Prevalence of borreliae in ixodid ticks from a floodplain forest ecosystem.

Host-seeking ixodid ticks were sampled in a floodplain forest ecosystem along the lower reaches of the Thaya (Dyje) river in South Moravia (Czech Republic) and Lower Austria during the period 1989-2002. The ticks were examined by dark-field microscopy for borreliae (Borrelia burgdorferi sensu lato, the agent of Lyme borreliosis), and attempts were made to culture the spirochetes in BSK-H medium from preparations containing their high numbers. Isolated borreliae were identified by PCR-RFLP analysis using probes directed against ribosomal spacer genes. A total of 797 nymphal and 719 adult (391 f

Social ecosystem health: confronting the complexity and emergence of infectious diseases.

The emergence and re-emergence of infectious diseases and their rapid dissemination worldwide are challenging national health systems, particularly in developing countries affected by extreme poverty and environmental degradation. The expectations that new vaccines and drugs and global surveillance would help reverse these trends have been frustrated thus far by the complexity of the epidemiological transition, despite promising prospects for the near future in biomolecular research and genetic engineering. This impasse raises crucial issues concerning conceptual frameworks supporting priority

Emerging infectious diseases in an island ecosystem: the New Zealand perspective.

Several unique features characterize infectious disease epidemiology in New Zealand. Historically, well-organized, government-run control programs have eliminated several zoonoses. More recently, however, communicable disease control has been mixed. Rates of rheumatic fever, tuberculosis, and enteric infectious are high, and rates of meningococcal disease are increasing. These diseases are over-represented in New Zealanders of Polynesian descent, who generally live in more deprived and overcrowded conditions than do those of European descent. Measles and pertussis epidemics are recurring becau

Advancing Equity for Global Health Author Teams within the Publishing Ecosystem.

The global health literature reflects the need to restructure systems that balance power, remove financial barriers, and ultimately enhance equity, but immediate strategies to enhance equity are often underappreciated, particularly with regard to navigating the publishing ecosystem. Our paper describes how researchers can navigate the publishing ecosystem to create greater equity within their author teams, including those related to communication, supporting junior investigators in building their writing and professional skills, selecting journals that prioritize equity, ensuring transparent r

The floor, not the ceiling: industry ties across the cancer ecosystem.

Industry financial ties to the US cancer ecosystem are everywhere. Kim et al.'s systematic review and meta-analysis finds that financial conflicts of interest exceed 50% prevalence across most oncology domains. These relationships are not always benign. A robust body of health services research, including a 2021 systematic review of 36 studies, demonstrates that 83% of analyses identify a positive association between industry payments and prescribing behavior. Even a single industry-sponsored meal costing <$20 has been linked to significantly higher odds of prescribing the promoted drug. As fe

MEDS - An Emerging Data Standard and Ecosystem for Health AI Research.

While data standards have been well adopted and highly impactful for observational health informatics, the emerging application of artificial intelligence (AI) to electronic health record (EHR) data - known broadly as health AI - still lacks broadly adopted data standards. This gap limits reproducibility and the development of open-source ecosystems for health AI, as well as limiting the ability to collaboratively develop and characterize emerging capabilities such as the use of foundation models. The Medical Event Data Standard (MEDS) is an open- source data standard and ecosystem designed an

Symphony of Digestion: Coordinated Host-Microbiome Enzymatic Interplay in Gut Ecosystem.

Digestion was once viewed as a host-driven process, dependent on salivary, gastric, pancreatic, and intestinal enzymes to break down macronutrients. However, new insights into the gut microbiota have redefined this view, highlighting digestion as a cooperative effort between host and microbial enzymes. Host enzymes initiate nutrient breakdown, while microbial enzymes, especially in the colon, extend this process by fermenting resistant polysaccharides, modifying bile acids, and transforming phytochemicals and xenobiotics into bioactive compounds. These microbial actions produce metabolites lik

Unveiling the role of phages in shaping the periodontal microbial ecosystem.

The oral microbiome comprises various species and plays a crucial role in maintaining the oral ecosystem and host health. Phages are an important component of the periodontal microbiome, yet our understanding of periodontal phages remains limited. Here, we investigated oral periodontal phages using various advanced bioinformatics tools based on genomes of key periodontitis pathogens. Prophages were found to encode various auxiliary genes that potentially enhance host survival and pathogenicity, including genes involved in carbohydrate metabolism, antibiotic resistance, and immune modulation. Direct and indirect ecosystem responses to vehicle compaction of soft sediments.

Change in an ecological community after a disturbance may be a direct impact or may be indirectly mediated through the response of a highly influential species. In northeastern Pacific tidal flats, a native bioturbator, ghost shrimp Neotrypaea californiensis, engineers soft-sediment habitat and interacts antagonistically with bivalve shellfish. Vehicle compaction has been used in pest control of ghost shrimp, but this disturbance lacks quantitative evidence of its efficacy and environmental impacts. Through three large (∼10 ha) experiments in Grays Harbor, Washington, USA, we tested the direct

Continued permafrost ecosystem carbon loss under net-zero and negative emissions.

The loss of ecosystem carbon (the sum of vegetation, litter, and soil carbon) may occur in a permafrost region under mitigation pathways, which could reduce the efficiency of carbon dioxide removal. Here, we investigate changes in permafrost under net-zero and negative emissions, based on idealized emission-driven simulations using a state-of-the-art Earth system model. While acting as a net ecosystem carbon sink during most of the positive emission phase, permafrost becomes a net ecosystem carbon source just before reaching net-zero and negative emissions. Permafrost slowly recovers, especial

Tire wear particles in aquatic environments: From biota to ecosystem impacts.

