In vitro filament-like formation upon interaction between lens alpha-crystallin and betaL-crystallin promoted by stress.

PURPOSE: To determine whether alpha-crystallin is capable of forming filament-like structures with other members of the crystallin family. METHODS: Water-soluble crystallins were isolated from calf lenses and fractionated into alpha-, betaH-, betaL-, and gamma-crystallins according to standard procedures. Chaperone-like activity of alpha-crystallin was determined in control and UV-A-irradiated lenses by the heat-induced aggregation assay of betaL-crystallin. Protein samples from this assay were analyzed by electron microscopy. In vitro filament formation was examined by transmission immunoele

Lens fiber cell differentiation and expression of crystallins in co-cultures of human fetal lens epithelial cells and fibroblasts.

Growth of the ocular lens is directed by the division and differentiation of a single layer of epithelial cells located at the equatorial region. It is conceivable that this region of the lens capsule presents a special microenvironment modulated by molecular cues emanating from the surrounding tissues. In an effort to investigate the source and nature of these molecular cues, we co-cultured human fetal lens epithelial cells and fibroblasts derived from the ciliary body. We observed morphological differentiation as evidenced by the appearance of differentiating lentoid structures associated wi

2-D electrophoresis distribution of stable 14C-glycation products from pig lens crystallins in relation to diabetic cataract formation.

We incubated pig lens crystallins with 14C-glucose and eliminated the unstable glycation products with posterior dialysis. All five soluble protein classes separable by Sepharose CL-6B column chromatography were radioactive, but the alpha-H fraction was five times more so than any of the others (14.2 vs. 3.1, 3.2, 1.6, and 2.3 x 10(3) cpm/mg protein, for alpha-H, alpha-L, beta-H, beta-L, and gamma proteins, respectively). However, the autoradiographs of our two-dimensional electrophoresis patterns revealed glycosyl adducts in all the soluble protein subunits except for gamma-a, and showed no e

A novel cataract-related mutation R10P in γA-crystallin increases susceptibility to thermal shock and ultraviolet radiation of γA-crystallin.

Congenital cataract is one of the most common causes of childhood blindness, typically resulting from genetic mutations. Over a hundred gene mutations associated with congenital cataract have been identified, with approximately half occurring in the Crystallin genes. In this study, we identified a novel γA-crystallin pathogenic mutation (c. 29G > C, p. Arg10Pro (R10P)), from a four-generation Chinese family with congenital cataract, and investigated its potential molecular mechanisms underlying congenital cataracts. We compared the protein structure and stability of purified the wild type (WT)

The mechanism for thermal-enhanced chaperone-like activity of α-crystallin against UV irradiation-induced aggregation of γD-crystallin.

Exposure to solar UV irradiation damages γ-crystallin, leading to cataract formation via aggregation. α-Crystallin, as a small heat shock protein, efficiently suppresses this irreversible aggregation by selectively binding the denatured γ-crystallin monomer. In this study, liquid chromatography tandem mass spectrometry was used to evaluate UV-325 nm irradiation-induced photodamage of human γD-crystallin in the presence of bovine α-crystallin, atomic force microscope (AFM) and dynamic light scattering (DLS) techniques were used to detect the quaternary structure changes of the α-crystallin olig

Polyamine Oxidase Is Involved in Spermidine Reduction of Transglutaminase Type 2-Catalyzed βH-Crystallins Polymerization in Calcium-Induced Experimental Cataract.

In an in vitro Ca2+-induced cataract model, the progression of opacification is paralleled by a rapid decrease of the endogenous levels of spermidine (SPD) and an increase of transglutaminase type 2 (TG2, EC 2.3.2.13)-catalyzed lens crystallins cross-linking by protein-bound N1-N8-bis(γ-glutamyl) SPD. This pattern was reversed adding exogenous SPD to the incubation resulting in a delayed loss of transparency of the rabbit lens. The present report shows evidence on the main incorporation of SPD by the catalytic activity of TG2, toward βH-crystallins and in particular to the βB2- and mostly in β

C3 glomerulonephritis with light chain crystalline podocytopathy and non-crystalline proximal tubulopathy: a case report and integrative clinicopathologic analysis.

