Plasma membrane damage by environmental materials enhances cell-cell fusion and impairs defense functions of macrophages.

INTRODUCTION: Many environmental materials can activate macrophages and trigger inflammatory responses. However, whether and how these exposures alter macrophage function in host defense remains elusive. METHODS: Bone marrow-derived macrophages were treated with various environmental materials, including laundry detergent, and their membrane damage was measured by Annexin V staining. Alterations in macrophage function were assessed by cell-cell fusion, in vitro phagocytosis, and in vivo clearance of Candida albicans. RESULTS AND DISCUSSION: Many environmental hazards damage the plasma membra

Separation-Free Extracellular Vesicle Microribonucleic Acid Profiling Using Structurally Oriented Membrane Fusion and Spatially Confined Amplification.

Current methods for profiling of extracellular vesicle (EV) microRNAs (miRNAs) involve either PCR or sequencing analysis of EV lysate or delivery of RNA probes into EVs for in situ hybridization, which suffer from complex protocols and low delivery efficiency/limited sensitivity, respectively. To overcome this limitation, we present a programmable, aptamer-mediated membrane fusion strategy that integrates aptamer-functionalized liposomes with nucleic acid nanostructures-Dual-color, Bone-shaped Biosensor-loaded Aptamer-Liposome (DBS-Alip) for rapid, one-step, in situ detection of multiple tumor

Lipid Disorder and PIP2-Regulated Clustering of Syntaxin-1 JMD-TMD Regions Govern Membrane Fusion Competence.

Syntaxin-1 (Stx1), a core neuronal SNARE protein, regulates membrane docking and fusion during neurotransmitter release. Although Stx1 clustering is widely observed at presynaptic active zones and in neuroendocrine cells, how Stx1 isoforms and lipid interactions organize the plasma membrane prior to fusion remains unclear. Here, we performed MARTINI coarse-grained molecular dynamics (cgMD) simulations of 1-10 copies of the juxtamembrane and transmembrane domains (JMD-TMDs) of Stx1A and Stx1B embedded in plasma membrane models to investigate membrane lipid disorder, Stx1 JMD-TMD clustering dyna

Bio-inspired suction cup microneedle loaded fusion membrane targeting nano-drug delivery system treat androgenetic alopecia.

Androgenetic alopecia (AGA) involves androgen-driven dermal papilla cell (DPC) dysfunction compounded by oxidative stress and inflammation. We engineered a leech-inspired suction-cup microneedle platform SC/MN@memPDA to deliver metformin precisely to the dermal papilla. The system integrates 300-µm short microneedles with suction-assisted negative pressure to increase insertion depth, mesoporous polydopamine nanoparticles (MPDA) for high-capacity, H₂O₂-responsive MET release, and a fusion membrane cloak derived from M2 marcophages and DPCs for inflammation targeting and dermal papilla niche ho

Plasma Membrane Damage by Environmental Materials Enhances Cell-Cell Fusion and Impairs Immune Functions of Macrophages.

Macrophages are the most abundant phagocytes and play an essential role in host defense. Previous studies have shown that many environmental materials can activate macrophages and trigger inflammatory responses. However, whether these exposures alter macrophage function in host defense remains unclear. This study found that many environmental materials (such as carbon nanotubes, tungsten carbide [WC], and detergents) can damage the plasma membranes of macrophages. This damage leads to decreased reactive oxygen species (ROS) production and phagocytosis but elevated cell-cell fusion. In vivo, ai

IFN-inducible human phospholipid scramblase 1 (PLSCR1) protein restricts HIV-1 infection by inhibiting membrane fusion.

