The influence of cyclobarbital and diazepam on drug metabolism in vitro and their binding to cytochrome P-450.

Cyclobarbital inhibited diazepam metabolism by rat liver supernatant only at concentrations of 1 mM. Ethylmorphine N-demethylation is strongly inhibited by diazepam in a non-competitive manner whereas cyclobarbital shows a competitive inhibition type. Both cyclobarbital and oxazepam are weak inhibitors. Hexobarbital- evokes typical type I spectral changes with rat liver microsomes whereas cyclobarbital and diazepam-induced spectral changes are similar to type II or inverse type I. Barbital evoked the same type of spectral changes as diazepam and cyclobarbital. With the usual concentration in v

[The dissimilar effects of the single and prolonged administration of melatonin on the hypnogenic action of hexenal in mice in relation to the dose and the time of day].

At the acute administration the pineal hormone melatonin depending on the dose (0.01-10 mg/kg) potentiated the hypnogenic effect of hexenal (hexobarbital sodium) in mice both in the daytime and at night. When low doses of the hormone were chronically administered in the light phase of the day, there was noted a decrease of the duration of barbiturate anesthesia. A similar result was found during its round-the-clock evaluation against the background of 0.1 mg/kg of melatonin (a single dose) in the late daytime hours and on the contrary a sharp enhancement of hexenal action before switching on t

Effect of Chinese herbal medicine "kanbaku-taiso-to" on neuropharmacological tests.

The "Kanbaku-taiso-to" (KT), mixture of three herbal drugs, Glycyrrhizae Radix, Tritici Semen and Zizyphi Fructus, sometimes shows a marked effect on insomnia and infantile convulsions. The ancient Chinese medical book "Kinkiyoryaku" described the effectiveness of KT on emotional irritability. To elucidate the mechanism of action of KT, neuropharmacological screening tests were performed and the following results were obtained. KT lengthened the hexobarbital sleeping time; KT showed a marked prolongation of time to death in pentylenetetrazole (PTZ)-induced convulsions; locomotor activity was i

Stimulatory effect of vegetables on intestinal drug metabolism in the rat.

The intestinal metabolism of hexobarbital, phenacetin, 7-ethoxycoumarin and benzo [a] pyrene in vitro was increased in rats fed either dried Brussels sprouts or dried cabbage in a nutritionally complete semisynthetic diet as compared to rats fed only the semisynthetic diet. Pretreatment of rats with several indoles present in Brussels sprouts and cabbage also stimulated intestinal drug metabolism, but the effect was smaller than when dried Brussels sprouts or dried cabbage was fed. The results obtained suggest a need for studies in man to determine whether vegetables and other dietary constitu

Central action of drugs acting on the cholinergic muscarinic receptor. II. Influence of ecothiopate administered into the lateral cerebral ventricle on behavior in rats.

Ecothiopate (Etp), 10 mug injected into the lateral cerebral ventricle (ivtr), diminished spontaneous motility and open-field performance, lowered body temperature, inhibited amphetamine-induced hypermotility and stereotypy, prolonged hexobarbital anesthesia and did not induce catalepsy. These behavioral symptoms were accompanied by lowered level of NA and elevated levels of 5-HT and 5-HIAA in the brain. No change in levels of DA were noted. Smaller doses (0-1 and 1 mug) had virtually no influence on behavior of the animals. The dose of 1 mug caused a rise in levels of 5-HIAA in the brain, but

A comparison of the potencies of a series of barbiturates at the neuromuscular junction and on the central nervous system.

The ability of a series of barbiturates to depress the depolarizing action of carbachol at the end-plate of guinea-pig lumbrical muscle was studied. The compounds studied were: amorbarbital, aprobarbital, barbital, barbituric acid, butabarbital, butalbital, dimethylbutylethyl barbituric acid, hexobarbital, mephobarbitak, secobarbital, thiamylal, and thiopental. The depressant activity was sensitive to small changes in structure of the compounds strongly suggesting that a specific receptor site was involved in the interaction of the drug with the tissue. The observed relative potencies on the m

STRESS MODIFICATION OF DRUG RESPONSE.

