Transactions of the American Ophthalmological Society1996eng
BACKGROUND/PURPOSE: The organoid nevus (sebaceous nevus) syndrome is characterized primarily by cutaneous sebaceous nevus, seizures, and epibulbar choristomas. On the basis of ophthalmoscopic and computed tomographic studies, a yellow fundus lesion recently observed in this syndrome has been called a coloboma by some investigators and a choroidal osteoma by others. This study was undertaken to review our personal experience with the organoid nevus syndrome, to review the English language literature on the subject, and to address some misconceptions regarding its ocular manifestations.

An unusual tumor mass of the scalp found in an elderly woman is presented. This lesion had many features simulating a verrucous squamous cell carcinoma of the scalp. Pathologic findings eventually revealed the tumor mass to be a proliferating trichilemmal cyst. Differentiation between a benign and malignant process in these lesions can sometimes be difficult. Clinical as well as histopathologic evaluation is necessary to make an accurate diagnosis.

Cardiac organoids are increasingly used to model human cardiac development and disease, but their small size often limits molecular characterization, especially if different approaches and protocols are necessary to extract and quantify metabolites and lipids. Here, we present a multistep workflow for combined targeted free amino-acid (FAA) based metabolomic and lipidomic profiling from a single pooled cardiac organoid sample. The protocol covers organoid harvesting, detergent-assisted lysis, and a modified Folch extraction that generates an organic phase for lipid analysis and an aqueous phas

Androgen signalling plays a central role in the development and function of the male urogenital system, including the adrenal gland, prostate, and testis. Disruption of the androgen axis underlies diseases ranging from infertility to castration-resistant prostate cancer, yet current models fail to fully display its multi-organ and hormone-driven complexity. Organoid systems provide a promising platform to model androgen-responsive tissues while preserving three-dimensional structure and cellular heterogeneity, with the ability for long-term culture and cryopreservation. Recent advances have en

Three-dimensional (3D) bioprinting enables the precise fabrication of complex tissue constructs by depositing living cells, biomaterials, and bioactive factors in a spatially controlled manner. Concurrently, organoids-self-organizing 3D cellular structures that recapitulate key aspects of organ architecture and function-have revolutionized biomedical research by providing physiologically relevant models for disease modeling, drug screening, and regenerative medicine. The convergence of 3D bioprinting and organoid technology represents a significant advancement, offering new opportunities for p

The human neocortex exhibits characteristic regional patterning (arealization) critical for higher-order cognitive function. Disrupted arealization is implicated in neurodevelopmental disorders (NDDs), but current neocortical organoid models largely fail to recapitulate this patterning, limiting mechanistic understanding. We establish a straightforward method for generating arealized organoids through short-term early exposure to anterior or posterior morphogens. These treatments created distinct anteroposterior (AP) signaling centers, supporting long-lasting polarization validated by spatial

The inaccessibility of human brain tissue limits the study of human development and function, a challenge that human stem-cell-derived neural models are beginning to address1,2. Transplantation of neural organoids into rodent hosts enables the in vivo study of aspects of human neurodevelopment and circuit function, alongside behavioural phenotyping of the host animals. However, spatial limitations and competition with host circuits constrain the integration of neural organoids, which is critical for studying disease. Here we establish a transplantation platform using a genetic strategy to effe

Liver is a complex organ, capable of incredible regenerative capacity in the mammalian body. In addition to well-known hepatocytes, liver is composed of a complex cell microenvironment, with each cell type contributing to homeostasis, regeneration and disease. Modelling of the liver in vitro has been hampered by 2D cell lines being too simple and whole animal models too complex to underpin the mechanisms of liver biology. Liver organoids are emerging as a great tool to study liver (patho)physiology. Increasing in complexity, organoids have successfully been used to understand the cellular inte

