Identification of trypanosomes isolated from reduviidae from north Chile.

Examination of 54 Triatoma infestans from a village near the European Southern Observatory La Silla in Chile and of 9 Triatoma spinolai from the territory of the observatory showed that 10 T. infestans were infected with trypanosomatids. Mice were infected with in vitro cultures initiated with five different trypanosomatid isolates and treated with the immunosuppressive drug cyclophosphamide to increase the parasitemia of the flagellates. Evidence of the presence of T. cruzi was provided by a comparative biometrical analysis of blood trypomastigotes and the occurrence of intracellular amastigo

Trypanosoma cruzi (Chagas, 1909) transmission among captive wild mammals, triatomines and free-living opossums from surrounding areas in the São Paulo Zoological Park, Brazil.

BACKGROUND: Trypanosoma cruzi is a multi-host parasite that infects dozens of mammalian species in the most variable wild environments in Americas. Between 2013 and 2018, autochthonous infections by T. cruzi were suspected in three European wolves (Canis lupus) and an orange dwarf porcupine (Coendou spinosus) from Fundação Parque Zoológico de São Paulo (FPZSP), Brazil, current Coordenadoria de Fauna Silvestre (CFS), which is inserted in a remnant of the Atlantic rainforest inside one of the biggest and most populous municipality of the world. This study aims to detect T. cruzi infections in ca

Modelling geospatial distributions of the triatomine vectors of Trypanosoma cruzi in Latin America.

Approximately 150 triatomine species are suspected to be infected with the Chagas parasite, Trypanosoma cruzi, but they differ in the risk they pose to human populations. The largest risk comes from species that have a domestic life cycle and these species have been targeted by indoor residual spraying campaigns, which have been successful in many locations. It is now important to consider residual transmission that may be linked to persistent populations of dominant vectors, or to secondary or minor vectors. The aim of this project was to define the geographical distributions of the community

Chagas disease vector blood meal sources identified by protein mass spectrometry.

Chagas disease is a complex vector borne parasitic disease involving blood feeding Triatominae (Hemiptera: Reduviidae) insects, also known as kissing bugs, and the vertebrates they feed on. This disease has tremendous impacts on millions of people and is a global health problem. The etiological agent of Chagas disease, Trypanosoma cruzi (Kinetoplastea: Trypanosomatida: Trypanosomatidae), is deposited on the mammalian host in the insect's feces during a blood meal, and enters the host's blood stream through mucous membranes or a break in the skin. Identifying the blood meal sources of triatomin

[Transmission of chagasic infection by oral route in the natural history of Chagas disease].

The enzootic cycle of Trypanosoma cruzi in the nature is maintained for millions of years, in great part by oral route, through the ingestion of infected triatomines by edentates(Xenarthra), marsupials and primates and by carnivores eating other infected mammals. The other phases of the natural history of Chagas disease--Anthropozoonosis, Zoonosis or Anphixenosis and Zooanthroponosis are more recent. The most ancient human mummies found with DNA of T. cruzi by PCR, from northern Chile and southern Peru, dated of 9.000 years and the infection must be acquired from the forest or in the caves as

Differences of susceptibility of five triatomine species to pyrethroid insecticides - implications for Chagas disease vector control.

As pyrethroids are presently the favored group of insecticides to control triatomines, we performed a series of bioassays to determine the intrinsic activity of some of the main compounds used in the control campaigns, against five of the main species of triatomines to be controlled. Comparing the insecticides it can be seen that lambdacyhalothrin is more effective than the other three pyrethroids, both considering the LD50 and 99 for all the three species with comparable results. On Triatoma infestans the LD50 of lambdacyhalothrin was followed by that of alfacypermethrin, cyfluthrin and delta

Studies in search of a suitable experimental insect model for xenodiagnosis of hosts with Chagas' disease. 4--The reflection of parasite stock in the responsiveness of different vector species to chronic infection with different Trypanosoma cruzi stocks.