Rats stressed by unilateral hind-leg ligation show a significantly decreased pharmacological response to the effects of hexobarbital, meprobamate, and pentobarbital. This effect is dependent upon an intact pituitaryadrenal axis, since it is absent in both hypophysectomized and adrenalectomized animals. It can be simulated by intravenous administration of adrenocorticotrophic hormone or corticosterone. The effect of stress on drug response is not noted with barbital, a compound which is not metabolized; this effect is inhibited by treatment of the animal with a drug metabolism inhibitor such as

Cardiovascular effects of two potential beta-hydroxylase inhibitors.

The purpose of this study was to evaluate the cardiovascular effects of two potential beta-hydroxylase inhibitors, a dithiocarbamate and a thiourea derivative of d-amphetamine, 1 and 2, respectively. These compounds when given intravenously elicited a significant dose-dependent depressor effect in normotensive rats and dogs anesthetized with chloralose-urethane. The hypotensive effect of these compounds appears to be the result of a combination of two action components: (a) the major component, which can be blocked by atropine, exerted through the parasympathetic division and (b) a decreased a

Pharmacodynamics and pharmacokinetics of some hypnotic drugs in spontaneously hypertensive rats (SHR).

Hypnotic action of barbiturates (hexobaribtal, cyclobarbital-Ca, phenobarbital, barbital) chloral hydrate, glutethimide was investigated in spontaneously hypertensive Okamoto-Aoki rats (SHR). Hypnotic effect of investigated drugs was decreased in hypertensive rats comparing with normotensive animals. The decrease in the hypnotic effect was related to the increase of the threshold hypnotic dose. No changes were observed in the pharmacokinetic parameters of hexobarbital and barbital and in the rate of crossing of blood-brain barrier by the latter drug in SHR rats. The results suggested that the

Central effects of AC-1 and TFG-1.

AC-1 and TFG-1 are both herb extracts from Avena Sativa and Trigonella Foenum Graecum respectively. In folk medicine, Trigonella Foenum Graecum is used as appetiser and general tonic as well as for pellagra treatment and for treatment of pulmonary disorders. Avena Sativa is another commonly used appetiser and tonic. In our study we examined the influence of AC-1 and TFG-1 on some of the major functions of the central nervous system. We divided the test animals in three groups and after administering the herb extracts we monitored different pharmacological parameters-Phenamine toxicity and ster

Pentobarbital quantification in the presence of phenobarbital by fluorescence polarization immunoassay.

We developed a method for measuring pentobarbital in samples that also contain phenobarbital. The phenobarbital is destroyed by adding sodium hydroxide and then heating at 95 degrees C for 60 min. Pentobarbital is not affected by this pretreatment and can then be measured with an Abbott TDx barbiturates kit. Using this method, we obtained an average analytical recovery of 98% of added pentobarbital and a correlation of y = 0.953x + 3.4 vs HPLC. The intra-assay CV was 3.7% at 25.8 mg/L; the interassay CV was 6.2% at 16.1 mg/L. Other long-acting barbiturates, e.g., hexobarbital, are also effecti

Normal levels of hepatic drug-metabolizing enzymes in neonatally induced, growth hormone-deficient adult male and female rats.

The effects of chronic growth hormone deficiency on the growth hormone-dependent, sexually dimorphic hepatic drug-metabolizing enzymes were studied in adult rats of both sexes. Neonatal administration of monosodium L-glutamate produced the expected syndrome characterized by stunted growth, obesity, prolonged vaginal estrus, and an inhibition in the growth of the pituitaries, adrenals, gonads, sexual accessory organs, and kidneys. Associated with these abnormalities was a 90% reduction in the concentration of serum growth hormone in male and female rats. In contrast, the activities of hepatic a

Age-dependent enhancement of diazepam sensitivity is accelerated in New Zealand Black mice.

Separate age groups of C57BL/6 and autoimmune New Zealand Black (NZB) mice were compared for diazepam-induced ataxia and barbiturate-induced loss of righting reflex. Between 1 and 3 months of age, both strains showed a similar age-related decrease in ED50 for diazepam-induced ataxia. However, between 3 and 12 months the decrease in ED50 was markedly greater in NZB mice. In contrast, age-related increases in the durations of loss of righting reflex following hexobarbital or barbital were similar in both strains. The results suggest that NZB mice show relatively accelerated age-related increases

Barbiturate and picrotoxin-sensitive chloride efflux in rat cerebral cortical synaptoneurosomes.

