Integrative Analysis of Arachidonic Acid Metabolism in the Pathogenesis and Immune Dysregulation of Psoriasis.

BACKGROUND: Psoriasis is a chronic inflammatory skin disorder with complex molecular mechanisms. While previous studies have demonstrated altered levels of arachidonic acid and its metabolites in psoriatic lesions, the specific roles of arachidonic acid metabolism (AAM) genes in the molecular pathogenesis and immune dysregulation of psoriasis remain poorly understood. This study aimed to investigate the role of AAM genes in the pathogenesis and immune dysregulation of psoriasis using an integrative bioinformatics approach. METHODS: Gene expression data from psoriasis patients and healthy cont

Role of SLC16A10 in Psoriasis Through the Regulation of Arachidonic Acid Metabolism in Keratinocytes.

Psoriasis is an inflammatory dermatological condition challenging to treat and prone to recurrence. The pathogenesis of psoriasis is closely associated with metabolic disorders, while therapies targeting the dysregulated metabolism in psoriasis remain limited. Therefore, exploring the pathogenesis of psoriasis and identifying potential metabolic therapeutic targets is imperative. In this study, potential biomarkers for clinically targeted metabolic therapies in patients with psoriasis are aimed to be identified. RNA-sequencing analysis is performed on metabolism-related genes to identify diffe

Gestational diabetes mellitus enhances arachidonic and docosahexaenoic acids in placental phospholipids.

In previous studies, we reported that neonates of women with gestational diabetes mellitus (GDM) have reduced blood levels of arachidonic acid (AA) and docosahexaenoic acid (DHA) that were unrelated to maternal status. Since both AA and DHA are selectively transferred from maternal to fetal circulation by the placenta, we have investigated whether the FA composition of the placenta is altered by GDM. Thirty-six women, 11 with and 25 without GDM, were recruited from Newham General Hospital, London. The women with GDM had higher levels of di-homo-gamma-linolenic (P < 0.05), docosatetraenoic (n-6

The effect of arachidonic acid on the hydroosmotic action of vasopressin in frog urinary bladder.

In experiments on frog urinary bladder it was found that 23 nM arginine-vasopressin (antidiuretic hormone, AVP), 10 min after addition, increased the content of free arachidonic acid (AA) in bladder tissue from 18.6 +/- 1.2 to 33.3 +/- 3.9 ng/mg protein. The exogenous AA (10(-8) - 10(-5) M) added to the serosal Ringer solution did not change the basal level of osmotic water flow, but significantly inhibited the hydroosmotic effect of 23 nM AVP in a dose-dependent manner. The inhibitory effect of AA was not eliminated in the presence of 1 microM indomethacin, 1 microM nordihydroguaiaretic acid,

Inhibitory effect of melatonin on platelet activation induced by collagen and arachidonic acid.

Melatonin, an indolamine synthesized in the pineal gland, is known to have antiprostanoid activity. The inhibition of platelet aggregation induced by melatonin has been proposed to take place through the cyclooxygenase pathway. In the present study, we found that melatonin has a marked inhibitory effect on collagen, arachidonic acid (AA), adenosine diphosphate (ADP), epinephrine, and A23187-induced aggregation in platelet-rich plasma. On the other hand, using metrizamide-filtered platelets resuspended in Tyrode's buffer, melatonin fails to suppress AA-induced platelet aggregation and 14C-5-HT

Thrombin-induced transfer of arachidonic acid in human platelets is not inhibited by trifluoperazine.

Human platelets have been shown to contain a Ca++- and CoA-independent transacylase enzyme that catalyzes the transfer of arachidonic acid from phosphatidylcholine (PC) to lysoplasmenylethanolamine. It has been suggested that this route may represent a major source for released arachidonic acid in stimulated platelets. In this study, we have shown using arachidonic-labelled human platelets that the thrombin-induced activation of a transacylase reaction was not affected by concentrations of trifluoperazine (TFP) (15 micrograms/2 X 10(9) cells) which abolished the accumulation of free [3H]arachi

The effect of aspirin on the metabolism of exogenous arachidonic acid in human polymorphonuclear leukocytes.

When human polymorphonuclear leukocytes (PMNL) were incubated with exogenous 14C-arachidonic acid (14C-AA), both lipoxygenase and cyclo-oxygenase metabolites were detected. The amount of the 5-lipoxygenase metabolites formed, including 5-hydroxy-6,8,11,14-eicosatetraenoic acid (5-HETE), was small. The amount of other mono-HETE's (migrating in the vicinity of 12-HETE) was greater, but this was obviously mainly due to the small amount of contaminating platelets. In the presence of calcium ionophore A23187 the rate of formation of 5-HETE was increased, but the formation of other metabolites remai

Biosynthesis of prostaglandin F2alpha from arachidonic acid and prostaglandin endoperoxides in the uterus.

Formation of prostaglandin F2Alpha in the cow and guinea pig uterus microsomes was studied using 14C-labeled arachidonic acid and prostaglandin H2. The total conversion of arachidonic acid was of a low order and underwent fluctuations during the estrous cycle of the guinea pig, being highest towards the end of the cycle. Injections of beta-estradiol-3-benzoate also resulted in higher activity of the uterine prostaglandin synthetase. The uterine prostaglandin synthesizing system appeared to differ in several respects from that present in seminal vesicles, with regard to the proportions of the p

Arachidonic acid promotes M2-like polarization of tumor-associated macrophages and glioma progression.

