Sox9 marks limbal stem cells and is required for asymmetric cell fate switch in the corneal epithelium.

Adult tissues with high cellular turnover require a balance between stem cell renewal and differentiation, yet the mechanisms underlying this equilibrium are unclear. The cornea exhibits a polarized lateral flow of progenitors from the peripheral stem cell niche to the center; attributed to differences in cellular fate. To identify genes that are critical for regulating the asymmetric fates of limbal stem cells and their transient amplified progeny in the central cornea, we utilized an in vivo cell cycle reporter to isolate proliferating basal cells across the anterior ocular surface epitheliu

A Novel Chitosan-Gelatin Scaffold and Cell Spray Therapy for Treating Limbal Stem Cell Deficiency.

Limbal stem cell deficiency (LSCD) is a severe ocular condition that impairs vision and causes chronic pain. Current therapeutic approaches, such as stem cell transplantation and biomaterial scaffolds, are under continuous refinement. This study evaluated a novel treatment strategy combining a biocompatible and biodegradable chitosan-gelatin (CS-Gel) electrospun scaffold with a topical limbal stem cell (LSC) spray for treating alkaline-induced LSCD in a rabbit model. The scaffold was fabricated via electrospinning, stabilized with water vapor, and characterized using SEM, ATR-FTIR, contact ang

Single-cell RNA sequencing in cornea research: Insights into limbal stem cells and their niche regulation.

The corneal epithelium is composed of stratified squamous epithelial cells on the outer surface of the eye, which acts as a protective barrier and is critical for clear and stable vision. Its continuous renewal or wound healing depends on the proliferation and differentiation of limbal stem cells (LSCs), a cell population that resides at the limbus in a highly regulated niche. Dysfunction of LSCs or their niche can cause limbal stem cell deficiency, a disease that is manifested by failed epithelial wound healing or even blindness. Nevertheless, compared to stem cells in other tissues, little i

Extracellular vesicles from adipose-derived mesenchymal stem cells promote colony formation ability and EMT of corneal limbal epithelial cells.

Corneal diseases are a leading cause of visual impairment, and their treatment remains challenging. Corneal epithelial stem cells exist in the limbus, the peripheral region of the cornea, and play an important role in corneal regeneration. Here, we evaluated the effects of extracellular vesicles from human adipose-derived mesenchymal stem cells (AdMSC-EVs) on limbal epithelial cells (LECs). Colony formation assays showed that the colony-forming efficiency of LECs significantly increased in the presence of AdMSC-EVs. We next demonstrated that AdMSC-EVs accelerated the migration of LECs in a scr

ABCB5+ Limbal Epithelial Stem Cells Inhibit Developmental but Promote Inflammatory (Lymph) Angiogenesis While Preventing Corneal Inflammation.

The limbus, the vascularized junction between the cornea and conjunctiva, is thought to function as a barrier against corneal neovascularization. However, the exact mechanisms regulating this remain unknown. In this study, the limbal epithelial stem cell (LESC) marker ABCB5 was used to investigate the role of LESCs in corneal neovascularization. In an ABCB5KO model, a mild but significant increase of limbal lymphatic and blood vascular network complexity was observed in developing mice (4 weeks) but not in adult mice. Conversely, when using a cornea suture model, the WT animals exhibited a mil

Comparative Study of Xenobiotic-Free Media for the Cultivation of Human Limbal Epithelial Stem/Progenitor Cells.

The culture of human limbal epithelial stem/progenitor cells (LSCs) in the presence of animal components poses the risk of cross-species contamination in clinical applications. We quantitatively compared different xenobiotic-free culture media for the cultivation of human LSCs. LSCs were cultured from 2 × 2 mm limbal tissue explants on denuded human amniotic membrane with different xenobiotic-free culture media: CnT-Prime (CnT-PR) supplemented with 0%, 1%, 5%, and 10% human serum (HS), embryonic stem cell medium (ESCM) alone or in combination with the standard supplemented hormonal epithelium

The Reliability of PCL/Anti-VEGF Electrospun Scaffolds to Support Limbal Stem Cells for Corneal Repair.

Since only few reported studies propose anti-vascular endothelial growth factor (anti-VEGF) delivery through electrospun scaffolds, this study greatly contributes to the potential prevention of patient's vision loss, as it explores electrospun polycaprolactone (PCL) coated with anti-VEGF for the blockage of abnormal cornea vascularization. In terms of physicochemical properties, the biological component increased the PCL scaffold fiber diameter (by ~24%) and pore area (by ~82%), while ut slightly reduced its total porosity as the anti-VEGF solution filled the voids of the microfibrous structur

Transcriptomic Landscape and Functional Characterization of Human Induced Pluripotent Stem Cell-Derived Limbal Epithelial Progenitor Cells.

