Polymorphism analysis of myosin 1H (G/A) and P561T (C/A) genes on class I, class II, and class III malocclusion.

CONTEXT: Besides environmental factors, genetic factors play an important role in the etiology of malocclusion. Polymorphisms of the Myosin 1H gene in orofacial muscle fibers are thought to influence the growth and development of the mandible. Growth hormone receptors are present on the growth of cartilage, especially the condyle of the mandible. The polymorphisms of the growth hormone receptor have an effect on the growth and development of the mandible. The potential of the Myosin 1H and P561T genes as bioindicators in aiding diagnosis of malocclusion is quite good based on the available lit

Overcoming multidrug- and extensively drug-resistant Salmonella Typhi with natural product-metal synergy: Evidence of DNA polymerase inhibition and cell membrane disruption.

The declining efficacy of azithromycin-the primary oral treatment for typhoid fever in XDR-endemic regions-highlights the urgency against drug-resistant Salmonella Typhi. In response, we developed two ƞ6-arene ruthenium(II) series (9-20 and 37-45) by integrating natural product-based antimicrobial motifs with the DNA-intercalating/ROS-generating features of Ru-scaffolds. Among these, complex 11 and 16 exhibited potent activity against MDR and XDR S. Typhi, with lead complex 16 showing MIC90 as low as 0.06 μg/mL and MBC values significantly superior to azithromycin. Time-kill kinetics showed ra

Aberrant splicing of the type III procollagen mRNA leads to intracellular degradation of the protein in a patient with Ehlers-Danlos type IV.

Ehlers-Danlos syndrome type IV (EDS IV) is an autosomal dominant disorder characterized by fragile skin, blood vessels, and internal organs and associated with decreased production, secretion, or thermal stability of type III procollagen. Mutations in the gene for type III procollagen have been identified in patients exhibiting decreased secretion or thermal stability of the protein, but no defect has been elucidated to explain the decreased production of type III procollagen in some patients with EDS IV. We report on a patient with a moderate case of EDS IV who produced decreased amounts of t

Molecular diagnosis of strongyloidiasis in tropical areas: a comparison of conventional and real-time polymerase chain reaction with parasitological methods.

This study aimed to evaluate the use of conventional polymerase chain reaction (cPCR) and real-time quantitative PCR (qPCR) in the diagnosis of human strongyloidiasis from stool samples in tropical areas. Stool samples were collected from individuals and were determined to be positive for Strongyloides stercoralis (group I), negative for S. stercoralis (group II) and positive for other enteroparasite species (group III). DNA specific to S. stercoralis was found in 76.7% of group I samples by cPCR and in 90% of group I samples by qPCR. The results show that molecular methods can be used as alte

A model for systemic lupus erythematosus based on chromatin disruption by polyamines.

Here it is hypothesized that some autoimmune disorders, such as systemic lupus erythematosus, result in part from overexpression of polyamines which leads to disruption of chromatin structure. Disruption of inactive chromatin, such as the inactive X chromosome, exposes sites of unrepaired DNA damage. Repair is then hampered by the polyamines. Disruption also facilitates transcription at previously sequestered sites. Especially interesting are RNA polymerase III sites in highly repeated sequences such as the Alu sequence. Transcription and translation from these sites could create RNA and polyp

[The application of molecular biology to thrombosis and hemostasis].

Current biological science has been revolutionized by a series of new investigative molecular techniques developed within the last 15 years. These techniques allow the detection of gene mutations responsible for congenital diseases including thrombophilia and hemorrhagic disorders. Here, we provide a short introduction to polymerase chain reaction (PCR), single-strand conformational polymorphism (SSCP), restriction fragment length polymorphism (RFLP) and northern blot techniques and present the results of DNA sequence analyses of 4 different types of antithrombin III abnormalities and one IIB-

Genetic analysis of the cause of endometrial osseous metaplasia.