Immunometabolism plays a crucial role in glioma progression, yet the metabolic mechanisms underlying glioma-associated macrophage (GAM)-mediated immunoregulatory remain poorly understood. Here, we analyzed metabolic profiles of gliomas at single-cell resolution and identified aberrant activation of the arachidonic acid (AA) metabolic pathway in GAMs. Further study reveal that AA promoted M2-like polarization of GAMs, and was associated with an immune-dampening tumor microenvironment in murine models. AA-reprogrammed GAMs downregulated MHC-I expression, impairing CD8+ T cell infiltration and ha

ACSL1 Contributes to Metabolic-Associated Fatty Liver Disease by Mediating Arachidonic Acid Metabolism.

The precise involvement of ACSL1 in metabolic-associated fatty liver disease (MAFLD) pathogenesis remains unclear. To this end, we analyzed the effects of long-chain acyl-CoA synthase (ACSL1) on MAFLD. Analysis of GEO datasets showed that ACSL1 was downregulated in MAFLD. To elucidate its mechanistic role, we generated BRL hepatocyte cell lines with stable ACSL1 knockdown or overexpression. These engineered cells were cultured with a lipid mixture containing 300 μM oleic acid, 150 μM palmitic acid, and 100 μM linoleic acid (OPL) to mimic MAFLD pathophysiology in vitro. The arachidonic acid metabolome reveals elevation of prostaglandin E2 biosynthesis in colorectal cancer.

Arachidonic acid metabolites are a family of bioactive lipids derived from membrane phospholipids. They are involved in cancer progression, but arachidonic acid metabolite profiles and their related biosynthetic pathways remain uncertain in colorectal cancer (CRC). To compare the arachidonic acid metabolite profiles between CRC patients and healthy controls, quantification was performed using a liquid chromatography-mass spectrometry-based analysis of serum and tissue samples. Metabolomics analysis delineated the distinct oxidized lipids in CRC patients and healthy controls. Prostaglandin (PGE

Arachidonic acid metabolism regulates the development of retinopathy of prematurity among preterm infants.

Extremely preterm infants are at risk of developing retinopathy of prematurity (ROP), characterized by neovascularization and neuroinflammation leading to blindness. Polyunsaturated fatty acid (PUFA) supplementation is recommended in preterm infants to lower the risk of ROP, however, with no significant improvement in visual acuity. Reasonably, this could be as a result of the non-consideration of PUFA metabolizing enzymes. We hypothesize that abnormal metabolism of the arachidonic acid (AA) pathway may contribute to severe stages of ROP. The present study investigated the AA-metabolizing enzy

[11 C]arachidonic acid incorporation measurement in human brain: Optimization for clinical use.

Arachidonic acid (AA) is involved in signal transduction, neuroinflammation, and production of eicosanoid metabolites. The AA brain incorporation coefficient (K*) is quantifiable in vivo using [11 C]AA positron emission tomography, although repeatability remains undetermined. We evaluated K* estimates obtained with population-based metabolite correction (PBMC) and image-derived input function (IDIF) in comparison to arterial blood-based estimates, and compared repeatability. Eleven healthy volunteers underwent a [11 C]AA scan; five repeated the scan 6 weeks later, simulating a pre- and post-tr

Control of cyclic AMP concentration in bovine endometrial stromal cells by arachidonic acid.

Second messenger signalling through cyclic AMP (cAMP) plays an important role in the response of the endometrium to prostaglandin (PG) E(2) during early pregnancy. Arachidonic acid, which is a by-product of the luteolytic cascade in ruminants, is a potential paracrine signal from the epithelium to the stroma. We investigated the effects of arachidonic acid on the response of the stroma to PGE(2). cAMP was measured in bovine endometrial stromal cells treated with agents known to activate or inhibit adenylyl cyclase, protein kinase C (PKC) or phosphodiesterase (PDE). PGE(2) increased the intrace

Arachidonic acid in mononuclear cells and its clinical significance in HCV cirrhotic patients.

OBJECTIVES: An abnormal fatty acid pattern in patients with advanced liver cirrhosis (LC) has been reported in plasma phospholipids and some other tissues. To elucidate the significance of arachidonic acid deficiency on the clinical pathophysiology of LC and hepatocellular carcinoma (HCC), we analyzed the fatty acid compositions of mononuclear cell phospholipids, plasma alpha-tocopherol, and thiobarbituric acid-reactive substances and serum tumor necrosis factor-alpha (TNF-alpha) in cirrhotic patients infected with the hepatitis C virus with and without HCC. METHODS: Twelve cirrhotic patients

Do lithium and anticonvulsants target the brain arachidonic acid cascade in bipolar disorder?

BACKGROUND: Lithium and certain anticonvulsants, including carbamazepine and valproic acid, are effective antimanic drugs for treating bipolar disorder, but their mechanisms of action remain uncertain. EXPERIMENTAL OBSERVATIONS: Feeding rats lithium chloride for 6 weeks, to produce a brain lithium concentration of 0.7mM, reduced arachidonic acid turnover within brain phospholipids by 75%. The effect was highly specific, as turnover rates of docosahexaenoic acid and palmitic acid were unaffected. Arachidonate turnover in rat brain also was reduced by long-term valproic acid administration. Lit

Cathepsin B mediates tumor necrosis factor-induced arachidonic acid release in tumor cells.

Arachidonic acid (AA) generated by cytosolic phospholipase A2 (cPLA2) has been suggested to function as a second messenger in tumor necrosis factor (TNF)-induced death signaling. Here, we show that cathepsin B-like proteases are required for the TNF-induced AA release in transformed cells. Pharmaceutical inhibitors of cathepsin B blocked TNF-induced AA release in human breast (MCF-7S1) and cervix (ME-180as) carcinoma as well as murine fibrosarcoma (WEHI-S) cells. Furthermore, TNF-induced AA release was significantly reduced in cathepsin B-deficient immortalized murine embryonic fibroblasts. Em
