JA-Responsive Transcription Factor SmMYB97 Promotes Phenolic Acid and Tanshinone Accumulation in Salvia miltiorrhiza.

Phenolic acids and tanshinones are active principles in Salvia miltiorrhiza Bunge administered for cardiovascular and cerebrovascular diseases. Jasmonic acid (JA) promotes secondary metabolite accumulation, but the regulatory mechanism is unknown in S. miltiorrhiza. We identified and characterized the JA-responsive gene SmMYB97. Multiple sequence alignment and phylogenetic tree analyses showed that SmMYB97 was clustered with AtMYB11, AtMYB12, and ZmP1 in the subgroup S7 regulating flavonol biosynthesis. SmMYB97 was highly expressed in S. miltiorrhiza leaves and induced by methyl jasmonate (MeJ

The Pentatricopeptide Repeat Gene Family in Salvia miltiorrhiza: Genome-Wide Characterization and Expression Analysis.

The pentatricopeptide repeat (PPR) gene family is one of the largest gene families in plants and plays important roles in posttranscriptional regulation. In this study, we combined whole genome sequencing and transcriptomes to systematically investigate PPRs in Salvia miltiorrhiza, which is a well-known material of traditional Chinese medicine and an emerging model system for medicinal plant studies. Among 562 identified SmPPRs, 299 belong to the P subfamily while the others belong to the PLS subfamily. The majority of SmPPRs have only one exon and are localized in the mitochondrion or chlorop

Continuing treatment with Salvia miltiorrhiza injection attenuates myocardial fibrosis in chronic iron-overloaded mice.

Iron overload cardiomyopathy results from iron accumulation in the myocardium that is closely linked to iron-mediated myocardial fibrosis. Salvia miltiorrhiza (SM, also known as Danshen), a traditional Chinese medicinal herb, has been widely used for hundreds of years to treat cardiovascular diseases. Here, we investigated the effect and potential mechanism of SM on myocardial fibrosis induced by chronic iron overload (CIO) in mice. Kunming male mice (8 weeks old) were randomized to six groups of 10 animals each: control (CONT), CIO, low-dose SM (L-SM), high-dose SM (H-SM), verapamil (VRP) and

Neuroprotection against Aβ25-35-induced apoptosis by Salvia miltiorrhiza extract in SH-SY5Y cells.

The neurotoxicity of β-amyloid protein (Aβ) contributes significantly to the pathogenesis of Alzheimer's disease (AD), and hence the attractive therapeutic strategies focusing on the modulation of Aβ-induced neurotoxicity are warranted. The present study aims to investigate the neuroprotection and underlying mechanisms by which Salvia miltiorrhiza Bunge (Lamiaceae) extract (SME) protects against Aβ25-35-induced apoptosis in SH-SY5Y cells. 2h Pre-treatment of SH-SY5Y cells with SME (0.01, 0.1 or 0.2mg raw herb/ml) concentration-dependently attenuated Aβ25-35-induced cell death, as evidenced by

Abietane diterpenoids from Salvia sclarea transformed roots as growth inhibitors of pathogenic Acanthamoeba spp.

Amoebae from the genus Acanthamoeba are known agents leading to various diseases such as granulomatous amoebic encephalitis (GAE), a chronic progressive disease of the central nervous system, amoebic keratitis (AK), chronic eye infection, amoebic pneumitis (AP), chronic lung infection, and skin infections. It is known that various synthetic anti-Acanthamoeba substances are ineffective. Therefore, other substances, e.g., natural plant compounds, are the focus of biological investigations regarding anti-parasite activity. In this work, the ability of four abietane diterpenoids (ferruginol, salvi

Salvia leriifolia Benth (Lamiaceae) extract demonstrates in vitro antioxidant properties and cholinesterase inhibitory activity.

The object of the present study was to investigate the in vitro antioxidant properties and cholinesterase inhibitory activity of Salvia leriifolia Benth extracts and fractions. The functional role of herbs and spices and their constituents is a hot topic in food-related plant research. Salvia species have been used since ancient times in folk medicine for cognitive brain function and have been subjected to extensive research. Thus, we hypothesize that S leriifolia, because of its functional properties, would be a good candidate to use as a nutraceutical product for improving memory in the elde

Tanshinone IIA isolated from Salvia miltiorrhiza elicits the cell death of human endothelial cells.

Tanshinone IIA, a major component extracted from the traditional herbal medicine, Salvia miltiorrhiza Bunge, is known to exhibit potent cytotoxicity against various human carcinoma cells in vitro. However, the mechanism by which tanshinone IIA produces this anti-tumor effect remains unknown. Since anti-neovascularization has generally been regarded as an effective strategy for anti-cancer therapy, we decided to investigate the mechanism underlying tanshinone IIA-mediated death of human endothelial cells. In this study, we demonstrate that tanshinone IIA elicits human endothelial cell death ind

Long-term administration of Salvia miltiorrhiza ameliorates carbon tetrachloride-induced hepatic fibrosis in rats.

