Prostate ageing across the adult lifespan: population MRI evidence from 30 000 men.

BACKGROUND: Prostate enlargement and lower urinary tract symptoms are common age-related conditions in men frequently encountered in primary care and geriatric medicine practice, yet population-level variation in prostate size and its biological drivers remain poorly defined due to limited scalable imaging. METHODS: We quantified prostate volume in 30 616 men using automated segmentation of whole-body MRI from the UK Biobank, establishing age-specific reference distributions across adulthood. RESULTS: Prostate volume increased continuously with age, and enlargement beyond commonly used clini

Climate signatures in the morphological differentiation of worldwide modern human populations.

The ability of cranial morphology to reflect population/phylogenetic history, and the degree to which it might be influenced by environmental factors and selection pressures have been widely discussed. Recent consensus views cranial morphology as largely indicative of population history in humans, with some anatomical cranial regions/measurements being more informative on population history, while others being under selection pressure. We test earlier findings using the largest and most diverse cranial dataset available as yet: 7,423 male specimens from 135 geographic human population samples

The brain's functional connectome in young children with prenatal alcohol exposure.

Prenatal alcohol exposure (PAE) can lead to altered brain function and structure, as well as lifelong cognitive, behavioral, and mental health difficulties. Previous research has shown reduced brain network efficiency in older children and adolescents with PAE, but no imaging studies have examined brain differences in young children with PAE, at an age when cognitive and behavioral problems often first become apparent. The present study aimed to investigate the brain's functional connectome in young children with PAE using passive viewing fMRI. We analyzed 34 datasets from 26 children with PAE

Factors Affecting Time-Varying Clearance of Cyclosporine in Adult Renal Transplant Recipients: A Population Pharmacokinetic Perspective.

AIM: The pharmacokinetic (PK) properties of cyclosporine (CsA) in renal transplant recipients are patient- and time-dependent. Knowledge of this time-related variability is necessary to maintain or achieve CsA target exposure. Here, we aimed to identify factors explaining variabilities in CsA PK properties and characterize time-varying clearance (CL/F) by performing a comprehensive analysis of CsA PK factors using population PK (popPK) modeling of long-term follow-up data from our institution. METHODS: In total, 3674 whole-blood CsA concentrations from 183 patients who underwent initial renal

Serum anti-Müllerian hormone concentration before and after salpingectomy for ectopic pregnancy.

RESEARCH QUESTION: Does salpingectomy for ectopic pregnancy affect the ovarian reserve measured by changes in pre- and post-operative levels of anti-Müllerian hormone (AMH)? DESIGN: This is a prospective observational multicentre study of 64 women treated with salpingectomy for an ectopic pregnancy. AMH was measured in serum samples collected at admission before salpingectomy and at follow-up (median time to follow-up [interquartile range] was 3 [3-4] months). Changes in serum AMH levels were investigated using Wilcoxon signed-rank test and the relationship between changes in AMH and age, tim

Ilium morphological variation during growth in forager populations.

Human pelvic morphology has often been described in terms of an evolutionary compromise between bipedalism, encephalization, and obstetrics; however, recent research has argued that the pelvis is more biologically plastic than previously thought. Variation in pelvic form exists among adult modern human populations, but the factors influencing this variation, and when it manifests during growth, remain to be understood. The aim of this study is to investigate patterns of growth and development of the ilium and to consider factors that may influence variation. The presence of variation in lower

Equivalence of in vitro blood clot mechanical properties using different anticoagulants.

The mechanical properties of blood clots are critical both to normal clot function and in thromboembolic diseases. Thus, assessing these properties is critical to our understanding of clot (patho-)physiology. Because in vivo clot assessment is practically difficult, we and others have resorted to studying clots in vitro. One unanswered question about these in vitro studies has been whether results are sensitive to the choice of anticoagulant. Therefore, the objective of our work is to determine whether the most common anticoagulants, acid citrate dextrose (ACD) and sodium citrate (SC), yield m

Phenotypic variability and gastrointestinal manifestations/interventions for growth in NAA10-related neurodevelopmental syndrome.

Our study of 61 children with NAA10-related neurodevelopmental syndrome, an X-linked disorder due to NAA10 gene variants, demonstrated a high prevalence of growth failure, with weight and height percentiles often in the failure-to-thrive diagnostic range; however, dramatic weight fluctuations and phenotypic variability is evidenced in the growth parameters of this population. Although never previously explored in depth, the gastrointestinal pathology associated with NAA10-related neurodevelopmental syndrome includes feeding difficulties in infancy, dysphagia, GERD/silent reflux, vomiting, cons

Variable phenotypes and penetrance between and within different zebrafish ciliary transition zone mutants.

Meckel syndrome, nephronophthisis, Joubert syndrome and Bardet-Biedl syndrome are caused by mutations in proteins that localize to the ciliary transition zone (TZ). The phenotypically distinct syndromes suggest that these TZ proteins have differing functions. However, mutations in a single TZ gene can result in multiple syndromes, suggesting that the phenotype is influenced by modifier genes. We performed a comprehensive analysis of ten zebrafish TZ mutants, including mks1, tmem216, tmem67, rpgrip1l, cc2d2a, b9d2, cep290, tctn1, nphp1 and nphp4, as well as mutants in ift88 and ift172. Inter and intra-generational phenotypic variability in a Portuguese family with DYT-GCH1.

DYT-GCH1 is the most common form of Dopa-responsive dystonia. We analysed a three-generation Portuguese family with the same variant of DYT-GCH1. Dystonia and parkinsonism were found, along with non-motor symptoms. There was a wide array of phenotypic manifestations. Further studies are needed to explain the wide clinical expression of this entity.

