Central inhibition of basal pancreatic and gastric secretions by beta-endorphin in rats.

The effects of intracerebroventricular injections of synthetic ovine beta-endorphin were tested in conscious rats with gastric and pancreatic fistulas. In the range of 0.8 to 25 microgram injected in the third ventricle of the brain, basal gastric and pancreatic secretions were strongly inhibited in a dose-dependent manner. Pancreatic volume, bicarbonate output, and total protein output were similarly inhibited, while the bicarbonate concentration was only slightly changed. Similar effects were noted after the administration of morphine. In the present model system, morphine was 20-30 times le

Non-opiate beta-endorphin fragments and dopamine-IV. gamma-Type endorphins may control dopaminergic systems in the nucleus accumbens.

Chronic treatment with des-enkephalin-gamma-endorphin (DE gamma E, beta-endorphin 6-17) twice daily for 10 days into the nucleus accumbens of rats resulted in hypoactivity, while similar treatment with gamma-endorphin antiserum led to a marked hyperactivity. This enhanced activity persisted for at least 3 days following discontinuation of treatment. Rats chronically treated with gamma-endorphin antiserum into the nucleus accumbens habituated at a slower rate when tested repeatedly for locomotor activity, as well as for nociception. Passive avoidance behaviour was attenuated in the treated rats

Immunologic localization of alpha- and beta-endorphins and beta-lipotropin in corticotropic cells of the normal and anencephalic fetal pituitaries.

The use of antibodies to alpha-(17--39) ACTH, beta-LPH, and alpha- and beta-endorphins has enabled the detection of immunoreactive cells as early as the eight week of fetal life. Immunoreactive cells are located in the anterior and intermediate lobes of the pituitary gland in normal human fetuses. The same cells reacted with different antisera used in the present study. No immunoreactive cells were observed in the posterior lobe. The specificity of the immunocytological reaction has been controlled. In seven anencephalic fetuses, the presence of immunoreactive cells suggests that ACTH and beta

Plasma met-enkephalin, beta-endorphin and leu-enkephalin levels in human hepatic encephalopathy.

To address the role of the opioid system in the pathogenesis of hepatic encephalopathy (HE) we measured plasma met-enkephalin, beta-endorphin and leu-enkephalin in patients with different grades of HE compared to control subjects and patients with cirrhosis. Plasma met-enkephalin levels were significantly higher in patients with cirrhosis and all grades of HE than controls. Plasma beta-endorphin levels were similar in the 3 groups. Plasma leu-enkephalin levels were significantly higher in HE grades II, III and IV than in controls, patients with cirrhosis and HE grade I patients. Our results su

A study of maternal plasma beta-endorphin in pre-eclamptic women during labour.

This prospective study was undertaken to determine the maternal plasma beta-endorphin levels in pre-eclamptic women during labour and to explore any correlation between the levels and the severity of pre-eclampsia. A total of 50 primiparous women at term were recruited over a period of 5 years. Twenty-five with pre-eclampsia and 25 healthy women considered as a control group. The mean concentration of maternal plasma beta-endorphin in pre-eclamptic group (22.73+/-8.02 pmol/l) was significantly lower (p<0.001) than that of the control group (48.38+/-5.94 pmol/l). A highly significant (p<0.001)

Chronic ethanol inhibits NK cell cytolytic activity: role of opioid peptide beta-endorphin.

The role of beta-endorphin (beta-EP) in ethanol-altered NK cell cytolytic activity is studied using male Fischer-344 rats as an animal model. Ethanol was administered for 1, 2, 3, or 4 wk in a liquid diet containing 8.7% ethanol (v/v), which means that 37% of the total calories were derived from ethanol. Rats treated with ethanol for 1 wk showed an increase in hypothalamic and plasma levels of immunoreactive (IR)-beta-EP, but displayed no significant effect on NK cell activity determined by (51)Cr release assay, as compared with those in pair-fed and ad libitum-fed animals. However, animals tr

Sex differences in locomotor activity following beta-endorphin in the ventrolateral periaqueductal gray.

