Lipopolysaccharide-induced anti-inflammatory acute phase response is enhanced in spermidine/spermine N1-acetyltransferase (SSAT) overexpressing mice.

Bacterial lipopolysaccharide (LPS) is an effective activator of the components of innate immunity. It has been shown that polyamines and their metabolic enzymes affect the LPS-induced immune response by modulating both pro- and anti-inflammatory actions. On the other hand, LPS causes changes in cellular polyamine metabolism. In this study, the LPS-induced inflammatory response in spermidine/spermine N(1)-acetyltransferase overexpressing transgenic mice (SSAT mice) was analyzed. In liver and kidneys, LPS enhanced the activity of the polyamine biosynthetic enzyme ornithine decarboxylase and incr

The Protective Effect of Topical Spermidine on Dry Eye Disease with Retinal Damage Induced by Diesel Particulate Matter2.5.

Air pollutants, especially ambient fine particulate matter2.5, may contribute to various ocular surface disorders, including dry eye disease, keratitis and conjunctivitis. A natural polyamine spermidine has a protective effect on the retina and optic nerve; however, no study has been conducted on the application of spermidine in particulate matter2.5-induced dry eye disease. In the present study, we investigated the effect of spermidine eye drops in topically exposed particulate matter2.5-induced dry eye models of Sprague-Dawley rats, by hematological, biochemical and histological evaluation.

Spermidine determines the sensitivity to the calmodulin antagonist, chlorpromazine, for the circadian conidiation rhythm but not for the mycelial growth in Neurospora crassa.

The gene that suppresses the phenotype of the cpz-2 mutation, which results in changing the sensitivity to chlorpromazine in relation to mycelial growth and circadian rhythms, was cloned in Neurospora crassa. This gene is not the cpz-2 gene itself but rather is identical to the spe-3 gene that encodes spermidine synthase in Neurospora. The intracellular content of spermidine was lowered in the cpz-2 strain compared to that of the wild-type strain. By integration of the spe-3 gene or by the addition of spermidine into culture medium, the temperature sensitivity of mycelial growth was lost and t

Reproductive aging drives deterministic microbiota assembly to mitigate uterine oxidative phosphorylation impairment via spermidine production in laying hens.

BACKGROUND: Reproductive aging represents a critical physiological bottleneck characterized by a progressive decline in tissue homeostasis and physiological function. While the gut microbiota is known to shift during host aging, the ecological forces governing the assembly of the reproductive microbiota and its functional feedback on uterine homeostasis remain poorly understood. RESULTS: We demonstrated that uterine aging drives a transition from stochastic to deterministic microbial community assembly, selecting for a microbiota enriched in Rhodococcus in aged laying hens. A Spermidine Derivative Ameliorates Dextran Sulfate Sodium-Induced Colitis in Mice by Inhibiting the MAPK4/AKT Signaling Pathway.

Ulcerative colitis (UC) is a chronic inflammatory bowel disease characterized by recurrent episodes and an inability to achieve a complete cure. The spermidine derivative (di-p-coumaroyl-caffeoyl spermidine, SPDD), as a key alkaloid, exhibits unique health benefits. However, it has not yet been reported whether SPDD can improve dextran sulfate sodium (DSS)-induced colitis in mice. Herein, we investigated the effects and mechanisms of SPDD on DSS-induced colitis in mice. SPDD was successfully purified from rose bee pollen and was found to have a protective effect on colitis, evidenced by reduce

Spermidine Prevents Polarity Loss of Absorptive Enterocytes in Jejunum of Lipopolysaccharide-Challenged Mice via 4D-DIA Proteomics Analysis.

Identifying effective compounds to restore the polarity of absorptive enterocytes (AEs) holds promise for mitigating the severity and duration of small intestinal disorders. Spermidine (SPD) is a natural polyamine; whether it can repair inflammation-induced loss of AE polarity remains unclear. In this study, we employed lipopolysaccharide (LPS)-challenged mice models combined with 4D data-independent acquisition (DIA) proteomics to investigate the mechanisms by which SPD alleviates polarity loss in AEs. Our results demonstrated that SPD supplementation enhanced the antioxidant capacity and imp

Spermidine alleviates sepsis-induced acute kidney injury by suppressing apoptosis and inflammation through modulation of TLR4-mediated signaling.

