a simultaneously delivered 50-Hz pure tone. Hydropic cochleas showed significantly less modulation of AP amplitude and latency than normal cochleas. This method may be useful in objectively evaluating Meniere's disease with electrocochleography.

The effects of continuous versus interrupted noise exposures on distortion product otoacoustic emissions in guinea pigs.

Distortion product otoacoustic emissions (DPOAE) were measured serially in guinea pigs before and following 4-h exposures to a half-octave band of noise centered at 6 kHz. Stimulus parameters used to elicit the DPOAE were f2/f1 = 1.26 and L2 = L1-10. The 80 dB SPL exposures resulted in attenuation of emissions, which was maximal at the frequency one-half octave above the exposure when referenced to the f2 stimulus, and which recovered back to baseline after 2 days. The 90 dB SPL exposures resulted in a permanent deficit in emissions elicited by high-frequency stimuli, as measured after 8 days

Binaural acoustic reflex activity following monaural noise exposure in decerebrate chinchillas.

Acoustic reflex thresholds, latencies and magnitudes were recorded binaurally in decerebrate chinchillas suffering monaural, temporary noise trauma. Simultaneous recordings of cochlear microphonics from both cochleae during monaural reflex stimulation served as an index for acoustic reflex dynamics. Baseline values of threshold, latency and magnitude were recorded from ipsilateral and contralateral ears prior to a monaural 2-hour exposure to a 0.5-kHz octave-band noise at 100 dB SPL. Reflex dynamics were measured immediately after exposure and again following 1 h of recovery. Cochlear micropho

Potentiating effects of cisplatin and ethacrynic acid in ototoxicity.

Seven milligrams per kilogram of body weight of cisplatin and 50 mg/kg of body weight of ethacrynic acid were given intravenously to guinea pigs. The threshold for Preyer's reflex, the cochlear microphonics (CM), and the endocochlear dc potential (EP) were measured during a four-day period after the injection. The changes in Preyer's reflex audiometry and the CM were more severe than that of the EP. These changes are greater than those that occur with the administration of each agent independently. The ototoxic interaction seemingly has a greater effect on the hair cells than on the stria vasc

Comparison of the non-adrenergic action of phentolamine with that of vanadate on cochlear function.

Two drugs, which upon superficial examination appeared to be acting on common processes, have been found upon closer investigation to act by quite different means. Both act primarily at the organ of Corti, causing a pronounced increase of the endocochlear potential and a depression of the cochlear microphonic (CM). These effects are accompanied by the elimination of a negative component of the EP; however, it was found that these three effects are produced by phentolamine in scala media (or, more slowly, in scala tympani) but by vanadate only in scala tympani. This difference in locus of actio

Auditory evoked responses under total spinal anesthesia in rats.

In order to investigate the function of the auditory pathway from the cochlea to the brain stem under total spinal anesthesia, the auditory brain stem response (ABR), compound action potential of the cochlear nerve (CAP), and cochlear microphonics (CM) were simultaneously recorded in rats. Total spinal anesthesia was induced by infusion of 2% lidocaine hydrochloride at a constant rate of 0.10 mL/min into the cerebrospinal fluid through the rats' skulls. The ABR completely disappeared within 1.5 to 4 minutes. After cessation of the injection, the ABR reappeared, starting from wave I and progres

Hearing impairment in patients with acoustic neuroma--analysis by electrocochleography.

OBJECTIVE: In patients with acoustic neuroma, the site and severity of hearing impairment are important in discussing surgical approaches. Since the effectiveness of conventional auditory psychological testing is limited, we studied objectively hearing impairment of the cochlea and the cochlear nerve due to the tumor. METHODS: Electrocochleography (ECochG) was carried out in 21 patients with acoustic neuroma. Cochlear microphonic potential (CM) and action potential (AP) in ECochG evoked with clicks and short tone bursts were recorded through a transtympanic needle electrode technique. Cochlea

Maintenance of cochlear function with artificial oxygen carriers.

By means of vascular perfusion via the anterior inferior cerebellar artery with a blood substitute containing the perfluorochemical FC 47 as oxygen carrier, it is possible to maintain normal or near normal levels of the cochlear microphonics and the endolymphatic potential of the guinea pig for perios of 90 min, or longer. Following 60 min. of perfusion with artificial blood, the levels of ATP and 5' AMP in the stria vascularis and the organ of Corti are comparable to those of nonperfused control animals maintained at optimal metabolic conditions. Following the same period of perfusion, the ap

Rupture of Reissner's membrane during acute endolymphatic hydrops in the guinea pig: a model for Ménière's disease?

CONCLUSION: The changes in cochlear function during a destructive acute endolymphatic hydrops were relatively small. This might be consistent with the hypothesis that an endolymphatic hydrops is a marker of disordered inner ear homeostasis rather than the cause of the clinical symptoms of Ménière's disease. OBJECTIVE: Assessment of cochlear function during induction of a destructive acute endolymphatic hydrops. MATERIALS AND METHODS: During repetitive microinjections of 0.5 microl of artificial endolymph at a rate of 50 nl/s the 2f1-f2 and f2-f1 cochlear microphonics distortion products (CMD

Electrocochleography in deaf subjects.

Electrocochleography using the transtympanic electrode technique was performed on 86 deaf ears. Auditory nerve action potential was detected in 9 ears, in 5 of which deafness was caused by cerebellopontine angle tumor or surgery. A low threshold of the cochlear microphonics' response was also observed in some of the cases with deafness caused by mumps or cerebellopontine angle tumor. Various pathophysiological states of the inner ear can be observed, even in cases showing the same total deafness when measured by audiometer.

Disruption of cochlear potentials by chemical asphyxiants. Cyanide and carbon monoxide.

While ischemia, hypoxic hypoxia, and carbon monoxide (CO) have received extensive study designed to characterize mechanisms by which they disrupt cochlear function, little data are available concerning cyanide's potential to disrupt auditory function. In this study, disruption of the compound action potential (CAP) and endocochlear potential (EP) by cyanide and CO was compared in rats treated with potassium cyanide (KCN) (7 mg/kg ip), saline, CO (35 ml/kg ip), and air. Acute KCN administration significantly suppressed CAP and EP transiently. The effect was seen initially on EP with CAP impairm

Electrocochleography in auditory neuropathy.

Auditory neuropathy (AN) is a disorder characterized by the absence or the severe impairment of the auditory brainstem responses (ABRs) together with the preservation of otoacoustic emissions and/or cochlear microphonic (CM). We recorded transtympanic electrocochleography (ECohG) evoked by 0.1 ms clicks in one young adult and in four children having distortion product otoacoustic emissions and absent ABRs. In all but one patient CM and summating potential (SP) were present with normal threshold, and their amplitudes appeared comparable to or higher than the values obtained from subjects with n

Changes in distortion of two-tone cochlear microphonic and otoacoustic emission signals during an acute endolymphatic hydrops in the guinea pig.

An acute endolymphatic hydrops was induced by the injection of 1.1 mul of artificial endolymph into the scala media of guinea pig cochleas. This volume corresponds with an acute endolymphatic hydrops of 23%. During and after the injection, cochlear function was assessed by measuring the 2f(1)-f(2 )and f(2)-f(1) distortion products in cochlear microphonics (CMDP) and the 2f(1)-f(2) distortion product otoacoustic emission (DPOAE). A reversible pressure increase of 23 Pa and a relatively stable endocochlear potential (EP) were accompanied by a mean decrease in 2f(1)-f(2) DPOAE of only 3.4 dB. Sim