Carbon tetrachloride (CCl4) is metabolized by cytochrome P450 to form a reactive trichloromethyl radical that triggers a chain of lipid peroxidation. These changes lead to cell injury, and chronic liver injury leads to excessive deposition of collagen in liver, resulting in liver fibrosis. The aim of this study was to evaluate the effects of long-term Salvia miltiorrhiza administration in CCl4-induced hepatic injury in rats. Salvia miltiorrhiza (10, 25 or 50 mg kg(-1) twice a day) was given for 9 weeks, beginning at the same time as the injections of CCl4. Rats receiving CCl4 alone showed a de

Network pharmacological prediction on metabolites of dominant endophytic strains from Salvia plebeia R. Br.

Salvia plebeia R. Br. exerts favorable therapeutic effects on multiple diseases such as bronchitis, urticaria and colorectal cancer. Endophytes play vital roles in the growth and development of medicinal plants. They can not only promote plant growth, but also synthesize active ingredients identical to those of host plants, as well as induce hosts to produce active components. For the first time, this study systematically investigated the endophytes of S. plebeia. We evaluated the antagonistic activity and plant growth-promoting traits of endophytes, explored the bacteriostatic effects of stra

[Cloning, expression pattern, and functional verification of phosphate transporter protein SmPht5;1 in Salvia miltiorrhiza].

Vacuolar phosphate transporters have recently been identified as key regulators of intracellular phosphate status in plant cells. However, related studies have not yet been reported on them in Salvia miltiorrhiza. In response, specific amplification primers were designed according to the open reading frame sequence of SmPht5;1 obtained from the S. miltiorrhiza genome, and the full-length SmPht5;1 gene was cloned. Bioinformatics analysis was conducted to characterize the physicochemical properties, subcellular localization, and conserved domains of its encoded protein. The secondary and tertiar

SmNAC68 transcription factor negatively regulate tanshinone biosynthesis under low-temperature conditions in Salvia miltiorrhiza.

Salvia miltiorrhiza Bge. (S. miltiorrhiza) is a traditional Chinese herbal medicine, primarily used for treating cardiovascular diseases. Tanshinones as the main medicinal components of S. miltiorrhiza, their content is an important factor affecting the medicinal and economic value. Reasonable post-harvest techniques are the critical means to improve the effective components, and short-term low-temperature can induce the synthesis of plant secondary metabolites. However, the molecular mechanisms of low-temperature regulation of tanshinones remain unclear. This study found that post-harvest low

Transcriptomic Insights Into Brassinolide-Mediated Control of Bioactive Compound Biosynthesis in Salvia miltiorrhiza Bunge.

BACKGROUND: Brassinolide (BR) plays a pivotal role in regulating plant secondary metabolism and can promote the accumulation of bioactive compounds. However, relatively few studies have explored the application of BR in Salvia miltiorrhiza, and the mechanistic basis for its function in this context remains poorly understood. METHODS: In this study, S. miltiorrhiza hairy roots were treated with BR at 1 μM and 5 μM. The treated samples were analyzed via high-performance liquid chromatography (HPLC)-based chemical quantification, transcriptomic sequencing, and quantitative real-time polymerase c

Salvia miltiorrhiza extract ameliorated ovarian damage induced by cyclophosphamide in mice by multidimensional mechanisms.

INTRODUCTION: Cyclophosphamide (CTX) can result in particularly severe ovarian damage by the broad range of damaging mechanisms, yet now single compounds struggle to provide comprehensive protection against CTX-induced ovarian damage. Salviae Miltiorrhizae Radix et Rhizoma (SM), as the traditional Chinese medicines (TCMs) with multi-component nature, offers hope for prevention and treatment against multi-mechanistic drugs causing ovarian damage. In this study, we aimed to investigate the protective effect of SM on ovarian hypofunction induced by CTX and explore its underlying mechanisms in mic

Oral Salvia officinalis phenolics abolish mortality and mitigate inflammation in acute TNBS murine colitis.

BACKGROUND: This study is the first to evaluate a phenolic extract of Salvia officinalis in the TNBS murine model of colitis. Inflammatory bowel disease (IBD) involves transmural oxidative and nitrosative stress that up-regulates inducible nitric-oxide synthase (iNOS) and cyclo‑oxygenase-2 (COX-2). Although phenolic constituents of S. officinalis are anti-inflammatory in vitro, their efficacy has never been tested in the trinitrobenzene-sulphonic acid (TNBS) model of colitis. METHODS: Aqueous-ethanolic sage leaves were extracted and standardized [(total phenolics = 6208 ± 200 mg gallic acid e