Human phospholipid scramblase 1 (PLSCR1) is an interferon-stimulated gene (ISG) that inhibits viral infections through various mechanisms. Here, we identify PLSCR1 as a host restriction factor that inhibits HIV-1 entry by impairing membrane fusion mediated by the envelope glycoprotein (Env). Using multiple cell types including the human SupT1 T cell line and purified CD4+ T cells, we demonstrate that PLSCR1 inhibits the replication of HIV-1 with diverse tropisms and subtypes, as well as HIV-2 and SIV. Mechanistically, we find that PLSCR1 blocks viral entry and cell-to-cell transmission, in par

Canine distemper virus and measles virus fusion glycoprotein trimers: partial membrane-proximal ectodomain cleavage enhances function.

The trimeric fusion (F) glycoproteins of morbilliviruses are activated by furin cleavage of the precursor F(0) into the F(1) and F(2) subunits. Here we show that an additional membrane-proximal cleavage occurs and modulates F protein function. We initially observed that the ectodomain of approximately one in three measles virus (MV) F proteins is cleaved proximal to the membrane. Processing occurs after cleavage activation of the precursor F(0) into the F(1) and F(2) subunits, producing F(1a) and F(1b) fragments that are incorporated in viral particles. We also detected the F(1b) fragment, inc

Enhancing TIM-3 immunotherapy with epirubicin-loaded pH sensitive fusion membrane nanoparticles for effective glioblastoma treatment.

Immune checkpoint blockade (ICB), particularly anti-programmed cell death protein 1 blockade (aPD-1), has demonstrated limited efficacy in glioblastoma (GBM) due to the tumor's highly immunosuppressive microenvironment and the restrictive blood-brain barrier (BBB). T cell immunoglobulin and mucin domain-3 (TIM-3), an emerging immune checkpoint widely expressed on various immune cells beyond just exhausted T cells, presents a promising target for GBM treatment. In this study, we engineered A2-TELM, a pH-sensitive macrophage membrane-coated nanoparticle conjugated with angiopep-2 peptide, to del

Galactose-modified erythrocyte membrane fusion liposomes enable the targeted delivery of drug nanoparticles to the liver.

The safe and efficient delivery of chemicals and biologics remains crucial for liver disease therapy. In this study, we developed a targeted drug delivery system utilizing a galactose-modified erythrocyte membrane coating technique and drug liposome nanoparticles, which were further optimized using orthogonal experiments and response surface analysis. The specificity, precision, accuracy, and stability exhibited satisfactory performance in bioanalytical analysis. Specifically, targeting ligands (Gal-DSPE-PEG3400) were efficiently inserted into red blood cell (RBC) membranes using a facile inse

Efficiency of membrane fusion inhibitors on different hemagglutinin subtypes: insight from a molecular dynamics simulation perspective.

The challenge in vaccine development, along with drug resistance issues, has encouraged the search for new anti-influenza drugs targeting different viral proteins. Hemagglutinin (HA) glycoprotein, crucial in the viral replication cycle, has emerged as a promising therapeutic target. CBS1117 and JNJ4796 were reported to exhibit similar potencies against infectious group 1 influenza, which included H1 and H5 HAs; however, their potencies were significantly reduced against group 2 HA. This study aims to explore the molecular binding mechanisms and group specificity of these fusion inhibitors agai

Hypoxia-induced factor 1α promotes osteotomy-induced regional acceleratory phenomenon via DC-STAMP mediated membrane fusion.

OBJECTIVES: The mechanisms of regional acceleratory phenomenon (RAP) induced by orthognathic osteotomy are unclear. It was not known if locally hypoxic microenvironment changes were involved in this phenomenon. METHODS: Hypoxia-induced factor-1α knockout mice harboring Cathepsin K (CTSK) Cre were used to investigate the effect of hypoxia-driven osteoclasts on alveolar bone remodeling. RAW264.7 cells were induced by CoCl2 to observe the effects of dendritic cell-specific transmembrane protein (DC-STAMP) on the fusion and differentiation of osteoclasts. RESULTS: We found mandibular osteotomy o

Myocyte-derived Myomaker expression is required for regenerative fusion but exacerbates membrane instability in dystrophic myofibers.