Both morphine and selective opioid receptor agonists produce typical biphasic effects upon activity in male rats with initial reductions in activity followed by subsequent increases in activity. Sex differences have been observed in a number of opioid-mediated responses, including the observation that microinjection of the opioid peptide, beta-endorphin into the ventrolateral periaqueductal gray (vlPAG) produces more pronounced analgesia in male rats relative to female rats. The present study evaluated whether beta-endorphin (5.2-26 microg) in the vlPAG produced biphasic activity effects in ma

Fetal ethanol exposure: hypothalamic-pituitary-adrenal and beta-endorphin responses to repeated stress.

Previous studies provide evidence that fetal ethanol exposure induces hypothalamic-pituitary-adrenal (HPA) and pituitary beta-endorphin (beta-EP) hyperresponsiveness to acute stressors. The present study demonstrates significant effects of in utero ethanol exposure on the parallel response patterns of the HPA axis and the pituitary beta-EP system to repeated exposures to a stressor, restraint stress, and indicates sex differences in response. Together, data from the two experiments indicate that, after repeated restraint exposures, fetal ethanol-exposed (E) males and females both show signific

[Activation of delta-opiate receptor simultaneously stimulates ACTH beta-endorphin and prolactin release].

To evaluate a possible physiological and pathological role of brain delta-opiate receptor and endogenous ligands in the control of ACTH, beta-endorphin (beta-EP) and prolactin (PRL) release, a specific agonist of the delta-opiate receptor--[D-Pen2, D-Pen5]-enkephalin (DPDPE) was microinjected into the third cerebral ventricle (1 microgram or 3 micrograms/3 microliters/rat) in conscious freely-moving male rats. Plasma ACTH, beta-EP and PRL concentrations were measured by radioimmunoassay (RIA). Our results clearly indicate that third ventricular injection of DPDPE significantly elevates plasma

Opioid receptors mediating antinociception from beta-endorphin and morphine in the periaqueductal gray.

beta-Endorphin and morphine produce an increase in the latency of the tail-flick reflex when administered into the PAG of awake rats. The antinociceptive effect of both opioid agonists was blocked by the sequential local injection of either CTP (D-Phe-Cys-Tyr-D-Trp-Lys-Thr-Pen-Thr-NH2), a selective mu opioid receptor antagonist, naltrexone, or beta-endorphin (1-27), a putative epsilon opioid receptor antagonist, with minimal selectivity. When either CTP or naltrexone was used as the antagonist, the dose-inhibition curves generated for beta-endorphin and morphine were not parallel, suggesting t

Beta-endorphin suppresses rat plaque-forming cell response by a non-opioid mechanism.

The opioid peptide beta h-endorphin 1-31 inhibited the plaque-forming cell (PFC) response to sheep red blood cells when rat splenocytes, immunized in vivo, were cultured in vitro with the peptide. The opioid antagonist naloxone failed to reverse the beta h-endorphin 1-31 suppression of the PFC response. Peptide fragments of beta h-endorphin 1-31, other opioid peptides, and alkaloids had no effect on the response. These data indicate that beta h-endorphin 1-31 suppresses antibody production or secretion by a specific, non-opioid receptor on the rat splenocytes.

beta-Endorphin augments the cytolytic activity and interferon production of natural killer cells.

We have investigated the in vitro effects of the neurohormone beta-endorphin (b-end) on natural killer (NK) activity and interferon (IFN) production mediated by large granular lymphocytes (LGL). LGL-enriched fractions from peripheral blood mononuclear cells (PBMC) from normal human volunteers were obtained by fractionation over discontinuous Percoll gradients. LGL were preincubated with or without various concentrations of b-end or the closely related peptides alpha-endorphin (a-end), gamma-endorphin (g-end), or D-ALA2-beta-endorphin (D-ALA2-b-end), a synthetic b-end analogue. Cyclo(Gly-Gln) inhibits the cardiorespiratory depression produced by beta-endorphin and morphine.

