Abnormal metabolism in melanocytes participates in the activation of dendritic cell in halo nevus.

A comprehensive analysis of spatial transcriptomics was carried out to better understand the progress of halo nevus. We found that halo nevus was characterized by overactive immune responses, triggered by chemokines and dendritic cells (DCs), T cells, and macrophages. Consequently, we observed abnormal cell death, such as apoptosis and disulfidptosis in halo nevus, some were closely related to immunity. Interestingly, we identified aberrant metabolites such as uridine diphosphate glucose (UDP-G) within the halo nevus. UDP-G, accompanied by the infiltration of DCs and T cells, exhibited correla

INTERPLAY OF LNCRNAS, METABOLIC CELL DEATH, AND IMMUNE MICROENVIRONMENT IN GENITOURINARY MALIGNANCIES.

Genitourinary cancers, including prostate, bladder, and renal cancers, represent a significant global health burden due to their high prevalence and resistance to conventional therapies. A critical aspect of cancer progression is metabolic reprogramming, which not only fuels uncontrolled growth but also profoundly influences programmed cell death pathways and the tumor immune microenvironment. This review synthesizes current research on the intricate roles of long non-coding RNAs (lncRNAs) in modulating three emerging forms of regulated cell death - cuproptosis, ferroptosis, and disulfidptosis

Diabetic retinopathy from the perspective of programmed cell death: Focusing on apoptosis, pyroptosis, and necroptosis.

Diabetic retinopathy (DR), characterized by vascular damage and chronic inflammation, poses a significant threat to vision. Programmed cell death (PCD) plays a crucial role in DR pathogenesis, with apoptosis, pyroptosis, and necroptosis being extensively implicated in retinal cell loss. Current research lacks a systematic analysis of the temporal dynamics of these PCD modalities. This review focuses on apoptosis, pyroptosis, and necroptosis to elucidate their stage-specific contributions to DR progression. Apoptosis primarily occurs in the nonproliferative DR (NPDR) phase and drives early micr

Multiomic Landscape of Primary Hypothyroidism Induced by Subchronic Exposure to Low-Dose Novel PFOS Substitute OBS in Human and Murine Models.

Sodium p-perfluorous nonenoxybenzenesulfonate (OBS) as a novel surrogate for perfluorooctanesulfonate (PFOS) has been extensively utilized in industrial manufacturing and daily life. However, studies on OBS-induced environmental health risks of obstructive biosynthesis (OBS) are currently limited, particularly the risk for thyroid diseases. Following the construction of in vivo (mouse) and in vitro (normal human primary thyrocytes) models of subchronic low-dose OBS exposure, we explored the thyroid-disrupting effects of OBS through multiomics approaches and experimental validations. Our result

Novel Regulatory Cell Death in Intervertebral Disc Degeneration.

Intervertebral disc degeneration (IDD) is a major cause of chronic low back pain. Its pathogenesis is highly complex and encompasses multiple pathological processes, including cellular dysfunction, extracellular matrix (ECM) degradation, inflammatory responses, oxidative stress, and aberrant mechanical loading. Traditionally, programmed cell death (PCD), particularly apoptosis and autophagy, has been regarded as a key contributor to IDD. However, with ongoing advances in this field, an expanding number of emerging forms of regulated cell death (RCD) have been identified and demonstrated to pla

Regulated cell death in cancer: Mechanisms, crosstalk, and opportunities for therapy.

Regulated cell death (RCD) is a fundamental biological process essential for tissue homeostasis, and the elimination of damaged or malignant cells. In cancer, dysregulation of RCD is closely linked to tumor initiation, progression, therapeutic resistance, and remodeling of the tumor microenvironment (TME). In this review, we classify RCD in cancer into three broad groups. Classical cell death types include apoptosis, autophagy, necroptosis, and pyroptosis, which have well-established roles in controlling cell fate. Metal-dependent pathways, represented by ferroptosis, and cuproptosis, highligh

Vav3, a potential diagnostic and prognostic marker of diabetes, regulates glycolipid metabolism.

Vav3 can significantly affect the energy metabolism of cancer cells by changing the cytoskeletal structure, involving the mechanism of disulfidptosis, while affecting the function of glucose transporters, regulating cellular glucose and lipid metabolism. This provides an interesting research idea for the exploration of the pathogenesis of diabetes and the targeted regulation of glucose and lipid metabolism. Vav3 protein not only participates in β-cell damage in type 1 diabetes but also extensively regulates insulin release and glucose metabolism in type 2 diabetes. Vav3 proteins directly or in

From sound waves to systemic cure: transformative nanotechnology-driven sonodynamic Strategies in liver cancer.

Liver cancer is the third leading cause of cancer-related deaths worldwide, with hepatocellular carcinoma (HCC) accounting for nearly 75-85% of total cases. As per recent global cancer estimates, over 900,000 new cases and more than 800,000 deaths occur annually, reflecting its high incidence and poor survival outcome. Despite advancements in surgical, systemic, and immunotherapeutic approaches, treatment efficacy for advanced HCC remains limited due to tumour hypoxia, recurrence, drug resistance, and off-target toxicities. Sonodynamic therapy (SDT), a non-invasive, ultrasound-activated therap

Metal-sulfur metabolic derangement and regulated cell death in age-related cardiovascular diseases: Mechanisms, biomarkers, and emerging interventions.

