Structure identification and analysis of the suspected chemical precursor of 2-fluorodeschloroketamine and its decomposition products.

In this work, 1-[(2″-fluorophenyl)(methylimino)methyl]cyclopentan-1-ol (2-fluorodeschlorohydroxylimine) was identified as a suspected chemical precursor of 2-fluorodeschloroketamine (2-FDCK) using gas chromatography-mass spectrometry (GC-MS) and gas chromatography-quadrupole/time-of-flight mass spectrometry (GC-Q/TOF-MS) and comparing the data with those of ketamine and its chemical precursor, hydroxylimine. Furthermore, the entire fragmentation pathway of 2-fluorodeschlorohydroxylimine was theorized from the GC-MS spectrum recorded using an electron ionization (EI) source, and the mechanisms

Thiolactone sulfoxides as new reactive metabolites acting as bis-electrophiles: implication in clopidogrel and prasugrel bioactivation.

The antithrombotics of the tetrahydrothienopyridine series, clopidogrel and prasugrel, are prodrugs that must be metabolized in two steps to become pharmacologically active. The first step is the formation of a thiolactone metabolite. The second step is a cytochrome P450 (P450)-dependent oxidation of this thiolactone resulting in the formation of a sulfenic acid that is eventually reduced into the corresponding active thiol. It has been postulated that the sulfenic acid metabolite resulted from a nucleophilic attack of water on a highly reactive thiolactone sulfoxide derived from P450-dependen

Chemoenzymatic synthesis of (1R,3R)-3-hydroxycyclopentanemethanol: An intermediate of carbocyclic-ddA.

The synthesis of carbocyclic-ddA, a potent antiviral agent against hepatitis B, relies significantly on (1R,3R)-3-hydroxycyclopentanemethanol as a key intermediate. To effectively produce this intermediate, our study employed a chemoenzymatic approach. The selection of appropriate biocatalysts was based on substrate similarity, leading us to adopt the CrS enoate reductase derived from Thermus scotoductus SA-01. Additionally, we developed an enzymatic system for NADH regeneration, utilising formate dehydrogenase from Candida boidinii. This system facilitated the efficient catalysis of (S)-4-(hy

Spraying calcium chloride helps to enhance the resistance of kidney bean plants to western flower thrips.

BACKGROUND: The western flower thrips (WFT), Frankliniella occidentalis (Thysanoptera: Thripidae), is a significant pest in horticulture and ornamental agriculture. While exogenous calcium (Ca) has been shown to confer plant immune responses against thrips, the detailed mechanisms of this interaction remain to be elucidated for improved thrips management strategies. This study aimed to assess the impact of exogenous Ca on WFT feeding behavior and to explore its role in enhancing the defense mechanisms of kidney bean plants against WFT attacks. We compared WFT feeding preferences and efficiency

Effects of elicitors on plant host selection by adult Leptinotarsa decemlineata (Coleoptera: Chrysomelidae).

The Colorado Potato Beetle, Leptinotarsa decemlineata Say, is the principal defoliator of potato crops globally. It is well known for its propensity to rapidly develop resistance. Thus, new control options which are resilient to the pest's resistance capabilities are a critical need. The use of chemical ecology in integrated pest management (IPM) programs has been proposed as a means to delay resistance. Elicitors are chemical growth regulators that activate plant defenses. These plant defenses provide numerous opportunities to integrate chemical ecology into IPM programs, including changes to

Contrasting plant transcriptome responses between a pierce-sucking and a chewing herbivore go beyond the infestation site.

BACKGROUND: Plants have acquired a repertoire of mechanisms to combat biotic stressors, which may vary depending on the feeding strategies of herbivores and the plant species. Hormonal regulation crucially modulates this malleable defense response. Jasmonic acid (JA) and salicylic acid (SA) stand out as pivotal regulators of defense, while other hormones like abscisic acid (ABA), ethylene (ET), gibberellic acid (GA) or auxin also play a role in modulating plant-pest interactions. The plant defense response has been described to elicit effects in distal tissues, whereby aboveground herbivory ca

CaCP15 Gene Negatively Regulates Salt and Osmotic Stress Responses in Capsicum annuum L.

Salt and osmotic stress seriously restrict the growth, development, and productivity of horticultural crops in the greenhouse. The papain-like cysteine proteases (PLCPs) participate in multi-stress responses in plants. We previously demonstrated that salt and osmotic stress affect cysteine protease 15 of pepper (Capsicum annuum L.) (CaCP15); however, the role of CaCP15 in salt and osmotic stress responses is unknown. Here, the function of CaCP15 in regulating pepper salt and osmotic stress resistance was explored. Pepper plants were subjected to abiotic (sodium chloride, mannitol, salicylic ac

Genome-wide characterization of the sorghum JAZ gene family and their responses to phytohormone treatments and aphid infestation.

Jasmonate ZIM-domain (JAZ) proteins are the key repressors of the jasmonic acid (JA) signal transduction pathway and play a crucial role in stress-related defense, phytohormone crosstalk and modulation of the growth-defense tradeoff. In this study, the sorghum genome was analyzed through genome-wide comparison and domain scan analysis, which led to the identification of 18 sorghum JAZ (SbJAZ) genes. All SbJAZ proteins possess the conserved TIFY and Jas domains and they formed a phylogenetic tree with five clusters related to the orthologs of other plant species. Similarly, evolutionary analysi

Changes in Essential Oil Profile of Thymus daenensis Celak., Thymus fedtschenkoi Ronniger., and Thymus vulgaris L. Under Methyl Jasmonate Treatment.

