Many secondary metabolites found in plants have a role in defence against herbivores, pests and pathogens. In this review, a few examples are described and discussed, and some of the problems in determining the precise role(s) of such metabolites highlighted. The role of secondary metabolites in defence may involve deterrence/anti-feedant activity, toxicity or acting as precursors to physical defence systems. Many specialist herbivores and pathogens do not merely circumvent the deterrent or toxic effects of secondary metabolites but actually utilize these compounds as either host recognition c

BACKGROUND: Chemical herbicides are essential for crop productivity, but increasing resistance demands new modes of action. Microbial metabolites offer a promising source of novel phytotoxins. Here, we elucidate the herbicidal mechanism of rubratoxin B (RB), a mycotoxin produced by Talaromyces purpureogenus XJ14C02 strain isolated from soil in Xinjiang, China.
RESULTS: RB exhibited potent root growth inhibition against Echinochloa crus-galli, Conyza canadensis, and Arabidopsis thaliana (IC₅₀ = 14.70, 9.45, and 0.21 μg mL-1, respectively). Integrated multi-omics profiling identified 2373 diffe

This study aimed to evaluate the effects of 4-hydroxyphenylacetic acid (4-HPAA), a phenolic acid compound from Yucca schidigera extract, in Caco-2 cells and artificial rearing of pigeons. In vitro experiments showed that 4-HPAA at different levels exhibited higher cell viability than shikimic acid and 3,4-dihydroxybenzoic acid, and with the highest viability observed at 600 μM. Additionally, 600 μM 4-HPAA alleviated LPS-induced upregulation of IL-1β and CASPASE1 expressions (P < 0.05). In vivo results demonstrated that supplemental 4-HPAA at 500, 1000, and 2000 mg/kg levels did not affect the

Three new shikimate-conjugated meroterpenes, tricycloalternarenes Z1-Z3 (1-3), were isolated from Alternaria alternate, an endophytic fungus residing in Apis mellifera. The structures of new compounds were characterized by analyses of their NMR and HRESIMS data. The absolute configurations were assigned through electronic circular dichroism (ECD) calculations and CD spectra. Bioassay results showed that tricycloalternarene Z1 (1) exhibited moderate anti-inflammatory activity against the LPS-induced production of NO in RAW 264.7 cells, with an IC50 value of 0.93 µM.

Salicylic acid (SA) is an active secondary metabolite that occurs in bacteria, fungi, and plants. SA and its derivatives (collectively called salicylates) are synthesized from chorismate (derived from shikimate pathway). SA is considered an important phytohormone that regulates various aspects of plant growth, environmental stress, and defense responses against pathogens. Besides plants, a large number of bacterial species, such as Pseudomonas, Bacillus, Azospirillum, Salmonella, Achromobacter, Vibrio, Yersinia, and Mycobacteria, have been reported to synthesize salicylates through the NRPS/PK

3-Deoxy-d-arabino-heptulosonate 7-phosphate synthase (DAH7PS) catalyses the first committed step of the shikimate pathway, which produces the aromatic amino acids as well as many other aromatic metabolites. DAH7PS catalyses an aldol-like reaction between phosphoenolpyruvate and erythrose 4-phosphate. Three phosphoenolpyruvate mimics, (R)-phospholactate, (S)-phospholactate and vinyl phosphonate [(E)-2-methyl-3-phosphonoacrylate], were found to competitively inhibit DAH7PS from Neisseria meningitidis, which is the pathogen responsible for bacterial meningitis. The most potent inhibitor was the v

Chorismate synthase is the last enzyme of the common shikimate pathway, which catalyzes the anti-1,4-elimination of the 3-phosphate group and the C-(6proR) hydrogen from 5-enolpyruvylshikimate 3-phosphate (EPSP) to generate chorismate, a precursor for the biosynthesis of aromatic compounds. Enzyme activity relies on reduced FMN, which is thought to donate an electron transiently to the substrate, facilitating C(3)-O bond breakage. The crystal structure of the enzyme with bound EPSP and the flavin cofactor highlighted two invariant serine residues interacting with a bound water molecule that is

Soybean cyst nematode (SCN, Heterodera glycines) causes significant soybean yield losses. The Rhg1 locus is a major contributor to SCN resistance and contains three genes that mediate this trait including Rhg1-GmAAT (Glyma.18G022400), which encodes the putative amino acid transporter AATRhg1. The molecular function of AATRhg1 in SCN resistance is not understood. In this study, rhg1-b soybean lines with Rhg1-GmAAT silencing demonstrated that Rhg1-GmAAT can contribute resistance against HG 0 SCN and also against problematic HG 2.5.7 populations that partially overcome rhg1-b-mediated resistance.