Cardiovascular ageing is accompanied by progressive impairment of redox buffering, mitochondrial quality control, proteostasis and metal homeostasis. Iron and copper homeostasis, together with cystine-glutathione and thiol/disulfide redox metabolism, are increasingly recognized as interconnected determinants of metabolic vulnerability and regulated cell death (RCD) in age-related cardiovascular diseases. Ferroptosis, defined by iron-dependent lipid peroxidation and impaired anti-peroxidative defence, has a substantial evidence base in myocardial ischaemia/reperfusion injury, atherosclerosis, h

BAY-876 restores lenvatinib sensitivity in resistant hepatocellular carcinoma cells by disrupting the GLUT1-SLC7A11-NADPH redox axis.

BACKGROUND: Acquired resistance limits the efficacy of lenvatinib in hepatocellular carcinoma (HCC). This study evaluated whether GLUT1 inhibition by BAY-876 reverses lenvatinib resistance by disrupting SLC7A11-associated redox buffering. METHODS: Lenvatinib-resistant HuH-7-LR and Hep 3B-LR cells were generated by stepwise drug exposure. Drug sensitivity, combination synergy, glucose uptake, colony formation, migration, ATP, NADPH/NADP⁺ and GSH/GSSG ratios, intracellular thiols, ROS, apoptosis, gene and protein expression, N-acetyl-L-cysteine (NAC) rescue, and SLC7A11 knockdown were evaluated

A context-dependent METTL1-m7G-SLC7A11 axis links metabolic stress to epithelial fate in ulcerative colitis.

Ulcerative colitis (UC) is characterized by chronic intestinal inflammation accompanied by epithelial barrier dysfunction and profound metabolic stress; however, how metabolic cues are integrated to determine epithelial cell fate remains incompletely understood. Here, we identify a context-dependent METTL1-m7G-SLC7A11 regulatory axis that links metabolic stress to intestinal epithelial outcomes during UC progression. By integrating analyses of human UC tissues, DSS-induced acute and chronic colitis mouse models, and mechanistic in vitro experiments, we demonstrate that METTL1 enhances N7-methy

A multi-programmed cell death-related LncRNA signature for prognosis and immune microenvironment evaluation in kidney renal clear cell carcinoma.

Kidney renal clear cell carcinoma (KIRC) shows substantial prognostic heterogeneity, motivating robust transcriptome-based risk stratification. Ferroptosis, cuproptosis and disulfidptosis are distinct stress-related cell-death programs whose gene-expression patterns may capture diverse tumour states. Using TCGA-KIRC RNA-seq data (n=533), tumour samples were randomly split 1:1 into a training set (n=267) and an independent test set (n=266); In the training cohort, lncRNAs associated with curated genes for each programmed cell death (PCD) modality were screened using EPIC-adjusted partial Spearm

Crosstalk of thioredoxin system and programmed cell death: from pathophysiology to novel therapy.

The thioredoxin (Trx) system, an integral component of cellular redox regulation, preserves protein dithiol-disulfide equilibrium through its conserved Cys-Gly-Pro-Cys active site and is involved in key cellular functions, including cell proliferation, apoptosis, and signal transduction. In cancer biology, the thioredoxin system plays dual roles: overexpression can suppress oxidative stress and promote tumor growth, whereas dysfunction can trigger programmed cell death (PCD). However, a critical area for future research is to delineate how Trx modulates the intricate networks of PCD, and to id

Cuproptosis in Cerebrovascular Diseases: Mechanisms and Advances in Targeted Therapeutic Strategies.

Cuproptosis is a recently identified form of regulated cell death. It is primarily driven by abnormal mitochondrial copper accumulation. Ferredoxin 1-dependent copper reduction promotes the pathological aggregation of lipoylated proteins involved in the tricarboxylic acid cycle and the loss of iron-sulfur cluster proteins. This process induces proteotoxic stress, ultimately causing the collapse of mitochondrial energy metabolism. Evidence from transcriptomic analyses, single-cell multi-omics studies, and in vivo and in vitro models suggests that copper dyshomeostasis may be involved in cerebro

Mechanistic insights and therapeutic implications of mitoxyperilysis: a spatially gated mitochondria-dependent lytic cell death.

Programmed cell death (PCD) has expanded to more than ten diverse lytic modalities such as pyroptosis, necroptosis, ferroptosis, and disulfidptosis. In 2025, Wang et al. reported in Cell a previously unrecognized form of lytic cell death, termed mitoxyperilysis, in which plasma membrane rupture is driven not by canonical pore-forming or membrane-disrupting enzymatic effectors, but by mitochondria-mediated oxidative injury localized to the cell periphery. This process is initiated by the synergistic combination of innate immune activation and metabolic disruption, referred to as IIAMD, which in

Metal-dependent regulated cell death: Molecular architecture and translational frontiers.

Intracellular metal dyshomeostasis has emerged as a key regulator of specialized regulated cell death (RCD) programs, challenging classical views that regard necrosis as entirely accidental. This review systematically delineates the molecular architecture and translational trajectories underlying metal-dependent RCD, including iron-driven ferroptosis, copper-mediated cuproptosis, and additional emerging modalities such as calcicoptosis, necrosis by sodium overload (NECSO), and the newly designated zincoptosis, mnoptosis, and coptosis. We examined distinct execution mechanisms, ranging from mem

Bioinformatics analysis of signature genes related to cell death in keratoconus.

Keratoconus is corneal disease in which the progression of conical dilation of cornea leads to reduced visual acuity and even corneal perforation. However, the etiology mechanism of keratoconus is still unclear. This study aims to identify the signature genes related to cell death in keratoconus and examine the function of these genes. A dataset of keratoconus from the GEO database was analysed to identify the differentially expressed genes (DEGs). A total of 3558 DEGs were screened from GSE151631. The results of Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) analysis sh
