Crevicular IgG antibodies and Streptococcus mutans on erupting human first permanent molars.

The specific IgG antibody activity against indigenous Streptococcus mutans in gingival crevicular fluid (GCF) was determined in 25, 6-7-yr-old children. Samples of plaque and GCF were collected from all erupting first permanent molars. Plaque samples were also collected from the adjacent second primary molars. The presence of clinical caries was scored as defs. In the same individual a highly significant correlation (p less than 0.005) was found between the level of specific antibody activity in GCF when comparing first permanent molars with each other. No correlation was found between the spe

Effect of Surface Modification of a Dental Composite on the Adhesion of Streptococcus mitis, Streptococcus mutans, and Candida albicans: An In Vitro Study.

Adhesion of different microorganisms to the surface of dental materials has generated significant interest since one of the most important requirements of biomaterials to be considered successful is their ability to withstand the damage caused by microorganisms that may lead to failure and the onset of different pathologies, such as caries. In vitro testing has demonstrated that surface modification is an alternative approach to reduce the adhesion of microorganisms to surfaces. The objective of this work was to assess the adhesion of Streptococcus mitis, Streptococcus mutans, and Candida albi

Susceptibility of Streptococcus mutans and Streptococcus sobrinus to cell wall inhibitors and development of a novel selective medium for S. sobrinus.

Representative strains of Streptococcus mutans and Streptococcus sobrinus showed differences in susceptibility to members of the monobactam group of beta-lactam antibiotics: S. sobrinus was less sensitive than S. mutans. The minimum inhibitory concentrations of aztreonam (AZT) and carumonam, both of which belong to this group, were 2,000 microg/ml for S. sobrinus and 125 microg/ml for S. mutans. Further addition of fosfomycin, bacitracin and sodium chloride to Mitis Salivarius agar (MS) supplemented with AZT resulted in growth inhibition of S. mutans and oral streptococci other than S. sobrinu

Anti-Cariogenic Potential of Lyophilized Cell-Free Supernatant of Probiotic Pediococcus pentosaceus GS4 against Streptococcus mutans and Streptococcus anginosus.

Streptococcus species, particularly Streptococcus mutans and Streptococcus anginosus, are recognized globally for their role in causing dental caries and other dental issues. Current treatments using fluoride products and conventional antibiotics are often ineffective due to the resilience of biofilms on tooth surfaces. To overcome these challenges, novel approaches that eliminate these pathogens are much needed. In this regard, we have prepared CFS (cell-free supernatant), a postbiotic preparation of our probiotic strain Pediococcus pentosaceus GS4, and investigated the anticariogenic propert

Streptococcus mutans-derived extracellular vesicles promote herpes simplex virus infection in oral epithelia.

The oral cavity contains the second most diverse bacterial community after the intestines, with bacteria and viruses coexist. Streptococcus mutans is a major pathogenic bacterium in the oral cavity, commonly associated with dental caries. We investigated the effects of S. mutans-derived extracellular vesicles (Sm EVs) on herpes simplex virus 1 (HSV-1) infection, which is prevalent in the oral cavity. We performed our experiments in human oral keratinocyte (HOK) cells and mucosal tissue-derived organoids, and analyzed human whole saliva (n = 50) for associations between S. mutans and HSV-1 enve

Nano-chitosan modified restorative materials suppress Streptococcus mutans biofilm and virulence gene expression.

Secondary caries and biofilm production remain a persistent concern, prompting the development of antibacterial restorative materials. However, worries about synthetic additives’ safety and long-term effects urge the use of natural, biocompatible alternatives. This study explored the antibacterial efficacy of chitosan (CH) and nano-chitosan (NCH) incorporated into different restorative materials: amalgam (Am), glass ionomer cement (G), and resin composite (R). This study employs a direct comparative analysis of both modified and unmodified restorative materials against Am to evaluate their eff

Food-grade Lacticaseibacillus paracasei postbiotics suppress oral Streptococcus mutans biofilm formation and cariogenicity.

Dental caries is a highly prevalent non-communicable disease driven by dysbiosis of the oral microbiota, in which Streptococcus (S.) mutans plays a keystone role. We discovered that cell-free supernatant (CFS) from food-associated Lacticaseibacillus (L.) paracasei disrupts S. mutans membranes, causing leakage, reduced viability, and decreased surface hydrophobicity. CFS also inhibits biofilms by decreasing biomass, metabolic activity, chain length, and exopolysaccharide (EPS) accumulation. Neutralization experiments revealed organic acids as the primary antibacterial factors: activity weakened

Berberine chloride hydrate impairs Streptococcus mutans biofilm formation via inhibiting sortase A activity.

Dental caries is a biofilm-associated chronic progressive disease, results from the dissolution of mineralized tooth tissue by acidic generated from bacterial sugar fermentation. S. mutans, a prominent pathogen of dental caries, is acknowledged for its role in cariogenic biofilm formation, utilizing Sortase A (SrtA) to catalyse surface proteins, thus promoting biofilm formation. In our previous studies, the inhibitory effect of the berberine chloride hydrate (BH) on S. mutans biofilms was confirmed. Here, we further investigate the influence of BH on S. mutans biofilm-induced bovine enamel car

Anandamide Alters Glycolytic Activity in Streptococcus mutans: Metabolomics and Stable Isotope Labeling Study.

Streptococcus mutans (S. mutans) is a cariogenic bacterium in the oral cavity that plays a significant role in plaque formation and dental caries. In previous research by our group, we showed that the endocannabinoid anandamide (AEA) has anti-bacterial and anti-biofilm activities against S. mutans. Here, we aimed to investigate its effects on S. mutans through metabolomics analyses. S. mutans was cultivated in the absence or presence of AEA at a sub-minimum inhibitory concentration (MIC), and changes in metabolites and metabolic pathways were assessed through liquid chromatography-mass spectro

Polyketides/nonribosomal peptides from Streptococcus mutans and their ecological roles in dental biofilm.

Streptococcus mutans is the major etiological agent of dental caries in humans. S. mutans overgrowth within dental biofilms can trigger biofilm dysbiosis, ultimately leading to the initiation or progression of dental caries. Polyketides and nonribosomal peptides (PKs/NRPs) are secondary metabolites with complex structures encoded by a cluster of biosynthetic genes. Although not essential for microbial growth, PKs/NRPs play important roles in physiological regulation. Three main classes of hybrid PKs/NRPs in S. mutans have been identified, including mutanobactin, mutanocyclin, and mutanofactin,

Targeted elimination of cariogenic Streptococcus mutans biofilms via Cu,Fe-doped chitosan nanozyme.

Human dental caries is an intractable biofilm-associated disease caused by the symbiotic cariogenic bacteria, but how to target effectively eliminate cariogenic bacterial and their biofilms without affecting normal bacteria still remains great challenges. To address this issue, we reported Cu,Fe-doped chitosan-based nanozyme (i.e. CS@Cu,Fe) that exhibits well peroxidase-like activity at acidic environment of caries, and kill S. mutans and S. sanguinis without impacting the normal S. oralis. The synergistic interaction between Cu and Fe could effectively enhance the efficiency of electron trans