BACKGROUND: TThymus plants are well-known medicinal plants and it is believed that the pharmaceutical and therapeutical properties of these plants are related to their essential oils. The quality and quantity of the essential oils, as a secondary metabolite of an aromatic plant, are directly related to the physiological state of the plant. The role of jasmonates in the plant as signal molecules in mediation and up-regulation of plant defense and secondary metabolism processes is well recognized. OBJECTIVE: With the aim of increasing the performance and stimulating secondary metabolites, this

Overexpressing the N-terminus of CATALASE2 enhances plant jasmonic acid biosynthesis and resistance to necrotrophic pathogen Botrytis cinerea B05.10.

Salicylic acid (SA) acts antagonistically to jasmonic acid (JA) in plant immunity. We previously reported that CATALASE2 (CAT2) promotes JA-biosynthetic acyl-CoA oxidase (ACX) activity to enhance plant resistance to necrotrophic Botrytis cinerea, and SA represses JA biosynthesis through inhibiting CAT2 activity, while the underlying mechanism remains to be further elucidated. Here, we report that the truncated CAT2 N-terminus (CAT2-N) interacts with and promotes ACX2/3, and CAT2-N-overexpressing plants have increased JA accumulation and enhanced resistance to B. cinerea B05.10, but compromised

DCPIB, an Inhibitor of Volume-Regulated Anion Channels, Distinctly Modulates K2P Channels.

K2P potassium channels stabilize the resting membrane potential in nearly all cells and have been implicated in several neuronal, cardiovascular, and immune diseases. DCPIB, a known specific and potent inhibitor of volume-regulated anion channels (VRAC), has been reported to activate TREK1 and TREK2 in astrocytes and in vitro recently. In the present study, we demonstrated DCPIB also voltage dependently activated TRAAK besides TREK1/TREK2, showing DCPIB activated all TREK subfamily members. In contrast, the compound potently inhibited several other K2P channels with no voltage dependence, incl

The beneficial effect of Trichoderma spp. on tomato is modulated by the plant genotype.

Rhizosphere-competent fungi of the genus Trichoderma are widely used as biofertilizers and biopesticides in commercial formulates because of the multiple beneficial effects on plant growth and disease resistance. In this work, we demonstrate that genetic variability among wild and cultivated tomato lines affects the outcome of the interaction with two 'elite' biocontrol strains of T. atroviride and T. harzianum. The beneficial response, which included enhanced growth and systemic resistance against Botrytis cinerea, was clearly evident for some, but not all, the tested lines. At least in one c

Background and Objective: Silybum marianum (milk thistle) is a valuable medicinal plant that produces silymarin, a complex of flavonolignans with well-documented hepatoprotective, antioxidant and anti-inflammatory properties. However, silymarin production from field-grown plants is limited by low yield, environmental variability and inconsistent metabolite accumulation. This study aimed to evaluate the effects of biotic and abiotic elicitors on biomass growth, silymarin biosynthesis and the expression of key biosynthetic genes in S. marianum</i

Single and Multiple Infestation by Coccoid Pests in Coffee Shape Plant Chemical Defenses and Interactions Across Trophic Levels.

Crop plants are attacked by multiple pests, inducing metabolic changes that shape interactions across trophic levels and influence the effectiveness of biological control. We examined how infestation by the coccoids Coccus viridis (scales) and Planococcus citri (mealybugs) modulates phytohormones, secondary metabolites, herbivore infestation, and behavior of the predator Cryptolaemus montrouzieri in Coffea arabica plants. Infestation by either scales or mealybugs activated the salicylate pathway but differentially regulated jasmonates and selected secondary metabolites. Scale infestation reduc

Jasmonate and hydrogen sulfide interaction ameliorates maize seed germination under heat stress through consolidating antioxidant defense system.

Jasmonate (JA) and hydrogen sulfide (H2S) are important signaling molecules, which play a crucial role in plant growth and stress resilience. However, JA and H2S interaction in ameliorating maize seed germination under heat stress is unclear. Therefore, in our study, the effect of seed priming with JA, NaHS (H2S donor), their inhibitors on germination and seedling growth, JA and H2S metabolism, and antioxidant defense activity under heat stress were investigated. The results showed that, as compared to the control without priming, seed priming with different concentrations (50, 100, 150, and 2

Jasmonate before salicylate in systemic acquired resistance.

Systemic acquired resistance (SAR) has long been viewed as a salicylate-driven process. Recent evidence by Gaikwad et al. reveals an earlier, jasmonate- and calcium (Ca2+)-dependent systemic phase that precedes salicylate accumulation. This temporal dynamic refines prevailing models of jasmonate-salicylate crosstalk and suggests a previously underappreciated early phase of SAR establishment.

Evaluating repellence properties of catnip essential oil against the mosquito species Aedes aegypti using a Y-tube olfactometer.

The mosquito species Aedes aegypti (L.) is known to act as a vector in the transmission of various diseases, including dengue fever and yellow fever. The use of insect repellents is one of precautionary measures used to mitigate the risk of these diseases in humans by reducing mosquito biting. Nepetalactone, a potent natural insect repellent primarily found in catnip (Nepeta cataria) essential oil, has emerged as a promising candidate for mosquito repellence. Here, we evaluated the potential of catnip essential oil (> 95% nepetalactone) for use as a mosquito repellent. Using a Y-tube olfactome