OBJECTIVE: To analyze solitary bone fragments from the uterine cavity through DNA genotyping, thus elucidating whether they originate from metaplasia, from previous abortion, or both. METHODS: We conducted a case series study on 14 patients, of whom eight yielded bone DNA. The patients selected had histopathologic diagnoses of bone fragments inside the uterine cavity or previously removed samples available for analysis. We extracted DNA from blood and bone fragments. To identify the bone tissue origin, these materials were genotyped using polymerase chain reactions for DNA loci. Six mini shor

Cidofovir, a new approach for the treatment of cervix intraepithelial neoplasia grade III (CIN III).

Cervix intraepithelial neoplasia grade III (CIN III) is an intraepithelial proliferative process with different levels of severity depending on both the extension of the proliferation in the epithelium and the presence of cellular atypia. Human papillomavirus (HPV) has been clearly associated with such lesions. The results of a preliminary study are described on the local application of cidofovir, an acyclic nucleoside phosphonate derivative with broad-spectrum anti-DNA virus activity for the treatment of CIN III. Cidofovir 1% in gel was applied three times, every other day, on the cervix of e

Human papillomavirus DNA integration and messenger RNA transcription in cervical low- and high-risk squamous intraepithelial lesions in Austrian women.

The human papillomavirus (HPV) plays an important role in the progression of cervical carcinoma. High-risk (HR) HPV types have been mainly identified in cytologic high-grade squamous intraepithelial lesions (HSILs) and histologic invasive carcinoma of the cervix. We examined cervical swabs of patients with abnormal Papanicolaou (Pap) smears, diagnosed as low-grade squamous intraepithelial lesions (LSILs) including atypical squamous cells of uncertain significance or HSILs. Low-risk (LR) HPV and HR-HPV types were identified by the Digene Hybrid Capture II test. Two-dimensional (2D) gel electrop

Genetic diversity of Apis mellifera filamentous virus in bees sampled in the USA and Jamaica.

Apis mellifera filamentous virus (AmFV) is a large filamentous virus with a ~500 kb double-stranded DNA genome identified in honeybees and a handful of other winged insects. Since its first identification almost 50 years ago, only a few nearly complete AmFV genomes have been sequenced. Genomic regions of AmFV have been identified using gene-specific primers as part of the viral screening surveys for honeybee viruses. Here, we assembled a complete AmFV genome from honeybees from the USA which shares 94.4-96.4% intergenomic similarity with previously published AmFV genomes. A distinct variation

Proteomic identification of potential biomarkers for cervical squamous cell carcinoma and human papillomavirus infection.

It is known that high-risk human papillomavirus infection is the main etiological factor in cervical carcinogenesis. However, human papillomavirus screening is not sufficient for early diagnosis. In this study, we aimed to identify potential biomarkers common to cervical carcinoma and human papillomavirus infection by proteomics for human papillomavirus-based early diagnosis and prognosis. To this end, we collected 76 cases of fresh cervical tissues and 116 cases of paraffin-embedded tissue slices, diagnosed as cervical squamous cell carcinoma, cervical intraepithelial neoplasia II-III, or nor

SRSF1 facilitates cytosolic DNA-induced production of type I interferons recognized by RIG-I.

BACKGROUND: Evidence has shown that psoriasis is closely associated with infection; however, the mechanism of this association remains unclear. In mammalian cells, viral or bacterial infection is accompanied by the release of cytosolic DNA, which in turn triggers the production of type-I interferons (IFNs). Type I IFNs and their associated genes are significantly upregulated in psoriatic lesions. RIG-I is also highly upregulated in psoriatic lesions and is responsible for IFN production. However, RIG-I mediated regulatory signaling in psoriasis is poorly understood. METHODS: We screened a cDN

Epidemiology of human papillomavirus infection in Greek asymptomatic women.

In an attempt to estimate the prevalence of human papilloma virus (HPV) positivity among asymptomatic, cytologically normal Greek women, and the possible associations between HPV infection and other demographic, sexual, behavioural and sociological parameters, we undertook an epidemiological study of 226 clinically normal women from an outpatient gynaecological clinic in Northern Greece. The polymerase chain reaction was used for detection of HPV DNA and dot blot hybridization analysis for HPV typing (only for the high-risk types 16 and 18). Eighty-two of the 226 women examined (36.3%) were po
