Kindler syndrome (OMIM 173650) is an autosomal recessive genodermatosis characterized by trauma-induced blistering, poikiloderma, skin atrophy, mucosal inflammation and varying degrees of photosensitivity. Although Kindler syndrome is classified as a subtype of epidermolysis bullosa, it has distinct clinicopathological and molecular abnormalities. The molecular pathology of Kindler syndrome involves loss-of-function mutations in a newly recognized actin cytoskeleton-associated protein, now known as fermitin family homologue 1, encoded by the gene FERMT1. This protein mediates anchorage between

BACKGROUND: Cesarean scar pregnancy (CSP) is a life-threatening condition with a rising incidence in China. The pathogenesis of CSP remains poorly understood, partly due to the limited availability of comprehensive datasets constrained by spatiotemporal factors.
OBJECTIVE: This study aimed to explore key regulatory molecules and mechanisms involved in CSP through a multi-omics approach.
METHODS: Proteomic analysis was performed on decidual and villous tissues from clinical patients (n = 6, including 3 CSP cases and 3 controls). Gene expression datasets (n = 9) were obtained from the GEO and

Essentials Acquired Glanzmann thrombasthenia (aGT) is generally caused by function-blocking antibodies (Abs). We demonstrated a unique aGT case due to marked reduction of αIIbβ3 with anti-αIIbβ3 Abs. The anti-αIIbβ3 Abs of the patient did not inhibit platelet function but reduced surface αIIbβ3. Internalization of αIIbβ3 induced by the Abs binding may be responsible for the phenotype. SUMMARY: Background Acquired Glanzmann thrombasthenia (aGT) is a bleeding disorder generally caused by function-blocking anti-αIIbβ3 autoantibodies. Aim We characterize an unusual case of aGT caused by marked red

We have employed echistatin, a 5.4 kDa snake venom disintegrin, as a model protein to investigate the paradox that small ligand-mimetics can bind to the resting alphaIIbbeta3 integrin while adhesive macromolecules cannot. We characterized the interactions between purified human alphaIIbbeta3 and two recombinant echistatin variants: rEch (1-49) M28L, chosen for its selectivity toward beta3-integrins, and rEch (1-40) M28L, a carboxy-terminal truncation mutant. While both contain an RGD integrin targeting sequence, only rEch (1-49) M28L was an effective inhibitor of alphaIIbbeta3 function. Electr

Urinary extracellular vesicles are emerging markers reflecting the disease status of diabetic kidney disease (DKD), but their types and functions in DKD disease are not yet fully understood. This study applied Astral-DIA proteome for the in-depth profiling of urinary migrasomes (umig, a novel type of extracellular vesicles) and serum from DKD patients, aiming to identify protein biomarkers indicative of disease status. Proteome analysis revealed that protein-protein interactions were identified among LAMA1, ITGB3, FGG, and FGB in the differentially expressed proteins of serum and umig in the N

BACKGROUND AND OBJECTIVES: High concentration of human platelet antigen-1a (HPA-1a) antibodies is reported to be associated with severe foetal and neonatal alloimmune thrombocytopaenia (FNAIT). The gold standard for quantification of anti-HPA-1a antibodies is the monoclonal antibody immobilization of platelet antigen (MAIPA) assay, which is a laborious method performed in only a few reference laboratories. The aim of this study was to evaluate the performance of the commercially available bead-based Luminex assay PakLx (Immucor) for quantitative measurement of anti-HPA-1a antibodies.

Several adhesion molecules have been shown to occur at the surface of endometrial cells. One of these is the integrin alpha v subunit which associates with various beta chains including beta 5. We demonstrate the presence of integrin beta 5 polypeptide in human endometrial epithelial cells throughout the menstrual cycle using immunocytochemistry with monospecific antibodies, and at the mRNA level by thermal amplification from endometrial cDNA. Integrin beta 5 is also found in a population of bone marrow-derived cells. A notable feature of the distribution of the beta 5 subunit in the glandular

Rhodostomin (Rho) is a snake venom protein isolated from Calloselasma rhodostoma. Rho is a disintegrin that inhibits platelet aggregation by blocking the binding of fibrinogen to the integrin alpha(IIb)beta3 of platelets. Rho produced in Escherichia coli inhibited platelet aggregation with a K(I) value of 263 nM. Although functional, Rho produced in E. coli is misfolded based on our 2D and 3D NMR studies. In order to correct the folding problem, Rho was expressed in Pichia pastoris. The recombinant Rho expressed in P. pastoris inhibited platelet aggregation with a resulting K(I) value of 70 nM

BACKGROUND: Fetal and neonatal alloimmune thrombocytopenia (FNAIT) is caused by the destruction of platelets in the fetus or newborn by maternal platelet alloantibodies, mostly against human platelet antigen (HPA)-1a. Recent studies indicate that two anti-HPA subtypes exist: Type I reacts with epitopes residing on the plexin-semaphorin-integrin (PSI) and type II with plexin-semaphorin-integrin/integrin epidermal growth factor 1 (I-EGF1) domains of the β3 integrin. Here, we evaluated whether a Cys460Trp mutation in the I-EGF1 domain found in a patient with Glanzmann thrombasthenia can alter the