Cardiovascular disease (CVD) remains the leading cause of global mortality, yet traditional in vitro and animal models often fail to recapitulate the integrated structural, cellular, and functional complexity of the human heart. Recent advances in human pluripotent stem cell technology, developmental bioengineering, and microphysiological systems have enabled the development of diverse three-dimensional cardiac tissue models, including self-organizing cardiovascular organoids and engineered cardiac platforms, three-dimensional constructs that partially recapitulate key myocardial features. Male infertility is increasing in prevalence due to cancer treatment, environmental exposures, and patient-specific syndromes. Traditional two-dimensional culture systems inadequately recapitulate the complex three-dimensional architecture, microenvironment, and endocrine milieu essential for spermatogenesis. Testicular organoid models aim to circumvent the challenges of two-dimensional culture, mimicking key aspects of the testicular microenvironment, while enabling controlled experimental manipulation and patient-specific disease modeling. Current testicular organoids demonstrate remarkable

Neurological injuries and neurodegenerative disorders, including spinal cord injury, traumatic brain injury, stroke, and Parkinson's disease remain largely incurable. In the central nervous system (CNS), a self-reinforcing cascade of neuroinflammation, oxidative stress, blood-brain barrier breakdown, and glial fibrotic scarring restricts long-distance axonal regrowth and graft survival. The peripheral nervous system (PNS) exhibits greater intrinsic regenerative potential, yet critical-length defects remain challenging and have driven the development of clinically relevant conduit designs. This

Hydrogel-based nano- and microfibers integrate the biomimetic functionality of hydrogels with the topological architecture of fibers, enabling a realistic emulation of the native extracellular matrix for various biomedical applications. Recent advances in nanofiber fabrication and stem cell engineering have positioned nanofibrous hydrogel scaffolds as compelling synthetic alternatives to conventional Matrigel for disease modeling and translational medicine. In this article, we examine hydrogel nanofiber architectures as a distinct class of material-structure systems. We first outline key desig

Organoids are powerful models for studying tissue dynamics across multiple cellular generations and offer key insights into organ homeostasis, cellular differentiation and disease. Live imaging of organoids is an essential tool for understanding dynamic processes on the cell level, while still in the tissue context. It allows for simultaneous quantification of gene expression and reconstruction of lineage trees, while tracking cells over space and time. Here, we present protocols for long-term imaging and quantitative analysis of intestinal organoids. Our workflow consists of sample preparatio

Retinal degenerative diseases (RDDs) comprise diverse genetic and phenotypic conditions that cause progressive retinal dysfunction and cell loss, leading to vision impairment or blindness. Most RDDs lack appropriate animal models for their study, which affects understanding their disease mechanisms and delays the progress of new treatment development. Recent advances in stem cell engineering, omics, and organoid technology are facilitating research into diseases for which there are no previously existing models. The development of retinal organoids produced from human stem cells has impacted t

Live-cell Ca2+ imaging is an important tool to detect activation of receptors by a putative ligand/drug and complements studies on transport processes, as intracellular Ca2+ changes provide direct evidence for substrate fluxes. Organoid-based systems offer numerous advantages over other in vitro systems such as cell lines, primary cells, or tissue explants, and in particular, intestinal organoid culture has revolutionized research on functional gastrointestinal processes. Calcium imaging using the fluorescent Ca2+ indicator Fura-2-AM can be applied to 3D intestinal organoids, which show an exc

Pituitary deficiencies, or hypopituitarisms, are defined as insufficient production of one or more adenohypophyseal hormones (growth hormone, TSH, ACTH, LH-FSH and prolactin). Congenital hypopituitarism, a rare disease with severely disabling consequences, often takes the form of isolated hormone deficiencies, such as isolated growth hormone deficiency (GHD) or isolated ACTH deficiency (ACTHD), or combined pituitary hormone deficiencies (CPHD), when several pituitary hormones are affected. They are mainly the result of genetic mutations, developmental malformations or the harmful action of env

Approximately 50% of patients with surgically resected early-stage lung cancer develop distant metastasis. At present, there is no in vivo metastasis model to investigate the biology of human lung cancer metastases. Using well-characterized lung adenocarcinoma (LUAD) patient-derived organoids (PDOs), we establish an in vivo metastasis model that preserves the biologic features of human metastases. Results of whole-genome and RNA sequencing establish that our in vivo PDO metastasis model can be used to study clonality and tumor evolution and to identify biomarkers related to organotropism. Inve
