The fate of glycogen in granular tubule cells of rat submandibular glands during secretory events.

Glycogen, studied electron microscopically in granular tubule cells by means of the periodic acid-thiocarbohydrazide-silver proteinase technique, was found to be scattered abundantly throughout the cytoplasm. Parasympathetic nerve stimulation caused no detectable change in the glycogen. Degranulation of the granular tubule cells after either sympathetic nerve stimulation or cyclocytidine injection caused loss of the glycogen from the cells. Study of tubule cells undergoing secretion in the early stages after cyclocytidine injection indicated that glycogenolysis was occurring. Glycogen had reac

Decrease of liver glycogen content in golden hamsters infected with plerocercoids of Spirometra erinacei.

Liver glycogen content decreased in golden hamsters experimentally infected with plerocercoids of Spirometra erinacei. The activity of glycogen synthase a decreased significantly in infected animals, whereas that of glycogen phosphorylase a was not significantly affected. These observations suggest that changes in glycogen content were not attributable to increased glycogenolysis, but rather resulted from suppressed glycogenesis. Plasma immunoreactive insulin (IRI) concentrations in infected animals were slightly lower than those in controls, but the differences were not statistically signific

Porcine malignant hyperthermia. VII: Hepatic metabolism.

Hepatic metabolism during porcine malignant hyperthermia (MH) was investigated in seven Pietrain pigs. The estimated hepatic blood flow decreased during MH, but an increase in oxygen extraction enabled the splanchnic oxygen uptake to be maintained. There was a large release of glucose and potassium from the liver in MH which made an important contribution to the hyperglycaemia and hyperkalaemia. Measurement of hepatic uptake of the precursors of gluconeogenesis, lactate, glycerol and alanine, showed that glucose efflux from liver was derived mainly from glycogenolysis. Lactate uptake by the li

Glycogen synthase (GYS1) mutation causes a novel skeletal muscle glycogenosis.

Polysaccharide storage myopathy (PSSM) is a novel glycogenosis in horses characterized by abnormal glycogen accumulation in skeletal muscle and muscle damage with exertion. It is unlike glycogen storage diseases resulting from known defects in glycogenolysis, glycolysis, and glycogen synthesis that have been described in humans and domestic animals. A genome-wide association identified GYS1, encoding skeletal muscle glycogen synthase (GS), as a candidate gene for PSSM. DNA sequence analysis revealed a mutation resulting in an arginine-to-histidine substitution in a highly conserved region of G

GSK3 involvement in amylin signaling in isolated rat soleus muscle.

Amylin can evoke insulin resistance by antagonizing insulin in a non-competitive manner. Here, we investigated the glycogenolytic effect of amylin in isolated skeletal muscle and compared it to the effects of a calcitonin gene-related peptide (CGRP). Amylin alone had no statistically significant effect on glucose transport. However, amylin decreased insulin-stimulated glucose transport by about 30%. The involvement of cAMP could not be detected at the concentrations shown to promote glycogenolysis. Previously, it has been shown that increased glycogen synthase kinase 3 (GSK3) activity plays a

Enzymatic characterization of four new mutations in the glucose-6 phosphatase (G6PC) gene which cause glycogen storage disease type 1a.

Glycogen storage disease type 1a (GSD1a) is caused by mutations in the gene of glucose-6 phosphatase (G6PC), encoding the last enzyme of gluconeogenesis and glycogenolysis. To study the effect of mutations previously identified, but not yet enzymatically characterized, in French GSD1a patients, we used an in vitro expression system of the human glucose-6 phosphatase (hGlc6Pase) cDNA. Wild type hGlc6Pase expressed in COS-7 cells exhibited kinetic features comparable to microsomal Glc6Pase from normal human liver and kidney. Four new mutations inducing aminoacid changes in the coding sequence, e

Fetal metabolic response to phloridzin-induced hypoglycemia in pregnant rats.

Phloridzin, an inhibitor of renal sugar transport, produces an important loss of glucose in urine of treated animals. In order to reduce severely the maternal glucose supply to the fetuses in short-term experiments, we have combined phloridzin administration to pregnant rats with 18 h starvation. Fetuses from starved phloridzin-treated mothers were compared with fetuses from starved mothers. Combined treatment markedly decreases fetal blood glucose concentration (-36%) and fetal liver glycogen stores (-76%). These changes are associated with a decrease in plasma insulin (-25%), a rise in plasm

Effects in skeletal muscle.

