Endogenous Glandular Chemistry and Methyl Eugenol-derived Metabolites in the Pheromone Communication of Bactrocera umbrosa.

Bactrocera umbrosa (Fabricius) is an oligophagous fruit fly of economic importance that infests mainly Artocarpus fruits (Moraceae). The sexual communication system of this species is poorly understood, partly due to difficulties in establishing laboratory colonies. Males of B. umbrosa are strongly attracted to and voraciously fed on methyl eugenol (ME), a naturally occurring plant secondary compound. Here, we elucidate the endogenously produced and ME-derived chemical signals involved in the species' communication. Gas chromatography-mass spectrometry analyses of pheromonal glands revealed th

Trichlorfon and spinosad resistance survey and preliminary determination of the resistance mechanism in Pakistani field strains of Bactrocera dorsalis.

The use of insecticides has been a primary tool to manage Bactrocera dorsalis in Pakistan; however, recent reports of field control failures necessitate mapping out the insecticide resistance problem. Therefore, eight field strains from Pakistan, were evaluated for their resistance against trichlorfon and spinosad. Compared with a reference strain, six field strains showed high levels of resistance to trichlorfon, while two field strains expressed intermediate resistance. In case of spinosad, five field strains fell in the susceptible range, whereas, the rest of the strains represented minor r

High hydrostatic pressure at low temperature as a quarantine treatment to improve the quality of fruits.

Application of high hydrostatic pressure has been proposed as an alternative quarantine process for the Mexican fruit fly, Anastrepha ludens Loew (Diptera: Tephritidae), one of the most important insects infesting mangoes, citrus fruits, and other fruits in Mexico and other Latin American countries. The present study was performed to determine the effect of high pressure treatments at 0 degrees C on the survivorship of eggs and larvae of this pest. The effect of time and pressure level at near-freezing temperatures on the egg hatch and survival of larvae is discussed. Eggs and larvae were pres

Clock genes regulate sex pheromone production and male mating ability in Bactrocera dorsalis.

Many animals display physiological and behavioral activities limited to specific times of the day. Certain insects exhibit clear daily rhythms in their mating activities that are regulated by an internal biological clock. However, the specific genetic mechanisms underlying this regulation remain largely unexplored. Mating in the fruit fly Bactrocera dorsalis exhibits a daily rhythm and is dependent on sex pheromones produced in the male rectum. We used transcriptome sequencing and clustered regularly interspaced short palindromic repeats (CRISPR) / CRISPR-associated nuclease 9 techniques to un

Dominance of an invasive fruit fly species, Bactrocera invadens, along an altitudinal transect in Morogoro, Eastern Central Tanzania.

Bactrocera invadens, a fruit fly from Asia, is an invasive pest species across Africa. It appears to continue spreading, not only in latitude but also in altitude. To assess its capacity to infest a large variety of hosts and its competition with other fruit fly species, a study along an altitudinal gradient was conducted. At low altitudes, the high abundance in the field and high infestation of B. invadens in different fruit species make it a serious pest. At high altitudes, colonization has started and B. invadens occurs in low numbers by reproducing successfully in high altitude fruits. Ove

Two heat shock cognate 70 genes involved in spermatogenesis regulate the male fertility of Zeugodacus cucurbitae, as potential targets for pest control.

The melon fly Zeugodacus cucurbitae Coquillett (Diptera: Tephritidae) is an agricultural quarantine pest threatening fruit and vegetable production. Heat shock cognate 70 (Hsc70), which is a homolog of the heat shock protein 70 (Hsp70), was first discovered in mice testes and plays an important role in spermatogenesis. In this study, we identified and cloned five Hsc70 genes from melon fly, namely ZcHsc70_1/2/3/4/5. Phylogenetic analysis showed that these proteins are closely related to Hsc70s from other Diptera insects. Spatiotemporal expression analysis showed that ZcHsc70_1 and ZcHsc70_2 ar

Follicular atresia during Dacus oleae oogenesis.