UNLABELLED: Recombinant attenuated Salmonella enterica serovar Typhimurium strains are believed to act as powerful live vaccine carriers that are able to elicit protection against various pathogens. Auxotrophic mutations, such as a deletion of aroA, are commonly introduced into such bacteria for attenuation without incapacitating immunostimulation. In this study, we describe the surprising finding that deletion of aroA dramatically increased the virulence of attenuated Salmonella in mouse models. Mutant bacteria lacking aroA elicited increased levels of the proinflammatory cytokine tumor necro

GC-MS-based metabolite profiling was used to analyse the response of Medicago truncatula cell cultures to elicitation with methyl jasmonate (MeJa), yeast elicitor (YE), or ultraviolet light (UV). Marked changes in the levels of primary metabolites, including several amino acids, organic acids, and carbohydrates, were observed following elicitation with MeJa. A similar, but attenuated response was observed following YE elicitation, whereas little response was observed following UV elicitation. MeJa induced the accumulation of the triterpene beta-amyrin, a precursor to the triterpene saponins, a

Pneumocystis carinii pneumonia remains a prevalent opportunistic disease among immunocompromised individuals. Although aggressive prophylaxis has decreased the number of acute P. carinii pneumonia cases, many patients cannot tolerate the available drugs, and experience recurrence of the infection, which can be fatal. It is now generally agreed that the organism should be placed with the fungi, but the identification of extant fungal species representing its closest kins, remains debated. Most recent data indicate that P. carinii represents a diverse group of organisms. Since the lack of method

Diseases caused by plant-parasitic nematodes in vegetables, among them Meloidogyne spp. root-knot nematodes (RKNs), lead to extensive yield decline. A molecular understanding of the mechanisms underlying plants' innate resistance may enable developing safe alternatives to harmful chemical nematicides in controlling RKNs. A tight relationship has been revealed between the WRKY transcription factors and RKN parasitism on tomato roots. We investigated the function role of tomato SlWRK3 and SlWRKY35 in regulating nematode disease development. Using promoter-GUS reporter gene fusions, we show that

Rosmarinus officinalis is an evergreen aromatic plant with important commercial interest as it contains numerous essential oils (composed of terpenoid compounds) and phenolic constituents (natural antioxidant compounds). This work aims at evaluating the concomitant effects of different inorganic and organic fertilization treatments and the subsequent increases in soil nutrient availability on terpenoids and other carbon-based secondary metabolites, e.g., flavonoids and phenolic compounds, in Rosmarinus officinalis leaves. The results showed that, as expected, the structural carbohydrate conten

Scientists are concerned by an ever-increasing rise in bacterial resistance to antibiotics, particularly in diseases such as malaria, toxoplasmosis, tuberculosis, and pneumonia, where the currently used therapies become progressively less efficient. It is therefore necessary to develop new, safe, and more efficient antibiotics. Recently, the existence of the shikimic acid pathway has been demonstrated in certain parasites such as the malaria parasite. These types of parasites cause more than a million casualties per year, and their effects are particularly strong in people with a compromised i

BACKGROUND: Preeclampsia is a severe pregnancy-related disorder characterized by hypertension and proteinuria, and affects approximately 2–8% of pregnancies worldwide. It poses substantial risks to both maternal and fetal health, including preterm birth, intrauterine growth restriction, and life-threatening complications, such as eclampsia and organ failure. However, the mechanisms underlying preeclampsia, especially those involving blood-associated proteins, are poorly understood. This study aimed to identify critical blood proteins linked to severe preeclampsia and explore their potential me

The recent recrudescence of Mycobacterium tuberculosis infection and the emergence of multidrug-resistant strains have created an urgent need for new therapeutics against tuberculosis. The enzymes of the shikimate pathway are attractive drug targets because this route is absent in mammals and, in M. tuberculosis, it is essential for pathogen viability. This pathway leads to the biosynthesis of aromatic compounds, including aromatic amino acids, and it is found in plants, fungi, bacteria, and apicomplexan parasites. The aroB-encoded enzyme dehydroquinate synthase is the second enzyme of this pa

Neuroinflammation is a key characteristic associated with neurological disorders, particularly depression and anxiety. This study aims to evaluate the neuroprotective and antidepressant-like effects of pine needle (PN) extracts in an LPS-induced neuroinflammation mouse model. Following seven days of oral administration of PN, the tail suspension test demonstrated a significant reduction in immobility time in PN-treated mice compared to LPS controls, surpassing the effect of the standard antidepressant bupropion. To elucidate the underlying mechanisms, we conducted a whole-genome microarray ana

Targeted enrichment of tocopherols in fungal fermentation systems remains a significant metabolic engineering challenge. This study presented a novel strategy employing homogentisic acid (HGA) modulation to efficiently enhance tocopherol production in the M. purpureus-fermented coix seed (MFCS) system. Three strategies of exogenous HGA supplementation (Exo-HGA), endogenous HGA accumulation (End-HGA), and the combination of exogenous HGA supplementation and endogenous HGA accumulation (Exo + End-HGA) were systematically investigated. While End-HGA optimally promoted lovastatin, pigment, and bio
