Background Trained immunity (TI) refers to a hyperresponsive functional state of innate immune cells, such as inflammatory monocytes, induced in response to a variety of stimuli including signals released from damaged tissue, and characterised by increased cytokine production. Inflammatory monocytes play a critical role in Duchenne muscular dystrophy (DMD) by driving chronic muscle inflammation and disease progression. In this study we examined the role of trained immunity in dystrophic muscle pathology by specifically targeting this process in monocytes. Methods We used the mdx mouse model of

INTRODUCTION: Salmonella enterica subsp. enterica serovar Choleraesuis is a pathogen that can cause severe systemic disease and remains a challenge in swine production due to its impact on animal health, economic losses, and zoonotic risk. While vaccination is a key control tool, current vaccines often provide limited protection to the targeted (and untargeted) serovar. Trained immunity has gained attention as a possible way to enhance non-specific protection.
METHODS: This study explored whether a porcine-derived BCG strain (dpB), administered live or inactivated, could improve the protectiv

Trained immunity enhances innate host defense by endowing monocytes with memory-like properties, yet the underlying integrated metabolic and epigenetic mechanisms remain elusive. Here, we demonstrate that coimmunization with Bacille Calmette-Guérin (BCG) and bacterial lipoprotein (BLP) induces a durable form of trained immunity that provides robust, long-term protection against polymicrobial sepsis from early life into adulthood. Single-cell RNA sequencing revealed that this effect is mediated by an expansion of CCR5hi memory-like monocytes with enhanced antimicrobial capacity. Mechanistically

Cancer progression involves systemic immune suppression and tumor recurrence facilitated by increased splenic protumoral myelopoiesis. To address this clinical challenge, we developed an engineered Trojan Mycobacterium as a trained immunity (TI)-mediated splenic myelopoiesis converter (t-SMC). This design provides an intravenously injectable anisotropic TI inducer with an aspect ratio of 3.6 for preferential spleen targeting and cellular uptake via a Mycobacterium "peeling-off and masking-up" strategy with human serum albumin incorporating an enzyme‑activatable Toll‑like receptor 7/8 agonist t

As one of the most burdensome oral diseases, periodontitis frequently leads to tooth loss and heightens the risk of systemic diseases. A major clinical challenge is that current strategies often fail to resolve chronic inflammation and bone resorption characteristic of its refractory state. Recent research implicates maladaptive trained immunity (TRIM) as a core mechanism, which promotes myeloid cell skewing and amplifies inflammatory responses and bone loss. Single-cell sequencing of peripheral blood mononuclear cells from periodontitis patients identified an elevated inflammatory stress stat

OBJECTIVE: This study investigated how β-glucan (BG)-induced trained immunity (TI) regulates osteoclast-driven bone resorption, with emphasis on the pivotal signaling axis governing this anti-resorptive memory.
METHODS: Bone marrow-derived macrophages (BMMs) were primed with BG for 24 h and subsequently challenged with LPS to establish a TI model, validated by cytokine secretion profiling (IL-1β, IL-6, TNF-α). Osteoclastogenesis was induced by M-CSF/RANKL and assessed via TRAP staining, F-actin ring formation, immunofluorescence for CTSK/MMP-9, and bone resorption assays. Histological evaluat

Trained immunity refers to memory-like responses of innate immune cells when they re-encounter pathogenic stimuli. Bacillus Calmette-Guérin (BCG) vaccination implies enhanced antiviral immunity, whereas the underlying mechanisms remain unclear. Herein, we have uncovered elevated expression of low-density lipoprotein receptor (LDLR) on BCG-trained macrophages with robust type I interferon (IFNI) production and antiviral effects both in vivo and in vitro. Consequently, cholesterol is accumulated in BCG-trained macrophages, leading to the augmentation of NFE2L1 expression and the formation of NFE

Older adults have decreased vaccine efficacy, but the adjuvanted recombinant VZV-gE zoster vaccine (RZV) is highly efficacious. We investigated memory-like innate immune responses after RZV and after the zoster vaccine live (ZVL), which is much less efficacious. RZV increased NK, monocyte, and DC activation in response to in vitro VZV-gE stimulation for up to 5 years post-vaccination, while ZVL increased only DC responses to VZV for up to 90 days. In purified monocyte and NK cell cocultures, RZV recipients showed increased responses to VZV-gE, HCMV and HSV antigenic stimulation post-vaccinatio

The measles, mumps, and rubella (MMR) vaccine protects against all-cause mortality in children, but the immunological mechanisms mediating these effects are poorly known. We systematically investigated whether MMR can induce long-term functional changes in innate immune cells, a process termed trained immunity, that could at least partially mediate this heterologous protection. In a randomized, placebo-controlled trial, 39 healthy adults received either the MMR vaccine or a placebo. Using single-cell RNA-Seq, we found that MMR caused transcriptomic changes in CD14+ monocytes and NK cells, but