Glycyl-L-glutamine (Gly-Gln; beta-endorphin 30-31) is an endogenous dipeptide that is synthesized through the post-translational processing of beta-endorphin. Previously, we showed that Gly-Gln inhibits the hypotension and respiratory depression produced by central beta-endorphin administration. In this study, we tested whether cyclo(Gly-Gln), a non-polar, cyclic Gly-Gln derivative, was similarly effective following intracerebro-ventricular (i.c.v.) or intra-arterial (i.a.) administration to pentobarbital-anesthetized rats pretreated with beta-endorphin (0.5 nmol i.c.v.). Glycyl-L-glutamine [beta-endorphin-(30-31)] attenuates hemorrhagic hypotension in conscious rats.

The profound hypotension caused by acute hemorrhage is thought to involve opioid peptide neurons. In this study, we tested whether glycyl-L-glutamine [Gly-Gln; beta-endorphin-(30-31)], a nonopioid peptide derived from beta-endorphin processing, prevents the cardiovascular depression induced by hemorrhage in conscious and anesthetized rats. Previously, we found that Gly-Gln inhibits the hypotension and respiratory depression produced by beta-endorphin and morphine but does not affect opioid antinociception. Hemorrhage (2.5 ml/100 g body wt over 20 min) lowered arterial pressure in conscious rat

Chronic intracerebroventricular administration of beta-endorphin augments natural killer cell cytotoxicity in rats.

We have studied the effect of chronic intracerebroventricular (i.c.v.) infusion of different opioid peptides on natural killer (NK) cell mediated cytotoxicity in vivo in the spontaneously hypertensive rat (SHR). The in vivo NK cell activity was measured as the clearance of 51Cr-labelled YAC-l lymphoma cells from the lung tissues. Further, the phenotype of lymphocytes in spleen and peripheral blood was analysed by flow cytometry (FACS). All opioid drugs were administered i.c.v. for 6 days with osmotic minipumps releasing 1.0 microliter/h. beta-Endorphin (10 or 20 micrograms/rat per day) signifi

Plasma corticotropin-releasing hormone, beta-endorphin and cortisol inter-relationships during human pregnancy.

To investigate the dynamic relationships among corticotropin-releasing hormone (CRH), beta-endorphin (beta EP), cortisol and obstetric events during pregnancy, blood samples were collected from 193 women at 28 weeks, 38 weeks, during labour and on the second postnatal day. Cord blood at delivery was also obtained. We found that: (1) Maternal plasma CRH, beta EP and cortisol rose from 28 to 38 weeks. (2) During the third trimester maternal plasma CRH and beta EP were correlated (r = 0.30, p < 0.001). (3) During labour, no correlations were found among maternal plasma CRH, beta EP and cortisol.

The role of pituitary beta-endorphin in mediating corticotropin-releasing factor-induced antinociception.

The hypothesis that blood-borne beta-endorphin modulates nociception was examined with corticotropin-releasing factor (CRF) as a potent and selective agent to stimulate its release from the pituitary gland. Intravenously administered CRF produced a dose-related antinociception in rats as determined by measuring paw-lick latencies on a 50 degrees C hot plate. A dose of 25 nmol/kg of CRF was comparable in both magnitude and duration of antinociception to a 7,500 nmol/kg (= 2.5 mg/kg) dose of morphine sulfate. The antinociceptive effect of CRF was blocked by both hypophysectomy and dexamethasone

Effect of sleep loss on beta-endorphin activity, epinephrine levels, and ventilatory responsiveness.

Sleep loss impairs ventilatory responsiveness to hypercapnia and hypoxia, and also interferes with performance on spirometry. To test the hypothesis that the decline in hypercapnic drive due to sleep loss is mediated by endorphin production, we measured loaded and unloaded CO2 response after injection of placebo and naloxone in 11 normal subjects who were alternately rested and sleep-deprived. Blood for beta-endorphin and epinephrine assay was drawn before testing each day. Unloaded CO2 response was lower after sleep loss than after sleep restoration; naloxone had no effect on this difference.