BACKGROUND: Non-invasive imaging strategies play a critical role for assessment of the efficacy of angiogenesis therapies. Hypoxia resulting from deficient blood flow is a potent stimulator of angiogenesis which is marked by upregulated alphavbeta3 integrin receptor.
METHODS AND RESULTS: The use of dual-isotope radiotracers targeted at alphavbeta3 and myocardial hypoxia has been demonstrated to non-invasively track hypoxia-induced angiogenesis in ischemic rat model of myocardial hypertrophy, which was induced by non-occlusive abdominal aortic banding followed by myocardial infarction at 6 wee

Cardiovascular and cerebrovascular diseases, the leading causes of death worldwide, are primarily driven by pathological thrombus formation resulting from uncontrolled platelet aggregation at sites of atherosclerotic plaque rupture. Current antiplatelet drugs are designed either to indirectly inhibit intracellular signaling pathways that activate integrin αIIbβ3 or to directly block the binding of the activated integrin to fibrinogen. However, because this interaction is also essential for normal hemostasis, these drugs inevitably increase the risk of serious spontaneous bleeding. In this stud

Fetal/neonatal alloimmune thrombocytopenia (FNAIT) is a life-threatening bleeding disorder caused by maternal antibodies targeting a paternally inherited alloantigen present on the surface of fetal platelets. The human platelet alloantigen HPA-1a/HPA-1b, also known as the PlA1/PlA2 alloantigen, is the most frequently implicated HPA for causing FNAIT in Caucasians. A single Leu33Pro amino acid polymorphism residing within the plexin/semaphorin/integrin (PSI) domain of the integrin β3 subunit (platelet membrane glycoprotein (GP), GPIIIa) is responsible for generating the HPA-1a and HPA-1b epitop

AIM: To generation of human Fab fragment against GP GPIIb/IIIa from phage display library and to study its effect on platelet aggregation.
METHODS: An antibody phage display library was constructed from spleen cells from a donor with idiopathic thrombocytopenic purpura(ITP) whose anti-GPIIb/IIIa antibody was positive. High-affinity human mAbs was selected by panning against GPIIb/IIIa expressed on CHO123 cells. The binding specificity of phage antibody to GPIIb/IIIa was detected by ELISA and Western blot. And the effect of antibody on platelet aggregation was analyzed.
RESULTS: After three r

The soluble urokinase plasminogen activator receptor (suPAR) has been implicated in a wide range of pathological conditions including primary nephrotic syndromes and acute kidney injuries. suPAR can trigger transduction cascades in podocytes by outside-in activation of αVβ3-integrin, but there is evidence that the functional cell surface response element is actually a complex of different types of receptors, which may also include the receptor for advanced glycation end-products (RAGE) and formyl peptide receptors (FPRs). Here we observed that ROS accumulation and Src activation could be evoke

: Glanzmann thrombasthenia is an inherited severe bleeding disease. Mutations associated with Glanzmann thrombasthenia are highly heterogeneous and occur across the two genes coding for the platelet αIIbβ3 integrin. This study was aimed at identifying Glanzmann thrombasthenia-associated novel mutations in Tunisian patients. Seven unrelated Glanzmann thrombasthenia patients issued from high consanguineous families (86%; 6/7 of the patients) were studied. Glanzmann thrombasthenia diagnoses were based on patients' bleeding histories and platelet aggregation tests. Screening of ITGA2B and ITGB3 ge

Thromboangiitis obliterans or Buerger's disease is an episodic and segmental inflammatory and thrombotic process of the medium and small arteries of the lower extremities. Even though the disease was described 90 years ago, the etiopathogenesis is still under consideration. Afflicted patients are mostly young male cigarette smokers without signs of atherosclerosis or other risk factors for peripheral arterial occlusive disease. This indicates that hereditary thrombophilic factors could play a role in the etiopathogenesis. Recently, increasing evidence shows that platelet receptor polymorphisms

Melanoma prognosis is based on histological criteria such as tumor thickness (measured by Breslow index), level of invasion (Clarck's level), presence of ulceration and number of mitoses per mm2. However, these parameters do not provide a precise prognosis in all cases: thin melanomas may develop metastases and thick melanomas may remain focalized for many years. For these reasons, the search for other prognostic factors is still ongoing. Many molecules play a part in the invasiveness and metastatic dissemination of melanoma have now been identified. Expression of these molecules has been stud

Objective To establish a stable cell line overexpressing integrin β3(ITGB3), aiming to overcome the limited replication efficiency and low viral yields of Hantaan virus (HTNV) in conventional cell culture systems. Methods A lentiviral vector encoding ITGB3 was constructed and packaged into viral particles. Vero E6 and A549 cells were transduced and selected with puromycin to generate stable lines. ITGB3 expression was verified by quantitative RT-qPCR, Western blot, and immunofluorescence staining, and cell viability was evaluated using the CCK-8 assay. Following HTNV infection, viral replicati

Human cytomegalovirus (HCMV) infection results in either productive or latent infection, the latter enabling life-long viral persistence. Monocytes support latent infection but become permissive to productive infection upon differentiation into macrophages. These differentiation-driven differences have been largely attributed to chromatin-mediated repression of the viral genome. Using metabolic labeling of newly synthesized RNA, we observe markedly lower viral transcription at early stages of infection in monocytes compared to macrophages. Unbiased comparison reveals that this difference is pa