Previous studies (1982, 1987) have emphasized the superiority of sylvatic vector species over domestic species as xenodiagnostic agents in testing hosts with acute or chronic infections by T. cruzi "Y" stock. The present study, which is unique in that it contains data on both infectivity rates produced by the same stock in 11 different vector species and also the reaction of the same vector species to seven different parasite stocks, establishes the general validity of linking efficiency of xenodiagnosis to the biotope of its agent. For example, infectivity rates produced by "São Felipe" stock

Differences in binding of lectin-gold conjugates by Trypanosoma cruzi and Blastocrithidia triatomae (Trypanosomatidae) in the intestine of Triatoma infestans (Reduviidae).

Two species of trypanosomatid flagellates, Trypanosoma cruzi and Blastocrithidia triatomae which develop in the intestine of Triatoma infestans, were incubated with lectin-gold conjugates to demonstrate sugar components of the cell surface with the electron microscope. T. cruzi, strain "Chile 5" (zymodeme 1), bound SBA-gold, revealing N-acetyl-galactosamine and/or galactose residues. WGA-BSA-gold, specific for N-acetylglucosamine, did not bind to the surface of this flagellate. On the other hand, N-acetyl-glucosamine bound WGA-BSA-gold on the cell membrane of epimastigotes and cysts of B. tria

Experimental transmission of Trypanosoma cruzi through the genitalia of albino mice.

Trypanosoma cruzi is usually transmitted by contact with the excreta of infected Triatominae; among non-vectorial infections, direct transmission through coitus has been proposed. We investigated this possibility by instilling, through the external meatus of the vagina and the penis of previously anesthetized NMRI albino mice, blood of mice infected with strains isolated from Didelphis marsupialis (opossum, strain CO57), Rattus rattus (rat, strain CO22) and human (strain EP). Some animals were allowed to copulate the same day of the instillation. In other experiments, the strains were inoculat

External Quality Assessment as a strategy for Chagas disease surveillance: a study in four municipalities of Pernambuco.

BACKGROUND: Triatomines are hematophagous insects that act as vectors of Trypanosoma cruzi, the causative agent of Chagas disease. External Quality Assessment (EQA) is a tool used for monitoring and improving taxonomic identification activities performed by the health laboratories in Brazil. METHODS: EQA were conducted in four municipalities in the state of Pernambuco. Boxes containing a panel of 12 adult Hemiptera specimens (male and female) were sent for species identification. RESULTS: The results revealed 68% accuracy, with difficulty for morphologically similar species, such as those of

Detection of Trypanosoma cruzi-infected triatomines in Iquitos: possible incipient colonisation in the largest metropolis of the Peruvian Amazon.

BACKGROUND: While Chagas disease (CD) has been controlled in many South American regions, the Amazon basin has emerged as a new focus of transmission. Metropolitan Iquitos (Loreto, Peru), has recently shown signs of potential disease emergence. OBJECTIVE: To assess the risk of CD transmission by evaluating triatomine presence and infection rates in households across Iquitos and nearby communities. METHODS: Entomological surveillance was conducted in domestic and peridomestic environments, following blood donor screenings (2011-2018) that confirmed local Trypanosoma cruzi cases. Triatomines w

Chagas disease in Andean countries.

The Andean Countries' Initiative (ACI) for controlling Chagas disease was officially created in 1997 within the framework of the Hipolito Unanue Agreement (UNANUE) between the Ministries of Health of Colombia, Ecuador, Peru, and Venezuela. Its objective was to interrupt transmission via vector and transfusion in the region, taking into account that there are 12.5 million people at risk in the four Andean countries forming the initiative in the area and around 3 million people are infected by Trypanosoma cruzi. The progress of control activities for the vector species present in the Andean sub-

Deforestation and human proximity influence Trypanosoma cruzi infection in palm-dwelling triatomines.

