Real-time fluorescent and microscopic uricase assay by monitoring the clearance of acridine orange-doped monosodium urate crystals under physiological conditions.

The accumulation of monosodium urate (MSU) crystals in joints plays a central role in the pathogenesis of gouty arthritis. While conventional treatments for acute gout focus on symptom relief, they do not address the root cause-crystal deposition. Recombinant uricase offers a promising long-term solution by catalyzing the conversion of poorly soluble uric acid into highly soluble allantoin, facilitating MSU crystal clearance. In this study, we developed a novel assay system using acridine orange (AO)-doped MSU crystals as a solid substrate and real-time sensing unit to monitor uricase activity

Brain tissue oxygen monitoring in traumatic brain injury: part I-To what extent does PbtO2 reflect global cerebral physiology?

BACKGROUND: The primary aim was to explore the association of global cerebral physiological variables including intracranial pressure (ICP), cerebrovascular reactivity (PRx), cerebral perfusion pressure (CPP), and deviation from the PRx-based optimal CPP value (∆CPPopt; actual CPP-CPPopt) in relation to brain tissue oxygenation (pbtO2) in traumatic brain injury (TBI). METHODS: A total of 425 TBI patients with ICP- and pbtO2 monitoring for at least 12 h, who had been treated at the neurocritical care unit, Addenbrooke's Hospital, Cambridge, UK, between 2002 and 2022 were included. Fiber-optic sensors for monitoring patient physiological parameters: a review of applicable technologies and relevance to use during magnetic resonance imaging procedures.

The issues involved with recording vital functions in the magnetic resonance imaging (MRI) environment using fiber-optic sensors are considered in this paper. Basic physiological parameters, such as respiration and heart rate, are fundamental for predicting the risk of anxiety, panic, and claustrophobic episodes in patients undergoing MRI examinations. Electronic transducers are generally hazardous to the patient and are prone to erroneous operation in heavily electromagnetically penetrated MRI environments; however, nonmetallic fiber-optic sensors are inherently immune to electromagnetic effe

An Evaluation of Real-world Smart Sock-Based Temperature Monitoring Data as a Physiological Indicator of Early Diabetic Foot Injury: Case-Control Study.

BACKGROUND: Lower extremity complications of diabetes represent major health care complications both in terms of cost and impact to quality of life for patients with diabetic peripheral neuropathy. Temperature monitoring has been shown in previous studies to provide a useful signal of inflammation that may indicate the early presence of a foot injury. OBJECTIVE: In this study, we evaluated the temperature data for patients that presented with a diabetic foot injury while using a sock-based remote temperature monitoring device. METHODS: The study abstracted data from patients who were enrolle

The Consistent Tick-Vertebrate Infectious Cycle of the Lyme Disease Spirochete Enables Borrelia burgdorferi To Control Protein Expression by Monitoring Its Physiological Status.

The Lyme disease spirochete, Borrelia burgdorferi, persists in nature by alternatingly cycling between ticks and vertebrates. During each stage of the infectious cycle, B. burgdorferi produces surface proteins that are necessary for interactions with the tick or vertebrate tissues it encounters while also repressing the synthesis of unnecessary proteins. Among these are the Erp surface proteins, which are produced during vertebrate infection for interactions with host plasmin, laminin, glycosaminoglycans, and components of the complement system. Erp proteins are not expressed during tick colon

The Longitudinal Physiological Reference Values of Middle Cerebral Artery Blood Flow Velocity in Extremely Preterm Infants: A Study Utilizing Multimodal Cerebral Hemodynamic Monitors.

BACKGROUND: Cerebral blood flow (CBF) is essential for maintaining cerebral metabolism in extremely preterm infants; however, no universally accepted reference standard exists for CBF velocity (CBFV). This study aimed to establish physiological reference patterns of middle cerebral artery CBFV under strictly defined physiological stability using multimodal monitoring. METHODS: This post-hoc analysis included extremely preterm infants (gestational age ≤ 28+6 weeks or birthweight 500-1000 g) from a prospective cohort study. Serial Doppler ultrasonography of the middle cerebral artery was perfor

Individualized monitoring of heat illness risk: novel adaptive physiological strain index to assess exercise-heat strain from athletes to fully encapsulated workers.

Objective. Exercise-heat strain estimation approaches often involve combinations of body core temperature (Tcore), skin temperature (Tsk) and heart rate (HR). A successful existing measure is the 'Physiological Strain Index' (PSI), which combines HR and Tcore values to estimate strain. However, depending on variables such as aerobic fitness and clothing, the equation's 'maximal/critical' Tcore must be changed to accurately represent the strain, in part because high Tsk (small Tcore-Tsk) can increase cardiovascular strain and thereby negatively affect performance. Here, an 'adaptive PSI' (aPSI)

Continuous multidimensional physiological monitoring redefines early warning and preemptive treatment for neutropenic sepsis following cisplatin chemotherapy in non-small cell lung cancer.

BACKGROUND: Despite the advent of targeted therapies, cisplatin remains a cornerstone in the management of non-small cell lung cancer Its systemic toxicity, however, often triggers neutropenic sepsis, a rapidly fatal oncological emergency. Traditional intermittent monitoring is inadequate here. Chemotherapy-induced "inflammatory silence" frequently masks the classical febrile response, leaving patients unprotected during outpatient windows. METHODS: In this Perspective, we mined global real-world pharmacovigilance data from the FAERS and Japanese Adverse Drug Event Report databases. We applie

Therapeutic Drug Monitoring of Vancomycin in Jordanian Patients. Development of Physiologically-Based Pharmacokinetic (PBPK) Model and Validation of Class II Drugs of Salivary Excretion Classification System (SECS).

Vancomycin is a commonly used antibiotic for multi-drug resistant gram-positive infections treatment, especially methicillin-resistant Staphylococcus aureus (MRSA). Despite that, it has wide individual pharmacokinetic variability and nephrotoxic effect. Vancomycin trough concentrations for 57 Jordanian patients were measured in plasma and saliva through immunoassay and liquid chromatography-mass spectrometry (LC-MS/MS), respectively. Plasma levels were within accepted normal range, with exception of 6 patients who showed trough levels of more than 20 μg/ml and vancomycin was discontinued. Baye

Study protocol for an exploratory interventional study investigating the feasibility of video-based non-contact physiological monitoring in healthy volunteers by Mapping Of Lower Limb skIn pErfusion (MOLLIE).

INTRODUCTION: Skin perfusion varies in response to changes in the circulatory status. Blood flow to skin is reduced during haemodynamic collapse secondary to peripheral vasoconstriction, whereas increased skin perfusion is frequently observed when haemodynamics improve with resuscitation. These changes in perfusion may be monitored using non-contact image-based methods. Previous camera-derived physiological measurements have focused on accurate vital signs monitoring and extraction of physiological signals from environmental noise. One of the biggest challenges of camera-derived monitoring is

Monitoring physiological processes of fast-growing broilers during the whole life cycle: Changes of redox-homeostasis effected to trassulfuration pathway predicting the development of non-alcoholic fatty liver disease.

In the broiler industry, the average daily gain and feed conversion ratio are extremely favorable, but the birds are beginning to approach the maximum of their genetic capacity. However, as a consequence of strong genetic selection, the occurrence of certain metabolic diseases, such as myopathies, ascites, sudden cardiac death and tibial dyschondroplasia, is increasing. These metabolic diseases can greatly affect the health status and welfare of birds, as well as the quality of meat. The main goal of this study was to investigate the changes in the main parameters of redox homeostasis during t