Phenotypic Variability Among Patients With D4Z4 Reduced Allele Facioscapulohumeral Muscular Dystrophy.

IMPORTANCE: Facioscapulohumeral muscular dystrophy (FSHD) is considered an autosomal dominant disorder, associated with the deletion of tandemly arrayed D4Z4 repetitive elements. The extensive use of molecular analysis of the D4Z4 locus for FSHD diagnosis has revealed wide clinical variability, suggesting that subgroups of patients exist among carriers of the D4Z4 reduced allele (DRA). OBJECTIVE: To investigate the clinical expression of FSHD in the genetic subgroup of carriers of a DRA with 7 to 8 repeat units (RUs). DESIGN, SETTING, AND PARTICIPANTS: This multicenter cross-sectional study

Phenotypic variance in monozygotic twins with SCA3.

BACKGROUND: Spinocerebellar ataxia type 3 (SCA3) is a hereditary neurodegenerative disorder with high clinical heterogeneity. Twin study is valuable to estimate the contributions of gene and/or environment to phenotypic variance. However, SCA3 twins were extremely sparse and rarely reported. METHODS: A pair of monozygotic twins with SCA3 was assessed using well-acknowledged scales. Genetic modifiers and methylation levels were determined by Sanger sequencing and pyrosequencing. RESULTS: Sharing identical CAG repeat lengths, the twins presented with similar symptoms, whereas, the younger sist

Dose-controlled tDCS reduces electric field intensity variability at a cortical target site.

BACKGROUND: Variable effects limit the efficacy of transcranial direct current stimulation (tDCS) as a research and therapeutic tool. Conventional application of a fixed-dose of tDCS does not account for inter-individual differences in anatomy (e.g. skull thickness), which varies the amount of current reaching the brain. Individualised dose-control may reduce the variable effects of tDCS by reducing variability in electric field (E-field) intensities at a cortical target site. OBJECTIVE: To characterise the variability in E-field intensity at a cortical site (left primary motor cortex; M1) an

Climatic adaptation in human inferior nasal turbinate morphology: Evidence from Arctic and equatorial populations.

OBJECTIVES: The nasal turbinates directly influence the overall size, shape, and surface area of the nasal passages, and thus contribute to intranasal heat and moisture exchange. However, unlike the encapsulating walls of the nasal cavity, ecogeographic variation in nasal turbinate morphology among humans has not yet been established. Here we investigate variation in inferior nasal turbinate morphology in two populations from climatically extreme environments. MATERIALS AND METHODS: Twenty-three linear measurements of the inferior turbinate, nasal cavity walls, and airway passages were collec

Clinical heterogeneity in patients with idiopathic blepharospasm: A cluster analysis.

BACKGROUND: Idiopathic blepharospasm is a clinically heterogeneous condition. It is not known whether the various manifestations become manifest sequentially during the course of the disease or aggregate in separate clusters identifying subpopulations of patients. METHODS: Eighty-nine patients with idiopathic blepharospasm were assessed using k-means cluster analysis to identify relatively homogeneous groups on the basis of low-intragroup/high-intergroup differences across a set of selected variables. RESULTS: The results suggest that there may be three groups of patients. Group 1 included p

Complex population structure of Lyme borreliosis group spirochete Borrelia garinii in subarctic Eurasia.

Borrelia garinii, a causative agent of Lyme borreliosis in Europe and Asia, is naturally maintained in marine and terrestrial enzootic cycles, which primarily involve birds, including seabirds and migratory passerines. These bird groups associate with, correspondingly, Ixodes uriae and Ixodes ricinus ticks, of which the latter species may bite and transmit the infection to humans. Studies of the overlap between these two natural cycles of B. garinii have been limited, in part due to the absence of representative collections of this spirochete's samples, as well as of the lack of reliable measu

The developmental genetics of biological robustness.

BACKGROUND: Living organisms are continuously confronted with perturbations, such as environmental changes that include fluctuations in temperature and nutrient availability, or genetic changes such as mutations. While some developmental systems are affected by such challenges and display variation in phenotypic traits, others continue consistently to produce invariable phenotypes despite perturbation. This ability of a living system to maintain an invariable phenotype in the face of perturbations is termed developmental robustness. Biological robustness is a phenomenon observed across phyla,

Exploratory proteomic characterization of Apis mellifera venom across an environmental gradient in central Chile.

BACKGROUND: Bee venom (Apis mellifera) is a complex biological secretion rich in peptides, enzymes, and bioactive molecules with recognized pharmacological potential. Environmental conditions may influence venom composition; however, geographic variation in the bee venom proteome remains insufficiently characterized, particularly in South American Mediterranean ecosystems, including central Chile. METHODS: Venom from multiple A. mellifera colonies within each sampling area of the Valparaíso Region, Chile, was pooled to generate one composite sample per geographic zone: Sea Level, Inland Valle

Quantitative analysis of the proteome and protein oxidative modifications in primary human coronary artery endothelial cells and associated extracellular matrix.

Vascular endothelial cells (ECs) play a key role in physiology by controlling arterial contraction and relaxation, and molecular transport. EC dysfunction is associated with multiple pathologies. Here, we characterize the cellular and extracellular matrix (ECM) proteomes of primary human coronary artery ECs, from multiple donors, and oxidation/nitration products formed on these during cell culture, using liquid chromatography-mass spectrometry. In total ∼9900 proteins were identified in cells from 3 donors, with ∼7000 proteins per donor. Of these ∼5300 were consistently identified, indicating