In the southwestern Brazilian Amazon, palm-dwelling triatomines maintain sylvatic transmission cycles of Trypanosoma cruzi, the causative agent of Chagas disease, and Trypanosoma rangeli, a related non-pathogenic parasite. Deforestation can reduce biodiversity and increase pathogen prevalence in triatomine populations; however, the effects of landscape structure on triatomine infection patterns remain poorly understood. Here, we address this knowledge gap by examining how forest cover and proximity to human dwellings influence triatomine infection patterns across gradients of deforestation. Fi

[About the use of "Maria" bio-sensors in the activities of epidemiological surveillance against Chagas' disease in the North of Minas Gerais State].

Eight hundred and sixty six bio-sensors for the passive detection of triatomine bugs, were tried in intra-domiciles of thirteen municipalities of an endemic area of Triatoma sordida in the North of Minas Gerais State, Brazil. This species is nowadays the most captured in Brazil and mainly in the above region. The sensors were examined every three months, corresponding to four times in the first seven municipalities (six hundred and forty two sensors, with a maximum positivity of 0.5%) and two times in the other six (two hundred and twenty two sensors, with a maximum positivity of 2.7%), showin

Vertebrate hosts and vectors of Trypanosoma rangeli in the Amazon Basin of Brazil.

A total of 46 Trypanosoma rangeli stocks were isolated from naturally infected mammals and triatomine vectors. Twenty-two stocks were from the common opossum (Didelphis marsupialis), one from the brown "4-eyed" opossum (Metachirus nudicaudatus), one from the anteater (Tamandua tetradactyla), one from the coati (Nasua nasua), seven from Rhodnius pictipes and 14 from Rhodnius robustus. Two stocks were also isolated from recently fed sandflies (Lutzomyia sp., Shannoni group). The stocks were identified as T. rangeli on the basis of natural or experimental salivary gland infections in Rhodnius, in

Trypanosoma cruzi (Kinetoplastida: Trypanosomatidae) in the Midwest: prevalence among vectors and medical provider awareness.

True bugs of the subfamily Triatominae may carry the protozoan Trypanosoma cruzi, the causative agent of Chagas disease. Endemic to South and Central America, the protozoan has been found in triatomines throughout the southern United States of America, yet it is unclear how far north Trypanosoma cruzi occurs in Triatoma species. The objective of this research was to determine if triatomines in the St. Louis metropolitan area carry this protozoan and to assess professionals across the One Health spectrum for knowledge of the vectors and Chagas disease. Triatomine bugs were collected using a var

Warm-region parasites invasion in temperate climate countries.

Long-term changes in weather conditions on Earth have a significant impact on the world around us. These include not only increasingly extreme weather events such as droughts and heatwaves. These effects can be felt throughout the natural environment, influencing the spread of parasites and the diseases they transmit. Climate change can alter the range and life cycles of parasites, and accelerate and lengthen the activity period of vectors. Four species are described in this manuscript: Balamuthia mandrillaris, Naegleria fowleri, Trypanosoma cruzi and Strongyloides stercoralis. Balamuthia mand

Allatotropic peptides modulate muscle contraction of the female reproductive system in Rhodnius prolixus (STÄL).

Allatotropin (AT) acts as a myoregulator at the level of the dorsal vessel (DV) and midgut (MG) in triatominae insects. Previous analyses of the expression of the AT receptor in Rhodnius prolixus showed that AT is expressed in the DV and MG, but also in the reproductive system in females. To further study the activity of AT on female reproductive organs we analyzed the response by adult females in different physiological conditions, including unfed (virgin and mated), and fed mated females (gravid), to doses ranging between 10-14 and 10-6M. Myoregulatory activity was evaluated in vivo, by reco

Harvesting Venom Toxins from Assassin Bugs and Other Heteropteran Insects.

Heteropteran insects such as assassin bugs (Reduviidae) and giant water bugs (Belostomatidae) descended from a common predaceous and venomous ancestor, and the majority of extant heteropterans retain this trophic strategy. Some heteropterans have transitioned to feeding on vertebrate blood (such as the kissing bugs, Triatominae; and bed bugs, Cimicidae) while others have reverted to feeding on plants (most Pentatomomorpha). However, with the exception of saliva used by kissing bugs to facilitate blood-feeding, little is known about heteropteran venoms compared to the venoms of spiders, scorpio
