Bevacizumab in advanced lung cancer: state of the art.

Despite recent advances in metastatic lung cancer treatment with the advent of immune checkpoint inhibitors and molecules targeting addictive genomic abnormalities, prognosis of most of the patients remains unfavorable. Combination approaches with older drugs, such as bevacizumab, should be thus envisioned. Bevacizumab is a monoclonal anti-VEGF antibody, approved by the US FDA and the EMA in first-line and maintenance settings of advanced nonsquamous non-small-cell lung cancer (NSCLC) treatment, in association with platinum-based chemotherapy. In the years to come, bevacizumab might be associa

Electrospun formulations of bevacizumab for sustained release in the eye.

UNLABELLED: Medicines based on vascular endothelial growth factor (VEGF) neutralising antibodies such as bevacizumab have revolutionized the treatment of age related macular degeneration (AMD), a common blinding disease, and have great potential in preventing scarring after surgery or accelerating the healing of corneal injuries. However, at present frequent invasive injections are required to deliver these antibodies. Such administration is uncomfortable for patients and expensive for health service providers. Much effort is thus focused on developing dosage forms that can be administered les

Direct Effects of Bevacizumab on Rat Conjunctival Fibroblast.

Successful cases of treatment of Bevacizumab for preventing scar after trabeculectomy in glaucoma patients encourage us to explore its mechanism. In this study, we primarily isolated conjunctival fibroblast from rat. RT-PCR analysis for the cells implicated that conjunctival fibroblast expressed vascular endothelial growth factor (VEGF) and its receptors. Immunofluorescence staining also showed positive staining for VEGFR-1. Furthermore, growth of fibroblast was significantly inhibited by Bevacizumab at dose of 2.5 mg/ml. Real time PCR results showed after 48 h intervention of 2.5 mg/ml Bevaci

Bevacizumab is safe in acute relapses of neuromyelitis optica.

OBJECTIVES: Neuromyelitis optica (NMO) is a relapsing autoimmune disease targeting the spinal cord and optic nerve leading to paralysis and blindness. Current treatment for acute NMO attacks is immunosuppression with high-dose corticosteroids and/or plasmapheresis. Preclinical animal studies suggest that bevacizumab might be beneficial in limiting the extent of inflammation during a NMO relapse by reducing the disruption of the blood-brain barrier. METHODS: We carried out an open-label phase 1b safety and proof-of-concept trial in 10 participants with NMO immunoglobulin G seropositive NMO, NM

[Bevacizumab: a new success in hereditary hemorrhagic telangiectasia].

INTRODUCTION: Hereditary hemorrhagic telangiectasia (HHT), is a rare, hereditary vascular dysplasia, characterized by recurrent epistaxis, mucocutaneous telangiectasias and visceral arteriovenous malformations. The vascular endothelial growth factor VEGF seems to play a crucial role in the pathogenesis of this disease. Recently bevacizumab, a humanized monoclonal VEGF inhibitor, has shown promise in treating patients with HHT. CASE REPORT: A 66-year-old man, having HHT since the age of 30 years with recurrent epistaxis related to telangiectasia at the nasal septum and chronic iron deficiency

Bevacizumab: a review of its use in advanced cancer.

The humanized monoclonal antibody bevacizumab (Avastin(®)) has been available in the EU since 2005. Results of phase III trials demonstrate that adding intravenous bevacizumab to antineoplastic agents improves progression-free survival and/or overall survival in patients with advanced cancer, including when used as first- or second-line therapy in metastatic colorectal cancer, as first-line therapy in advanced nonsquamous non-small cell lung cancer, as first-line therapy in metastatic renal cell carcinoma, as first-line therapy in metastatic breast cancer, and as first-line therapy in epitheli

Bevacizumab and micrometastases: revisiting the preclinical and clinical rollercoaster.

The use of bevacizumab, a monoclonal antibody against vascular endothelial growth factor (VEGF), in combination with standard therapeutic approaches, has offered clinical benefit for patients with advanced colorectal, breast, ovarian, renal, non small-cell lung cancer and glioblastoma. However, the strategy of administering bevacizumab until disease progression has been challenged by certain preclinical evidence, suggesting that prolonged exposure to anti-VEGF treatment may elicit an adaptive-evasive response, resulting in a more aggressive tumor phenotype. Moreover, the use of bevacizumab in

Acute and severe acne in a patient treated with bevacizumab.

BACKGROUND: Bevacizumab is a recombinant, humanized monoclonal antibody against vascular endothelial growth factor (VEGF) that inhibits angiogenesis. Bevacizumab is typically well tolerated; its major side effects include hypertension, proteinuria, bleeding, gastrointestinal perforation, and arterial thrombotic events, among others. Cutaneous side effects associated with the use of bevacizumab are rare and involve mainly itching and exfoliative dermatitis. Nonspecific skin rash and acneiform eruptions have recently been described in patients following infusion of bevacizumab. METHODS: Finding

Intrastromal injection of bevacizumab in patients with corneal neovascularization.

Corneal neovascularization (NV) not only reduces visual acuity, but it also causes loss of the cornea's immune privilege, strongly contributing to a worse prognosis in penetrating keratoplasty. Several mediators participate in corneal angiogenesis, and the role of vascular endothelial growth factor (VEGF) has been extensively proven. Anti-VEGF agents have been shown to be effective in slowing the growth of corneal neovessels. Bevacizumab, an anti-VEGF agent, has been successfully used in the treatment of corneal neovascularization. In this paper, we report a series of patients who underwent in

Intravitreal Bevacizumab for the Treatment of Central Serous Chorioretinopathy.

The benefit of intravitreal bevacizumab injection in the long-standing idiopathic central serous chorioretinopathy is described. Intravitreal injection of 1.25 mg of bevacizumab was given to a patient, who complained of reduced vision for 10 months due to idiopathic central serous chorioretinopathy. Within a week, his visual acuity improved from 20/80 to 20/25; 3 months later, the visual acuity reached 20/20 with complete resolution of the serous retinal detachment, as was demonstrated by optical coherence tomography. No recurrences or complications were found for a period of 6 months.