The first biological action of amylin to be described was the inhibition of insulin-stimulated incorporation of radiolabeled glucose into glycogen in the isolated soleus muscle of the rat. This antagonism of insulin action in muscle was non-competitive, occurring with equal potency and efficacy at all insulin concentrations. Amylin inhibited activation of glycogen synthase, partially accounting for the inhibition of radiolabeled glucose incorporation. However, this did not account for a low rate of labeling at higher amylin concentrations, wherein the radioglycogen accumulation was even less t

Glycogen storage disease type Ia (GSD Ia) during pregnancy: report of a case complicated by fetal growth restriction and preeclampsia.

Glycogen storage disease type Ia (GSD Ia) leads to disturbed glycogenolysis and gluconeogenesis due to a deficiency in the enzyme glucose-6-phosphatase. A patient with GSD Ia showed hypoglycemia and proteinuria without dietary management since early pregnancy. The patient's condition was complicated by hypertension with increase in proteinuria at 22 weeks of gestation. In spite of administration of antihypertensive drugs and dietary management, the disease became more severe with deterioration in the fetal status and inhibition of fetal growth. Thus, a cesarean section was performed at 26 week

[Metabolic effect of coffee and caffeine].

Coffee and its most important constituent, caffeine, may not only stimulate the function of many organs but also increase the metabolism in the body. These effects require a higher energy production which is mainly obtained from striated muscles by glycogenolysis and from fat tissue by lipolysis. Sutherland and Butcher were able to demonstrate that these degradation processes are primarily caused by an increase of cyclic 3,5-AMP. - In this connection caffeine and other methylxanthines are of special interest because these compounds also increase the intracellular amount of cyclic 3,5-AMP. This

McArdle disease and carbohydrate ingestion before exercise: Timing on exercise tolerance, clinical relevance, and application to real world settings.

PURPOSE: McArdle disease (MD) is a rare (1:100000) genetic myopathy whereby the enzyme glycogen phosphorylase is not expressed in skeletal muscle, resulting in the blockage of glycogenolysis and presenting as physical activity intolerance. To circumvent the blockage in glycogenolysis, current clinical practice guidelines advise individuals with MD (IWMD) to ingest 37 g of sucrose 5-10 minute (min) before undertaking exercise. METHODS: The current investigation undertook randomized repeated measure clinical exercise trials on five IWMD to compare exercise tolerance under different timing of

Interaction of free fatty acids and epinephrine in regulating hepatic glucose production in conscious dogs.

To determine the effects of an increase in lipolysis on the glycogenolytic effect of epinephrine (EPI), the catecholamine was infused portally into 18-h-fasted conscious dogs maintained on a pancreatic clamp in the presence [portal (Po)-EPI+FFA, n = 6] and absence (Po-EPI+SAL, n = 6) of peripheral Intralipid infusion. Control groups with high glucose (70% increase) and free fatty acid (FFA; 200% increase; HG+FFA, n = 6) and high glucose alone (HG+SAL, n = 6) were also included. Hepatic sinusoidal EPI levels were elevated (Delta 568 +/- 77 and Delta 527 +/- 37 pg/ml, respectively) in Po-EPI+SAL

A Case of Glycogen Storage Disease Type 1a Mimicking Familial Chylomicronemia Syndrome.

Glycogen storage disease type 1a (GSD1a) is an autosomal recessively inherited inborn error of metabolism caused by a mutation in the G6PC gene, which encodes the catalytic subunit of glucose-6-phosphatase-α (G6Pase-α) enzyme. This enzyme plays a role in the final step of gluconeogenesis and glycogenolysis. Patients carrying GSD1a show growth retardation, hypoglycemia, hepatomegaly, hepatic steatosis, hyperlipidemia, hyperuricemia and lactic acidemia. Long-term symptoms include gouty arthritis and uric acid stones, osteoporosis, renal failure, intestinal impairment, cirrhosis and hepatic adeno

Management of Diabetic Ketoacidosis in Children and Adolescents with Type 1 Diabetes Mellitus.

