Safety of Ketorolac in Primary Total Hip Arthroplasties: An Analysis of Over 7,000 Cases.

BACKGROUND: Ketorolac is commonly utilized perioperatively in primary total hip arthroplasty (THA), either intravenously or via periarticular infiltration analgesia. While effective for pain control, concerns remain regarding its potential to cause acute kidney injury (AKI), particularly in patients who have chronic kidney disease (CKD). This study evaluated the association between perioperative ketorolac administration and postoperative AKI, stratified by baseline renal function and administration route. METHODS: We retrospectively analyzed 7,211 inpatient THAs performed between 2011 and 202

An analysis of harms reporting in systematic reviews regarding ketorolac for management of perioperative pain.

BACKGROUND: Owing to the frequent perioperative use of ketorolac tromethamine and its ability to minimise postoperative opioid requirements, it is important to continually reassess harms associated with its use. Our primary objective was to investigate the extent of harms reporting in systematic reviews (SRs) on ketorolac for perioperative pain. METHODS: In May 2022, we conducted a search of major databases, MEDLINE (PubMed and Ovid), Embase, Epistemonikos, and the Cochrane Database of Systematic Reviews to identify eligible SRs on ketorolac for perioperative pain. [Influence of simulated microgravity on the threshold of pain sensitivity in humans with single dose of ketorolac].

The data supporting the influence of simulated microgravity effects on pain sensitivity were obtained in the series of experiments involving human. In conditions of antiorthostatic hypokinesia (ANOH) and <<dry>> immersion revealed no reduction in pain sensitivity in the morning, which is typical for normal conditions. Ketorolac has no effect on pain sensitivity, when determining the pain threshold (PT) by method of thermoalgometry. However, the conditions of simulated microgravity substantially alter the pharmacokinetics of ketorolac, increasing the rate of absorption of the drug and reduce it

Locally administered ketorolac and bupivacaine for control of postoperative pain in breast augmentation patients.

With recent developments in the field of analgesia, the question arises whether there is a role for placing local anesthetics, nonsteroidal anti-inflammatory drugs, or both into the breast implant pocket. The objective of this study was to test the effectiveness of locally administered intraoperative ketorolac and bupivacaine with epinephrine at reducing pain in the postoperative period. The study was a prospective, randomized, double-blind clinical trial. One hundred consecutive retropectoral breast augmentation patients were enrolled, and informed consent was obtained. A real-world signal detection analysis uncovering the safety profile of ketorolac tromethamine.

Ketorolac tromethamine is a potent nonsteroidal anti-inflammatory drug commonly used to treat moderate-to-severe pain, especially in the context of postoperative and acute injury. However, real-world studies on the adverse events (AEs) associated with ketorolac tromethamine remain limited. A disproportionality analysis was performed using data from the FDA Adverse Event Reporting System between January 2004 and September 2024 to evaluate the safety profile of ketorolac tromethamine by analyzing the demographic characteristics and timing of AEs. A total of 3655 AEs associated with ketorolac tro

Evaluating the Effects of Perioperative Ketorolac Use on Uncemented Total Hip Arthroplasty Outcomes.

Background/Objectives: Ketorolac is commonly used for pain management after orthopedic surgery, but concerns regarding its effects on postoperative complications remain. This study evaluates the impact of ketorolac use on short- and long-term outcomes in adult patients undergoing uncemented primary total hip arthroplasty (THA), where implant stability relies on biological fixation through bone ingrowth into a porous-coated prosthesis rather than bone cement. Methods: A retrospective cohort study was conducted using the TriNetX Research Network. Patients aged 18 years or older who underwent unc

R-ketorolac ameliorates cancer-associated cachexia and prolongs survival of tumour-bearing mice.

BACKGROUND: Cancer-associated cachexia (CAC) is a debilitating syndrome associated with poor quality of life and reduced life expectancy of cancer patients. CAC is characterized by unintended body weight reduction due to muscle and adipose tissue loss. A major hallmark of CAC is systemic inflammation. Several non-steroidal anti-inflammatory drugs (NSAIDs) have been suggested for CAC treatment, yet no single medication has proven reliable. R-ketorolac (RK) is the R-enantiomer of a commonly used NSAID. The effect of RK on CAC has not yet been evaluated. METHODS: Ten- to 11-week-old mice were in

The effects of ketorolac injected via patient controlled analgesia postoperatively on spinal fusion.

Lumbar spinal fusions have been performed for spinal stability, pain relief and improved function in spinal stenosis, scoliosis, spinal fractures, infectious conditions and other lumbar spinal problems. The success of lumbar spinal fusion depends on multifactors, such as types of bone graft materials, levels and numbers of fusion, spinal instrumentation, electrical stimulation, smoking and some drugs such as nonsteroidal anti-inflammatory drugs (NSAIDs). From January 2000 to December 2001, 88 consecutive patients, who were diagnosed with spinal stenosis or spondylolisthesis, were retrospective