Sepsis-associated acute kidney injury (AKI) remains a critical clinical challenge with limited therapeutic options. This study investigated the renoprotective effects of spermidine using sepsis-associated AKI models. In vitro, HK-2 cells were challenged with lipopolysaccharide (LPS) to establish an inflammatory injury model, followed by spermidine treatment (25-100 μM). In vivo, a cecal ligation and puncture (CLP)-induced septic AKI mouse model was employed. Key methodologies included CCK-8 viability assay, flow cytometry apoptosis analysis, western blotting (apoptotic proteins: Bcl-2/Bax/clea

Exogenous spermidine enhances the photosynthetic and antioxidant capacity of rice under heat stress during early grain-filling period.

High temperature has adverse effects on rice growth by inhibiting the flag leaf photosynthetic and antioxidant capacity, which can be alleviated by various exogenous chemicals such as spermidine (Spd). However, the role of Spd in conferring heat tolerance in rice is not well documented. Conventional japonica rice varieties Wuyunjing 24 and Ningjing 3 were treated with high temperatures at 37.5/27.0°C (day/night) and foliar sprayed with 1mmolL-1 Spd after flowering. Results showed activities of superoxide dismutase (SOD) and peroxidase (POD) activities were deceased during high temperature trea

Spermidine ameliorates 3-nitropropionic acid (3-NP)-induced striatal toxicity: Possible role of oxidative stress, neuroinflammation, and neurotransmitters.

AIM AND BACKGROUND: Spermidine, a naturally occurring polyamine, is involved in various internal biological functions, due to its antioxidant and anti-inflammatory properties. A decreased level of polyamines is associated with aging and neurodegenerative disorders including Huntington disease (HD). 3-Nitropropanoic acid (3-NP) induces a spectrum of HD-like neuropathologies in rat striatum and thus serves as a good experimental model of HD. Therefore, the aim of the present study was to investigate whether spermidine confers neuroprotection against 3-NP-induced striatal toxicity and to explore

Hepatocyte-specific ablation of spermine/spermidine-N1-acetyltransferase gene reduces the severity of CCl4-induced acute liver injury.

Activation of spermine/spermidine-N(1)-acetyltransferase (SSAT) leads to DNA damage and growth arrest in mammalian cells, and its ablation reduces the severity of ischemic and endotoxic injuries. Here we have examined the role of SSAT in the pathogenesis of toxic liver injury caused by carbon tetrachloride (CCl(4)). The expression and activity of SSAT increase in the liver subsequent to CCl(4) administration. Furthermore, the early liver injury after CCl(4) treatment was significantly attenuated in hepatocyte-specific SSAT knockout mice (Hep-SSAT-Cko) compared with wild-type (WT) mice as deter

VDR-Spermidine Axis Protects Against Age-Related Granulosa Cell Dysfunction and Follicular Decline via DNMTs-Mediated p53 Methylation.

Ovarian aging, marked by a decline in follicle quantity and quality, is a complex process whose underlying mechanisms remain elusive. Here, we identify the vitamin D receptor (Vdr) as a key anti-aging transcription factor whose expression in granulosa cells (GCs) declines with age. Using GCs-specific Vdr knockout (cVKO) mice and a Vdr-knockout (VKO) human granulosa-like cell line, we demonstrate that loss of Vdr triggers GCs aging and disrupts ovarian function. Integrated transcriptomic and metabolomic analyses from VKO and WT cells revealed that Vdr loss downregulates the de novo spermidine (

Spermidine suppresses liver fibrosis by remodeling the communication signal between liver sinusoidal endothelial cells and hepatic stellate cells.