Limbal stem cell deficiency (LSCD) is a complex, multifactorial disease affecting limbal epithelial progenitor cells (LEPC), which are essential for maintaining corneal stability and transparency. Human induced pluripotent stem cell-derived (hiPSC-) LEPC are a promising cell source for the treatment of LSCD. However, their similarity to native tissue-derived (T-) LEPC and their functional characterization has not been studied in detail. Here, we show that hiPSC-LEPC and T-LEPC have rather similar gene expression patterns, colony-forming ability, wound-healing capacity, and melanosome uptake. I

Bone marrow-versus adipose tissue-derived mesenchymal stem cells for corneal failure in an experimental model of limbal stem cell deficiency.

Ocular limbal stem cell deficiency (LSCD) occurs because of corneal epithelial stem cell destruction or dysfunction at the limbal niche. LSCD can cause corneal blindness, and the current therapy based on limbal stem cell transplantation is continuously improving. The aim of this work was to compare the safety and efficacy of human mesenchymal stem cells (hMSCs) derived from bone marrow (hBM-MSCs) and adipose tissue (hAT-MSCs) when transplanted to a rabbit model of LSCD. Both hMSC types expressed the corneal and limbal epithelial cell markers CK3, CK12, ZO-1, and ABCG2 under standard culture co

Therapeutic Effect of Human Adipose Tissue-Derived Mesenchymal Stem Cells in Experimental Corneal Failure Due to Limbal Stem Cell Niche Damage.

Limbal stem cells are responsible for the continuous renewal of the corneal epithelium. The destruction or dysfunction of these stem cells or their niche induces limbal stem cell deficiency (LSCD) leading to visual loss, chronic pain, and inflammation of the ocular surface. To restore the ocular surface in cases of bilateral LSCD, an extraocular source of stem cells is needed to avoid dependence on allogeneic limbal stem cells that are difficult to obtain, isolate, and culture. The aim of this work was to test the tolerance and the efficacy of human adipose tissue-derived mesenchymal stem cell

Automated quantification of central corneal epithelial thickness for the diagnosis of limbal stem cell deficiency.

Limbal Stem Cell Deficiency (LSCD) is a corneal disorder characterized by conjunctivalization of the cornea. Central corneal epithelial thickness, measured using Anterior Segment Optical Coherence Tomography (AS-OCT), is a biomarker for LSCD. Manual measurement of epithelial thickness remains the gold standard for diagnosis because existing automated commercial software is unreliable in severe cases. Standard machine learning approaches have limited applicability for automating this measurement due to the difficulty of distinguishing corneal epithelium from scar tissue, and imprecise clinical

Surface modified electrospun scaffold supports iPSC-derived limbal stem cell function.

This study focuses on the development of an electrospun Poly Lactide-co-Glycolic Acid (PLGA) scaffold for ocular surface regeneration, specifically for the treatment of Limbal Stem Cell Deficiency (LSCD). The scaffold is designed to support the attachment, proliferation, and maintenance of induced pluripotent stem cell-derived limbal stem cells (iPSC-LSCs). To address the hydrophobic nature of PLGA and enhance biocompatibility, the scaffold surface was functionalized with extracellular matrix proteins, specifically Collagen IV and Laminin-521, through atmospheric plasma treatment. Ocular Surface Inflammation as a Driver of Cornea Limbal Stem Cell Deficiency: Mechanisms and Implications.

Ocular surface inflammation is a major disruptor of corneal epithelial homeostasis and a key driver of limbal stem cell deficiency (LSCD). Limbal stem cells (LSCs), residing within the specialized limbal niche, maintain corneal transparency through continuous epithelial renewal and by preventing conjunctival encroachment onto the corneal surface. Chronic or severe inflammatory insults-stemming from systemic autoimmune disorders, ocular surface diseases, infections, trauma, or environmental stressors-can damage both LSCs and their microenvironment, ultimately leading to limbal insufficiency. Treatment of vernal keratoconjunctivitis simulating total limbal stem cell deficiency: a case report.

BACKGROUND: Severe vernal keratoconjunctivitis (VKC) can lead to total limbal stem cell deficiency (LSCD), a condition characterized by conjunctivalization of the cornea and neovascularization that often results in severe vision loss and typically requires surgical intervention. This report presents a novel case where a condition clinically simulating total LSCD, induced by chronic inflammation from VKC, was reversed using only anti-inflammatory therapy. CASE PRESENTATION: A 15-year-old male with a 7-year history of inadequately managed VKC presented with typical signs of clinically apparent t