Trained immunity refers to the long-lasting memory traits of innate immunity. Recent studies have shown that trained immunity is orchestrated by sustained changes in epigenetic marks and metabolic pathways, leading to an altered transcriptional response to a second challenge. However, the potential heterogeneity of trained-immunity induction in innate immune cells has not been explored. In this study, we demonstrate cellular transcriptional programs in response to 4 different inducers of trained immunity in monocyte populations at single-cell resolution. Specifically, we identified 3 monocyte

β-Glucan-induced trained immunity in myeloid cells leads to long-term protection against secondary infections. Although previous studies have characterized this phenomenon, strategies to boost trained immunity remain undefined. We found that β-glucan-trained macrophages from mice with a myeloid-specific deletion of the phosphatase SHIP-1 (LysMΔSHIP-1) showed enhanced proinflammatory cytokine production in response to lipopolysaccharide. Following β-glucan training, SHIP-1-deficient macrophages exhibited increased phosphorylation of Akt and mTOR targets, correlating with augmented glycolytic me

Understanding the mechanisms implicated in the establishment of trained immunity could aid the design of novel therapeutic approaches. By studying Bacille Calmette-Guerin (BCG) vaccination in a human cohort, Ziogas and colleagues demonstrate the role of lactate generation and its use for histone lactylation as a key mechanism for establishing innate immune memory.

Trained immunity is a concept that is currently in development and refers to the long-term functional reprogramming of innate immune cells in response to microbial or inflammatory stimuli. This process serves a dual purpose in the gastrointestinal tract, contributing to chronic inflammatory conditions like inflammatory bowel disease and maintaining host defense. The production of pro-inflammatory mediators is augmented by epigenetic and metabolic changes that are induced by the persistent activation of innate immune cells, which is triggered by microbial components and damage-associated signal

INTRODUCTION: The paradigm of trained immunity (TI), a de facto innate immune memory mediated by metabolic and epigenetic reprogramming of innate leukocytes, has expanded the conceptual framework for rational vaccine adjuvant design. Moving beyond conventional empiricism, TI offers a mechanistic basis that may enhance protection against certain heterologous pathogens, particularly in specific conditions.
AREAS COVERED: This review provides a comprehensive analysis of next-generation adjuvants engineered to induce TI. We outline the core mechanistic foundations encompassing the PI3K/Akt/mTOR/H

Although trained immunity has been explored in various experimental models, the effects of oral β-glucan administration in mice and in vivo comparisons of different training intervals remain unexplored. First, this study evaluated the ability of β-glucan from Debaryomyces hansenii CBS 8339 (βG-Dh) to induce trained immunity in mice through oral administration at different doses. BALB/c mice received oral βG-Dh (0.5, 1, or 2 mg), oral zymosan (1 mg), or intraperitoneal zymosan (1 mg; positive control), followed by ex vivo stimulation of splenocytes with LPS. Low (0.5 mg) and medium (1 mg) doses

Myalgic encephalomyelitis/chronic fatigue syndrome (ME/CFS) is a complex chronic disease of which the underlying (molecular) mechanisms are mostly unknown. An estimated 0.89% of the global population is affected by ME/CFS. Most patients experience a multitude of symptoms that severely affect their lives. These symptoms include post-exertional malaise, chronic fatigue, sleep disorder, impaired cognitive functions, flu-like symptoms, and chronic immune activation. Therapy focusses on symptom management, as there are no drugs available. Approximately 60% of patients develop ME/CFS following an ac

Trained immunity, an innate immune memory, has emerged as a promising strategy to enhance host defense against sepsis, a leading cause of mortality in critical care. While Bacillus Calmette-Guérin (BCG) is the most widely used vaccine for tuberculosis prevention, its broader use as an inducer of trained immunity is limited by adverse reactions. Here, it is reported that BCG-derived outer membrane vesicles (B-OMVs) effectively trigger trained immunity to protect against experimental polymicrobial sepsis. Comprehensive characterization and safety assessments confirmed that B-OMVs exhibited no si

Clostridium perfringens (C. perfringens) infection is recognized as one of the most challenging issues threatening food safety and perplexing agricultural development. To date, the molecular mechanisms of the interactions between C. perfringens and the host remain poorly understood. Here, we show that stimulator of interferon genes (STING)-dependent trained immunity protected against C. perfringens infection through mTOR signaling. Heat-killed Candida albicans (HKCA) training elicited elevated TNF-α and IL-6 production after LPS restimulation in mouse peritoneal macrophages (PM). Although HKCA

BackgroundHeterologous effects of vaccines are mediated by "trained immunity," whereby myeloid cells are metabolically and epigenetically reprogrammed, resulting in heightened responses to subsequent insults. Adenovirus vaccine vector has been reported to induce trained immunity in mice. Therefore, we sought to determine whether the ChAdOx1 nCoV-19 vaccine (AZD1222), which uses an adenoviral vector, could induce trained immunity in vivo in humans.MethodsTen healthy volunteers donated blood on the day before receiving the ChAdOx1 nCoV-19 vaccine and on days 14, 56, and 83 after vaccination.