Hepatic fibrosis is a pivotal stage in which chronic liver disease progresses from reversible injury to decompensation. Liver sinusoidal endothelial cells (LSECs) play a regulatory role in hepatic stellate cells (HSCs) activation through paracrine signaling; therefore, maintaining the physiological phenotype of LSECs is critical for antifibrotic intervention. Spermidine (SPD) has been recognized for its antifibrotic properties; however, its impact on LSECs' function and the underlying mechanism remains largely unknown. In this study, analysis of NHANES data revealed an inverse association betw

Roe deer uterine fluid metabolome reveals elevated glycolysis, fatty acid breakdown, and spermidine synthesis upon reactivation from diapause†.

The blastocyst of the European roe deer (Capreolus capreolus) undergoes a period of decelerated growth and limited metabolism. During this period known as embryonic diapause, it floats freely in the uterus encircled by the histotroph. Prior to implantation, reactivation is marked by rapid embryonic growth and conceptus elongation. We hypothesized that the uterine fluid, which is known to undergo changes in its composition to support early embryonic development, contributes to controlling embryonic growth during diapause and elongation. We therefore characterized the pre-implantation uterine fl

Spermidine mitigates ferroptosis in free fatty acid-induced AML-12 cells through the ATF4/SLC7A11/GCLM/GPX4 pathway.

Non-alcoholic fatty liver disease (NAFLD) is a prominent cause of chronic liver disease worldwide. Spermidine (SPD), a naturally occurring polyamine, has shown potential in alleviating the accumulation of hepatic lipids and reducing NAFLD symptoms in overweight mice. Nonetheless, the specific mechanisms through which SPD exerts its effects remain largely unknown. This study seeks to explore the protective effects of SPD on NAFLD and to clarify the underlying mechanisms. An in vitro model of NAFLD was established by inducing steatosis in AML-12 cells through the use of free fatty acids (FFAs).

Hyaluronic acid methacrylate/Pluronic F127 hydrogel enhanced with spermidine-modified mesoporous polydopamine nanoparticles for efficient synergistic periodontitis treatment.

Bacterial infection, reactive oxygen species (ROS) accumulation, and persistent inflammation pose significant challenges in the treatment of periodontitis. However, the current single-modal strategy makes achieving the best treatment effect difficult. Herein, we developed a double-network hydrogel composed of Pluronic F127 (PF-127) and hyaluronic acid methacrylate (HAMA) loaded with spermidine-modified mesoporous polydopamine nanoparticles (M@S NPs). The PF-127/HAMA/M@S (PH/M@S) hydrogel was injectable and exhibited thermosensitivity and photocrosslinking capabilities, which enable it to adapt

Exogenous spermidine alleviates diabetic cardiomyopathy via suppressing reactive oxygen species, endoplasmic reticulum stress, and Pannexin-1-mediated ferroptosis.

Diabetic cardiomyopathy (DCM) is a serious complication and death cause of diabetes mellitus (DM). Recent cardiology studies suggest that spermidine (SPD) has cardioprotective effects. Here, we verified the hypothesis of SPD's protective effects on DCM. Therefore, db/db mice and primary neonatal mouse cardiomyocytes were used to observe the effects of SPD. Immunoblotting showed that ornithine decarboxylase (ODC) and SPD/spermine N1-acetyltransferase (SSAT) were downregulated and upregulated in the myocardium of db/db mice, respectively. We found that diabetic mice showed cardiac dysfunction in

Identification of two natural compound inhibitors of Leishmania donovani Spermidine Synthase (SpdS) through molecular docking and dynamic studies.

Visceral leishmaniasis caused by the protozoan Leishmania donovani is the most severe form of leishmaniasis and it is potentially lethal if untreated. Despite the availability of drugs for treating the disease, the current drug regime suffers from drawbacks like antibiotic resistance and toxicity. New drugs have to be discovered in order to overcome these limitations. Our aim is to identify natural compounds from plant sources as putative inhibitors considering the occurrence of structural diversity in plant sources. Spermidine Synthase (SpdS) was chosen as the target enzyme as it plays a vita