Diabetic ketoacidosis (DKA) is the end result of insulin deficiency in type 1 diabetes mellitus (T1D). Loss of insulin production leads to profound catabolism with increased gluconeogenesis, glycogenolysis, lipolysis, and muscle proteolysis causing hyperglycemia and osmotic diuresis. High levels of counter-regulatory hormones lead to enhanced ketogenesis and the release of 'ketone bodies' into the circulation, which dissociate to release hydrogen ions and cause an overwhelming acidosis. Dehydration, hyperglycemia, and ketoacidosis are the hallmarks of this condition. Treatment is effective rep

Exercise and Preexercise Nutrition as Treatment for McArdle Disease.

McArdle disease is due to an inborn defect in the muscle isoform of glycogen phosphorylase (or "myophosphorylase"), the enzyme that catalyzes the first step of glycogenolysis. This condition is still not fully understood, and although advances in research would help patients immeasurably, these would also enhance our understanding of exercise metabolism. It has been 10 yr since the first published report demonstrating the benefits of regular aerobic exercise for these patients. However, misconceptions remain and the value of exercise prescription for patients with McArdle disease is still over

Involvement of endocrine system in a patient affected by glycogen storage disease 1b: speculation on the role of autoimmunity.

Glycogen storage disease type 1b (GSD1b) is an inherited metabolic defect of glycogenolysis and gluconeogenesis due to mutations of the SLC37A4 gene and to defective transport of glucose-6-phosphate. The clinical presentation of GSD1b is characterized by hepatomegaly, failure to thrive, fasting hypoglycemia, and dyslipidemia. Patients affected by GSD1b also show neutropenia and/or neutrophil dysfunction that cause increased susceptibility to recurrent bacterial infections. GSD1b patients are also at risk for inflammatory bowel disease. Occasional reports suggesting an increased risk of autoimm

Metabolic effects of the delay in obliteration of the umbilical cord in the newborn rat.

The effect of delay in obliteration of the umbilical cord on the time-courses of liver glycogen, blood glucose and lactate concentrations and blood PO2, PCO2 and pH during the first 6 h of extrauterine life were studied. Obliteration of the umbilical cord 2 h after delivery resulted in an increase in liver glycogenolysis without significantly affecting the other parameters studied. A 6-hour delay in obliteration of the umbilical cord increased lactiacidemia and decreased blood PO2 and pH without significantly affecting the other parameters studied. A supply of pure oxygen to the newborns decre

Glucose-resistant hypoglycemia in inanition.

Three patients had starvation-related hypoglycemia that was not responsive to the infusion of glucose. The patients resembled fatal cases of hypoglycemia seen very rarely in victims of kwashiorkor. None of the patients had a history of drug ingestion, nor was there any evidence of endocrine insufficiency. Two of the patients had low serum insulin levels. Complete postmortem examinations were done for all three patients and there was no evidence of islet cell abnormalities or of malignant neoplasms. A syndrome of severe hypoglycemia refractory to therapy, that was occasionally seen, in years pa

Influence of mercuric chloride induced uremia on intramolecular structure of muscle glycogen in rats.

Long term maintenance of male rats of Long Evans variety following mercuric chloride induced uremia has shown an altered intramolecular structure of muscle glycogen with no significant difference in muscle glycogen content. The structure of muscle glycogen in these uremic rats when compared to sham control rats differed significantly from the structure of liver glycogen observed earlier in nephrectomized rats (Mannan et al., 1975). Biochemical analysis of muscle glycogen revealed a significant inverse relationship between the muscle and liver glycogen structure in terms of number of segments a

Beyond energy storage: roles of glycogen metabolism in health and disease.

Beyond storing and supplying energy in the liver and muscles, glycogen also plays critical roles in cell differentiation, signaling, redox regulation, and stemness under various physiological and pathophysiological conditions. Such versatile functions have been revealed by various forms of glycogen storage diseases. Here, we outline the source of carbon flux in glycogen metabolism and discuss how glycogen metabolism guides CD8+ T-cell memory formation and maintenance. Likewise, we review how this affects macrophage polarization and inflammatory responses. Furthermore, we dissect how glycogen m