This report describes a novel renal triad consisting of C3 glomerulonephritis (C3GN), light chain crystalline podocytopathy (LCCP), and non-crystalline light chain proximal tubulopathy (NC-LCPT) in a 41-year-old male presenting with nephrotic syndrome and rapidly progressive renal failure. Investigations revealed monoclonal IgG kappa paraproteinemia and isolated C3 hypocomplementemia. Renal histopathology demonstrated three distinct lesions: (1) C3GN with mesangial and capillary wall C3 deposits; (2) LCCP, featuring intralysosomal κ-restricted crystalline deposits in podocytes; and (3) NC-LCPT

[When the retina sparkles-Crystalline deposits in the retina].

BACKGROUND: Crystalline retinopathies are characterized by refractile deposits within different retinal layers and may be associated with genetic, systemic, toxic, or iatrogenic causes. Talc retinopathy represents a well-described toxic entity related to intravenous drug abuse. CASE PRESENTATION: A 44-year-old male presented with intermittent bilateral visual impairment over four weeks. Best-corrected visual acuity was 0.8 in both eyes. Funduscopic examination revealed multiple fine, glistening crystalline deposits in the central retina without signs of intraocular inflammation. Optical coher

A Crystalline Mono-Coordinate Indium(I)-Phosphaalkenyl.

The synthesis of the mono-coordinate indium(I) compound In[C(Ad)═PTer] (Ad = 1-adamantyl, Ter = 2,6-Dipp2-C6H3; Dipp = 2,6-diisopropylphenyl) is reported. Key to the formation of this monomeric species is the steric protection offered by the supporting phosphaalkenyl ligand which hinders aggregation. The title compound can be accessed from the reaction of (InTer)2 with the phosphaalkyne AdC≡P in an unusual redox-neutral transformation in which the In─C bond of the (InTer)2 precursor, known to dissociate in solution, adds across the C≡P triple bond of the phosphaalkyne. This insertion reaction

Heterogenous electromediated depolymerization of highly crystalline polyoxymethylene.

Post-consumer plastic waste in the environment has driven the scientific community to develop deconstruction methods that yield valued substances from these synthetic macromolecules. Electrocatalysis is a well-established method for achieving challenging transformations in small molecule synthesis. Here we present the first electro-chemical depolymerization of polyoxymethylene-a highly crystalline engineering thermoplastic (Delrin®)-into its repolymerizable monomer, formaldehyde/1,3,5-trioxane, under ambient conditions. We investigate this electrochemical deconstruction by employing solvent sc

Pressure induced topological and topological crystalline insulators.

Research on topological and topological crystalline insulators (TCIs) is one of the most intense and exciting topics due to its fascinating fundamental science and potential technological applications. Pressure (strain) is one potential pathway to induce the non-trivial topological phases in some topologically trivial (normal) insulating or semiconducting materials. In the last ten years, there have been substantial theoretical and experimental efforts from condensed-matter scientists to characterize and understand pressure-induced topological quantum phase transitions (TQPTs). In particular,

Pathological characteristics of light chain crystalline podocytopathy.

Monoclonal immunoglobulin light chain (LC) crystalline inclusions within podocytes are rare, poorly characterized entities. To provide more insight, we now present the first clinicopathologic series of LC crystalline podocytopathy (LCCP) encompassing 25 patients (68% male, median age 56 years). Most (80%) patients presented with proteinuria and chronic kidney disease, with nephrotic syndrome in 28%. Crystalline keratopathy and Fanconi syndrome were present in 22% and 10%, respectively. The hematologic condition was monoclonal gammopathy of renal significance (MGRS) in 55% and multiple myeloma

µ-Crystallin: A thyroid hormone binding protein.

µ-Crystallin is a NADPH-regulated thyroid hormone binding protein encoded by the CRYM gene in humans. It is primarily expressed in the brain, muscle, prostate, and kidney, where it binds thyroid hormones, which regulate metabolism and thermogenesis. It also acts as a ketimine reductase in the lysine degradation pathway when it is not bound to thyroid hormone. Mutations in CRYM can result in non-syndromic deafness, while its aberrant expression, predominantly in the brain but also in other tissues, has been associated with psychiatric, neuromuscular, and inflammatory diseases. CRYM expression i