Programmed cell death, constitutes a common fundamental incident that occurs during oogenesis in a variety of different animals. It plays a significant role in the maturation process of the female gamete and also in the removal of abnormal and superfluous cells at certain checkpoints of development. In the present study, we demonstrate the existence of follicular atresia during mid-oogenesis in the olive fruit fly Dacus oleae (Tephritidae). The number of atretic follicles increases following the age of the fly, suggesting for the presence of an age-susceptible process. The atretic follicles co

Predicting range expansion of three tropical fruit flies and risks to pome and stone fruit production in temperate regions.

Human activities and climate change are driving the northward expansion of invasive fruit flies (Diptera: Tephritidae), posing increasing threats to temperate fruit production. Here, we combine the MaxEnt model with direct field observations to assess the potential range expansion of three tephritid species-Bactrocera dorsalis, Zeugodacus tau, and Z. cucurbitae-under climate change scenarios through 2060, and to evaluate the associated risks to temperate fruit production. Model projections indicate that the suitable habitat area for all three species will expand significantly in the future and

Targeting Trehalose-Glucose Metabolism to Disrupt Symbiont-Mediated Pyrazine Sex Pheromone Synthesis in Bactrocera dorsalis (Oriental Fruit Fly).

Bactrocera dorsalis is an invasive pest causing severe economic losses. Its reproduction depends on sex pheromone-mediated courtship. We previously found that symbiotic Bacillus in the male rectum produce pyrazine sex pheromones (2,3,5-trimethylpyrazine/2,3,5,6-tetramethylpyrazine, TMP/TTMP) in a glucose-dependent manner, but whether host trehalose-to-glucose conversion regulates this remains unknown. Here, we show that mature males have higher rectal glucose and lower trehalose than females. Trehalase (Treh), which hydrolyzes trehalose to glucose, is enriched in the male gut and rectum, where

Dual mechanisms of olfactory plasticity: Transient modulation by phosphatidylcholine and D-chiro-inositol and sustained regulation via the sex peptide-juvenile hormone system.

Insects exhibit olfactory plasticity to optimize their survival and reproduction, which ensures that their sensory preferences flexibly meets their needs. However, a comprehensive understanding of the sequential regulatory mechanisms driving these sensory adjustments is lacking. Addressing this knowledge gap is crucial for clarifying how the olfactory system of insect flexibly meets their reproductive needs. Using electroantennogram (EAG) and single-sensillum recordings, we found that female Bactrocera dorsalis (Hendel) (Diptera: Tephritidae) displayed distinct olfactory preferences for ovipos

Antibiotics and Pesticides Enhancing the Transfer of Resistomes among Soil-Bayberry-Fruit Fly Food Chain in the Orchard Ecosystem.

While substantial amounts of antibiotics and pesticides are applied to maintain orchard yields, their influence on the dissemination and risk of antibiotic resisitome in the orchard food chain remains poorly understood. In this study, we characterized the bacterial and fungal communities and differentiated both antibiotic resistance genes (ARGs) and virulence factor genes (VFGs) in the soil, Chinese bayberry (matured and fallen), and fruit fly gut, collected from five geographic locations. Our results showed that fruit fly guts and soils exhibit a higher abundance of ARGs and VFGs compared wit

A Laboratory Bioassay Method to Assess the Use of Toxic Bait on Anastrepha fraterculus (Weidemann 1830).

The lack of standardization of bioassays for the evaluation of toxic bait toxicity on the South American fruit fly, Anastrepha fraterculus (Weidemann 1830), has led to erroneous interpretations of assay results. The objective of this study was to develop a methodology for the standardization and validation of toxicological tests on A. fraterculus toxic bait using the Success™ 0.02CB formulation (80 mg L-1 of spinosad). Anastrepha fraterculus adults, obtained from larvae reared on an artificial diet, showed higher susceptibility (LT50 = 48.96 h) than adults from larvae reared on cattley guava (

A PCR-based diagnostic assay for detecting DNA of the olive fruit fly, Bactrocera oleae, in the gut of soil-living arthropods.