Muscle progenitor cell fusion is required for the formation and regeneration of multinucleated skeletal muscle fibers. Chronic muscle regeneration in Duchenne muscular dystrophy (DMD) is characterized by ongoing fusion of satellite cell (SC) progeny, but the effects of fusion on disease and the mechanisms by which fusion is accomplished in this setting are not fully understood. Using the mdx mouse model of DMD, we deleted the fusogenic protein Myomaker in SCs or myofibers. Following deletion in SCs, mice displayed a complete lack of myocyte fusion, resulting in severe muscle loss, enhanced fib

Glycoprotein 5 Is Cleaved by Cathepsin E during Porcine Reproductive and Respiratory Syndrome Virus Membrane Fusion.

Porcine reproductive and respiratory syndrome (PRRS) is a serious viral disease affecting the global swine industry. Its causative agent, PRRS virus (PRRSV), is an enveloped virus, and therefore membrane fusion between its envelope and host cell target membrane is critical for viral infection. Though much research has focused on PRRSV infection, the detailed mechanisms involved in its membrane fusion remain to be elucidated. In the present study, we performed confocal microscopy in combination with a constitutively active (CA) or dominant negative (DN) mutant, specific inhibitors, and small in

HIV-1 anchor inhibitors and membrane fusion inhibitors target distinct but overlapping steps in virus entry.

HIV-1 entry into cells is mediated by the envelope glycoprotein (Env) and represents an attractive target for therapeutic intervention. Two drugs that inhibit HIV entry are approved for clinical use: the membrane fusion-inhibitor T20 (Fuzeon, enfuvirtide) and the C-C chemokine receptor type 5 (CCR5) blocker maraviroc (Selzentry). Another class of entry inhibitors supposedly target the fusion peptide (FP) and are termed anchor inhibitors. These include the VIRIP peptide and VIRIP derivatives such as VIR165, VIR353, and VIR576. Here, we investigated the mechanism of inhibition by VIR165. We show

Regulatory Role of the Morbillivirus Attachment Protein Head-to-Stalk Linker Module in Membrane Fusion Triggering.

Morbillivirus (e.g., measles virus [MeV] and canine distemper virus [CDV]) host cell entry is coordinated by two interacting envelope glycoproteins, namely, an attachment (H) protein and a fusion (F) protein. The ectodomain of H proteins consists of stalk, connector, and head domains that assemble into functional noncovalent dimer-of-dimers. The role of the C-terminal module of the H-stalk domain (termed linker) and the connector, although putatively able to assume flexible structures and allow receptor-induced structural rearrangements, remains largely unexplored. Here, we carried out a nonco

H1N1 Swine Influenza Viruses Differ from Avian Precursors by a Higher pH Optimum of Membrane Fusion.

UNLABELLED: The H1N1 Eurasian avian-like swine (EAsw) influenza viruses originated from an avian H1N1 virus. To characterize potential changes in the membrane fusion activity of the hemagglutinin (HA) during avian-to-swine adaptation of the virus, we studied EAsw viruses isolated in the first years of their circulation in pigs and closely related contemporary H1N1 viruses of wild aquatic birds. Compared to the avian viruses, the swine viruses were less sensitive to neutralization by lysosomotropic agent NH4Cl in MDCK cells, had a higher pH optimum of hemolytic activity, and were less stable at

Intersubunit interactions modulate pH-induced activation of membrane fusion by the Junin virus envelope glycoprotein GPC.

The mature arenavirus envelope glycoprotein GPC is a tripartite complex comprising a stable signal peptide (SSP) in addition to the receptor-binding (G1) and transmembrane fusion (G2) subunits. We have shown previously that SSP is a key element in GPC-mediated membrane fusion, and that GPC sensitivity to acidic pH is modulated in part through the lysine residue at position 33 in the ectodomain loop of SSP (J. York and J. H. Nunberg, J. Virol. 80:7775-7780, 2006). A glutamine substitution at this position stabilizes the native GPC complex and thereby prevents the induction of pH-dependent membr