The effects of various barbiturates and picrotoxin in modifying the efflux of chloride (36Cl-) was studied in a novel subcellular preparation from rat cerebral cortex, the 'synaptoneurosome'. Dilution of synaptoneurosomes pre-loaded with 36Cl- resulted in rapid efflux of 36Cl- that could be measured as early as 10 s following dilution. In the presence of barbiturates such as pentobarbital and hexobarbital there was a significant increase in 36Cl- efflux which was not observed with the pharmacologically-inactive barbiturate, barbital. The effect of barbiturates in enhancing 36Cl- efflux was als

[The inhibition of lidocaine metabolism by various barbiturates in rat hepatic microsome].

To evaluate the effects of various barbiturates on lidocaine metabolism by cytochrome P-450 (P-450), enzyme kinetics were analyzed in an in vitro study using rat hepatic microsomes. Phenobarbital, amobarbital, hexobarbital, pentobarbital, and thiamylal showed the mixed type inhibition of lidocaine metabolism with inhibition constants being 4.89, 1.08, 2.76, 0.77 and 0.65 mM, respectively. Same as lidocaine, all barbiturates used in the present study, corresponding to binding with P-450, induced the I type of spectral change of P-450. Since these did not affect cytochrome C reductase activity,

[Effect of the diethylamide of nicotinic acid (cordiamine) on the hydroxylating function of the liver].

Diethylamide of nicotinic acid (subcutaneously, 40 and 120 mg/kg, once a day for 4 days) was shown to exert no influence on p-hydroxylation of aniline and to increase the rate of N-demethylation of amidopyrine and ethylmorphine in the rat liver microsomal fraction by 21 and 47% as compared with the control. At the same dose of 40 mg/kg and the same schedule of administration the drug was found to increase urine excretion of antipyrine metabolites: nor-antipyrine, 4-hydroxy-antipyrine and the sum of metabolites by 229, 89 and 80%, respectively, during the first 90 min of the experiment. [Changes in testosterone oxidation in rat liver microsomes after prolonged administration of ethylmorphine].

Ethylmorphine administration to male rats during 3 weeks markedly decreased the cytochrome P-450 content and the rate of oxidation of ethylmorphine, hexobarbital and testosterone in liver microsomes. The decrease of the substrate oxidation rate was correlated with the decrease in the maximal spectral changes induced by the substrate binding to cytochrome P-450. Calculation of the substrate oxidation and binding data per mole of cytochrome P-450 showed that considerable changes were observed in the case of testosterone only. A combined administration of testosterone and ethylmorphine did not pr

Liquid chromatographic determination of barbiturates using a hollow-fibre membrane for postcolumn pH modification.

A sensitive, high-performance liquid chromatographic method is described for the determination of barbiturates by postcolumn pH modification. The barbiturates (barbital, phenobarbital, hexobarbital and amobarbital) were separated on a C18 column using a mixture of methanol and water as an eluent. Then the pH of the eluent was raised to 10 by introducing ammonia or ammonium ion through a sulphonated hollow-fibre membrane inserted between the column and the detector. The detection was based on the primary ionized barbiturates at 240 nm. At barbiturate concentrations of 2.0 micrograms/ml, the wit

Effects of a single administration of 6-aminonicotinamide on hepatic microsomal drug metabolism in rats.

Pretreatment of rats with 6-aminonicotinamide (6-AN) induced a significant increase in aminopyrine (AM) N-demethylation and hexobarbital (HX) hydroxylation in rats. In this study, the concentrations of AM, 4-monomethylaminoantipyrine (MAA), aminoantipyrine (AA) and further metabolites were measured by using the GC/MS method. The increase in AM N-demethylation induced by 6-AN treatment was confirmed by measuring the area under the plasma concentration-time curve (AUC) for AM, MAA and AA in rats after AM administration. Of the first two steps of AM metabolism mediated by cytochrome P-450-coupled