Bactrocera oleae (Rossi) (Diptera: Tephritidae) is considered the most devastating pest of the olive tree worldwide. In an effort to develop management and biological control strategies against this pest, new molecular tools are urgently needed. In this study, we present the design of B. oleae-specific primers based on mitochondrial DNA sequences of cytochrome oxidase subunit I (COI) gene. Two pairs of B. oleae-specific primers were successfully designed and named as SBo1-F/SBo1-R and SBo2-F/SBo1-R, being able to amplify 108 and 214 bp COI fragments, respectively. The specificity of designed p

Effects of the olfactory environment and nutrition on the ability of male Mediterranean fruit flies to endure starvation.

The Mediterranean fruit fly, Ceratitis capitata (Wiedemann) (Diptera: Tephritidae), is targeted for control using the sterile insect technique (SIT). For this technique to succeed, released males must be able to compete with wild males for copulations. Male success is mediated by survival in the field often in adverse conditions. Manipulation of the postteneral environment experienced by sterile males before release has been shown to affect male sexual success and survival. The objectives of this study were to determine how various diets, combined with exposure to volatiles containing alpha-co

The Gene Editing of Eukaryotic Translation Initiation Factor Binding Protein 3 and Its Potential Role in Rapid Cold Hardening of Two Invasive Fruit Flies.

Bactrocera dorsalis (Hendel) and Bactrocera correcta (Bezzi) are two globally concerning quarantine pests within the genus Bactrocera (Tephritidae). Both species inflict severe damage on the agricultural industry and international export trade. Their distribution range has expanded due to global warming, posing an increasing threat to fruit production. Heat shock proteins (HSPs), functioning as molecular chaperones, are known to contribute to temperature adaptation in insects. However, the cold adaptation regulatory mechanisms in these two Bactrocera species remain unclear. In this study, eIF4

ChIP-seq profiling of H3K4me3 and H3K27me3 in an invasive insect, Bactrocera dorsalis.

Introduction: While it has been suggested that histone modifications can facilitate animal responses to rapidly changing environments, few studies have profiled whole-genome histone modification patterns in invasive species, leaving the regulatory landscape of histone modifications in invasive species unclear. Methods: Here, we screen genome-wide patterns of two important histone modifications, trimethylated Histone H3 Lysine 4 (H3K4me3) and trimethylated Histone H3 Lysine 27 (H3K27me3), in adult thorax muscles of a notorious invasive pest, the Oriental fruit fly Bactrocera dorsalis (Hendel) (

Insecticidal Activity of Ricinus communis Leaf Extracts Against Bactrocera zonata and Bactrocera cucurbitae: Identification of Potential Bioactive Compounds.

The melon fruit fly, Bactrocera zonata, and the pumpkin fruit fly, Bactrocera cucurbitae (Diptera: Tephritidae), are important agricultural pests that cause significant losses in a wide range of fruit and vegetable crops. Increasing concerns over pesticide resistance and environmental contamination have driven interest in plant-derived insecticide alternatives. This study investigated the insecticidal activity of Ricinus communis L. leaf extracts against both species. The crude leaf extract was fractionated using solvents of increasing polarity (n-hexane, methanol, and ethyl acetate). The resu

Spatio-temporal temperature variations in MarkSim multimodel data and their impact on voltinism of fruit fly, Bactrocera species on mango.

Fruit flies are the most serious economic insect pests of mango in India and other parts of the world. Under future climate change, shifts in temperature will be a key driver of ecosystem function especially in terms of insect pest dynamics. In this study, we predicted the voltinism of the three economically important fruit fly species viz., Bactrocera dorsalis (Hendel), Bactrocera correcta (Bezzi) and Bactrocera zonata (Saunders) of mango from 10 geographical locations in India using well established degree day approaches. Modeling the cost-effectiveness of insect rearing on artificial diets: A test with a tephritid fly used in the sterile insect technique.

We modeled the cost-effectiveness of rearing Anastrepha ludens, a major fruit fly pest currently mass reared for sterilization and release in pest control programs implementing the sterile insect technique (SIT). An optimization model was generated by combining response surface models of artificial diet cost savings with models of A. ludens pupation, pupal weight, larval development time and adult emergence as a function of mixtures of yeast, a costly ingredient, with corn flour and corncob fractions in the diet. Our model revealed several yeast-reduced mixtures that could be used to prepare d