Tire wear particles (TWPs), mainly generated through friction between tires and road surfaces, represent a major source of traffic-related microplastic pollution, posing threats to biota and ecosystem functions. These particles are a complex mixture of toxic compounds, including heavy metals (e.g., zinc) and organic compounds (e.g., 6-PPD), and their diverse leachates exacerbate their ecological impacts. This review collates current knowledge on the occurrence of TWPs and their leachates in aquatic systems, emphasizing their toxicological effects on species and cascading ecological consequence

Microbiota Ecosystem in Recurrent Cystitis and the Immunological Microenvironment of Urothelium.

Urinary tract infections (UTIs) represent one of the most frequent low genital tract diseases in the female population. When UTIs occur with a frequency of at least three times per year or two times in the last six month, we speak of recurrent UTI (rUTI) and up to 70% of women will have rUTI within 1 year. It was previously thought that antibiotic resistance was principally responsible for the recurrence of UTIs, but nowadays new diagnostic technologies have shown the role of microbiota in the pathophysiology of these diseases. Much research has been conducted on the role of gut microbiome in

Mobilizing the clinical trial ecosystem to drive adoption of master protocols.

Master protocol studies typically use an overarching protocol to answer several questions by guiding a variety of sub-studies. These sub-studies can incorporate multiple diseases, therapies, or both. Although this innovative approach offers many benefits, including the ability to deliver clinical research that is more patient-centric and efficient, several common barriers curtail widespread adoption. The Clinical Trials Transformation Initiative (CTTI) convened industry representatives, regulatory agencies, patient groups, and academic institutions to identify emerging best practices and devel

Methanotrophy by a Mycobacterium species that dominates a cave microbial ecosystem.

So far, only members of the bacterial phyla Proteobacteria and Verrucomicrobia are known to grow methanotrophically under aerobic conditions. Here we report that this metabolic trait is also observed within the Actinobacteria. We enriched and cultivated a methanotrophic Mycobacterium from an extremely acidic biofilm growing on a cave wall at a gaseous chemocline interface between volcanic gases and the Earth's atmosphere. This Mycobacterium, for which we propose the name Candidatus Mycobacterium methanotrophicum, is closely related to well-known obligate pathogens such as M. tuberculosis and M

The child ecosystem and childhood pulmonary tuberculosis: A South African perspective.

INTRODUCTION: This study investigates drivers of childhood pulmonary tuberculosis (PTB) using a childhood ecosystem approach in South Africa. An ecosystem approach toward identifying risk factors for PTB may identify targeted interventions. METHODS: Data were collected as part of a prospective cohort study of children presenting at a primary care facility or tertiary hospital with possible TB. Characterization of the childhood ecosystem included proximal, medial, and distal determinants. Proximal determinants included child characteristics that could impact PTB outcomes. Medial determinants i

Dissecting esophageal squamous-cell carcinoma ecosystem by single-cell transcriptomic analysis.

Esophageal squamous-cell carcinoma (ESCC), one of the most prevalent and lethal malignant disease, has a complex but unknown tumor ecosystem. Here, we investigate the composition of ESCC tumors based on 208,659 single-cell transcriptomes derived from 60 individuals. We identify 8 common expression programs from malignant epithelial cells and discover 42 cell types, including 26 immune cell and 16 nonimmune stromal cell subtypes in the tumor microenvironment (TME), and analyse the interactions between cancer cells and other cells and the interactions among different cell types in the TME. A Novel Arthropod Host of Brucellosis in the Arid Steppe Ecosystem.

Brucellosis is a severe public health problem in the Inner Mongolia regions of China. The recent prevalence of brucellosis outbreaks may be attributed to an increase in the activity of ticks and other air-borne vectors. Dermacentor nuttalli (D. nuttalli) is a native tick species of Inner Mongolia; similar to other tick species, D. nuttalli carries a variety of pathogens that can be transmitted to a wide range of animals. In this study, we have investigated the potential of D. nuttalli in transmitting brucellosis. From 2015 to 2019, 2,256 ticks were collected from 23 different pastoral areas of

AMPK May Play an Important Role in the Retinal Metabolic Ecosystem.

Evidence suggests that metabolic dysregulation plays an important role in disease etiology of retinal degenerations. Several studies suggest that preserving the retinal metabolic ecosystem may be protective against retinal degenerations. We investigated whether activation of 5' adenosine monophosphate protein kinase (AMPK) is protective to the retina in several preclinical mouse models of retinal degeneration and found that metformin-induced activation of AMPK was able to delay or prevent retinal degeneration in the rd10 model of retinitis pigmentosa, the NaIO3 model of RPE and retinal injury,

Optimal control of irrupting pest populations in a climate-driven ecosystem.

Irruptions of small consumer populations, driven by pulsed resources, can lead to adverse effects including the decline of indigenous species or increased disease spread. Broad-scale pest management to combat such effects benefits from forecasting of irruptions and an assessment of the optimal control conditions for minimising consumer abundance. We use a climate-based consumer-resource model to predict irruptions of a pest species (Mus musculus) population in response to masting (episodic synchronous seed production) and extend this model to account for broad-scale pest control of mice using
